Roles of Nucleosides on Amino Acid Transport Systems of E. Coli

E. coli를 배양하여 삼투에 의한 충격으로 막 내의 단백질을 추출시킨 후, 이들 E. coli의 아미노산 수송에 관여하는 Nucleoside의 영향을 관찰하였다. Pyrimidine Nucleoside인 Cytidine과 Uridine이 아미노산 수송을 크게 증가시켰는데 Purine Nucleoside들은 효과가 조금 뒤졌다. 증가된 아미노산 수송은 주로 세포내의 아미노산 pool에 도착되었고, 단백질이나 RNA합성을 저지하여도 이 수송은 영향을 받지않았다. 이런 자극 효과는 Nucleoside의 독특한 효과에 의한 것으로 밝혀졌다. Transport of a number of amino acids in osmotically shocked cells of Escherichia coli after growth in enriched media has been found to be enhanced by nucleosides, particularly cytidine and uridine which are more effective than purine ribonucleosides. Deoxyribonucleosides have activity approximately equivalent to that of the corresponding ribonucleosides. The stimulatory activity is involved primarily, in increasing the expandable pool of amino acid within the cell and is not significantly affected by inhibitors of protein or RNA synthesis. The stimulatory effect is specific to nucleosides because neither ribose, deoxyribose and free bases nor pseudouridine had any effect.

Evidence for Alkaline Phosphatase Involvement in tRNA-Stimulated Amino Acid Transport Systems

세포막 단백질 중의 하나인 periplasmic 단백질이 적절한 tRNA의 존재하에 아미노산 수송기작에 관여함이 알려졌는데, 이 단백질이 alkaline phosphatase로 밝혀졌다. 이 단백질의 분자량은 gel여과 방법으로는 88,000, SDS전기 영동으로는 43,500으로 나타났는데 두 개의 동일한 단일체로 존재함을 나타내었다. 아미노산 분석과 효소 역할에 의거하여 alkaline phosphatase임이 알려졌고, 적절한 tRNA와 더불어 아미노산 수송을 크게 증가 시킴을 볼 수 있었다. Purified alkaline phosphatase or tRNA alone did not exert any significant effect on leucine, isoleucine or valine transport, whereas simultaneous addition of these two molecules increased the amino acid transports drastically.

The apparent molecular weight of the purified protein was estimated to be 88,000 by gel filtration and 43,500 by SDS-polyacrylamide gel electrophoresis, indicating this protein is a dieter composed of two identical monomers. This paper describes the design of the ossicular replacement prosthesis for the people who have problem of hearing because of middle ear disease. Also the simulation results of the designed product for verification by finite element analysis software (ABAQUS) are presented. New model is applied to middle ear FE analysis which was generated in previous study. The full replacements of ossicular prosthesis for ossicles (malleus, incus and stapes) are made of Hydroxyapatite and Titanium. Although other existing prosthesis models consider only sound amplification effect, current type has damper system which is operating on the audible frequencies. High frequency sound transmitted to inner ear can be reduced and the prosthesis and inner ear can be prevented from damage.

A gastrointestinal metastasis from lung cancer is rare and is quite often found during an autopsy. A 58-year-old man was admitted with left upper back pain and a mass in the left upper lobe on the chest radiograph. The transbronchial lung biopsy revealed an adenocarcinoma of the lung. A gastroduodenoscopy was carried out to evaluate the indigestion and epigastric discomfort. The duodenofibroscopy revealed bulging mucosa on the minor papilla of the duodenum. A metastasis to the duodenum from an adenocarcinoma of the lung was diagnosed by a duodenofibroscopic biopsy of the bulging lesion. We report this rare case of an adenocarcinoma of lung with a metastasis to the duodenum. (Tuberc Respir Dis 2007; 63: 283-288)

성과감사의 개념화

우리나라는 책임추궁 방식의 합법성감사의 반발과 부작용 극복 차원에서 합법성감사의 토대 위에 성과감사를 도입함으로써 성과감사와 합법성감사의 개념적 혼란 문제가 지속적으로 제기되어 왔다. 이는 성과감사를 체계적으로 발전시키고 그 실효성을 확보하는 데 큰 장애요인으로 작용한다. 이러한 문제인식하에 본 논문은 성과감사의 이론적 기반과 제도적 적용을 분석하여 현대 감사환경에 적합한 성과감사의 개념을 정립하는 데 그 목적이 있다. 이를 위해 Wittgenstein(1968)의 ‘언어게임(language game)’ 이론에 기초하여 성과감사 도입의 환경적 요인에 대한 검토, 그리고 성과감사라는 용어의 구성요소인 ‘성과’와 ‘감사’의 사전적 정의와 그 이론적 의미에 대한 고찰을 기반으로, INTOSAI와 미국과 영국 등 해외 주요국 최고감사기구를 대상으로 감사종류 및 성과감사의 개념, 기준 및 범위, 접근방법 등 제도적 적용실태를 분석함으로써 제도운영의 공통적인 규칙을 탐색하였다. 이를 통해 현대 감사환경에 적합한 성과감사의 개념적 요건으로 ① “목표달성과 관련된 핵심사항을 대상으로 하는 감사”, ② “간결하고 명확한 메시지를 전달하는 감사”, ③ “판단기준에 따른 감사결과를 균형있게 제시하는 감사”, ④ “미시적·개별적 조치보다는 거시적·구조적 개선을 추구하는 감사”를 제시하는 한편, 그 개념적 기초에 기반하여 우리나라 감사원의 향후 성과감사 발전방향을 제안하였다. In Korea, the problem of conceptual confusion between performance audit and compliance audit has been continuously raised by introducing performance audit on the basis of compliance audit in order to overcome negative side effects of compliance audit. This is a major obstacle to the development of performance audit. The purpose of this paper is to analyze the theoretical basis and institutional application of performance audit, and to conceptualize of performance audit to make it suitable for recent audit environment. It applied ‘language game’ theory (Wittgenstein, 1968) to conceptualization methodology, and reviewed the environmental factors of performance audit introduction, the dictionary definition and the theoretical meaning of 'performance' and 'audit' which are components of performance audit. Based on this reviews, it searched the common rules in reality by analyzing the institutional application of INTOSAI and the supreme audit institutions of major countries such as USA and UK. According to the analysis, the conceptual requirements suitable for recent audit environment include: 1) an audit that focuses on key issues in terms of achieving the goal; 2) an audit that conveys a concise and clear message; 3) an audit that presents audit results in a balanced manner based on audit criteria; and 4) an audit that pursues structural improvement rather than individual correction. Based on the conceptual basis, it proposes the direction of future performance audit.
