목적 : 엘라스타제는 폐의 탄성 섬유를 파괴하여 폐기종과 폐고혈압을 유발하는 역할을 한다. 이에 본 연구는 문합약침액(文蛤藥鍼液)의 엘라스타제 억제효능과 항산화효능을 측정하여 폐기종과 폐고혈압 치료의 가능성을 확인하고자 실시하였다. 방법 : 문합약침액(文蛤藥鍼液)의 엘라스타제에 대한 억제효능과 di(phenyl)-(2,4,6-trinitrophenyl) iminoazanium (DPPH) 및 nitric oxide(NO) 자유기 소거능을 측정하였다. 결과 : 문합약침액(文蛤藥鍼液)은 엘라스타제 활성을 통계적으로 유의하게 억제하였고, DPPH 및 NO 자유기 소거능이 있음을 통계적으로 유의하게 확인하였다. 결론 : 본 연구를 통해 문합약침액(文蛤藥鍼液)의 엘라스타제 억제효능과 항산화 효능이 확인되었다. 본 연구 결과를 근거로 시험관 내 실험 이외의 추가적인 연구를 통해 폐기종과 폐고혈압 치료제로서 문합약침액(文蛤藥鍼液)의 활용을 고려해 볼 수 있을 것으로 기대한다.
주자독서환(朱子讀書丸)의 아밀로이드베타로 유발된 생쥐 알츠하이머모델에 대한 효과

Object: This research investigated effects of Jujadokseo-hwan on mice with Alzheimer's Disease induced by $amyloid-{\beta}$. According to Dongyibogam, Jujadokseo-hwan can cure amnesia. Amyloid-B is believed to induce oxidative stress and inflammation in the brain, postulated to play important roles in the pathogenesis of Alzheimer's disease. In this way $Amyloid-{\beta}$ induces Alzheimer's Disease. Methods : In order to make an efficient prescription and cope with dementia, learning and memory functions of mice were tested on passive avoidance test and V-maze task. $NF-{\kappa}B$ were measured from protein derived from the brain. RT-PCR was done for !gene analysis. Primers were protein kinase Band $NGF-{\alpha}$. Results : 1. Jujadokseo-hwan was effective for memory capacity on passive avoidance test. but noneffective for spatial memory capacity and locomotor activity on Y -maze task. 2. The measurement of $NF-{\kappa}B$ showed upward tendancies and the result of RT-PCR showed up-regulation when given Jujadokseo-hwan by mouth. Conclusion: Results suggest that Jujadokseo-hwan is effective on mice with Alzheimer's Disease induced by $amyloid-{\beta}$.
개심산 가미방과 구성 약물의 아세틸콜린에스터레이즈 효소활성에 대한 효과

開心散은 鄕藥集成方에서 健忘症 치료에 사용하는 처방이다. 본 연구는 開心散의 효능을 현재 서양의학에서 치매치료약의 치료기전 중의 하나인 아세틸콜린에스터레이즈(AChE) 효소활성에 미치는 효과를 이용하여 연구하였다. 목적 : 健忘治療를 목적으로 구성된 처방인 開心散의 AChE에 대한 작용을 구성약물의 효과와 비교하여 연구하고, 새로운 처방을 구성하기 위해 본 실험이 구상되었다. 방법 : 생쥐 대뇌신경세포의 균질 혼합액으로부터 효소의 활성을 Ellman의 방법을 개선한 엘리자(ELISA)법을 이용하여 측정하였다. 결과 : 실험결과 開心散의 50% 효소활성저해농도(ID_50)를 구한 결과, 0.5 mg/ml이었고, 開心散 加味方의 ID_50은 0.2 mg/ml로 우수한 효소활성저해작용을 나타내었다. 또한 Lineweaver-Burk 식으로부터 계산한 결과 開心散 加味方의 효과는 비경쟁적으로 저해함을 알 수 있었다. 결론 : 健忘 치료를 위해 전통적으로 사용해 온 開心散은 치매치료의 작용기전 중 AChE 효소활성억제효과를 보였으며, 開心散 加味方은 더 높은 효소활성억제효과를 나타내었다. Bee venom has been clinically used to control the pain of inflammation disease etc. in general. Although the effect of bee venom herbal acupuncture has been reported, its mechanism has not been fully elucidated yet. Free radical metabolism seems to occupy a remarkably common position in the mechanisms of inflammation and ageing related disease. Oxidative damage to DNA, lipids, proteins and other molecules may contribute to the development inflammation disease. NO or DPPH is one of the free radicals and a mediator in inflammation diseases. The objective of this study is to investigate the scavenging effect of bee venom herbal acupuncture against NO and DPPH. The followings are the summary of the results: (1) There is no significant scavenging effect of bee venom herbal acupuncture on NO in BVS and BVP group. (2) There is a significant scavenging effect of bee venom herbal acupuncture on DPPH in BVS-1 and BVP-1 group. These results suggest that bee venom herbal acupuncture can be used for inflammation diseases such as rheumatoid arthritis. This study would provide important basic data on the possibility of the clinical treatment of bee venom herbal acupuncture. Further studies are required to investigate the antioxidative effects of it.
동의보감(東醫寶鑑) 중 고마요법(膏摩療法)에 사용된 방제(方劑)에 대한 고찰(考察)

