        한국의학사 강의 개발과 강의 사례 연구

        This research will explore how to develop and teach Korean medical history in Korean universities. In 2009-2010, I participated in a project called “Developing the Module of the Korean Medical History,” which was funded by the Korean Research Foundation. For this project, done at Yonsei Medical College, five specialists worked together to develop the module: Yeo In-sok (Director of the project, Contemporary period), Lee Hyun-sook (Ancient period and the Goryeo dynasty), Kim Seong-su (Joseon dynasty), Sihn Kyu-whan (Modern 1: the late 19th century and early 20th century), and Park Yun-jaeoe (Colonial period). One of the results of this project was the publication of the book History of Medicine in Korea. As a case study, I analyzed the results of my experiences teaching Korean medical history to graduate students in medical college in 2010 and freshmen in nursing college in 2013. In the graduated school of medical college, all of the five members gave lectures, however, it seems like too many lecturers for five students in eight classes. I could not give any previous reading material and assignment. It means we need more time to teach properly. In 2013, I taught Korean medical history by myself as the Korean History class in a nursing college. I invited Prof. Kim and Sihn as a guest teacher. According to my survey of the nursing college`s freshmen students, my students selected the forensic medicine of the Joseon dynasty and the military medicine of Silla Unification War in the 7th Century as the best subjects. The next favorite subjects were the comparison of the Modernization and Exploitation theories in colonial Korea, as well as Ancient Medicine, specifically trephination and skull modification. They selected these because the visual media, made by KBS, about the forensic medicine of the Joseon dynasty and about the artificial cranial deformation of the Gaya, which we had watched in the class. From this, I discovered that watching visual images was the most efficient method of learning for the class. Another assignment I gave to my classes concerned the military medicine of ancient Korea, and my students were responsible for reading my article “Military Medicine of the Silla Unification War,” summarizing the article, and recording their thoughts and impressions. This subject was also a favorite. I determined that self-study was a very efficient way to develop students` ideas and passions. The results of the survey about the comparison of the Modernization and Exploitation theories revealed that my students supported the Exploitation theory more than the Modernization theory. In the 1st class, 17 students supported the Modernization theory, while 49 students subscribed to the Exploitation theory in the Colonial medicine. On the other hand, only 7 students agreed with the Modernization theory at the 2nd class, while 55 students supported the Exploitation theory. I believe this is the result of the influence of Prof. Sihn, who had provided a special lecture to my students as a supporter of the Exploitation theory. I also recognized that my way of questioning had influenced the answers of the students. In the question of the 1st class, I had explained the Modernization theory at first, and then I had them choose “agree” or “disagree” and explain their answer. In the 2nd class, on the contrary, I introduced the Exploitation theory in advance and had them select “agree” or “disagree.” The 2nd class might have been influenced by the method of questioning, and they selected the Exploitation theory as an easier answer. Most of the nursing college`s students were content learning about the history of Korean medicine as a Korean history class. The text book History of Medicine in Korea played a considerable role in guiding my students, even though it has many weaknesses. In conclusion, the history of Korean medicine is a good subject for teaching Korean history at a university, and it also develops the identification of medical college students.

        High-Pressure Synthesis of $SmFeAsO_{1-x}F_x$(x=0.2) Single Crystals

        Fluorine-doped $SmFeAsO_{1-x}F_x$ single crystals with the nominal value of x=0.2 were grown at $1350-1450^{\circ}C$ under the pressure of 3.3 GPa by using the self-flux method. Plate-shaped single crystals in the range of a few-150 ${\mu}m$ in their lateral size were obtained. The detailed crystal structure was analyzed by using the x-ray diffractometry. Superconducting transition temperature, determined by the resistive transition, of a single crystal was about 49 K with a narrow resistive transition width of ${\sim}1$ K. A relatively sharp transition, a low residual resistivity, and a large residual resistivity ratio compared with those reported for $REFeAsO_{1-x}F_x$(RE=Sm, Nd) single crystals indicate the high quality of our single crystals.

