해양에서 분리한 Exiguobacterium sp. SC2-1의 항산화 활성 및 특성

제주도 연안의 해수로부터 항산화물질을 생산하는 세균을 분리하였으며 항산화물질의 특성을 조사하였다. 분리된 항산화 생성 균주는 그람양성의 통성혐기성균으로 운동성을 가진 단간균이었으며 생육시 NaCl를 필요로 하였다. 분리균주는 형태학적, 배양적, 생화학적 특성 및 165 rDNA 염기서열 분식결과 Exiguobacterium 속으로 동정되어 Ekiguobaoteriurn sp. SC2-1로 명명하였다. 항산화물질 생산균주 배양액의 radical 소거활성은 DPPH (1,1-diphenyl-2-picrylhydrazyl) 법을 사용하였으며, 항산화물질 생성 최적조건은 $25^{\circ}C,\;NaCl\;4\%,\;pH\;6\~8$ 이었으며, 최적 탄소원은 $1\%$ maltose였다. 배양액의 hydroxyl radical 소거활성은 $73\%$였으며 superoxide radical 소거활성은 $35\%$를 나타내었다. For the reseach of the natural marine antioxidant, an antioxidant-producing bacterium was isolated from seawater in Jeju costal area. The isolated strain SC2-1 was Gram-positive, catalase positive, oxidase negative, motile and small rods. The strain utilized sucrose, dextrose, fructose, mannitol and maltose as a sole carbon and energy source and NaCl required for growth. The radical scavenging activity of the culture supernatant was detemined by DPPH method. This bacterium was identified based on morphological, biochemical characteristics and 16S rDNA sequence analysis, and then was named Exiguobacterium sp. SC2-1. The optimum conditions of culture for production antioxidnat were $25^{\circ}C$, pH 6-8 and $4\%$ NaCl. The stain showed the highest activity and growth cultured in medium which added $1\%$ maltose. Hydroxyl radical activity of the supernatant of Exiguobacterium sp. SC2-1 was $73\%$. The SOD activity of the culture supernatant was estimated about $35\%$.
염색공단폐수의 색도처리를 위한 염료분해 균주의 분리와 특성

To treat chromaticity contained in effluents of dyeing wastewater efficiently, potent dye-degrading microorganisms were isolated from influent water, aeration- tank sludge, recycle water and settling-tank sludge located in leather and dyeing treatment plant. Six potent strains were finally isolated and identified as Comamonas testosteroni, Methylobacteriaceae bacterium, Stenotrophomonas sp., Kluyveromyces fragilis, Ascomycetes sp. and Basidiomycetes sp. When Basidiomycetes sp. was inoculated into ME medium containing basal mixed-dyes, 93% of color was removed after 8 days incubation. In the same experiment, the 1:1 mixed culture of Basidiomycetes sp. and photosynthetic bacterium exhibited 88% of color removal; however, it showed better color removal for single-color dyes. The aeration-tank and settling-tank samples revealed higher color removal (95-96%) for black dyes. The settling-tank sample also revealed higher color removal on basal mixed-dyes, which resulted in 90% color removal after 6-h incubation. From the above results, it is expected to achieve a higher color removal using the mixed microorganisms that were isolated from aeration-tank and settling-tank samples.
