        Biomarkers could be critical and useful tools for assessing the biological effects of smoking and detecting differences between potentially reduced exposure product (PREP) and conventional cigarettes. Smoking-related biomarkers can be classified into three categories as biomarkers of exposure, biomarkers of effects, and biomarkers of potential harm. When compared with the biomarkers of effects or harm, the biomarkers of exposure for chemical constituents of cigarette smoke are well established and characterized. In addition, they could offer the important information in understanding how cigarette smoke interacts with biological molecules and causes the disease to human. Therefore, we provide an overview of 6 biomarkers of exposure (Nicotine and nicotine metabolites, Carboxyhaemoglobin, NNAL (4-(methylnitrosoamino)-1-(3-pyridyl)-1-butanol) and NNAL - glucuronide, 3-Hydroxypropyl-mercapturic acid, and Monohydroxy-butenyl-mercapturic acids, and Urine mutagenicity) which were validated through extensive research and clinical experience. These reliable biomarkers could help identify the efficacy of PREP by predicting early toxicological effects and lead to improve it.

      • 옥수수 수염의 물 추출물이 혈압에 미치는 영향

        Zea mays Linne, which belongs to Graminae family, has been ordinarily used as edibles and fodder. Its stigma, Maydis Stigma, has been discarded without any special application with the exception of being used as diuretic in the sphere of Chinese medicine. Moreover, the pharmacological research toward Maydis Stigma as a medicinal herb has not been regretfully undertaken at all. The authors hereby paid attention to this point of view and made experiment to find out whether the water extract of Maydis Stigma might possess some pharmacological effect on the blood pressure of the rats or not. $LD_{50}$ to the mice was determined and the experiment was performed making the use of several agents related to the blood pressure such as atropine, Avil, dichloroisoproterenol and hexamethonium. The blood pressure of the rats and rabbits was recorded with the equipment of polygraph (Grass model 7 DAB) and its accessory, Statham pressure transducer. The results of the experiment are as follows. 1. $LD_{50}$ to the mice weighing 20g is determined 1,125 mg/kg. 2. The blood pressure manifests the gradual decrease by the rate of $-15.50{\pm}3.32,\;-22.14{\pm}8.73$ and $-46.25{\pm}5.17\;mmHg$ in proportion to ZW 25, 50 and 150mg/kg administered to the rats. 3. The blood pressure also manifests the same gradual decrease in proportion to ZW administered to the rabbits. 4. It is of significant difference to manifest the increase of $+5.14{\pm}4.65\;mmHg$ in the rats pretreated with atropine as compared with $-22.14{\pm}8.73\;mmHg$ of control group when administered each ZW 50mg/kg. 5. The administration of ZW 50 mg/kg to the rats pretreated with Avil, dichloroisoproterenol and hexamethonium does not manifest the significant difference in comparison with the control group.

      • 양채송화(洋菜松花)의 알코홀 추출물이 적출 조개 심장운동에 미치는 영향

        Lampranthus spectabilis is an ornamental plant which belongs to one of the Aizoaceae family as an one-year herb. There are no illustrations on the references that Lampranthus spectabilis has been made use for the purpose of the medicinal plant among the traditional herb doctors and the laymen. However, it has the specific characteristics to be analogous to the Cactaceae family in the botanical classification. Besides, mescaline, the main ingredient of Peyote cactus, demonstrates the sympathomimetic effect as well as the psychotomimetic effect. The authors called our attention to the resemblance to the botanical family between Aisoaceae and Cactaceae, and made a survey whether the alcohol extract of Lampranthus spectabilis might possess any pharmacological effect on the isolated heart of clam or not. The results of the experiment were as follows. 1. Administration of LX to the isolated heart of clam produced an inhibitory effect on the cardiac motility in proportion to its concentration 10<sup>-4</sup>, 5 X 10<sup>-4</sup> and 10<sup>-3</sup> g/ml respectively. 2. Administration of LX 5 X 10<sup>-4</sup> and 10<sup>-3</sup> g/ml produced the blockade effect on the cardiac motility pretreated with serotonin 10<sup>-7</sup> and 10<sup>-4</sup> g/ml, as well as with methysergide 10<sup>-6</sup> and 10<sup>-5</sup> g/ml. 3. Administration of LX 5 X 10<sup>-4</sup> g/ml produced no effect on the cardiac motility pretreated with acetylcholine 10<sup>-7</sup> g/ml. 4. Administration of LX 5 X 10<sup>-4</sup> g/ml produced the blockade effect on the cardiac motility pretreated with barium chloride 10<sup>-4</sup> g/ml, as well as with prostaglandin 20μg. Consequently, it is concluded that the alcohol extract of Lampranthus spectabilis has an effect of depression on the motility of the isolated heart of clam. Its mechanism of action might be due to a direct action on the heart. the heart.

