화랑곡나방의 발생에 따른 Esterase Isozymes의 Pattern변화

Changes in activity and classification of esterase isozymes during the tire cycle or Plodia inteipunctella (Hiibner) were investigated by the native polyacrylamide gel electrophoresis. The stage specificity in esterase activity and isozyme pattern was observed throughout the larvalpupal-adult transformation. The activity esterase was highest at the 3-day old adult stage, and the lowest level at the prepupal stage. A total of 12 esterase bands were identified throughout the development, and the bands showing high enzyme activity was observed in the middle part of gel. Twelve esterases on the basis of inhibition by the three types of inhibitors (organophosphates, eserine sulfate and sulfhydryl reagents) were classified into three class, namely, carboxylesterase (CE), arylesterase (ArE) and cholinesterase (ChE), and these classes contained 7, 3 and 2 isozymes, respectively.

The binding affinity of the differentially induced cytochrome P-450 (P-450) in rat liver microsomes with testosterone and 16α-hydroxytestosterone (16α-OH-T) was compared. Interaction of testosterone with all three microsomel P-450 of normal, MC- or PB-treated rats, gave type I spectral changes and their spectral binding constants (Ks) to P-450s were 35, 33, and 25 μM at 25℃, respectively. PB-inducible P-450s among three of these showed the highest affinity to testosterone, and the ratio of low to high spin conversion of P-450 by this compound showed also a similar tendency as the results of binding affinity. 16α-OH-T, one of testosterone metabolites, binding to all three P-450 exhibited a reversed type I spectral changes and their Ks values to normal, MC- and PB-inducible P-450 were 41, 28, and 23 μM, respectively, but binding of it with PB-inducible P-450 showed another Soret band near 460 nm. These results indicate that 16α-OH-T also binds to P-450s with a high affinity like testosterone, and it may be further metabolized to more polar compounds by P-450.

The purpose of this study is to focus on specified activity to realize strategy-physical-management model, extension of partnership , role assignment method through Area Management System appeared from large development project in Japan. As a result of analysis, key points are suggested to apply Area Management System suitable to our urban environment as followings: First, it is important to have everything organized at the beginning of project. Having participation system for local residents is important element to realize succession of development, and after process. Second, participation of variety groups of people is needed. The movement of participation expanding to citizens becomes motive to maintain, and develop charm and value of district. Third, fair assignment is essential between people to guarantee continuous activity in an organization. Forth, activity needs to be happened continuously with assignment, partnership between participated organizations. It is recommended that realization of Strategy-Physical-Management Model needs to be processed step by step with cooperation, fair assignment between Coordinator to unify efforts from common people, Cooperator to guarantee realization of development plan, and Manager deriving citizen participation to improve value and charm of area. Fifth, role of public is important as an equal partner with private. Public sector needs to support private sector to make them activated, and to draw participation from citizen. The problem of Area Management System in Korea is that public sector lead principles and organization is not sustained, neither volunteered. To overcome such matters, it is essential to build a partnership which is not in public sector lead, but in creativity of private sector so that it is possible for private sector to take part in.
