        단일 태아세포에서의 PEP-PCR을 이용한 성의 결정과 Dystrophin 유전자 분석

        Recently, through the development of the primer extension preamplification(PEP) method which amplifies the whole genome, simultaneous multiple DNA analysis has become possible. Whole genome from each single cell can be amplified using 15 base oligonucleotide random primer. The greatest advantage of PEP-PCR is the ability to investigate several loci simultaneously and confirm results by analysing multiple aliquots for each locus. This technique led to the development of preimplantation genetic disease diagnosis using blastomere from early embryo, sperm, polar body and oocyte. In this study, we applied PEP-PCR in 20 cases of single amniocyte and 20 cases of single chorionic villus cell for the clinical application of the prenatal and preimplantational genetic diagnosis. We analysed 7 gene loci simultaneously which are 46, 47 exons related to dystrophin gene, two VNTR (variable number tandem repeat) markers using 5'dysIII, 3'CA related to dystrophin gene and DYZ1, DYZ3, DYS14 regions on chromosome Y. In all the tests, 97.5% of PEP-PCR amplifications with single cells were successful. We obtained 38/40 (95%) accuracy in gender determination through chromosome analysis comparison. Therefore, these results have significant implications for a sperm or oocyte analysis and prenatal or preimplantational genetic diagnosis.

        근이양증 가계에서의 PEP-PCR을 이용한 착상전 유전자진단

        General PCR technique alone has a limitation for preimplantation genetic diagnosis(PGD) using single blastomere. Recntly developed primer extension preamplification(PEP) technology amplifies the whole genome and thus, simultaneous multiple locus analysis became possible. In this study, we report the efficacy of PEP-PCR for PGD in three muscular dystrophy carriers undergoing IVF-ET. A total of 37 blastomeres were obtained from 40 embryos at six to eight cell stage in three IVF cycles in two DMD and one BMD carriers. Whole genome from single blastomeres were amplified using I5-base oligonucleotide random primers. PCR amplified products of exon 45 in the dystrophin gene and alphoid X/Y loci for gender determination were analysed by 2% metaphor gel electrophoresis. A total of 37 PEP-PCR replicates from 37 single blastomeres from 40 embryos and 37 blanks were performed. We obtained the reliable results for exon 45 and alphoid X/Y. Transfer of female embryos and unaffected male embryo was attempted in three couples. Unfortunately, pregnancy was not achieved in these cases. PEP-PCR is a reliable and efficient PGD method in multiple locus analysis using single blastomere.

        반복유산을 경험한 384부부의 세포유전학적 연구

        During the years 1984 to 1989, in order to determine of chromosome abnormalities are associated with recurrent spontaneous abortions, cytogenetic studies were performed 384 couples. Abnormal karyotypes were found in 51(13.3%) couples. There was no apparent relation with the number of abortions. The abnormalities were as follows: 17(4.4%) balanced translocation; 15(3.9%) mosaicisms; 17(4.4%) pericentric inversion; 2(0.5%) addition or isochromosome. Chromosome abnormalities were observed in 34(67%) of the wives and 17(33%) of the husbands. In addition, we detected polymorphic variants of chromosomes in 89(23.2%) subjects. Reciprocal translocations(13/17) were more common than the robertsonian type(4/17). All of the mosaicisms were associated with the sex chromosomes in 10 females and 5 males subjects. Pericentric inversions were most common in chromosome 9. Compared to previously studied general populations, significantly higher frequencies of translocations, mosaicisms and inversions were found in couples with repetitive spontaneous abortion. This suggests that couples should have chromosome studies after two or more abortions.