        The objective of this study was to determine the effect of smoking conditions on the in vitro toxicological activity of mainstream smoke. The 2R4F reference cigarette was machine-smoked by International Organization for Standardization(ISO) and Canadian Intense(CI) conditions. Smoke was analysed for chemical composition and in vitro toxicity. The cytotoxic potencies of both the total particulate matter(TPM), which were collected in Cambridge filter pad, and gas/vapor phase(GVP), which was bubbled through in phosphate-buffer saline in a gas-washing bottle, were assessed neutral red up take assay with chinese hamster ovary(CHO) cells. The assessment for genotoxicity of TPMs generated under ISO and CI conditions was determined using Salmonella mutagenicity assay and in vitro micronucleus assay. When calculated on an equal TPM basis, in vitro toxicity of TPM obtained under CI condition was decreased compared to TPM generated under ISO condition. The results of chemical composition analyses revealed that the lower toxicological activity under CI condition than that of ISO condition could be explained by the decreased in the contents of phenols, N-nitrosoamines and aromatic amines of TPM on an equal TPM basis.

        The objective of this study was to investigate the effect of nitrogen compounds such as protein on the chemical composition and toxicity of cigarette mainstream smoke. BSA protein was treated into the tobacco leaf of original 2R4F cigarette at 1~4 % level. The studies were performed which included a bacterial mutagenicity assay and a mammalian cell cytotoxicity assay for total particulate matter(TPM), and glutathione(GSH) consumption assay for gas/vapor phase(GVP) and determination of smoke chemical constitute. Cigarettes treated with protein were observed dose-dependent increase in yield of volatiles, semi-volatiles and aromatic amines compared with control cigarette. However, carbonyl compounds such as acrolein was lower than that of control cigarette when calculated on an equal TPM basis. The cytotoxicity of TPM obtained from the protein-added cigarettes was not different from that of control cigarette. However, the mutagenicity of the TPM from protein-treated cigarettes(1~4 %) was up to 10-27 % higher than that of control. On the other hand, toxicity of GVP from protein-treated cigarette(4 %) was significantly decreased compared with control cigarette. An overall assessment of our data suggests that nitrogen compounds such as protein should be important for the chemical composition and biological activity of cigarette mainstream smoke.

        The aim of this study is to compare the sensitivity of both two NCI-H292 and A549 cell types to acute inflammatory responses induced by cigarette smoke. For this, we treated two kinds of smoke fractions derived from 2R4F reference cigarettes: total particulate matter(TPM) collected onto a Cambridge filter pad and gas/vapor phase(GVP) prepared by bubbling through in buffer solution. When we measured cellular cytotoxicity by neutral red uptake assay after treatment for 24 hours, TPM and GVP induced cytotoxic effect in a dose-dependent manner in the range of 10-$100{\mu}g$/mL and 60-$300 {\mu}g$/mL., respectively, in both cell types without any cellular difference. Additionally, when we examined acute inflammatory responses by analyzing cytokines secreted into culture media including tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-8(IL-8), and transforming growth factor-$\alpha$(TGF-$\alpha$) as well as matrix metalloproteinase-1(MMP-1), the treatment with smoke fractions increased those marker proteins in a dose-dependent manner in NCI-H292. Meanwhile, in A549 cells only MMP-1 was observed to be increased in a dose-dependent fashion. Collectively, our data indicate that NCI-H292 cell type is more sensitive to cigarette smoke-induced inflammatory response than A549 cells. This suggests that NCI-H292 could be useful as an in vitro evaluation tool to assess harmful effects of cigarette smoke.

        한국표준형 원전 증기발생기 Stay 용접부 자동검사시스템 및 현장 검증

        The stay cylinder weld at the steam generator of Korean Standard Nuclear Power Plants is safety class I component and is subjected to be inspected by the volumetric examination such as ultrasonic method. As accessibility of this area is limited due to the narrow space and high radiation, the existing manual inspection method involves various difficulties. Moreover operators may be exposed to internal contamination by contaminated dust during the surface buffing process to improve the inspection reliability of this area. Recently the new automatic inspection system for stay cylinder welds has been developed. The inspection system basically consists of a driving assembly, data acquisition device and signal processing units. The driving assembly is classified by 1) the scanner for inspecting and buffing the weld, 2) pillars for guiding the scanner and 3) the base frame for loading and supporting pillars. The scanner has 4 sensor modules to inspect in 4 refracted angles and 4 incident directions. These components can be inserted into the skirt of the stay cylinder through the manway hole and assembled easily by one-touch in the skirt. Data acquisition device and signal processing units developed in previous works are also newly upgraded for better processing of data analysis and evaluation. The system has been successfully demonstrated not only in the mock-up but also in the field. In this paper, newly developed inspection system for the stay cylinder weld of the steam generator is introduced and their field applications are discussed.