The purpose of this study is to expand the prescription usage from internal treatment to external treatment by applying the compatibilities of herbal ingredient for ointment massage therapy. For this, specific cases of usage were investigated in Donguibogam, which is now inscribed on the UNESCO Memory of the World Register, has been not only one of the classics of Korean medical asphere, but has compiled all the contents of the Oriental medical traditions including ointment massage therapy that was completed through different Chinese dynasties, such as Song (宋), Jin(金) and Won(元) by the 17th century. Research results of this study is : 1. Ointment Massage Therapy was used to treat the symptoms of diseases such as ACHI-mun(dental range : 牙齒門), PI-mun(dermal range : 皮門), JECHANG-mun (sore range : 諸瘡門), SO A-mun(pediatrics range : 小兒門). 2. Out of all the manipulations, it is a scrubbing manipulation that used most. 3. Out of all the formulations of ointment massage therapy, it is powder types and ointment types that most representative formulation.
Epigallocatechin-3-gallate의 사람 비점막 섬유아세포 케모카인발현에 대한 효과

Epigallocathechin-3-gallate (EGCG), the main polyphenol component in green tea, inhibits angiogenesis, urokinase, and matalloproteinases, and EGCG also has the antioxidative property. Recent reports proposed that EGCG may modulate the immune response on allergy or asthma. Human nasal mucosal fibroblasts are a rich source of cytokines, inflammatory mediators, and chemokines. Chemokines are important for the recruitment of leukocytes to sites of infection, which is essential in host defense. The objective of this study was to investigate the effect of EGCG on the expression of the chemokines such as RANTES (regulated upon activation, normal T cell expressed and presumably secreted), eotaxin, and interleukin-8 (IL-8) in human nasal mucosal fibroblasts after stimulation with cytokines like IL-4, tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, and $interferon-{\gamma}\;(IFN-{\gamma})$. To detect the expression of chemokine genes, RT-PCR was performed. Expressions of RANTES, eotaxin, and IL-8 mRNA stimulated with IL-4 and $TNF-{\alpha}$ were increased, respectively, while the expression of those genes incubated with $IFN-{\gamma}$ was similar pattern compared to control group. Analyses of chemokine genes of cells pretreated with EGCG showed that the expressions of eotaxin, and IL-8 genes stimulated $IFN-{\gamma}$ were higher compared with those not pretreated with EGCG.

본 연구는 4종의 재래닭인 연산오계, 현인흑계, 횡성약닭, 황봉의 일반성분, 카르노신, 안세린 함량 및 면역활성을 비교하기 위하여 실시하였고, 대조구로 백색 레그혼을 사용하였다. 시험용 닭을 모두 동일한 조건에서 13주간 사육하여 도계한 후 화학적 성분조사를 실시하였다. 일반성분 분석에서는 4종의 재래닭이 백색레그혼에 비하여 조지방 함량이 높았으며, 콜레스테롤 함량은 현인흑계가 다른 품종에 비하여 높았고 횡성약닭이 유의적으로 낮았다(p<0.05). 면역증진 효능을 측정하기 위하여 대식세포를 이용하여 실험한 결과는 재래닭 4종의 가슴육에서만 대식세포 증식 및 NO 억제효능이 있었다. 대식세포 탐식작용은 현인흑계와 횡성약닭가슴육에서 유의적으로 증가되었고, 염증 관련 사이토카인(IL-4, IL-10, IFN-γ) 유전자 발현은 백색레그혼에 비하여 4종의 재래닭에서 억제효과가 컸다. 결론적으로 4종류의 재래닭이 백색레그혼에 비하여 면역증진 효능이 높았으며, 특히 가슴육에서 효능이 있었다. 재래닭 품종 간에도 일반성분 및 면역조절능에 차이가 있는 것으로 나타났다. This study was conducted to compare the general composition and immunomodulatory activity of breast and thigh meats from four lines of Korean native chickens: Yeonsan Ogye, Hyunin Black, Hwangbong, and Hoengseong Yakdak. White Leghorn was used as a control. Fifteen male chickens (three chickens in each line) were grown under the same conditions and slaughtered at 13 weeks old. The four lines of Korean native chickens, regardless of the part, had higher contents of crude fat (p<0.05) than White Leghorn. The cholesterol contents were significantly higher in Hyunin Black and significantly lower in Hoengseong Yakdak than those of other chickens (p<0.05). The immunomodulatory effect, assessed by macrophage cell proliferation and nitric oxide production, was only observed in the breast meat of the four lines of Korean native chickens. The phagocytic activity of macrophage cells was significantly augmented by the breast meat of Hyunin Black and Hoengseong Yakdak. Furthermore, the mRNA expressions of pro-inflammatory cytokines, such as IL-4, IL-10 and IFN-γ, was significantly suppressed by Korean native chickens compared with White Leghorn. These results suggested that the four lines of Korean native chickens exhibited greater immune-enhancing activity than White Leghorn.