        C-분염(分染)에 의한 사람의 염색체 다형성에 관한 연구

        사람에 있어서 C-이질염색질의 다형성을 관찰하기 위하여, 일반환자 234명(여자 165, 남자 69)의 말초혈액백혈구을 배양하였다. 슬라이드 제작후 C-분염법(NOR-염색법, GC-분염법 포함)에 의해 염색체 1, 9, 16번 그리고 Y의 이질염색질(qh)과 D와 G군의 부수체와 단완의 변이를 관찰한후 변이를 나타낸 환자와 임상적 고찰을 함께 실시 하였다. I. 234명중 변이세포를 갖는 사람은 91명(여 68, 남 23)이었으며, 변이세포를 갖고 있지않은 사람은 143명(여97, 남 46)이다. 변이세포를 갖고있는 91명에 대한 총변이수는 125예로서 다음과 같다. 1) 염색체 1, 9, 16번과 Y의 qh변이의 수는 99예 였고, 2) D와 G군의 부수체(Satellite)와 단완의 변이의 수는 21예 였으며, 3) 염색체 9번의 편동원체 역위인 경우가 5예였다. II. 염색체 1번, 9번, Y에서 이질염색질이 크게 증가한 환자 12명의 병력을 조사하여 다음과 같은 결과을 얻었다. 1) 생식기관의 이상(reproductive failure)인 경우가 3예 (1qh+)였고, 2) 3회이상의 반복유산(habitual abortion)인 경우가 3예, 즉 inv(9)이 2예, 1qh+가 1예 이고, 3) 암의 초기상태(precancerous state)가 2예, 즉 inv(9)이 1예, 1qh+가 1예, 4) 결핵(tuberculosis)은 1예(1qh+), 5) 정상(normal)은 3예, (1qh +2예, Yqh+1예)였다. 위와같은 결과를 통하여, 본 실험에서의 염색체 1, 9, 16번 그리고 Y의 qh의 변이중 이질염색 질의 감소(qh-)는 단순한 다형현상이라 생각되며, 이질염색질의 증가(qh+)와 9번염색체의 편동원체 역위현상은 임상적 질환과 상당한 연관성이 있다고 사료된다. 추후 이러한 C-이질염색질의 다형성과 임상적 이상과의 관계에 대한 지속적인 연구가 필요하다고 사료된다. Lymphocyte chromosome preparations obtained by the micromethod (Arakaki and Sparkes, 1963) from 234 our patients (165 females and 69 males) were analysed by C-, NOR-and GC-bandings for chromosome heteromorphisms. The centromeric regions of chromosomes 1,9,16 and the long arm of the Y chromosomes were tested for C heteromorphism. Minor variations found in this study such as inv(9), prominant short arms and large satellites of acrocentrics were also examined by appropriate banding techniques. Of the 234 probands, a total of 125 different C-variants were detected, and the average frequency of the variants per individual was estimated to be 0.53. The observed variations were as follows : 99 qh variants, 5 pericentric inversions of chromosome 9, and 21 satellite and/or short arm variants.

        인간이 경량벽체에 가하는 수평하중의 크기에 관한 연구

        The purpose of this research is to comprehend experimentally the characteristics of human strength for using as the basic data of impact resistance test method of lightweight wall. Human motions exerting static load are classified to 4 types. Pushes with two hands or shoulder are defined as the instantaneously forcing motions with hands or shoulder put on the force plate. Leanings back or one-hand against the wall are defined as motions of taking a rest in their respective comfortable posture. Human motions exerting dynamic load are classified to 3 types. Selecting 3 levels of motion strength (weak, middle, strong), 3 levels of force plate stiffness (A: 20kN/cm, B: 4.7kN/cm, C: 2.2kN/cm), and 30 male subjects, load was measured when they applied strength to the force plate. Results of this research are as follows: (1) The maximum load ratio (Pmax/W) of static load for each motion was 1.17-1.25 in two hands pushing, 0.95-0.99 in shoulder pushing, 0.16-0.18 in back leaning, and 0.12-0.15 in one hand leaning. (2) Human dynamic load and object collision were different in the load characteristics. (3) The maximum load ratio of dynamic load for each motion was 10.07 in heel kick, 4.46 in shoulder hitting, and 5.58 in fist blow. 공동주택의 구조형식이 점차 기둥식 구조로 바뀌면서 비내력 경량벽체의 수요가 증가하고 있다. 경량벽체는 구조적 안전을 위해 소정의 내력을 확보할 필요가 있다. 본 연구에서는 경량벽체의 정적 수평하중저항성 및 내충격성 시험방법의 기초자료로서 활용하기 위해 인간이 벽체에 가하는 힘을 실험적으로 파악하였다. 정적하중을 가하는 동작으로는 양손 밀기, 어깨 밀기, 등 기대기, 한 손 기대기의 4종류를 설정하였다. 동적하중을 가하는 동작으로는 발뒤꿈치차기, 어깨 부딪치기, 주먹치기의 3종류를 설정하였다. 하중해석 장치의 하중판 강성은 20kN/cm, 4.7kN/cm, 2.2kN/cm의 3종류로 설정하였다. 정적하중 해석결과로부터, 동작별 최대하중비(Pmax/W)는 양손 밀기의 경우 1.17~1.25, 어깨 밀기의 경우 0.95~0.99, 등 기대기의 경우 0.16~0.18, 한 손 기대기의 경우 0.12~0.15인 것을 알 수 있었다. 또한 동적하중 해석결과로부터, 동작별 최대하중비(Pmax/W)의 상한 값은 발차기의 경우 약 10.07, 어깨 부딪치기의 경우 4.46, 주먹치기의 경우 약 5.58인 것을 알 수 있었다.

