        Analysis of coenzyme Q<sub>10</sub> in human plasma by high performance liquid chromatography

        코엔자임 큐텐은 비타민과 유사한 물질로 체내 중 미토콘드리아에 주로 분포 되어 있으며 항산화 작용을 나타낸다고 알려져 있다. 본 연구는 27-44세의 건강한 성인 남 녀 24명을 대상으로 하였고 혈장내 코엔자임 큐텐의 농도를 정량 평가하기 위하여 UV 검출기가 장착된 고성능액체 크로마토그라피를 사용 하였다. 혈장 내 코엔자임 큐텐을 분리하기 위해서 메탄올로 지방단백질을 제거하였고 노말헥산을 이용하여 액체-액체 추출법으로 코엔자임 큐텐을 용해 시켰다. 그 후 원심분리를 실시하여 노말헥산과 제단백된 물질을 층분리 시키고 상층액을 다른 유리 시험관에 옮겨 담았다. 옮겨진 노말헥산 층은 질소 가스를 이용하여 증발 농축을 시켰으며 남은 잔사에 순수한 에탄올로 재용해 시켜 검사 장비 내로 주입 하였다. 이 때 사용된 컬럼은 C18 역상 컬럼이었고 275 nm의 파장이 이용되었으며 메탄올과 에탄올을 85:15로 섞은 이동상을 1.7 mL/min의 유속으로 흘렸다. 본 검사 방법의 정량한계(limit of quantitation : LOQ, N/S=10)는 0.02 mg/L로 평가 되었고 0.1-2.0 mg/L의 농도 범위에서 검량선을 작성 하였다. 대상군 24명의 혈장 중 코엔자임 큐텐의 농도는 0.41-0.98 mg/L의 범위에서 분포 하고 있었으며 평균 농도는 $0.62{\pm}0.13mg/L$ 였다. 본 연구의 결과 본 검사 방법은 특수성을 가지고 있었으며 혈장 내 $CoQ_{10}$을 분석하는데 충분한 정량한계를 가지고 있었기에 임상에 적용이 가능하고 연구 기관에서 사용하기에 적합하다고 사료 된다.

        DMBA로 유도된 햄스터 협낭암종에서 p53 유전자 변이와 mdm-2 단백의 발현에 관한 연구

        Cellular proliferation is an intricately regulated process mediated by the coordinated interactions of critical growth control genes. Two of these factors in mammalian cells are the p53 and mdm-2 genes. A protein product of the mem-2 oncogene has been recently shown to associate with the protein encoded by the tumor suppressor gene p53. The p53 tumor suppressor protein is stabilized in response to DNA damage and other stress signals and causes the cell to undergo growth arrest or apoptosis, thus preventing the establishment of mutations in future cellular generations. Mutation or loss of p53 is a very common event in tumor progression. It occurs in about 50% of all tumors analysed including of colon, lung, breast and liver. The cellular mdm-2 gene, which has potential transforming activity that can be activated by overexpression, is amplified in a significant percentage of human sarcoma and in other mammalian tumors. Proteins encoded by the mdm-2 gene are able to bind to the p53 protein and, when overexpressed, can inhibit p53's transcriptional activation function, thus mdm-2 can act as a negative regulator of p53 function. Experimental study was performed to observe the relationship between p53 gene mutation and mdm-2 protein expression and apply the results to the clinical activity. 36 golden syrian hamster each weighing $60{\sim}80g$ were used and painted with 0.5% DMBA by 3 times weekly on the right buccal cheek(experimental side) for 6, 8, 10, 12, 14 and 16 weeks. Left buccal cheek(control side) was treated with mineral oil as the same manner to the right side. The hamsters were sacrificed on the 6, 8, 10, 12, 14 & 16 weeks. Normal and tumor tissues from paraffin block were examined for histology and immunohistochemistry observation, and were completely dissected by microdissection and DNA from both tissue were isolated by proteins K/phenol/chloroform extraction. Segments of the hamster p53 exons 5, 6, 7 and 8 were amplified by PCR using the oligonucleotide primers, and then confirmational change was observed by SSCP respectively. The results were as follows : 1. Dysplasia at 6 weeks, carcinoma in situ at 8 weeks and invasive carcinoma from 10 weeks could be observed in experimental groups. 2. p53 mutations were detected in 10 of the 36(28%) and the exons 6(6 of the 10 : 60%) was the most hot spot area among the highy conserved region(exons 5, 6, 7 & 8). 3. Immunohistochemical study confirmed 22 of the 36(61%) of p53 expression involving 10 of p53 mutations. 4. mdm-2 expression of was showed in 3 of the 36(8%) involving 1 of the 22 of p53 expression and 2 of the 14 of p53 non-expression. From the above results, mutation of p53 gene or expression of p53 protein may have the influence of the DMBA induced carcinoma of hamster buccal pouch but the expression of mdm-2 protein may not have relationship with tumorigenesis.

