      • In Vitro Translation of Glucose-6-Phosphate Dehydrogenase Isozyme mRNA and In Vivo Isotope Labeling of the Enzyme from Peking Duck

        오리의 uropygial gland와 간에서 $poly(A)^+mRNA$를 분리하여 그들의 in vitro translation mixture로부터 pI 6.6인 glucose-6-phosphate dehydorgenase를 분리 하고 in vivo isotope labeling된 같은 효소를 분리하여 SDS polyacrylamide disc gel electrophoresis를 통하여 비교하였다. 그 결과 in vitro translation mixture로 부터 분리한 효소와 in vivo labeling된 효소는 그것이 uropygial gland의 것이거나 간의 것이거나 모두가 uropygial gland에서 직접 정제한 효소와 같은 크기임을 알 수 있었다. 이러한 사실은 uropygial gland에 존재하는 pI 6.6인 glucose-6-phosphate dehydrogenase가 갖는 효소적 특성은 소수의 아미노산의 차이에 기인하는 것으로 예측된다. One glucose-6-phosphate dehydrogenase with pI of 6.6 was isolated from in vitro translation mixture and was compared with in vivo isotopically labeled enzyme by SDS polyacrylamide gel electrophoresis. The molecular size of the enzymes, isolated by antibody prepared against the purified enzyme and protein A agarose, are very similar to each other. This result suggests that the specific catalytic properties of the enzyme from the uropygial gland may depend on the minor difference in amino acid sequence.

        전산화단층사진을 이용한 타액선의 정량분석에 관한 연구

        Purpose: The purpose of this study was to calculate the size and CT number of both normal parotid and submandibular gland. and evaluate their relation to sex, age and obesity using computed tomography. Materials and Methods: The computed tomography was performed parallel to the Frankfurt plane in 46 subjects with healthy salivary gland. The subjects were divided into the three groups (young, middle. old) according to their ages. The size of salivary gland was determined as maximum cross-sectional area and the CT number of salivary gland was determined as the mean CT number of three ROI's. The body mass index was calculated from weight and height. Results: The mean maximum cross-sectional area was 7.79(±1.25)cm² on parotid gland and 4.12(±0.83) cm² on submandibular gland. The mean CT number was -4.43(±23.87) HU on parotid gland and 50.01(±15.63) HU on submandibular gland. There was decreasing pattern of the maximum cross-sectional area of submandibular gland and the CT number of both parotid and submandibular gland according to age(p<0.05). As the body mass index increased. the maximum cross-sectional area of parotid gland increased and CT number of both parotid and submandibular gland decreased(p<0.05). The maximum cross-sectional area of submandibular gland in male was larger than that in female(p<0.05). As the maximum cross-sectional area and CT number of left salivary gland increased. those of right gland increased(p<0.05). Conclusion : Intra-individual differences in salivary gland size and CT number is considered in the age and individual obesity.

        석회암 공동지역의 교량기초 보강에 관한 연구

        국내 강원도 지역에 광범위하게 분포하고 있는 불규칙한 석회암 공동(空洞)은 구조물의 사용성 저하 및 붕괴 등 예기치 못한 사고를 유발할 수 있다. 따라서 석회암 공동분포지역에서는 구조물의 기초지지력 증진을 위한 적절한 지반보강공사가 요구되며, 환경오염 문제를 최소화할 수 있어야 한다. 일반적으로 석회암 공동의 기초지지력 확보를 위해 시공하는 R.J.P(Rod Jet Pile) 공법은 주입재료를 시멘트모르타르를 사용하지만, 석회암 공동의 특성상 공동을 통한 시멘트성분의 유출로 인근지역의 식수오염과 양어장의 송어폐사가 발생하였다. 따라서 이러한 문제점을 해결하기 위해서 국내 외 적용가능한 보강공법들을 비교 검토하였으며, 실내시험을 통해 주입재료를 선정하였다. 또한, 1, 2단계로 구분하여 시공하고 선정된 보강재료 주입을 통해 저유동성 재료를 사용하는 비배출압밀공법을 한단계 향상시킨 몰탈압입공법을 제안하고 시험시공을 하였다. 시험시공 동안 인근지역 양어장의 수질변화가 거의 발생하지 않았으며, 시공위치에서 코어를 채취하여 압축강도를 측정한 결과 기준강도 5 MPa 이상으로 구조물의 기초지반 지지력을 확보하는 것으로 분석되었다. 따라서 몰탈압입공법은 주변 수질환경에 대한 친환경성을 확보함과 동시에 기초지반 지지력을 확보하는 것으로 검토 되었다. Irregular distributions of limestone cavity in Gang-Won province area may cause unexpected accidents from reduced serviceability or failure of structure. It is requested that an appropriate ground reinforcement method should be used to improve bearing capacity of structure, and the method should also be satisfied with environmental requirements. Among several methods used for foundation constructions in cavity area, Rod Jet Pile(RJP) method has been widely used. While the RJP method was used to improve bearing capacity for the railway bridge foundations, water pollutions of drinking water as well as fishery located adjacent to this project area were occurred. The main reason of the water pollution was cement runoff used in cement mortar during injecting material in RJP method. Laboratory tests were performed to prevent water pollution. The compaction mortar method using low movable material was selected for this project. The quality of water at a fishery adjacent to the site and the compressive strength of cores taken from the construction site were measured. Test results show that the water pollutions was minimized, and the average compressive strength of foundation material was over 5 MPa. As a result of this study, compaction mortar method can be used to ensure the bearing capacity of foundation and to prevent environment pollutions.

