N/A Oxidants from cigarette smoke or those produced by phagocytes are implicated in the pathogenesis of emphysema, We reasoned that augmentation of antioxidant enzymes in cigarette smokers may be important in restricting direct and indirect oxidant damage to alveolar structures. Accordingly, we studied catalase, superoxide dismutase (SOD) and glutathione in alveolar macrophages (AM) from smoke-exposed hamsters and nonsmoking control hamsters. Hamsters were exposed to six cigarettes/day, 6 days/week, for 8 weeks. The results were as follows; 1) AM from smoking hamsters compared to controls showed significant increase in SOD (p<0,01) and catalase (p<0,01), 2) AM from smoking hamsters compared to controls showed no significant change in glutathione(p>0.05) and filtration of smoke does not affect glutathione activity. We conclude that chronic smoke exposure induces augmentation of selective antioxidant enzymes in the lung macrophages which may limit oxidant-mediated damage to alveolar structures, We suggest that this induction of antioxidant enzymes may be a mechanism to escape unchecked oxidant injury to the lower respiratory tract of cigarette smokers.