        중고생의 인구학적 특성, 건강, 인터넷 중독과 약물사용과의 관계

        본 연구는 제5차 청소년건강행태 온라인조사 자료를 통해 중고생의 인구학적 특성, 건강, 인터넷 중독 요인에 따른 약물사용의 차이를 비교하고자, 첫째, 각 변인에 대한 빈도분석, 백분율, 평균 등의 기술 통계분석을 진행하였으며, 둘째, 각 독립변인에 따른 약물사용 여부의 차이를 고찰하고자 카이자승 검증을 진행하였다. 연구결과를 정리하면, 인구학적 특성 요인에 있어서는 남학생이 여학생보다, 부모와 동거하지 않는 경우에, 가계의 경제수준이 높은 경우에, 중학생의 경우에, 성적이 낮은 경우에, 건강 요인에서는 주관적으로 건강하지 못하다고 느끼는 경우에, 주관적으로 행복하지 못하다고 느끼는 경우에, 스트레스정도가 낮은 경우에, 우울을 경험한 경우에, 그리고 인터넷 중독 요인에 있어서는 인터넷 중독인 경우에 그렇지 않은 경우에 비해 통계학적으로 유의한 수준에서 높은 약물사용 빈도를 보이는 것으로 나타났다. 이상의 결과를 토대로 본 연구에서는 중고생의 약물 사용에 대한 예방과 대책에 대해 논의를 진행하였다. This study aimed to examine the relationships between demographic characteristics, health, and internet addiction factors and drug uses among the Korean high school students. The 2009 Korean Youth Health Risk Behavior On-line Survey data was used for this study. It selected 73,238 cases of junior and senior high school students from the raw dataset and conducted a secondary data analysis. Frequency analysis and chi-square test were conducted for this study. Gender, living with parents, economic status, school level, academic record, subjective health status, subjective well-being, level of stress, level of depression and internet addiction were significantly associated with their drug uses. Findings may be used for developing strategies to cope with drug uses among the Korean high school students.

        Ultrastructural Localization of GABAergic Neuronal Components in the Dog Basilar Pons

        An immunocytochemical study of GABA-positive neuronal elements was performed at the electron microscopic level to examine subcellular distribution of the inhibitory neurotransmitter in the dog basilar pons. Electron-dense reaction product was observed in neuronal somata and dendritic processes. One or more unlabeled axon terminals made asymmetric synaptic contacts with these GABAergic somatic and dendritic profiles. A large number of GABA-positive axon terminals were also observed. They made symmetric as well as asymmetric synaptic contacts with unlabeled dendritic profiles. In axo-axonic synapses, postsynaptic axon-like processes were consistently GABA-immunoreactive. These observations suggest that the inhibitory local circuit neurons in the dog basilar pons play a major role in cerebro-ponto-cerebellar circuitry by integrating various afferent inputs and conveying them into the cerebellar cortex and the deep cerebellar nuclei. 개의 교핵내의 GABA를 함유한 억제성 신경세포의 연접부위를 전자현미경을 이용한 면역세포화학적 방법으로 조사하였다. 반응산물은 신경원의 핵부위나 가지돌기에서 관찰되었으며, 반응산물을 포함하고 있지 않은 신경종말이 이들과 비대칭형 연접을 형성하였다. 그 외에도 많은 수의 GABA성 신경종말이 관찰되었으며, 이들은 반응산물을 함유하지 않은 가지돌기와 대칭형 또는 비대칭형 연접을 형성하였다. 한편 축삭돌기-축삭돌기 연접에서는 연접후 유축사돌기 (axon-like processes)가 GABA-양성이었다. 이와 같은 관찰은 개의 교핵에 존재하는 억제성 개재신경원이 여러 수입계로부터의 정보를 통합하고, 이를 소뇌피질이나 소뇌핵에 전달함으로써, 대뇌-교핵-소뇌계의 신경경로에서 조절기능을 담당함을 뒷받침하는 것이다.

