        HL-60 세포주, 제대혈 및 골수 단핵구 세포의 interferon-gamma에 대한 감수성에 관한 연구

        Background: Finding of the regulation of various gene expression by cytokine including $IFN-{\gamma}$ in hematopoietic stem cell will light up the understanding of pathogenesis of aplastic anemia in various aspects. To study on aplastic anemia, however, we have to circumvent the difficulty of directly obtaining bone marrow stem cells from the patient. Therefore, we tried to find out a cell can replace the bone marrow stem cells for study on cell signaling pathway and regulation of gene expression by $IFN-{\gamma}$. Materials and Methods: HL-60 cells, of 20 ng/mL of $IFN-{\gamma}$. Total RNA was isolated from the cells and RT-PCR of the indoleamine 2,3-dioxygenase (IDO), $IFN-{\gamma}$, TNF-${\alpha}$, $MIP-1{\alpha}$, and $TGF-{\beta}2$ was carried out for the estimation of the gene expression. Results: $IFN-{\gamma}$ induced IDO gene expression of mononuclear cells from umbilical cord blood showed similar pattern as compared to that of bone marrow. Whether $INF-{\gamma}$ was treated or not, $TNF-{\alpha}$ was expressed in both mononuclear cells from umbilical cord blood and bone marrow. However, HL-60 cells showed different expression patterns. HL-60 cells would express neither IDO nor $TNF-{\alpha}$ even under the culture with 20ng/mL of $IFN-{\gamma}$. Conclusion: Our results showed bone marrow can be replaced with mononuclear cells from umbilical cord blood in the study on the relation between aplastic anemia and $IFN-{\gamma}$ including $IFN-{\gamma}$ cell signaling pathway.

        OLAP 분야에서 지금까지 연구되어온 영역 질의는 주어진 영역에 대한 집단 연산의 결과를 구하는 단순한 형태이다. 그러나 실제 데이타 분석 과정에서는 이러한 단순한 형태의 영역 질의뿐만 아니라, 집단 연산이 포함된 특정 조건을 만족하는 데이타 큐브 내의 영역을 찾는 형태의 확장된 영역 질의에 대한 필요성이 존재한다. 본 논문에서는 이러한 확장된 영역 질의 유형의 일반적인 형태를 정의하고, 이에 대한 대표적인 예인 ‘MAX-of-SUM 질의’의 효율적인 처리 기법을 제안한다. MAX-of-SUM 질의는 데이타 큐브 상에서 영역합(SUM)이 최대(MAX)가 되는 영역을 찾는 질의를 의미한다. 본 논문에서는 MAX-of-SUM 질의 처리 시 검색의 대상이 되는 영역들에 대한 SUM 연산의 결과값이 취할 수 있는 범위를 미리 예측하는 기법을 제안한다. 즉, 영역에 대한 SUM 값의 범위를 예측함으로써, 이들 중에서 최대값을 찾기 위해 실제로 계산하여야 하는 영역의 개수를 줄여 빠른 질의 처리를 보장한다. Recent researches about range queries in OLAP are only concerned with applying an aggregation operator over a certain region. However, data analysts in real world need not only the simple range query pattern but also an extended range query pattern that finds ranges which satisfy a special condition specified by using several aggregation operators. In this work, we define the general form of the extended range query and propose an efficient processing method for the ‘MAX-of-SUM’ query, which is the representative form of the extended range query pattern. The MAX-of-SUM query finds the range which has the maximum range sum value in data cube where the size of the range is given. The proposed query processing method is based on the prediction of the scope of the range sum values. That is, the search space on the query processing can be reduced by using the result of the prediction, and hence, the query response time is also reduced.

