유기산전해질을 이용한 접촉 글로우 방전 전기분해공정에서 후코이단의 저분자화

접촉글로우방전 전기분해(CGDE)에 의한 후코이단의 저분자화에 대해 연구하였다. CGDE 공정 후에 후코이단을 기능성 식품재료로 사용하기 위해 유기산을 전해질로 사용하였다. 실험결과 유기산을 이용한 전기분해에 의해 후코이단의 분자량을 효과적으로 감소시킴을 보였다. 전해질의 농도가 증가하면 글로우방전 시작 전압이 감소하고 글로우방전 시작 전류는 증가하였다. 반응시간에 따른 후코이단의 분자량 변화로부터 저분자화반응은 1차 반응속도 식을 따름을 보였다. 유기산을 이용한 CGDE에 의해 후코이단의 분자량이 처음의 약 1/77로 감소하였으며, CGDE 저분자화 과정에서 유리된 황산기 함량이 작았다. Depolymerization of fucoidan induced by contact glow discharge electrolysis(CGDE) was investigated. To utilize fucoidan as a functional food material after CGDE, organic acids were used as a electrolyte in CGDE process. Experimental results showed that CGDE using organic acid reduced the molecular weight of fucoidan effectively. As electrolyte concentration increased, onset voltage of glow discharge decreased and onset current of glow discharge increased. From the variation of molecular weight of fucoidan with the reaction time, it was demonstrated that the degradation of fucoidan followed a first-order rate law. Molecular weight of fucoidan treated with CGDE using organic acid was about 77 times lower compared to initial fucoidan with little free sulfate.

Properties of fucoidan used for functional cosmetic ingredients and the effect of fucoidan molecular weight on the cosmetic functions were studied. Fucoidan was extracted from Undaria pinnatifida sporophylls and molecular weight (35~160kDa) of fucoidan was controlled by contact glow discharge electrolysis (CGDE). To test possibility of fucoidan as a cosmetics material, tyrosinase inhibition property, water-holding property, elastase activity inhibition property and DPPH free radical scavenging property were measured. Water-holding property of fucoidan was higher than that of hyaruronic acid, which is known as the one of the best water-holding material. The water-holding strength of fucoidan slightly increase as molecular weight of fucoidan decrease, Elastase activity inhibition (anti wrinkle effect) of fucoidan was higher than that of adenosine using standard material for anti wrinkle test. Optimum molecular weight of fucoidan to obtain highest tyrosin inhibition effect, elastase inhibition effect and radical scavenger effect is 100kDa.

Laminaria roots have not been practically used in Korea. In this study, the extraction process of carbohydrates and minerals from Laminaria roots was investigated and the properties of extracted components were measured. Hydrochloric acid generally used in carbohydrate extraction from seaweeds in order to obtain high extraction yield. But in this work, to utilize extracted components as a functional food material, organic acids such as citric acid were used, Organic acid as extraction solvent has low extraction yield compared to strong acids, Therefore optimum condition for maximum yield was investigated in carbohydrate extraction from Laminaria roots using organic acid, We measured the extraction yields of carbohydrate with variation of extraction temperature, extraction time, concentration of organic acid and particle size of samples. The extraction yield increased as the particle size decreased and temperature became high. The extraction yield was 19.0 wt% after 4.0 hours extraction with 0.2 wt% citric acid at 100˚C. Potassium concentration was high compared other minerals in extraction solution, that is, the ratio of K/Na was about 3.0. Fucoidan from Laminaria roots had same carbohydrate composition and lower molecular weight compared that of Undaria pinnatifida.
