        사람의 골수 줄기 세포로부터의 골세포 분화 과정에서 BMP-2가 미치는 영향과 그에 따른 분화 유전자의 발현 비교 연구

        It is commonly acknowledged that bone morphogenic protein (BMP-2) functions as a potential osteogenic inducer in bone formation. Recently, several papers reported that bone marrow-derived stem cell (BMSC) from human is not responsive to BMP-2 in comparison to high capacity of BMP-2 in the osteoinduction of stromal cell derived from bone marrow of rodent animals such as rat or mouse. In this study, we characterized BMSC derived from 11 years old donor for the responsiveness to rhBMP-2, dexamethasone (Dex) and 1,25-dihydroxyvitamin D (vitamin D), in order to analyze their function in the early osteogenesis. The effect of over mentioned agents was evaluated by means of assessing alkaline phosphatase (ALP) activity/staining, RT-PCR analysis and von Kossa staining. In addition, we analyzed the meaning of expressed several osteoblastic markers such as alkaline phosphatase, collagen typeI, osteopontin, bone sialoprotein and osteocalcin with relation to either differentiation or mineralization. Only in the presence of Dex, human BMSC could commit osteoblastic differentiation and matrix mineralization, and either BMP-2 or vitamin D treatment was not able to induce. But BMP-2 or Vitamin D showed potential synergy effect with Dex. ALP and bone sialoprotein were clearly expressed in response of Dex treatment compared to weak expression of osteopontin in early osteogenesis. Therefore, we expect that this study will contribute partly to elucidiating early osteogenesis mechanism in human, but variations among bone marrow donors must be considered through further study.

        쥐의 골수로부터 추출한 줄기세포를 이용한 조골세포로의 분화 유도과정에서 나타난 문제점에 관한 분석 연구

        This study was aimed to characterize osteogenic potential of rat bone marrow stromal cells (BMSC) isolated with standard flushing method and investigate the plasticity of transdifferentiation between osteoblastic and adipocytic lineage of cultured BMSC. Unlike aspiration method in human, rat bone marrow was extracted by means of irrigation with culture media that elevates the possibility of co-extraction of committed osteoprogenitor, or preosteoblast or other progenitor cells of several types present inside bone marrow. The cultured stromal cells showed high ALP activity which is representative marker of osteoblast without any treatment. Osteogenic inducers such as Dex and BMP-2 were examined for the evaluation of their effect on osteogenic and adipocytic differentiation of stromal cells, because they function as osteoinductive agent in stromal cells, but simultaneously induce adipogenic differentiation. Osteogenic differentiation was evaluated by measuring alkaline phosphatase activity or mRNA expression of osteoblast markers such as osteopontin, bone sialoprotein, collagen type I and CbfaI, and in vitro matrix mineralization by von Kossa staining. Oil red staining method was used to detect adipocyte and adipocytic marker, aP2 and $PPAR{\gamma}2$ expression was examined using RT-PCR. It can be supposed that irrigation procedure resulted in high portion of already differentiation-committed osteoprogenitor cell showing elevated ALP activity and strong mineralization only under the supplement of $100{\mu}M$ ascorbic 2-phosphate and 10mM ${\beta}$-glycerophosphate without any treatment of osteogenic inducers such as Dex and BMP-2. Dex and BMP-2 seemed to transdifferentiate osteoprogenitor cells having high ALP activity into adipocytes temporarily, but continuous treatment redifferentiated into osteoblast and developed in vitro matrix mineralization. This property must be considered either in tissue engineering for bone regeneration, or in research of characterization of osteogenic differentiation, with rat BMSC isolated by the standard irrigation method.

