        마우스 소뇌과립층의 apoptosis를 지표로 한 진단용 초음파의 안전성 검증

        We have studied, by a nonisotopic in situ end-labeling(ISEL) technique, frequency of apoptosis in the external granular layer(EGL) of the cerebellum of immature mice by ${\gamma}$-rays irradiation from $^{60}Co$ or diagnostic ultrasound exposure. The total number of normal cells and cells showing morphological features of apoptosis were counted. The frequency of apoptotic cells was expressed as a percentage of the total number of cells in EGL. The extent of changes following 200 cGy(1090 cGy/min) was studied at 2, 4, 6, 8, 12, or 24 hours after exposure. The maximal frequency was found 6~8 hours after exposure. The immature mice that received 18, 36, 54, 108, 198, 396 cGy of ${\gamma}$-rays or diagnostic ultrasound(7.5MHz, 4.2mW, $I_{SPTA}=7.9mW/cm^2$, $I_{SPTA}=114.3W/cm^2$) for 10 or 30 minutes were examined 6 hours after irradiation. Measurements performed after ${\gamma}$-ray irradiation showed a dose-related increase in apoptotic cells in each of the mice studied. The dose-response curves were analyzed by a linear-quadratic model ; frequency of apoptotic cell in the EGL was y = $(0.1349{\pm}0.01175)D$+$(-0.0001522{\pm}0.0000334)D^2$+0.048($r^2$ = 0.981, D = dose in cGy). In the experiment of ultrasound exposure, the frequency of apoptotic cell was $0.106{\pm}0.130$(10 minutes exposure) and $0.167{\pm}0.220$(30 minutes exposure). We estimated the relative dose of the yield from the experiment with ultrasound by substituting the yield from ultrasound exposure into the curve from the ${\gamma}$-irradiation. The relative dose of ultrasound exposure compared with ${\gamma}$-irradiation were 0.432 cGy(10 minutes exposure) and 0.885 cGy(30 minutes exposure). We have found that there is no evidence to indicate that diagnostic ultrasound involves a significant risk.

        We have studied, by a nonisotopic in situ DNA end-labeling (ISEL) technique, frequency of apoptosis in the external granular layer (EGL) of the cerebellum after whole-body irradiation of newborn mice and intestinal crypt cell of adult mice by gamma-rays from $^{60}Co$. The extent of changes following 2 Gy(10.9 Gy/min) was studied at 2, 4, 6, 8, 12, or 24h after exposure. The maximal frequency was found 4-8h after exposure. The mice that received 0.18, 0.36, 0.54, 1.08, 1.98, or 3.96 Gy were examined 6h after irradiation. Measurements performed after irradiation showed a dose-related increase in apoptotic cells in each of the mice studied. The dose-response curves were analyzed by a linear-quadratic model; frequency(%) of apoptotic cell in the newborn mice cerebellum was ($13.49{\pm}1.175$)D+$(-1.52{\pm}0.334)D^2$+0.048($r^2=0.981$, D = dose in Gy) and frequency(number per crypt) of apoptotic cell in the intestinal crypt of adult mice was ($3.857{\pm}0.420$)D+$(-0.535{\pm}0.120)D^2$+0.155($r^2=0.952$, D = dose in Gy). It provides the basis required for a better understanding of results which will be obtained in any further studies for biological responses of radiation using newborn and adult mice.

        단세포 전기영동법을 이용한 인체, 마우스 및 랫드 림프구의 방사선에 의해 유발된 DNA 손상 측정

        The alkaline single-cell gel electrophoresis (SCGE) assay, called the comet assay, has been applied to detect DNA damage induced by a number of chemicals and biological factors in vivo and in vitro. The DNA damage was analysed by tail moment (TM) and tail length (TL), which were markers of DNA strand breaks in SCGE. Human, mouse and rat peripheral blood lymphocytes (PBLs) were irradiated with different doses of $^{60}Co$ ${\gamma}$-rays, e.g. 1, 2, 4, and 8 Gy at a dose rate of 1 Gy/min. A dose-dependent increase in TM (p<0.01) and TL (p<0.01) was obtained at all the radiation doses (1-8 Gy) in human, mouse and rat PBLs. Mouse PBLs were more sensitive than human PBLs which were in turn more sensitive than rat PBLs when the treated dosages were 1 and 2 Gy. However, human PBLs were more sensitive than mouse PBLs which were in turn more sensitive than rat PBLs when the irradiation dosages were 4 and 8 Gy. Data from all three species could be fitted to a linear-quadratic model. These results indicated that there may be inherent differences in the radio-sensitivity among PBLs of mammalian species.

