        EBM 기반 구축을 위한 팔물탕 문헌 연구 분석

        Objective : The purpose of this report was to provide the information activity and safety of Palmul-tang by analyzing domestic/international papers and theses about Palmul-tang, Methods: Domestic/international papers and theses related to Palmul-tang were reviewed and analyzed, These papers were then classified by year, experimental method and subject, Results: The following results were obtained in this study. 1. The study of Palmul-tang started from 1985 and was continuously increased. The study was mainly forcused on experimental model rather than clinical study. 2. As these studies were classified by subject, papers related to immune intensification were most abundant by 20 papers, Besides there were several papers related to cardiovascular activity, reproductive activity, anti-apoptotic activity and cerebral hemodynamics. 3. Among the papers related to immune intensification. the studies on recovery of fatigue were mostabundant by 10 papers and the studies of on immune cell and cytokine express were six. In addition to. several studies were associated with anti-cancer activity and anti-allergic activity. Recovery of fatigue was determined by measurement of fatigue markers in a living body such as lactate. CPK, pyruvate and triglyceride and assessment of exercise capability of animals such as swimming test. slopped plate test. Rota-rod test, and activity cage test after Palmul-tang treatment. 4. According to experimental data. it is supported that Palmul-tang has been used as Qi and Blood intensifier with immune intensification and recovery of fatigue. 5. The paper related to safety of Palmul-tang was only one paper which is studied on acute toxicity of Palmul-tang with experiment with ICR mouse. There was no study on evaluating safety by observing liver and kidney functions after Palmul-tang treatment Conclusion: Palmul-tang is being used in various ways associating with immune intensification. cardiovascular activity and reproductive activity. However. studies on efficacy and mechanism of Palmul-tang should be conducted at the molecular biology level and studies on safety of Palmul-tang need to be completed at the clinical level.

        글루타메이트에 의해 산화적 스트레스를 받은 HT22 세포에서 핑거루트의 신경세포 보호활성

        Excessive glutamate causes oxidative stress in neuronal cells, which can cause degenerative neurological disorders. We tried to find medicinal plant showed neuroprotective activity by using glutamate-injured HT22 cell as a model system. In this study, we found that Boesenbergia rotunda methanol extract showed neuroprotective activity against glutamate induced neurotoxicity in mouse hippocampal HT22 cells. B. rotunda methanol extract suppressed the formation of reactive oxygen species and decreased intracellular Ca<sup>2+</sup>concentration. Also, B. rotunda made mitochondrial membrane potential maintain to normal levels. In addition, B. rotunda increased total glutathione amount and activated antioxidative enzyme such as glutathione reductase and glutathione peroxidase compared to glutamate-treated groups. These results suggested that B. rotunda decreased neuronal cell death damaged by high concentrations of glutamate treatment, via antioxidative mechanism and might be one of candidate of development of new drug to treat neurodegenerative disease such as Alzheimer's disease.

        ICR 마우스를 이용하여 사물탕(四物湯)의 급성 독성에 관한 연구

        Objectives: Samul-tang(Siwu-tang) has been traditionally prescribed a medicine as a restorative. Methods: In this study, we investigated the acute toxicity about water-extracted Samul-tang(Siwu-tang). Twenty-five mice completed 14 days of oral Samul-tang(Siwu-tang) at the respective doses of 0(control group), 2560, 3200, 4000 and 5000mg/kg. Results: We observed survival rates, general toxicity, change of body weight, and autopsy. Conclusions: Compared with the control group, we could not find any toxic alteration in all treated groups (2560, 3200, 4000 and 5000mg/kg). $LD_{50}$ of Samul-tang(Siwu-tang) was over 5000mg/kg and it is very safe to ICR mice.