        토양에서 분리된 Aeromonas hydrophila AM-28이 생산하는 단백질 가수분해효소의 특성

        토양으로부터 탈지대두박을 잘 분해하는 단백질 가수분해효소를 생산하는 미생물을 분리하여 형태적, 생화학적 특성과 세포벽 지방산조성을 조사한 결과, 분리된 세균은 Aeromonas hydrophila로 동정되었다. 분리된 세균은 glycerol을 탄소원으로, tryptone 및 $(NH_4)_2HPO_4$을 진소원으로, $CaCl_2$를 무기원으로 사용한 배지에서 효소를 가장 많이 생산하였으며, 생산된 효소의 반응최적 pH는 8.0이고, 반응최적온도는 $65^{\circ}C$였다. 이 효소는 $55^{\circ}C$ 이하의 온도에서, pH 7.0에서 13.0의 범위에서 비교적 안정한 것으로 나타났다. 또한 이 효소는 금속이온 $Cu^{2+},\;Hg^{2+}.\;Mn^{2+}$ 및 $Zn^{2+}$에 의해서 효소활성이 저해를 받고, 효소저해제 phenylmethylsulfonyl fluoride (PMSF)와 ethylenediaminetetraacetic acid (EDTA)에 의해서 효소활성이 강력히 저해되는 성질로부터 효소활성부위에 serine기와 금속이온을 갖고 있는 것으로 밝혀졌다. A bacterial strain NO. AM-28, showing proteolytic activity against defatted soybean was isolated from domestic soil. The isolated strain was identified as Aeromonas hydrophila by both the biochemical tests using API kit and the analysis of cellular fatty acid profile with MIDI system. The protease production from A. hydrophila AM-28 was highly enhanced when it was cultivated in the medium containing glycerol as a carbon source, tryptone or $(NH_4)_2HPO_4$ as a nitrogen source, and $CaCl_2$ as a mineral source. The optimal pH and temperature for the enzyme was 8.0 and $65^{\circ}C$, respectively. The enzyme was stable up to $55^{\circ}C$ and at pH values ranging from 7.0 to 13.0. The enzyme activity was inhibited by phenylmethylsulfonyl fluoride and EDTA, indicating that serine residue and metal ions be involved in enzyme activity.