The extent of changes following 200 cGy (1090 cGy/min) was studied at 2, 4, 6, 8, 12, or 24 hours after exposure. The maximal frequency was found 4~6 hours after exposure. The mice that received 18, 36, 54, 108, 198, 396 cGy of y-rays or diagnostic ultrasound (7.5 MHz, 4.2 mW, ISPTA =7.9 mW/㎠, ISPPA= 114.3 W/㎠) for 10 or 30 minutes were examined at 6 hours after irradiation. Measurements performed after y-ray irradiation showed a dose-related increase in proportion of apoptotic cells in each of the mice studied. The dose-response curves were analyzed by a linear-quadratic model; frequency (number per crypt) of apoptotic cells in the intestinal crypt of adult mice was (0.0386±0.004204)D+(-0.0000535±0.00001120)D²+0.15475 (r²=0.952, D=dose in cGy). In the experiment of ultrasound exposure, the frequencies of apopototic cells were 0.181±0.055 (10 minutes exposure) and 0.225±0.137(30 minutes exposure). We estimated the relative dose of the yield from the experiment with ultrasound by substituting the yield from ultrasound exposure into the curve from the y-irradiation. The relative doses of ultrasound exposure compared with y-irradiation were 0.692 cGy (IO minutes exposure) and 1.334 cGy (30 minutes exposure).

        앞으로 다가올 유비쿼터스 컴퓨팅 환경에서는 일상생활 어디에서든지 컴퓨팅이 가능해질 것으로 예상되며, 원격지의 상대방과의 상호작용이 다양한 형태로 발전할 것으로 예상된다. 이를 지원하기 위하여 유비쿼터스 환경에서의 다자간 인터랙티브 협업 시스템들이 연구되고 있다. 그런데, 현재 개발된 다자간 협업 환경 시스템에서는 자료를 공유하는데 있어서, 데이터에 대한 인덱싱과 검색에 비중을 두지 않고 있다. 공유 데이터는 늘어나는 반면, 그것을 효과적으로 검색할 수 있는 방법이 마련되지 않아 데이터를 공유하고 활용하는 효율성이 떨어질 것이 예상됨에 따라서 키워드 검색 기반으로 데이터를 검색할 수 있는 방법을 마련하고자 한다.

        방사선 피폭 마우스에서 보중익기탕 및 구성단미의 효과

        We performed this study to determine the effect of Bu-Zhong-Yi-Qi-Tang, as a prescription of traditional Oriental medicine, and its major ingredients on jejunal crypt survival, endogenous spleen colony formation, apopotosis in jejunal crypt cells, lethality and hematological change of mice irradiated with high and low dose of Y-radiation. Bu-Zhong-Yi-Qi-Tang administration before irradiation protected the jejunal crypts (p<0.0001), increased the formation of endogenous spleen colony (p<0.05) and reduced the frequency of radiation-induced apoptosis (p<0.05). The survival rate and mean survival time of the groups treated with Bu-Zhong-Yi-Qi-Tang within 30 days after the treatment were far better than the irradiation control group. In the experiment on the effect of ingredients of Bu-Zhong-Yi-Qi-Tang, the result indicated that the extract of Rensan (Panax ginseng), Danggui (Angelica sinensis), Shengma (Cimicifuga heracleifolia) and Chaihu (Bupleurum falcatnosa) might have a major radioprotective effect. Although the mechanisms of this inhibitory effect remain to be elucidated, these results indicated that BU-Zhong-Yi-Qi-Tang might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the protective nature of Bu-Zhong-Yi-Qi-Tang extract and its ingredients.

        어류양식용 사료의 위생화. II. 어류양식용 사료의 조성분과 지방산화에 대한 방사선 조사 및 butylated hydroxyanisole의 효과

        The chemical composition of fish feeds(mackerel, gizzard shad, shrimp and formula feed) were analyzed and compared with those of untreated and irradiated (5 kGy) feeds, and the effects of gamma irradiation (5 kGy) and butylated hydroxyanisole (BHA, 200ppm) on feeds spoilage were investigated by determination of thiobarbituric acid (TBA) value and peroxide value for 14 days at $20^{\circ}C$, $4^{\circ}C$ or $-20^{\circ}C$. Crude protein, crude lipid, crude fiber, crude ash and nitrogen free extract were not significantly changed by gamma irradiation with the dose of 5 kGy. Immediately gamma irradiation increased the initiative oxidation of feeds as expected. But BHA was found to be greatly effective in minimizing the radiation-induced peroxidation (p<0.05). The TBA value and peroxide value increased with the elapse of the storage period. The level of lipid oxidation was increased depending on the rise in storage temperature. But the additions of BHA inhibited the lipid oxidation (p<0.05).

