제주시 (濟州市) 일원 4개호 해수의 분변성 오염세균의 분포

The present study was performed from August November 1978 to investigate the degree of the bacterial pollution at 4 sites sea water around Jeju city: Dongbudu, Seobudu, Yongyeon and Yongduam. Total coliforms (TC), fecal coliforms (FC), and Total Bacteria (TB) were examined together with the measurement of pH and water temperature. pH values ranged from 7.1 to 8.4 at all sites. The TC levels at Yongduam were 130∼350/100 ml which showed the lowest value among all sites, 1403∼3,500/100ml at Seobudu, 1,600-over 24,000/100ml at Yongyeon and over 2.4x 10^6/100ml at Dongbudu which showed the highest value. The FC levels at Yongduam were 49∼240/100ml, 70∼1,400/100mI at Seobudu, 280∼2,400/100ml at Yongyeon and the highest values were 3.5X10³∼1.4X10^6/100ml at Dongbudu also. FC:TC ratios were 21∼68% at Yongduam, 10∼34% at Yongyeon, I4∼58% at Dongbudu and 22∼58% at Seobudu. According to the criteria in America, the water quality of Yongduam is comparatively good sea water in both considerations of the TC and FC levels. The sea water of Yongyeon has over those values, so it is considered that posibility of the occurrence of Salmonella is much. The FC levels at Seobudu has also the posibility of containing Salmonella. The degree of the bacterial pollution at Dongbudu is very high. This means that there are severe inflow of fecal sewege from Sanji-brook and other sewers to Dongbudu sea water.

The Physical, chemical and bacteriological investigations on the three sites of each two streams, Yeonhee-chun and Donghong-chun which are the sources of two waterfalls, Chonjiyon-Fall and Chongbang-Fall respectively in Seogwipo City, Korea, were carried out monthly from October 1990 to Septermber 1991. The ranges of DO, BOD, SS and SiO_2-Si concentration of Yeonhee-chun were 6.61∼12.82mg/l, 0.1∼4.5mg/l, 0.10∼17.10mg/l and 5.11∼16.22 mg/l respectively, and those of Donghong-chun were 5.30∼11.92mg/l, 0.4∼17.2mg/l, 0.51∼48.67mg/l and 5.16∼14.80mg/l respectively. The result of bacterial investigations for the pollution indicator bacteria such as total coliforms, fecal coliforms, fecal streptococci and others showed that the bacterial quality of Yeonhee-chun was somewhat better than that of Donghong-chun. The MPNs of total coliforms of Yeonhee-chun and Donghong-chun were 750-24×10^4/100 ml and 1100-24×10^5/100 ml respectively, and those of fecal coliforms were 23-24x10^3/100 ml and 9-24×10^5/100 ml respectively. The numbers of fecal streptococci of Yeonhee-chun and Donghong-chun were 2-1,324/10 ml and 3-13,233/10 ml respectively.

제주도 양식장 배출수 퇴적층에서 단백질 분해 효소를 생산하는 세균을 분리하였으며, 각각 SK-2, 및 SK-125라고 명명하였다. 분리균주 SK-2의 16S rDNA의 염기서열 분석 결과 Bacillus licheniformis와 Bacillus subtillis의 염기서열과 99%의 상동성을 보였으며, BIOLOG를 이용한 생화학적 분석에서도 Bacillus family와 유사한 특성을 보여 최종적으로 분리균주 SK-2를 Bacillus sp. SK-2으로 명명하였다. 또한 분리균주 SK-125의 16S rDNA의 염기서열 분석 결과 Pseudoalteromonas haloplanktis의 염기서열과 99%의 상동성을 보였으며, BIOLOG를 이용한 생화학적 분석에서도 Pseudoalteromonas family와 유사한 특성을 보여 최종적으로 분리균주 SK-125를 Pseudoalteromonas sp. SK-125라고 명명하였다. Bacillus sp. SK-2와 Pseudoalteromonas sp. SK-125균주의 온도별 실험에서는 40℃에서 가장 높은 생육도를 보였으며, 효소활성은 Bacillus sp. SK-2와 Pseudoalteromonas sp. SK-125 두 균주 모두 30℃에서 가장 높은 활성을 보였다. In this study, protease-producing bacteria were isolated from the marine sedimentary layer in coastal Jeju. We isolated 2 protease producing strains (SK-2 and SK-125) and tested their protesase producing activities. Gram staining and BIOLOG of isolated strains revealed that strains SK-2 and SK-125 belong to Bacillus and Pseudoalteromonas families, respectively. The 16S rDNA nucleotide sequences analyses of the isolated strains showed 99% sequence homology with those of Bacillus sp. and Pseudoalteromonas sp.; therefore, the isolated strains SK-2 and SK-125 were named Bacillus sp. SK-2 and Pseudoalteromonas sp. SK-125, respectively. The optimum conditions for the cell growth of protease activities were obtained when the both isolates were cultured at 30℃, 96 hrs and pH 7~8.