      • 폐폴리올레핀 폼과 탄성 포장재로 구성된 어린이 놀이터 바닥의 안전성에 관한 실험적 연구

        본 연구에서는 폐폴리올레핀 폼을 완충재로 이용한 어린이 놀이터 바닥의 안전성을 알아보기 위하여 한계하강높이, 보행적합성 및 장기피로예측, 미끄럼저항성에 대하여 시험하고 평가하였다. 시험체는 두께 30mm, 50mm, 70mm의 폐폴리올레핀 폼에 각각 탄성 포장재를 12mm, 15mm 두께로 포설한 6종류로 하였다. 한계하강높이 시험은 KS G 5758:2009에 의거하여 실시하였다. 보행적합성 및 장기피로예측 시험에는 바닥 경도 시험장치(O-Y HMA)를 이용하였으며, 미끄럼저항성 시험에는 휴대형 미끄럼 시험기(ONO PPSM)을 이용하였다. 시험결과, 폐폴리올레핀 폼과 탄성 포장재로 구성된 바닥은 충격저감성 관점에서 상당히 안전한 것으로 나타났다. 바닥의 경도는 남성에게는 보행감이나 피로 측면에서 우수한 성능을 발휘하지만, 여성에게는 구두(미들 힐)를 신고 보행하기에 적합하지 않고 오래 사용할 경우 쉽게 피로감을 느낄 수 있는 것으로 나타났다. 미끄럼저항성은 표면에 물기 등의 이물질이 있더라도 일상적인 동작에 대해서는 비교적 안전한 것으로 나타났다. This study is purposed to verify the safety of the floor of the children's playground using polyolefin foam waste and rubber paving. The critical fall height, walking suitability, long term physical fatigue expectation and slipperiness were tested. Polyolefin foam wastes in thickness of 30mm, 50mm and 70mm were prepared with paving the rubber paving materials in 12mm and 15mm thickness respectively. The test on the critical fall height was carried out according to KS G 5758:2009. The floor hardness test equipment (O-Y HMA) was used for the test on hardness of the floor from a viewpoint of walking suitability and fatigue. A portable slipperiness tester (ONO PPSM) was used for slipperiness test. It was revealed from the test that the floor made of the polyolefin foam waste and rubber paving were considerably safe from a viewpoint of impact absorption. With regards to the hardness of the floor, it was shown the excellent performance in the aspects of walking and fatigue for male. But it was not suitable with walking on the shoes(middle heels) for female. And they will be very fatigue if they were in a long time walking or standing. As far as the slipperiness is concerned, it was shown that it was comparatively safe for the ordinary motions even though the surface was wet.