        고선량 및 저선량 방사선 조사 마우스에서 누에동충하초(Paecilomyces japonica)의 효과

        Cordyceps has a reputation for its broad biological activities and as a tonic which replenishs vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on radiationinduced damages were investigated. We performed this study to determine the effect of Dongchongxiacao (Paecilomyces japonica) on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells and hair follicles of mice irradiated with high and low dose of gamma-radiation. Treatment with Dongchongxiacao showed no significant modifying effects on the jejunal crypt survival and endogenous spleen colony formation. The frequency of radiationinduced apoptosis was reduced by pretreatment of Dongchongxiacao (i.p.: 50 mg/kg of body weight, at 12 and 36 hours before irradiation, p<0.01). The spontaneous levels of apoptotic cells are $0.082{\pm}0.041$ in intestinal crypts and $0.231{\pm}0.084$ per hair follicle section of skin. Pretreatment of Dongchongxiacao was associated with decreases of 26.86% in intestinal crypt and 66.36% in hair follicle decrease in the number of cells with nuclei positively stained for apoptosis compared with the irradiation control group. We demonstrated for the first time that Dongchongxiacao administration could reduce the extent of apoptosis produced by radiation in the hair follicle. The results presented herein that Dongchongxiacao given before irradiation is capable of reducing the severity of cell loss as a result of apoptosis.

        대표적 근류 생약의 방사선 방호효과

        We performed this study to determine the effect of Jiegeng(Platycodon grandiflorum), Danggui(Angelica sinensis), Gancao(Glycyrrhiza glabla), Chaihu(Bupleurum falcatnosa), Shoudehuang(Rehmannia glutinosa), Huangqi(Satragalus membranaceus), Muxiang(Saussurea lappa), Yuanzhi(Polygala tenuifolia), Rensen(Panax ginseng) and Baishaoyao(Paeonia lactiflolia), as Oriental radices herbs, on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of ${\gamma}-radiation$. Jiegeng(p<0.005), Danggui(p<0.0005), Gancao(p<0.005), Chaihu(p<0.05), Muxiang(p<0.05), Rensen(p<0.005) and Baishaoyao(p<0.005) were effective in intestinal crypt survival. Danggui(p<0.05), Chaihu(p<0.05), Shoudehuang(p<0.05), Huangqi(p<0.05), Rensan(p<0.005) and Baishaoyao(p<0.05) increased the formation of endogenous spleen colony. The frequency of radiation induced apoptosis was also reduced by pretreatment with Chaihu(p<0.05), Muxiang(p<0.005), Yuanzhi(p<0.05), Rensan(p<0.05) and Baishaoyao(p<0.05). Although the mechanisms of this effect remain to be elucidated, these results indicated that Danggui, Chaihu, Muxiang, Rensan and Baishaiyao might be a useful radioprotector, especially since it is a relatively nontoxic natural product.

        태아시기의 방사선 노출에 의한 마우스 뇌 발달에 미치는 영향에 관한 형태학적 연구

        The present study analyze the morphological aspects of the cerebrum of mice with prenatal exposure to high and low dose (0.5, 1, 2 Gy) of $\gamma$-radiation on gestation day 12 or 16. The animal were allowed to give birth and the offspring were sacrificed at postnatal days 28 for gross and microscopic examination of cerebrum. Their body weight, brain weight, brain length, brain width, cortical thickness and area of cingulum bundle were examined. The histological and planimetric analysis were performed observing coronal sections. The gross malfomation (microcephaly) and abnormality of cortical architecture were prominent after exposure to 2 Gy on day 12 of gestation. significant dose-related reductions in body weight, brain weight, brain size were found in all irradiated groups. A significant change was found in thickness of the cerebral cortex and area of the cingulum bundle in the groups exposed to 0.5 Gy or more. There was no difference a lamina patter of six layers in cerebral cortex between the control and irradiated groups, but cell packing density increased significantly in the group exposed to 1 Gy or more. These results suggested that dose as low as 0.5 Gy could cause a morphologically reduce change in developing mouse cerebrum and exposure on day 12 of gestation to $\gamma$-irradiation is a particularly sensitive phase in causing malformation and abnormality of central nerve system.