        십전대보탕 발효물의 성분 분석 및 뇌신경 세포 보호 활성

        Sipjundaebo-tang was a well-known restorative traditional herbal prescription that used to treat anemia, anorexia, fatigue and inflammation. In this study, we examined the bioconversion of compounds in the Sipjundaebo-tang (SJ) and fermented Sipjundaebo-tang with Lactobacillus fermentum KFRI 164 (FSJ) using established HPLC-DAD method. The chromatogram of FSJ has shown that the contents of six bioactive compounds 5-HMF, paeoniflorin, ferulic acid, cinnam aldehyde, decursin, glycyrrhizin in SJ has decreased. And the contents of unknown compounds (1), (2), (3), (4) and (5) in FSJ were higher than each contents of SJ. The antioxidant activities of SJ and FSJ were conducted by DPPH free radical and Hydrogen peroxide (H2O2) scavenging assay. Electron donating activity (EDA, %) value of FSJ has shown higher than 21.9% and 14.5% at a concentration of 0.5 mg/ml for DPPH radical scavenging activity and H2O2 scavenging activity, respectively. Also, the neuroprotective activities of SJ and FSJ against glutamate-induced oxidative stress in a mouse hippocampal cell line (HT22) were evaluated by MTT assay. As a result, FSJ has shown higher neuroprotective activity than 56.5% comparing with SJ. In conclusion, the fermented SJ using microorganism could convert compounds in SJ and enhance antioxidant activity and neuroprotective activity.

        황련해독탕 중 3종 생리활성 물질의 HPLC-DAD 동시 정량분석법 확립

        In this study, a high performance liquid chromatography-diode array detector method was established, for simultaneous determination of three compounds, berberine, palmatine and geniposide in Hwangryunhaedok-tang, To develop and validate method, C18 column (5 μm, 4.6 mm×250 mm) was used with gradient mobile phase, water containing 0.1% trifluoroacetic acid (TFA) and MeOH at the column temperature of 30oC. UV wavelength was set at 230 and 280 nm. Validation of the chromatography method was evaluated by linearity, precision and accuracy test. Calibration curve of standard components showed good linearity (R2>0.9999). The limits of detection (LOD) and limits of quantification (LOQ) varied from 0.05 to 0.17 μg/ml and 0.15 to 0.53 μg/ml, respectively. The relative standard deviations (RSDs) data of intra-day and inter-day test were in less than 2.99% and 1.90%, respectively. The results of the accuracy test were in the range of 98.36 to 102.52% with RSDs values 0.32 to 1.98%. The results of validation indicated that this method was a very accurate and sensitive assay.

        증숙 과정을 통한 소청룡탕의 뇌신경세포 보호 활성 변화에 관한 연구

        So Cheong Ryong Tang (SCRT) has been used as a traditional herbal medicine for the treatment of the bronchial asthma. In this study, the effect of steaming process on changes in 11 major compounds (homogentisic acid, ephedrine HCl, paeoniflorin, cinnamic acid, cinnam aldehyde, glycyrrhizin, 6-gingerol, schizandrin, methyleugenol, gomisin A and gomisin N) content and neuroprotective activity of SCRT were evaluated. Major compound content was slightly different with steaming produce. The contents of paeoniflorin, cinnamic acid, cinnam aldehyde, 6-gingerol and methyleugenol were decreased and homogentisic acid, ephedrine HCl, glycyrrhizin, schizandrin and gomisin A were increased by steaming process. The neuroprotective activity of steaming SCRT was determined in mouse hippocampal HT22 cells by MTT assay. As a result, neuroprotective activity of a steaming SCRT was higher than that of a non steaming SCRT. This study demonstrated that neuroprotective activity of SCRT can be improved through steaming process.

        서장경으로부터 분리한 Homosyringaldehyde의 뇌신경세포 보호 활성

        Homosyringaldehyde was isolated and identified from the 80% methanol extract of roots of Cynanchum paniculatum. C. paniculatum has been widely used for the treatment of various diseases such as neurasthenia, insomnia, dysmenorrheal and toothache. This compound exerted significant neuroprotective activities against glutamate-induced neurotoxicity in hippocampal HT22 cell line by 37.53% (at the concentration of 100 μM). We investigated mode of action of this compound. Homosyringaldehyde (100 μM) significantly decreased the ROS level and Ca2+ concentration in the oxidative stress induced HT22 cells by oxidative glutamate toxicity. Thus, our results suggest that homosyringaldehyde significantly protect HT22 cells against glutamate-induced oxidative stress, via antioxidative activities. As the results, we suggest that homosyringaldehyde may be useful in the treatment of neurogenerative disorders.