        신체장애자의 안전성 관점에서 경사로 바닥의 미끄럼 평가방법에 관한 기초적 연구

        이 연구의 목적은 신체적 약자와 건강한 사람의 경사로 바닥에 대한 미끄럼 평가척도의 변동 경향을 비교 분석하는 것이다. 이 연구를 통해서 반신마비 장애인과 건강한 사람에 대한 중심동요 비교 실험을 통하여 신체적 약자의 중심동요 특성을 항상 재현하는 장애 고령화 체험장비가 개발하였다. 그리고 신체적 약자(건강한 사람이 장애 고령화 체험장비 착용)와 건강한 사람을 대상으로 한 관능검사결과로부터 수평바닥 및 경사로 바닥에 대한 미끄럼 평가척도를 구성하였다. 미끄럼 평가척도와 미끄럼저항계수(C.S.R')의 관계를 분석한 결과로부터 경사로의 기울기나 미끄럼저항 차이에 따른 미끄럼 평가척도의 변동 경향은 남성과 여성이 서로 유사하다는 사실을 명확히 규명하였다. 경사로 바닥에 대한 미끄럼의 평가는 건강한 사람은 C.S.R'가 0.30 이상이면 안전하다고 평가하지만, 신체적 약자는 C.S.R'가 $0.45{\sim}0.50$ 이상인 경우에도 위험하다고 평가하였다. 이러한 평가의 차이는 주로 건강한 사람과 신체적 약자의 보행 메커니즘이 다르기 때문인 것으로 생각된다. The purpose of this research is to compare and analyze the change that must be applied to the evaluation of the slipperiness of an inclined floor, when considering its use by the physically handicapped and by the able-bodied. For this study, a simulator that can reproduce the characteristics of the body sway of a physically handicapped person was developed through a comparative experiment between able-bodied people and people with hemiplegia. In addition, as cale for the evaluation of the slipperiness of flat floors and inclined floors was composed by the sensory test result targeting physically handicapped persons (able-bodied persons wore the body sway for the handicapped and ageing in the experiment) and the able-bodied. In terms of the evaluation of the slipperiness of an inclined floor, it was evaluated that it is safe when the C.S.R' (Coefficient of Slip Resistance) of a healthy person is over 0.30, while it was evaluated that it is dangerous when the C.S.R' of a physically handicapped person is over $0.45{\sim}0.50$. Concerning this difference, it is judged that the reason for this difference is that the walking mechanism differs between an able-bodied person and a physically handicapped person.

        폐경 중년여성을 위한 심뇌혈관질환 예방교육프로그램의 효과

        Purpose: This study was conducted to examine effects of a cardiocerebrovascular disease (CVD) prevention education program on knowledge, self-efficacy and health behavior among postmenopausal middle-aged women. Methods: A non-equivalent control group pretest-posttest design was used. Participants were 53 postmenopausal middle-aged women who registered in two community culture centers in G metropolitan city. Experimental group (n=26) received a CVD prevention education program 8 times over 8 weeks. Knowledge, self-efficacy and health behavior of the participants were examined with self-report structured questionaries. Data were collected between October 15 and December 11, 2013, and were analyzed using chi-square test, Fisher’s exact test, independent t-test, and analysis of covariance with SPSS/PC version 21.0. Results: After the intervention the experimental group showed significant increases in the knowledge of CVD symptoms (p<.001) and CVD risk factors (p<.001), level of self-efficacy (p=.028) and health behavior (p<.001) compared to the control group. However, no significant difference was found between groups for knowledge of CVD prevention (p<.133). Conclusion: Results suggest that a CVD prevention education program can be an effective strategy to improve knowledge on CVD symptoms and risk factors, self-efficacy and health behavior for postmenopausal middle-aged women.

        비내력 경량벽체의 수평하중저항성 및 내충격성 시험방법 표준화에 관한 연구

        As the structural type of apartment houses has been changing from wall column structures to Rahmen structures, demand for lightweight walls has been increasing. Although lightweight walls reduce self-weight of the building and enable efficient use of interior spaces, to apply them as building walls, the structural safety of them should be secured without fail. The lateral load resistance test method and impact resistance test method for the verification of lightweight walls’ structural safety have not yet been established. The purpose of this study is to standardize lateral load resistance test method, impact resistance test method using large soft body, and impact resistance test method using small hard body. Most of the configuration of the test methods and testing device were made by modifying the content of BS 5234:1992. However, the conditions of loads applied to specimens were reestablished reflecting the characteristics of loads acting on actual walls. In the lateral load resistance test, load conditions were divided into three levels; 500N, 1000N, and 3000N so that the conditions could be selected and applied according to the uses of walls. In the impact resistance test using large soft body, the length of the rope was set to be at least 2.5m and the drop heights were divided into three levels; 200mm, 400mm, and 600mm. In the impact resistance test using small hard body, the drop heights were divided into three levels; 0.1m, 0.2m, and 0.5m. The performance test methods standardized in this study will be objective evaluation tools when optimum lightweight walls are designed or selected in construction sites. In addition, they will be usefully utilized when setting performance goals for development of new lightweight walls.