        재생불량성 빈혈의 병태생리에서 Fas 항원과 Apoptosis의 역할

        Background: Clinical observations and laboratory studies have supported an immune basis for most acquired aplastic anemias, with the majority of patients responding to immunosuppressive therapy. Fas, a member of the tumor necrosis factor (TNF) receptor superfamily is a critical downregulator of cellular immune responses. Proinflammatory cytokines like interferon gamma (IFN-${\gamma}$) and TNF-${\alpha}$ can induce Fas expression and render hematopoietic progenitor cells susceptible to Fas-induced growth suppression and apoptosis. Methods: In order to investigate the involvement of apoptosis in the pathogenesis of aplastic anemia (AA), we measured the expression of Fas antigen and caspase-3 on bone marrow (BM) mononuclear cells (MNCs) of AA in the presence or absence of IFN-${\gamma}$, TNF-${\alpha}$, or macrophage inflammatory protein 1-${\alpha}$ (MIP-$1{\alpha}$). Results: We confirmed that AA BM MNCs were more apoptotic and highly expressed Fas antigen than normal donors. Stimulation by IFN-${\gamma}$, TNF-${\alpha}$, or MIP-$1{\alpha}$ increased Fas antigen and caspase-3 expression in AA BM MNCs than BM MNCs of normal donors. Anti-Fas monoclonal antibody enhanced IFN-${\gamma}$, TNF-${\alpha}$, or MIP$1{\alpha}$ mediated caspase-3 expression in BM MNCs of normal donors. Among these three cytokines, IFN-${\gamma}$ enhanced apoptosis most strongly via Fas-caspase-3 pathway. Conclusion: These results suggest that Fas signal pathway may play a role in the pathophysiology of aplastic anemia and negative hematopoietic regulators like IFN-${\gamma}$ can induce apoptosis of bone marrow progenitors in part by Fas induction.

        자활사업의 유형별 고용촉진 효과 분석

          In this study we attempt to assess the effectiveness of Workfare approach of Korean public assistance program, and thus to draw practical policy implications. In order to achieve our research goal, we first measure and analyze employment rate and qualification attainment rate of the program participants. The analyses results indicate that the average employment rate is turned out to be 33.5%, and the average qualification attainment rate is 30.46%, which shows low level of program effectiveness. Such a low level of effectiveness is found to be due both to the internal factors including insufficient training and job search services and to the external factors including participants health status and socioeconomic status of households. The policy prescriptions we draw are; first, Korean government should make an effort to enrich the content of training program regardless of the types of programs, and, second, they need to expand the welfare infrastructure in order to promote rapid reemployment of the program participants.

        생체 외 제대혈 배양에서 거대핵세포 조혈에 대한 Interleukin-11 (IL-11)의 효과

        Background: The megakaryopoiesis and platelet production is regulated by several hematopoietc factors such as thrombopoietin (TPO), interleukin-11 (IL-11) and interleukin- 3 (IL-3). IL-11 is a potent stimulator of megakaryopoiesis in vivo, and acts primarily as a megakaryocyte maturation factor in vitro and it can act synergistically with IL-3 and TPO. We performed this study to investigate the effects of recombinant human IL-11 (rhIL-11) with other hematopoietic factors on megakaryocyte colony formation in vitro. Methods: CD34+ cells were separated from umbilical cord blood and megakaryocyte colonies using MegaCult Assay Kit were cultured with rhIL-11, recombinant human IL-3 (rhIL-3), and recombinant human TPO (rhTPO) for 7 and 14 days. The number and percentage of CD34+ and CD41a+ cells were determined by flowcytometry. Results: The number of CD41a+ cells were $0.54{\pm}0.05{\times}10^4$ (rhIL-11 100 ng/ml), $5.32{\pm}0.23{\times}10^4$ (rhIL-3 100 ng/ml), and $8.76{\pm}0.15{\times}10^4$ (rhTPO 50 ng/ml) of total expanded cells during the culture of the purified CD34+ cells in liquid phase for 7 days. The number of CD41a+ cells were increased to $7.47{\pm}0.69{\times}10^4$ (rhIL-3+ rhIL-11), $11.92{\pm}0.19{\times}10^4$ (rhTPO+rhIL-11) of total expanded cells, respectively, during the culture of the purified CD34+ cells in liquid phase for 7 days in the presence of rhIL-11 (100 ng/ml). When the purified CD34+ cells were cultured in semisolid mediaincluding various concentration of rhIL-11, the megakaryocyte colonies were not formed. When the purified CD34+ cells were cultured with rhIL-11 and rhTPO or with rhIL-11 and rhIL-3, the number of megakaryocyte colonies were increased compared with rhTPO or rhIL-3 alone. Conclusion: These results indicate that IL-11 exerts a potent proliferative activity to colony forming unit-megakaryocyte from human umbilical cord blood, and it acts with other hematopoietic factors synergistically.

        기능적 장애 개념과 중증 장애인 기준의 타당성

        One of the most frequently discussed problems regarding employment quota system for disabled workers in Korea is that this system severely handicapped people are much less likely to be employed than less severly handicapped people are. Regarding this matter, active debates are made in academia focusing on establishing a new disability standard with taking account of vocational disability. In this study, we attempt to analyze the validity of disability standard in current law. We used the data from Survey on Disabled Workers for 2005 and compared the severe disability defined with functional standard and that defined with medical standard. The analysis findings indicate that severely disabled workers under the definition of functional disability are less likely to conduct their tasks without difficulties compared to those under the definition of medical disability. As the findings imply, Korean government should consider setting up new disability assessment system based on the notion of vocational or functional disability.