        인체 α 1 - antitrypsin cDNA 의 분리 및 E . Coli 내에서의 발현

        Human α1-AT cDNA was screened from human liver cDNA library, by use of oligonucleotide probes. DNA insert of 1.3 kbp from a positive clone was subcloned into pUC9 plasmid and was analyzed by restriction enzyme digestion and partial DNA sequencing. The obtained gene was identical to human αl-AT cDNA reported previously. In order to express αl-AT cDNA under regulation of tac promoter, expression vector pT-AT was constructed. E. coli JM109 cells transformed with pT-AT showed αl-AT activity upon induction of recombinant protein synthesis. The recombinant αl-AT was immunologically identical to authentic αl-AT that was purified from human blood. When the recombinant cell lysates were analyzed on SDS-polyacrylamide gel electrophoresis, the expressed α1-AT was not detected by staining with Coomassie Brilliant Blue. However Western blotting of the same gel, using rabbit IgG against αl-AT and peroxidase-conjugated goat anti-rabbit IgG antibody, showed the existence of αl-AT at the expected molecular weight region. Molecular weight of recombinant αl-AT was slightly lower than that of authentic αl-AT; which presumably was due to lack of glycosylation inside E. coli cells.

        Robotized MIG welding of large aluminium components for high speed trains is state-of-the-art. The implementation of online laser cameras enables seam tracking and adoptive modification of welding parameters. A constant fill is achieved regardless of the gap tolerances. Friction Stir Welding has been introduced to the market as a reliable and fast joining technology. The advantages of high welding speeds and the elimination of arc light, fumes and liquefaction in the welding spot lead to economical realisation of heavy-duty gantry systems. FSW robots offer a high flexibility with regard to welding of curved parts, and can be equipped with laser cameras for exact joint tracking.

        분석자동화를 위한 용액 조제 시스템 설계에 관한 연구

        석유화학 단지에서는 보다 완성도 높은 제품을 위한 많은 연구 개발이 진행되고 있으며 이 과정에서 제품의 중간 및 완성 제품에 대한 분석은 필수적이다. 그러나 이와 같은 분석 작업들이 아직도 많은 부분에서 수작업에 의한 분석 시료가 제조되고 있고 위험한 화학물질에 노출되어 열악한 환경에서 분석이 이루어지고 있다. 본 연구에서는 이와 같은 문제점을 해결하기 위하여 분석 자동화를 위한 멀티 제어 시스템을 개발하였다. 또한 이들 장치의 유기적인 동작과 자동화를 위한 프로그램을 개발하여 적용하고 이들 장치의 신뢰성 검증을 위한 실험을 통하여 표준 용액 조제를 위한 정량 펌프의 정확도가 ${\pm}0.01m{\ell}$의 오차 범위를 갖는 매우 우수한 결과를 얻을 수 있었다. Petrochemical complex has been a lot of research for the development of a more mature product and analysis for mid-process and finished products is essential in these process. But these analyzes are still by hand work samples being manufactured in many parts. Moreover they are exposed to hazardous chemical and such as the analysis is being made in a very poor working conditions. In this paper, in order to solve such problems the multi control system has been developed for the automated analysis. In addition, the organic behavior of these systems and the development of a program for the automated applied, and throughout the experiment to verify the reliability of this device for the accuracy of the dosing pumps for the standard solution prepared with a range of error of ${\pm}0.01m{\ell}$ was able to get a very good experimental results.