        가미청폐탕, 가감정기탕, 가미사물탕, 가미이중탕이 호흡기 뮤신의 분비 및 뮤신 유전자의 발현에 미치는 영향

        Objectives In this study, the author intended to investigate whether Gami-cheongpetang (GCP), Gagam-jeongkitang (GJG), Gami-samooltang (GSM) and Gami-ijoongtang (GIJ) significantly affect in vivo (animal model) and in vitro (cultured cells) mucin secretion and MUC5AC gene expression in airway epithelial cells. Methods For in vivo experiment, the author induced hypersecretion of airway mucin in rats by introducing SO2 for 3 weeks. Enzyme-linked immunosorbent assay (ELISA) was used to assess the effects of orally-administered GCP, GJG, GSM and GIJ in vivo mucin secretion from tracheal goblet cells of rats after 1 week. Also, the effects of the agents on TNF- or EGF-induced MUC5AC gene expression in human airway epithelial cells (NCI-H292) were investigated. Possible cytotoxicities of the agents were assessed by examining the rate of survival and proliferation of NCI-H292 cells. Results (1) GCP and GJG significantly inhibited hypersecretion of in vivo mucin, although GSM and GIJ did not affect hypersecretion of in vivo mucin; (2) GCP and GJG significantly increased in vitro mucin secretion from cultured HTSE cells. However, GSM and GIJ did not affect in vitro mucin secretion from HTSE cells; (3) GCP and GJG significantly inhibited the expression levels of EGF-induced MUC5AC gene in NCI-H292 cells. However, GSM and GIJ increased the expression levels of EGF-induced MUC 5AC gene in NCI-H292 cells; (4) GCP, GJG, GSM and GIJ did not significantly inhibit the survival and proliferation of NCI-H292 cells. Conclusions These results suggest that GCP, GJG, GSM and GIJ can not only affect the secretion of mucin but also affect the expression of mucin gene. The author suggests that the effects of GCP, GJG, GSM and GIJ with their components should be further investigated by using animal experimental models that simulate the diverse pathophysiology of pulmonary diseases.

        Nonlinearity in the Somatosensory Cortex Response to Vibrotactile Stimulator in fMRI

        기능성 자기공명영상을 이용하여 진동자극에 대한 감각피질의 비선형성에 대하여 연구하였다. 진동자극은 25 Hz의 주파수로 5개의 각기 다른 진동 자극 기간, 2초, 4초, 8초, 12초 그리고 16초와 함께 20초의 무자극 주기와 pseudo-random순서로 구성되었다. 감각피질에 대한 자극 기간의 변화에 따른 선형성을 이해하기 위하여 두 다른 방법(시스템의 선형성 조사법과 gamma-variate 함수를 적용하여 impulse response함수를 구하는 방법)을 사용하여 혈류반응 함수를 분석하였는데, 그 결과는 거의 비슷하였다. 감각피질에서의 BOLD 반응은 8초보다 작은 자극 기간 동안에는 비선형이고 8초 이상일 때는 선형성을 보였다. 자극 기간의 함수로 Impulse response 함수의 진폭, 면적, 피크까지의 시간, FWHM을 계산하였고 진폭과 면적의 크기는 자극 기간이 증가할 때 감소함을 보여줌으로써 자극 기간 8초에서 BOLD 반응이 비선형성에서 선형성으로 변함을 뒷받침해 주었다. The nonlinearity of hemodynamic response in the somatosensory cortex was investigated with vibrotactile stimulation. The stimuli consisted of a train of 25 Hz, each tasting five different duration periods, 2 s, 4 s, 8 s 12 s, or 16 s with 20 sec periods of no vibration in a pseudo-random order. In order to understand the linearity on the change of stimulus duration for somatosensory cortex, two different tests- checking the linearity of system and finding the impulse response function from gamma-variate function were applied to analyze the hemodynamic response functions. They have produced nearly same results. The BOLD response in the somatosensory cortex Is nonlinear for stimuli of less than 8 seconds, but nearly linear for stimuli greater than 8 seconds. The amplitude, area, TTP, and FWHM as functions of the stimulus duration were calculated and showed a significant downward trend with Increasing stimulus duration for the amplitude and the area. It supports the ranges of nonlinearity are less than 8 seconds.

