인진청간탕 및 와송 어성초 가미방의 간섬유화억제에 미치는 효과

Objectives : This study was performed to investigate the anti-fibrogenic effect and the effect on cell growth and apoptosis in YJCGT, YJCGT YSO and YJCGT YSCO on thioacetamide-induced rat liver tissue and the immortalized human hepatic cell line LX2. Materials and Methods : LX2 cells were treated with various concentrations (0, 50, 150, 300 ug/ml) of YJCGT, Y+YSO, and Y+YSCO extract for 24, 48 and 72 hours. After the treatment, cell viability was measured by using MTT assay. Caspase inhibitor assay, and cell viability were determined by a colorimetric assay with PMS/MTS solution. Rat liver fibrosis was induced by intraperitoneal thioacetamide injection 150 mg/kg 3 times a week for 5 weeks. After the treatment, body weight, liver & spleen weights, liver function test, the complete blood cell count and the change of portal pressure were studied. After YJCGT, Y+YSO, and Y+YSCO treatment, percentages of collagen in thioacetamide-induced rat liver tissue were measured. Results : The viability of the LX2 cell decreased in a dose- and time-dependent manner. Exposure of LX2 cells to YJCGT, YJCGT+YSO and YJCGT+YSCO induced caspase-3 activation, but co-treatment of YJCGT, YJCGT+YSO and YJCGT+YSCO with the pan-caspase inhibitor Z-VAD-FMK, and the caspase-3 inhibitor Z-DEVE-FMK, blocked apoptosis. There was no difference in rat body weight between the thioacetamide only group and the YJCGT, YJCGT+YSO and YJCGT+YSCO groups. In the YJCGT, YJCGT YSO and YJCGT YSCO groups, the serum level of GPT significantly went down compared with the thioacetamide only group. In the YJCGT, Y+YSO, Y+YSCO groups, white blood cell elevated by thioacetamide injection decreased but RBC, Hgb, and Hct increased. In the Y+YSO group, the portal pressure elevated by thioacetamide injection significantly decreased. In the histological finding, thioacetamide injections caused severe fibrosis, but YJCGT, Y+YSO, and Y+YSCO treatment significantly reduced the amounts of hepatic collagens. Conclusions : YJCGT, Y+YSO, and Y+YSCO inhibit the growth of LX2 cells by inducing apoptosis through caspase activity. YJCGT, Y+YSO, and Y+YSCO have beneficial effects on the treatment of cirrhotic patients as well as patients with chronic hepatitis.

Objectives : The aim of the study was to evaluate the effect of WS on HepG2 cell cycle and expression of related genes. Methods : The MTT assay, Cell counting analysis, $[^3H]-Thymidine$ Incorporation Assay, Flow cytometric analysis, Quantitative RT-PCR were studied. Results : WS inhibited HepG2 cell proliferation in low concentration$(1-10\;{\mu]g/ml)$ which did not cause direct cytotoxicity, with dose-dependant manner. WS in-hibited DNA synthesis as well. Flow cytometric analysis on the HepG2 cell showed G2/M phase arrest. Conclusion : These results suggest that WS inhibits HepG2 cell proliferation not by the gene regulation but by G2/M phase arrest in the cell cycle. Thus further studies on the effect of WS in G2/M phase regulation are thought to be needed.

An upper bound solution is obtained to perform the process analysis of hot strip rolling process. The material flows within the roll bite at various geometries and frictional conditions are obtained from finite element analysis and the typical flow pattern which is necessary to determine the kinematically admissible velocity field is assumed. From the kinematically admissible velocity field, the upper bound energy is calculated and the rolling load, angle of neutral point and forward slip ratio at various operational conditions are obtained from upper bound energy. The process analysis of above mentioned parameters at various operational conditions have provided valuable information which is hard to obtain during rolling operation and the predicted ranges of quantitive values from these analyses lie whthin the bound of actual operational data.
레이저 직접금속성형기술을 이용한 금형재 표면보수 특성 연구

Direct laser metal forming technology was applied to restore the damaged mold surface. In order to estimate melting characteristics of the 20 ㎛ Fe-Cr-Ni powder, single layer experiments were performed at various levels of heat input. The process window of the 20 ㎛ Fe-Cr-Ni powder provided feasible process parameters for the smooth regular surface. The cross hatching scanning strategy on the multiple layer experiment was performed to reduce the thickness non-uniformity of edge portions compared with the one direction scanning. To estimate the coherence between the melted powder and the basematal, the tendency of hardness distribution has been observed. The hardness of the melted and the remelted zone was distributed from 400HV to 600HV. It is over 2 times compared of the hardness of the basemetal. Experimental results show that the mold restoring process using direct laser metal forming can be successfully applied in the mold repair industry.
FMEA for the reliability of hydroformed flanged part for automotive application

Tube hydroforming technology is widely used in the automotive industries due to its advantages such as weight reduction, increased strength, improved quality, and reduced tooling cost compared to conventional manufacturing technology. The hydroformed parts often have to be structurally joined at some point. Therefore, the hydroformed automotive parts which have a localized attachment flange are useful. In many cases, the parts formed by hydroforming process are directly related with structural safety, reliability of hydroformed parts must be considered. In this study, hydroforming process of flanged rectangular parts was designed and process reliability was investigated. Finite element analysis was performed to optimize tool geometry considering process parameters such as die aspect ratio and pressure conditions with Dynaform 5.5. Hydroforming experiments to fabricate a flanged rectangular part were performed with optimized tool geometry. The relationship between process parameters and defect was analyzed by FMEA (failure modes and effects analysis). The result shows that process condition was optimized and reliability of rectangular part was increased.

During the tube hydropiercing, the region adjacent to the pierced hole will be deformed and will be drawn away from the die block as the punch advances through the wall of the tube. The deformation in the region may range from a substantially flat form to a countersunk form, so called rollover. In this study, the effects of material properties, shape of piercing punches, roundness of tube surface and internal pressure within the tube during piercing on the rollover have been investigated experimentally. The results provide the quantitative variation of rollover at given hydropiercing parameters, and a relationship between the deformed radius and the rollover caused by the deformation has been established.

adaption of the model predictions is highly desirable. In general, the width deviation at the head and tail ends of strip may be different from that of the steady state region. Therefore, the dynamic edger corrections can be used to compensate the width deviations which would otherwise occur. For the precise width control, the effect of edger roll gap and rolling conditions on the width deviation of head and tail ends of strip has been investigated and the effective method to decrease width deviation has been proposed. On-line application of dynamic edger control method in this study shows about 50% width compensation at the head end of the strip, and near perfect compensation at the tail end of strip.
