Suckling piglets and weaned pigs showed anorexia, dehydration, severe abdominal breathing, emaciation and paresis from Oct. 1993. to Nov. 1993. Five 2-week-old piglets were submitted for diagnosis in Kangwon National University. At necropsy, the pin-point well demarcated yellowish white foci were scattered on the surface of the lung, heart, liver, spleen and kidney. Histologically, multifocal areas of necrosis with mononuclear cells infiltration were found in the lung, heart, liver, lymph node, spleen, kidney and small intestine. These lesions tended to be associated with blood vessels. Variable round to ovoid tachyzoites were located at the periphery of the lesions. The organisms were demonstrated as Toxoplasma gondii by immunohistochemical staining method. Ultrastructurally, this parasite was surrounded with parasitophorous vacuole in alveolar macrophage. The parasite was crescent-shaped and $6{\sim}8{\times}1{\sim}2{\mu}m$ in size. It was enclosed by an thick outer membrane and an underlying thin inner membrane. Several club-shaped paired organelles and conoids lay in the cytoplasm at the anterior. Numerous round body and one to several mitochondria were presented in the cytoplasm. Based on the gross findings, histopathology, immunohistochemical and electron microscopic findings, this case was diagnosed as toxoplasmosis in piglets.

This study surveyed the prevalence of traumatic reticular diseases (TRD) of slaughter cattle in Korea, the typology of the causative foreign bodies and the effects on beef quality and carcass weight. The overall prevalence of TRD in 3,121 slaughter cattle was 5.5%. However, the prevalence was significantly higher in Korea indigenous cattle Hanwoo (5.8%, p < 0.05) and female cattle (20.75%, p < 0.001). The prevalence significantly increased in aged cattle (p < 0.001). Major lesions related to foreign bodies were reticulitis (96%) and peritonitis (86%). Most causative foreign bodies were made of iron including nails, wires, steel rods, screw nails, and syringe needles. Cattle affected with TRD produced significantly lower grade quality of beef compared to normal cattle (p < 0.0001), but TRD did not affect carcass weight. The data will be useful in the management of TRD, with the aim of increasing beef productivity in Korea.

Embryonic stem cells are derived from inner cell mass of blastcyst. They are pluripotent in differentiation and show self-renewal ability. Therefore, the cells can differentiate into cell types which can be derived from all the three germ layers. In this report, we screened 100 compounds for their potential effects on the differentiation of KH2 mouse embryonic stem cells to cardiomyocytes. KH2 cells are developed in our lab and express enhanced green fluorescence protein under the direction of α-myosin heavy chain gene promoter. This cell line shows no fluorescence in undifferentiated condition. However, cardiomyocytes derived from the cells show green fluorescence. Test compound was added during the differentiation period of the cells. After 15 days of differentiation, the cells were trypsinized and measured by flow cytometry for their fluorescence protein expression. All tested compound did not show any significant increase in the rate of fluorescent cells compared with that of their control group. However, we could confirm that our cell screening system is very stable.

Mouse embryonic stem cells are pluripotent stem cells that can be differentiate into all the cell types derived from three germ layers in vitro. We aimed to confirm the neuronal cell types derived from the embryonic stem cells by two different differentiation protocols, which would guide us which protocol is useful for in vitro neuronal toxicity test. The mouse embryonic stem cells derived from 129 mouse strain, TC-1 cells, were differentiated according to 30-day differentiation protocol or 15-day differentiation protocol. At the end of the differentiation period, neuronal cells (neuron, astrocyte and oligodendrocyte) were identified by immunocytochemistry using marker antibodies. According to the results, the numbers of astrocytes and oligodendrocytes were much higher than that of neurons in 30-day differntiation protocol. However, oligodendrocytes were overwhelming compared to astrocytes and neurons in the 15-day differntiation protocol. These results indicated that the neuronal cell types and the cell numbers derived from the embryonic stem cells should be considered when selecting in vitro neuronal cell toxicity models using embryonic stem.

There were eight Korean indigenous cattles affected with bovine tuberculosis(BTB) detected by inspectors at slaughterhouses located in Chungbuk province from May 2006 through July 2007. Postmortem finding of BTB cases was characterized by the presence of several caseous or calcified nodules encapsulated by connective tissue from the pleural/peritoneal surface, livers, lungs and regional lymph nodes. On microscopic examinations, the characteristic lesion of BTB was the formation granulomatous nodules, which contains central calcified necrotic zone surrounded by epithelioid cells, macrophages and a few Langhans` type giant cells. In addition, mononuclear cells and fibroblasts were also infiltrated. At the periphery, encapsulation was formed that protect the neighboring healthy tissues.

This study was conducted to assess the chemopreventive effects of chitosan in a rat colon carcinogenesis induced by azoxymathane (AOM). Ninety, 5-week-old, male F344 rats were divided into three groups. THe animals in group 1 received subcutaneous injections of 15㎎/㎏ AOM three times for two weeks, then were placed on powdered basal diet containing 2% chitosan for 37 weeks from weeks 3 to 40. The animals in group 2 were given AOM alone. The animals in group 3 were given 2% chitosan without prior carcinogen treatment. All animals were sacrificed at week 12 for quantitative analysis of aberrant crypt foci (ACF) and at week 40 for analysis of tumor induction. Total numbers of ACF and AC per colon of group 1 were not significantly different from those of group 2. Tumor incidences and multiplicities of small intestine in the group 1 were significantly decreased compared with those of the group 2 (P<0.05). According to pathological diagnoses, adenocarcinoma incidence and multiplicity in the small and large intestine in the group 1 were significantly decreased compared with those of the group 2 (p<0.05). No toxic effects were observed in animals given chitosan in terms of body weights, and liver or kidney histology. These results indicate that chitosan may have a potential as chemopreventive agents of colon carcinogenesis during the postinitiation stage.