        Ultrastructural Observations of Glutamatergic Synaptic Components in the Basilar Pontine Nuclei of the Dog

        The distribution of glutamatergic synaptic structures in the dog basilar pons was investigated at the ultrastructural level using monoclonal antibodies against fixative-modified glutamate. Electron-dense reaction product was densely localized at the perinuclear region in the neurenal somata and often observed along the microtubules located within the dendritic processes. One or more unlabelled axon terminals made asymmetric synaptic contacts with glutamate-immunoreactive dendritic profiles. In audition, reaction product was observed either within axonal processes surrounded by myelin sheath or axon terminals. Immunoreactive axon terminals made asymmetric synaptic contact either with unlabelled or labelled dendritic profiles. These observations provided an anatomic evidence of how this excitatory neural element might perform its function in a multisynaptic pathway involving glutamatergic afferents to the basilar pons, glutamate-immunoreactive pontocerebellar projection neurons, and the glutamate-positive granule cells of the cerebellar cortex. 개의 교핵내 glutamate를 함유한 연접 구조의 분포를 fixative-modified glutamate에 대한 monoclonal antibody를 사용하여 미세구조적 차원에서 조사하였다. 반응산물은 신경세포체의 핵주위 영역에 위치하였으며, 가지돌기내의 미세소관을 따라서 존재하였다. 반응산물을 함유하지 않은 한 개이상의 신경종말이 glutamate에 면역반응을 나타낸 가지돌기와 비대칭형 연접을 형성하였다. 그 외에도 수초로 둘러싸인 축삭돌기나 신경종말내에서도 반응산물이 관찰되었다. 이들 신경종말은 표지된 또는 표지되지않은 가지돌기와 비대칭형 연접을 형성하였다. 이상의 관찰은 glutamate에 면역반응을 나타내는 흥분성 교핵 신경원이, glutamate를 함유한 교핵으로의 여러 수입계나, 교핵으로부터 소뇌로의 투사 신경세포, 그리고 소뇌피질내의 glutamate성 과립세포를 포괄하는 다연접 신경경로에서 어떤 역할을 수행하는지에 관한 형태학적 근거를 제시한다.

        A Reliable Protocol for transfection of mature primary hippocampal neurons using a neuron-glia co-culture system

        형질전환은 유전자의 기능을 이해하는데 매우 중요한 기법이다. $Ca^{2+}$-인산 침전법은 시간과 비용이 저렴하여 가장 흔히 사용된다. 그러나 성숙 신경세포는 어린 신경세포나 다른 세포종에 비하여 형질전환이 어렵고 쉽게 죽는다. 본 연구에서는 Clontech사의 $CalPhos^{TM}$ Mammalian Transfection 방법을 수정하여 성숙한 신경세포를 효율적으로 형질전환할 수 있는 방법을 고안하였다. 대뇌 신경교세포를 DMEM/10% 말혈청에서 70-80% confluence까지 키우고 배지를 혈청이 첨가되지 않은 Neurobasal/Ara-C로 바꾸어 주어 더 이상 신경교세포가 분열하지 않게 한 다음, 여기에 E19 해마신경세포를 접종하여 배양하였다. $DNA/Ca^{2+}$-인산 침전물은 Clontech사의 $CalPhos^{TM}$ Mammalian Transfection Kit을 이용하여 크기($0.5-1\;{\mu}m$ in diameter) 및 농도(약 10 particles/$100\;{\mu}m^2$)를 배지에서 배양시간을 변화시켜 적당히 조절하였다. 이렇게 하면 in vitro에서 2주 이상 배양한 신경세포도 24-well plate 한 well당 10-15개의 형질전환된 건강한 신경세포를 얻을 수 있었다. 이 방법의 효용성을 검증하기 위하여 연접단백질인 $EGFP-CaMKII{\alpha}$ 융합단백질과 RFP 단백질 유전자(각각 $pEGFP-CaMKII{\alpha}$ 및 pDsRed2)를 형질전환한 결과 전자는 점박이 모양, 후자는 세포전체에 퍼진 양상의 표현을 관찰할 수 있었다. 따라서 본 연구는 성숙한 신경세포를 효율적으로 형질전환할 수 있는 방법을 제공한다. DNA transfection is a powerful tool for studying gene functions. The $Ca^{2+}$-phosphate precipitation remains one of the most popular and cost-effective transfection techniques. Mature neurons are more resistant to transfection than young ones and most other cell types, and easy to die if microenvironment changes. Here, we report a transfection protocol for mature neurons. The critical modifications are inclusion of glial cells in culture and careful control of $Ca^{2+}$-phosphate precipitation under microscope. Cerebral glial cells were grown until ${\sim}70-80%$ confluence in DMEM/10% horse serum, which was thereafter replaced with serum-free Neurobasal/Ara-C, and 319 hippocampal neurons were plated onto the glial layer Formation of fine $DNA/Ca^{2+}$-phosphate precipitates was induced using Clontech $CalPhos^{TM}$ Mammalian Transfection Kit, and the size ($0.5-1\;{\mu}m$ in diameter) and density(about 10 particles/$100\;{\mu}m^2$) were carefully controlled by the time of incubation in the medium. This modified protocol can be reliably applied for transfection of mature neurons that are maintained longer than two weeks in vitro, resulting in 10-15 healthy transfected neurons per a well of 24-well plates. The efficacy of the protocol was verified by punctate expression of $pEGFP-CaMKII{\alpha}$, a synaptic protein, and diffuse expression of pDsRed2. Our protocol provides a reliable method for transfection of mature neurons in vitro.