        만성신부전환자에서 혈청 Adenosine Deaminase 활성도에 관한 연구

        N/A To evaluate the clinical availability of serum adenosine deaminase (ADA) activity as an immunological parameter in chronic renal failure patients undergoing hemodialysis, serum ADA activities and other immunologica1 parameters, such as total lymphocyte count and its subpopulations and concentrations of immunoglobulin G, A and M, were measured simultaneously and analyzed for their correlations in 20 normal adults and 30 chronic renal failure patients. The results were as follows: 1) In normal adults, the mean value of serum ADA activities was 18.22±1.58U/L with no age and sex differences. 2) In two grous of long-term hemodialysis (over 12months) and short-term hemodialysis (under 1 month), mean values of serum ADA activities were 31.4±10.1U/ L and 33.6±12.3U/L respectively. There were significant differences between normal adults and the two hemodialysis groups, but there was no significant difference between the two hemodialysis groups. 3) There was no significant difference among various renal diseases. 4) Mean values of serum ADA activities decreased after hemodialysis in each group. 5) There were no significant correlations between serum ADA activity and BUN, creatinine, total lymphocyte count and subpopulations, and concentrations of immunoglobulin G, A and M. As mentioned above, serum ADA activities in chronic renal failure patients undergoing hemodialysis were higher than in normal adults, and there was a decreasing tendency of serum ADA activity immediately after hemodialysis, but serum ADA activity seems of little use as an immunological parameter in chronic renal failure patients.

        지방공무원의 개인특성과 직무특성이 경력몰입에 미치는 영향

        경력은 조직구성원들에게 있어서 장기적이며 평생에 걸쳐 직업과 관련된 것으로 고용관계가 불확실한 현시점에서 그 중요성이 더욱 커져가고 있다. 본 연구의 목적은 개인특성과 직무특성이 경력몰입에 미치는 영향을 검증하는데 있다. 이를 위해 선행연구를 토대로 가설을 설정하여 실증연구를 실시하였다. 356명을 대상으로 분석한 결과 개인특성인 자기효능감과 승진욕구는 경력몰입에 정(+)의 영향을 미치는 것으로 나타났다. 또한 직무특성의 경우 직무적합성은 통계적으로 유의한 정(+)의 영향을 미치는 것으로 나타났으나, 직무자율성은 유의한 관계가 없는 것으로 나타났다. 분석결과를 바탕으로 연구의 한계점과 향후 연구방향을 제시하였다. As today’s organizations are no longer lifelong employment, individuals tend to put stress on psychological satisfaction with the need to manage one’s career and employability in the open labor market rather than pursuing rapid promotion or high compensation in a organizations. Traditionally, the job of local public servant was a relatively stable occupation. In recent years, however, local public servants should acquire new competency and develop their competency to respond to the changes of administrative environment. The purpose of this study is to examine the effects of individual and job characteristics on career commitment among local public officials in Korea. The data was obtained from public officials serving for the regions of Kangwon and Kyeong-In in Korea. A total of 500 questionnaires was distributed and 410 results were collected, finally, 356 questionnaires were used for the actual analysis. The major findings of the study are summarized as follows: Career commitment was positively related to self-efficacy, need for promotion, person-job fit while it was not related to job autonomy. This study discussed implications suggested by these results.