        심리적 유인과 사이트 품질, 공동체의식이 온라인게임에 미치는 영향

        The purpose of this research is to identify if psychological temptation, site quality and sense of community influence user's flow and addiction and to identify causalities among flow, addiction, customer satisfaction and customer loyalty in online industry. Many previous researches in the area of online games have been carried out about addiction by psychologist and about the development of related technologies by scientists. There are only a few studies about the customer satisfaction from the online business perspective. However, this research is different from the previous ones in the sense that both flow and addiction are considered in the study of the relationship between customer satisfaction/loyalty and flow/addiction in the area of online industry. The empirical results of high-order factor analysis indicate that six independent variables such as design, information, feedback, impulsiveness and motivation have converge three second-order variables such psychological temptation, site quality and sense of community. Consequently, site quality and sense of community have impacts on the flow, while on the other hand, psychological temptation has impacts on the addiction. Conclusively, customer satisfaction and loyalty are positively related not with the addiction but with the flow. Besides, customer loyalty is significantly influenced by the flow and the customer satisfaction. This indicates that companies in the online game industry have to develop a strategy for the flow which is more socially and ethically allowable than the addiction.

        Pyridoxal - 5 ' - Phosphate 에 의한 Rhizobium trifolii Malonyl - CoA Synthetase 의 화학변형

        Pyridoxal-5`-phosphate inactivated malonyl-CoA synthetase from Rhizobium trifolii reversibly. The second-order rate constant, K_i was 2975 M^(-1) · min^(-1) at pH 7.0, 30℃. Inactivation of malonyl-CoA synthetase by PLP was reversed by dilution but not after reduction with NaBH₄. The spectra of malonyl-CoA synthetase, inactvated by PLP and reduced by NaBH₄, showed a characteristic Schiff base absorption change at 325 nm. ATP protected the enzyme from inactivation by PLP. These studies indicated that an essential amine is located at or near ATP binding region of the enzyme active site.

        염화나트륨 농도가 느타리 자실체의 아미노산과 향기성분에 미치는 영향

        본 연구는 염화나트륨에 의한 비생물적 스트레스가 생육과정 중의 느타리에게 미치는 영향을 자실체의 생육특성, 그리고 구성아미노산함량과 향기패턴 분석을 통해 확인하였다. 농도별로 염화나트륨을 처리한 톱밥배지에서 수확한 느타리 자실체의 생육특성을 조사한 결과, 처리구별 수량은 무처리구 대비 염화나트륨 0.5%처리구에서의 값은 비슷했으며, 염화나트륨 처리 농도가 1.0%에서 2.0%으로 증가하면서 수량이 확연히 감소하였다. 자실체의 구성아미노산함량 분석결과, 무처리구 대비 모든 염화나트륨 처리구에서 glutamic acid과 proline을 제외한 aspartate, threonine, serine, glycine, alanine, methionine, valine, isoleucine, leucine, phenylalanine, tyrosine, lysine, histidine, arginine의 14종 구성아미노산의 함량이 낮음을 확인하였다. 자실체의 향기패턴 분석결과, 염화나트륨 처리구에서 버섯고유의 향을 나타내는 octane compound로 추정되는 물질의 intensity가 감소하였음을 chromatography상에서 확인하였다. We investigated the effect of sodium chloride-associated abiotic stress on the development of Pleurotus ostreatus. We examined the growth characteristics of fruiting bodies, constituent amino acids, and fragrance pattern to determine the effect of culturing Pleurotus ostreatus on a sawdust substrate supplemented with sodium chloride in a dose-dependent manner. Pleurotus ostreatus fruiting bodies exhibited an increasing tendency towards augmented yields when grown in the presence of 0.5% sodium chloride as compared with that grown in the control group. However, increasing the supplementation of sodium chloride from 1.0 % to 2.0% resulted in significantly decreased yields of Pleurotus ostreatus fruiting bodies in these groups as compared with that in control groups. Further assessment revealed the presence of 14 types of amino acids in the fruiting bodies, including aspartate, threonine, serine, glycine, alanine, methionine, valine, isoleucine, leucine, phenylalanine, tyrosine, lysine, histidine, and arginine, at lower levels in all the sodium chloride-treated groups than in the control group; except for glutamic acid and proline. Similarly, fragrance pattern analysis of the Pleurotus ostreatus fruiting body by chromatography confirmed that the intensity of the substances presumed to be octane compounds, to which the unique flavor of mushrooms is attributed, was lower in all the sodium chloride-treated groups than in the control group.