        Modification of Tomato Aspermy Virus Symptom by Cucumber Mosaic Virus- Associated Satellite RNA

        Tomato aspermy virus(TAV-B)는 두 개의 다른 CMV균주 D 및 K에서 분리한 satellite RNA의 증식 및 encapsidation에 대하여 helper virus의 역할을 하였다. 이 두 satellite RNA는 역시 ??? system에서 보이는 바와 같이 TAV가 감염된 담배에서 TAV 병징을 크게 감소시켰으며, 이는 감염된 식물내의 Virus 함량의 감소와 관계가 있었다. 또한 CMV가 감염된 토마토에서 lethal necrosis를 유발시킴을 볼 수 있었다. A tomato Aspermy Virus (TAV-B) served as a helper virus for multiplication and encapsidation of satellite RNAs which were isolated from two different CMV isolates, D and K. These two satellite RNAs induced renarkable attenuation of TAV symptoms in infected tobacco, which was correlated with a reduction of virus content in the plant. The CMV satellite RNAs also caused lethal necrosis in TAV-infected tomato as in the case of CMV system.

        중학생들의 대기압에 대한 개념을 향상시키기 위한 실험방법 개선안

        본 연구는 중학교 교과서의 대기압 단원을 학습하였을 때 학생들의 개념유형이 어떻게 변하는지 조사하고, 대체개념을 개선해나갈 수 있는 실험방법을 모색하는데 목적이 있다. 중학교 2학년을 대상으로 ‘대기압’ 에 관한 사전 개념을 질문지와 면담을 통하여 조사하고 두 집단으로 나누어 6차 교육과정의 중학교 2학년 과학교과서 중 8종의 교과서에 나오는 대기압에 관한 실험을 모두 한 집단과 교과서 외의 제안실험을 한 집단으로 구분하여 2주 동안 실시하였다. 두 집단간의 수업방식은 차별화하지 않았고 실험소재만 다르게 하였으며, 기압단원 학습 일주일 후 동일한 질문지를 사용하여 개념변화를 알아보았다. 결론적으로 학생들의 기압에 대한 대체개념이 학습 후에도 쉽게 변화되지 않았지만, 제안실험을 한 집단이 기압에 대한 개념형성이 효과적으로 나타난 점으로 보아 대체개념을 개선시킬 수 있는 다양한 학습방법과 과학적 개념획득을 위한 실험손재가 지속적으로 연구되어져야 할 필요성이 있다. The purpose of this study is to investigate the types of concepts about atmospheric pressure of middle school students and to design a new method to improve students' understandings on atmospheric pressure. The second grade students of the middle school has been selected and examined their pre-concepts on atmospheric pressure through questionnaires and interviews. Two groups of students have been taught using the second grade science textbooks and experimental methods designed from this study, respectively. The results suggest that we need to develope various teaching methods that can improve alternative concept as well as experimental tools that can help students understand difficult scientific concepts. The reason is that once alternative concept is occupied, it is extremely difficult to fix it. In addition, the proposed experimental methods are effective in establishing the concept and understanding on atmospheric pressure.