        웨이블릿 방법을 이용한 수치표고모형 압축 포맷 변환

        In this paper, improved wavelet algorithm is proposed for an effective digital elevation model compressive format using JPEG2000 compressive format. This improved algorithms presents for reducing root mean square error of digital elevation model reconstruction using separations between satellite image data and digital elevation model. It is shown that the proposed wavelet algorithm achieves low RMSE and it also demonstrates higher PSNR under per same bit rate whereas standard JPEG2000 format. The experimental results imply the possibility that the improved method can be applied to digital elevation model data which require better data accuracy. 본 논문에서는 JPEG2000 압축 포맷을 사용하여 수치표고모형 자료의 효율적인 압축을 위한 개선된 웨이블릿 알고리즘을 제시하였다. 개선 알고리즘은 위성 영상 자료와 수치표고모형 자료를 분리하여 수치표고모형 자료의 재구성에 있어서 발생되는 RMSE를 줄일 수 있었다. 개선 웨이블릿 알고리즘은 낮은 RMSE를 보였으며, 표준 JPEG2000 포맷에 비하여 동일한 비트율에서도 높은 PSNR을 나타냈다. 실험 결과를 통해 개선 웨이블릿 알고리즘이 데이터의 정확도가 높아야 하는 수치표고모형 자료의 압축에 유용할 것으로 판단되다.

      • The Tip-Tilt Correction System in AO System for Small Telescope

        We are developing Adaptive Optics (AO) system for 24 inch telescope at Seoul National University Observatory. It consists of the tip-tilt correction system and the residual wavefront error correction system with a deformable mirror and a wavefront sensor. We present the construction and performance measurements of the tip-tilt correction system. The tip-tilt component is the single largest contributor to wavefront error, especially for small telescope. The tip-tilt correction system consists of a quadrant photodiode, a tip-tilt mirror and a feed back loop. The collimated He-Ne laser beam is used for input light source and is artificially disturbed by air turbulence generated by a heat gun. Most of the turbulence is of low frequency less than 20 Hz, but extends to a few hundreds Hz. It is found that the closed loop system using proportional-integral-derivative (PID) control successfully corrects tip-tilt error at a rate as high as 300~400 Hz.

        항해안전 및 업무효율을 위한 ECDIS 사용자측면에서의 개선사항 연구

        전자해도 시스템은 2012년 7월 1일 부로 국제항해에 종사하는 선박에 단계별로 설치가 강제화 되었다. 전자해도시스템은 백업장치 를 비치할 경우 종이해도를 대체하여 사용할 수 있는 설비로 선교에서 사용의존도가 높은 핵심항해설비라고 할 수 있다. 현재 전 세계적으로 전자해도 시스템 이용 시 다양한 오류가 발생하고 있으며, 국제해사기구 및 국제수로기구에서도 문제를 인식하고 오류해결을 위한 방안을 모색 하고 있는 중이다. 본 논문에서는 선장 항해사를 포함하는 사용자들을 대상으로 전자해도시스템의 이용 시 오류 발생현황과 정보표시 및 주요 기능 이용 시 개선 요구사항에 관한 설문을 시행하였다. 그 결과를 기반으로 항해안전과 효율측면에서의 ECDIS 기능 개선사항을 제시하였다. 조사에서 사용자들은 ECDIS 기능사용에 있어 대부분 효율성 측면에서는 긍정적으로 평가하였으나 항해안전성에 대한 불안감을 많이 느끼는 것으로 나타났다. On J uly 1 2012, the carriage requirement for ECDIS became mandatory in a phased-in manner. The system has since been widely used as a core bridge navigation system and as a replacement for paper charts with a back up arrangement. However operational anomalies have been found in the system internationally. IMO and IHO have recognised these anomalies and seek for solution. This study has been conducted on a survey of seafarers on the use of ECDIS with regard to anomalies identification, level of the seafarer satisfaction and requirements for further functional improvement. From the results, the ECDIS user focused requirements suggested on strengthening the level of navigational safety and efficiency. The efficiency of using main functions of ECDIS was positively evaluated. However, there were concerns regarding its operational safety.

        서울전파천문대(SRAO) 6M 망원경의 ON-THE-FLY 관측 시스템 구축

        On-The-Fly (OTF) observation method is developed for the efficient use of 6 M radio telescope at Seoul Radio Astronomy Observatory (SRAO). This technique, in which data and information of antenna position are recorded synchronously while driving a telescope regularly and rapidly across a field, provides more efficient use of telescope time and better calibration of the acquired data than the traditional point-to-point observation method does. For the realization of the method, we (1) added RT-Linux modules to the existing operating system, (2) replaced digital voltmeter with voltage-to-frequency converter, and (3) modified many SRAO observation programs. By observing Moon and G78.2+2.7 using this method and comparing them with previous observations, we verify the successful operation and efficiency of the OTF observation mode.