      • 제대혈 조혈모세포 분리를 위한 적혈구 제거 방법에 관한 연구

        연구배경 :현재까지 제대혈 은행에서 적혈구 제거후에 냉동 보관 방법이 보편화되지 못한 것은 Ficoll, lysis, starch와 중력에 의한 방법 등이 약 50-90%에 이르는 조혈모세포가 손실된다는 이유 때문이다, 따라서 대부분의 제대혈 은행에서 전체 제대혈을 냉동보관 후에 이식에 사용하고 있다. 지금까지의 보고들은 서로 다른 제대혈에서 여러 가지 방법으로 적혈구제거를 실시한 것으로 결과가 서로 큰 차이가 난다. 방법 : 저자는 같은 제대혈에서 Ficoll, gelatin, methylcellulose, RBC lysis 등의 방법으로 적혈구를 제거하여 다음과 같은 결과를 얻었다. 결과: 1) 채취한 평균 제대혈량은 105±23ml,단핵구수는 18.6±4.09×10^(6)/ml이었다. 2) 각각의 방법으로 적혈구를 제거한 다음의 단핵구수는 Ficoll, 방법 4.43±1.50×10^(6)/ml, gelatin방법 7.71±0.37×10^(6)/ml, methylcellulose 방법 5.90±0.51×10^(6)/ml, RBC lysis방법10.72±2.74×10^(6)/ml개 였으며 이 중 RBC lysis방법에서 단핵구 수가 가장 많았다. 3) 각각의 방법으로 적혈구를 제거한 다음 flow cytometry로 측정한 CD34 양성세포 수는 Ficoll 방법 2.65±0.52×10⁴/ml, gelatin방법 3.50±0.43×10⁴/ml, methylcellulose방법 2.71±0.23×10⁴/ml, RBC lysis방법 9.36±1.62×10⁴/ml개 였으며 이 중 RBC lysis방법에서 CD34양성세포 수가 가장 많았다. 전체 세포에 대한 CD34양성세포의 비율을 보면 Ficoll방법 0.72±0.14, gelatin 방법 0.50±0.08%, methylcellulose 방법 0.44±0.03%, RBC lysis방법 0.79±0.04%였다. 4) CFC수는 Ficoll 방법 21.73±11.77×10³/ml, methylcellulose 방법 15.33±7.14×10³/ml, RBC lysis 방법 26.50±4.26×10³/ml개 였으며 이 중 RBC lysis 방법에서 CFC수가 가장 많았다. 결론 : 이상의결과로 보나 Ficoll, nethylcellulose, gelatin, RBC lysis 등의 방법을 이용하는 경우 모두 적혈구 제가가 잘 되었으며 특히 RBC lysis방법을 이용한 경우 가장 많은 CD34양성세포 수와 CFC수를 얻어 적혈구 제거 방법으로 가장 좋았다. 하지만 다른 보고들에서는 gelatin에 의한 방법이 좋다고 하였는데 이는 실험 방법의 차이에 의한 것으로 보여 이에 대한 연구가 좀 더 많은 례에서 이루어져야 할 것으로 사료된다. Background : Human umbilical cord blood (UCB) has been successfully used as an althemative source of allogeneic hematopoietic stem cells for pediatric transplantation. A bank of cryopreserved UCB has the advantage over existing volunteer marrow donor registries because time consuming donor tracing can be avoided. Efficient clinical banking of UCB requires volume reduction of cord blood collections to decrease storage space and costs. The simplest way to achieve this is no deplete cord blood red cell before cryopreservation. Methods : We compared four different red cell depletion techniques, Ficoll, gelation, methylcelulose and red cell lysis method in terms of mononuclear cell count, CD34-positive cell count and colony forming cell (CFC) count. Results : 1) Mean volume and mononuclear cell count of collected UCB were 105±23mL and 18.63±4.09×10^(6)/mL. 2) Among different red cell depletion techniques, red cell lysis mothod showed the highest yeild of mononuclear cell counts, CD34-positive cell counts and CFC counts. Conclusion : Cord blood collection processed with red cell lysis could provide enough stem cells for engraftment but other studies revealed different result. This may be due to the different study method. We concluded that RBC lysis method is effective for the red cell depletion of cord blood without major losses of CFC or CD34-positive cells.