        홍경천 뿌리 및 캘러스 초음파 추출물의 항암활성 비교

        홍경천은 높은 약리작용에도 불구하고 특수한 배양 조건 및 낮은 번식율로 인해 홍경천 활성 성분의 대량생산이 불가능한 실정이다. 따라서 본 연구에서는 환경에 구애받지 않고 대량생산 및 품질유지가 가능한 식물조직배양 공정을 통해 홍경천 조직으로부터 유도된 callus를 대상으로 초음파 추출공정을 적용하여 활성성분의 용출을 극대화 및 독성저감을 가능하게 하고 callus에 대한 항암활성을 탐색하여 홍경천 callus의 기능성 식품 소재로서 가치를 발견하고자 하였다. 인간 정상 세포인 HEK293과 HEL299에 대한 세포독성을 측정한 결과, 홍경천과 callus의 독성이 유의적인 차이를 나타내지 않았으며, 홍경천 callus 초음파 추출물이 전체적으로 낮은 독성을 나타내는 것으로 미루어 보아 초음파 공정을 통한 세포독성 저감효과를 확인할 수 있었으며, 조직배양 공정을 통해 홍경천의 독성 저감이 이루어지지 않은 것을 미루어 볼 때 callus가 홍경천과 같거나 비슷한 정도의 활성 성분을 함유하고 있을 것이라 사료된다. 홍경천 및 callus의 소화기, 호흡기 계통의 암세포 생육억제 활성 및 selectivity를 측정한 결과, 특히 간암 세포(HEP3B)에 대해 홍경천 및 callus 추출물의 고농도 첨가 시 암세포 저해 활성이 급격히 증가하는 것을 확인할 수 있었으며, callus 초음파 추출물은 최고농도 1.0 mg/mL에서 73.19%의 암세포 생육저해 활성과 최고 5.94의 selectivity를 나타내었다. 전체적인 항암활성 측정 결과, callus 추출물에서 높은 항암활성을 나타냈으며 특히 위암 및 간암에 대한 탁월한 항암효과가 확인됨에 따라 앞으로 홍경천 및 홍경천 callus의 소화기계 암 또는 특정 암에 대한 항암활성 연구가 필요할 것으로 생각된다. 암의 예방 및 억제에 중요한 기전으로 작용할 수 있는 항산화 활성 평가를 위해 DPPH radical 소거능을 측정한 결과, 초음파 공정을 통한 callus 추출물에서 홍경천의 높은 항산화 작용을 확인할 수 있었다. 또한 인간 면역세포 생존율의 측정을 통해 홍경천 및 callus 추출물에 의해 면역세포가 자극을 받는 것을 확인할 수 있었으며 대식세포의 NO-생성량이 증가하는 것을 확인할 수 있었다. 이러한 결과를 통해 면역계 세포들의 면역체계의 항상성 유지 및 antibody와 cytokine과 같은 면역물질 생성을 통한 암세포 억제 및 제거작용이 가능할 것이라 사료된다. 홍경천 조직으로부터 유도된 callus에 초음파 추출 공정을 적용할 시 세포독성 저감과 활성 성분 용출량의 극대화에 기인한 항암 및 암 발생과 억제 기작에 영향을 주는 항산화 및 면역증강 활성의 증가를 확인함에 따라 홍경천 callus가 기능성 항암 소재로서의 가치가 매우 높다고 사료된다. 하지만 이와 같은 홍경천 callus의 항암 소재로서 활용을 위해서는 활성성분의 용출을 극대화 할 수 있는 추출 공정의 확립과 홍경천 본래의 독성은 줄이고 암세포에 선택적인 활성을 나타낼 수 있는 활성 성분의 규명 및 정상세포 독성을 배제할 수 있는 추출물 농도의 최적화에 대한 연구가 이루어져야 하겠다. In this study, the anticancer activity of the water extract at 100oC was compared to that of the callus extracts via a ultrasonification extraction process. All the extracts were utilized to evaluate cytotoxicity, antioxidant and immune activities. The callus extracted via ultrasonification extraction showed relatively low cytotoxicity on normal human cell lines, HEK293 and HEL299, showing 13.17% and 21.78%, respectively. The callus extract has 59.82% which was similar to 61.70% for water extracts. It was also found that callus extract yielded higher nitric oxide secretion form macrophage than other extracts. The growths of both human stomach adenocarcinoma (AGS) cell and human lung carcinoma (A549) were inhibited up to 70% by adding 1.0 mg/mL of the callus extracts with ultrasonification extraction. This inhibition ratio (70%) was almost close to that of water extract. Human hepatoma carcinoma (HEP3B) cell growth was most significantly inhibited up to 75% by adding 1.0 mg/mL of callus extracts, and its selectivity was highest compared to other extracts. It indicates that the callus extracts could selectively inhibit growth of digestive system-related cancer cells. It can be also concluded from the results of this study that the callus extracts associated with ultrasonification extraction process have the potential for anticancer activity.