Physical and chemical analyses of water discharged from 4 crowded farms (Sungsan, Pyosun, Wimi and Daejung) in Jeju island were performed from July, 2006 to Dec, 2006, and the result of the analyses showed that hydrogen ion concentrations (pH) for water discharged from Sungsan farm was 7.74, Pyosun was 7.68, Wimi was 7.68 and Daejung was 7.7. Salinity levels for Sungsan, Pyosun and Wimi had an average of 31~33 ‰, indicating characteristics of far distance areas, whereas that of Daejung was 28.81%, which was far lower compared to regular sea water salinity. As the result of measuring dissolved oxygen (DO) for each area, each area showed first graded DO for each discharged water based on water quality level for each sea district. The result of measuring the temperature for discharged water showed that water temperatures for summer were 23~25℃, and those for winter were 16~18℃. Nitrogen concentrations for discharged water exceeded each sea area's water quality level in all farms. In the case of phosphate, its average value was 0.48 ㎎/ℓ for Sungsan, 0.55 ㎎/ℓ for Pyosun, 0.66 ㎎/ℓ for Wimi, and 0.44 mg/l for Daejung, and chemical oxygen demand (COD) was shown to be 1.5 ㎎/ℓ, 1.8 ㎎/ℓ, 1.6 ㎎/ℓ and 2.3 ㎎/ℓ for Sungsan, Pyosun, Wimi and Daejung respectively. For suspended solids (SS), the average concentration was 19.3 ㎎/ℓ, 21.2 ㎎/ℓ, 21.3 ㎎/ℓ and 18.5 ㎎/ℓ for Sungsan, Pyosun, Wimi and Daejung respectively. The results of physical and chemical analyses for discharged water in farms based on time showed that almost all items were shown to increase in the forenoon and decrease, overall, in the afternoon.
제주지역 양식넙치의 연쇄구균증 발생동향 및 원인균에 대한 분자적 동정

제주지역 양식넙치의 주요 세균성 질병의 일종인 연쇄구균증 발병동향과 특성을 분석하였다. 제주지역 양식넙치의 연쇄구균증은 연중 발생하는 양상을 보였으나 상대적으로 고수온기에 발생율이 높았으며, 30cm 이상의 개체에서 주로 관찰되었다. 2003년 6월부터 2005년 5월 사이에 분리된 연쇄구균 균주를 multiplex PCR assay 기법을 이용하여 종 조성을 분리한 결과 Streptococcus iniae와 S. parauberis로 동정되었으며 그 비율은 각각 46%와 54%이었다. S. iniae는 상대적으로 9월과 10월에 높은 분리율을 나타내었으나 S. parauberissms 3월과 4월에 분리율이 높게 나타났다. S. iniae와 S. parauberis 감염개체의 병리학적 중상은 일부분 유사하였으나 S. iniae 분리개체는 복부팽만, 탈장, 복수저류증상이 주로 관찰되는 반면, S. parauberis 분리개체는 체색흑화, 무안측 발적 중상이 두드러지게 관찰되었다. 이 두 종의 병원체 모두 ampicillin과 amoxicillin에 감수성이 높았으며, S. iniae 균주들은 S. parauberis 균주들보다 상대적으로 doxycycline, erythromycin, oxytetracycline에 감수성이 높았다. Streptococcosis of olive flounder(Paralichthys olivaceus) is an important bacterial disease in Jeju island. In this study, we investigated monthly infection pattern of this disease in different size of the flounder fish. Even though the disease occurred throughout the year, the infection ratio was relatively higher in the months with warm water season. The infection was more prevalent in adult flounder over 30 cm total length compare to these of small size fish. Two infectious species of streptococcosis pathogens were detected by multiplex PCR assay. Detection ratios of Streptococcus iniae and S. parauberis reached up to 46% and 54%, respectively, from June 2003 to May 2005 in Jeju island. S. iniae occurred intensively from September to October, whereas S. parauberis reported from March to May. S. iniae and S. parauberis infections of cultured flounder share some common features, but clinical findings showed considerable differences between two diseases. Distended abdomen, protruded anus and ascitic fluid in the peritoneal cavity are evident lesions detected in S. iniae infection, whereas, flounders infected by S. parauberis showed prominent lesions such as darkened surface and haemorrhaging in the non-ocular side. Both streptococcosis pathogens were sensitive to antibiotics, such as ampicillin and amoxicillin. However, S. iniae strains were more sensitive to doxycycline, erythromycin and oxytetracycline than S. parauberis strains.