      • 조직배양에 의한 위도상사화(Lycoris uydoensis M. Kim)의 효율적 기내번식

        위도상사화(Lycoris uydoensis M. Kim)의 효율적인 기내 급속 대량 증식체계를 확립시키고자 잎과 인편을 배양하였다. 5℃에서 60일간 저온 처리한 모구를 구고를 중심으로 상부, 중부 및 하부로 구분하여 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA, 0.5 mg·L-1 NAA와 1.0 mg·L-1 BA 또는 3.0mg·L-1 IBA와 3.0 mg·L-1 BA 혼용처리된 MS 배지에 배양한 결과 식물생장조절물질과는 관계없이 디스크가 부착된 인편의 하부가 캘러스 및 자구 형성에 가장 효과적인 부위였다. 캘러스는 잎절편이나 인편조직에서 쉽게 형성되었으나 소자구 및 신초는 인편에서만 형성되어 기내번식에 적합한 절편체는 인편이었다. 자구 형성 및 신초분화는 배지내 첨가된 식물생장조절물질에 따라 치상체에서 직접 자구를 형성하거나 캘러스를 통해 식물체로 분화되는 2가지 경로로 형성되었는데 디스크가 부착된 인편을 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA, 0.5 mg·L-1 NAA와 1.0 mg·L-1 BA 또는 3.0 mg·L-1 IBA와 3.0 mg·L-1 BA 혼용처리된 MS 배지에 배양하여 30일 이내에 60%-100%까지 유도하였다. 다량의 자구형성 및 유지는 MS배지에 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA가 혼용되거나 또는 3.0 mg·L-1 IBA와 3.0mg·L-1 BA 혼용처리 되었을 때 0.5 mg·L-1 NAA와 1.0mg·L-1 BA 혼용처리 되었을 때 보다 효과적이었다. 6% sucrose는 자구형성 및 비대를 촉진시켰으며 특히 3.0 mg·L-1 IBA와 3.0 mg·L-1 BA 혼용처리배지에 6% sucrose를 첨가시키므로 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA가 혼용 배지보다 구의 비대가 1-2배 증가하였다. 인편유래 캘러스로부터 기관분화 또는 인편에서 직접 분화된 자구에서 성공적으로 식물체가 재분화되었다. This study was conducted to establish an effective in vitro mass propagation system using leaf and bulb scale explant sources of Lycoris uydoensis M. Kim. For the bulb scale culture, the bulbs treated at 5℃ for 60 days were dissected into three parts according to bulb height (upper, middle, and bottom), and allowed for a culture on the MS medium with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA, 0.5 mg ? L-1 NAA plus 1.0 mg ? L-1 BA, and 3.0 mg ? L-1 IBA plus 3.0 mg ? L-1 BA. Regardless of plant hormone treatments, the bottom part of the bulb including the disk proved to be most effective in forming the callus and bulblets. Callus was readily produced on the young leaf segment and bulb scale tissues, whereas, bulblets and shoots were formed only on bulb scale explants. Bulb scales with disk appeared to the best optimal explant for in vitro propagation. Two distinct pathways, either organogenesis through callus or direct bulblet formation, could be recognized in bulb scale culture. Bulblet formation and shoot organogenesis from the bulb scales with disk were effectively induced up to the 60%~100% on the MS medium supplemented with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA, 0.5 mg ? L-1 NAA plus 1.0 mg ? L-1 BA, and 3.0 mg ? L-1 IBA와 3.0 mg ? L-1 BA within 30 days of culture. On MS media supplemented with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA or 3.0 mg ? L-1 IBA와 3.0 mg ? L-1 BA were more effective than 0.5 mg ? L-1 NAA plus 1.0 mg ? L-1 BA for induced and maintained a large number of bulblets. Six percent of sucrose enhanced bulblet formation and expanded the bulblets. Especially, adding 6% of sucrose on MS media supplemented with 3.0 mg ? L-1 IBA와 3.0 mg ? L-1 BA was one or two times of bulblets expanted than on MS mediun with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA plus 6% of sucrose. Plantlet regeneration was successfully achieved from bulb scale-derived callus, via shoot bud induction or direct bulblet formation.

        코로나19 팬데믹 불안을 겪는 노인들의 불안감 완화를 위한 대면 비대면 융합 차문화치료 프로그램

        본 연구는 코로나19 팬데믹 불안과 디지털 소외를 경험하는 노인들의 불안감 완화를 위한 대면 비대면 융합차문화치료 프로그램을 개발하고, 프로그램의 효과성을 검증하기 위하여 P시 소재 K노인주간보호센터를 이용하는 70세 이상의 노인들 8명을 연구대상으로 하여 개발된 프로그램을 적용하고 그 효과를 살펴보고자 한다. 코로나19 팬데믹 불안을 겪는 노인들의 불안감은 프로그램 참여 전 참가자들의 심리불안 점수 평균은 3.02(SD 0.25)였고, 참여 후 평균은 2.79(SD 0.15)로 나타나 프로그램 참여 전과 후의 유의한 차이로 심리불안 점수가 완화되는 것으로 나타났다(Z=4.245, P=.004). 이러한 분석결과를 바탕으로 코로나19 팬데믹 불안을 겪는 노인들의 불안감 완화를 위한 대면 비대면 융합 차문화치료 프로그램의 확대를 위한 실천적 제언을 제시하였다. This study aims to develop a face-to-face & non-face-to-face convergence tea culture therapy program for elderly people experiencing CORONA 19 pandemic anxiety and digital alienation, and apply the program to eight senior citizens aged 70 or older who use the K Senior Citizens' Day Care Center in P City to verify its effectiveness. Anxiety among elderly people experiencing coronavirus anxiety was 3.02 (SD 0.25) before participating in the program and 2.79 (SD 0.15), indicating a significant difference between before and after participating in the program (Z=4.245, P=.004) Based on this analysis, we present practical suggestions for the expansion of face-to-face & non-face-to-face convergence tea culture therapy programs to alleviate anxiety among elderly people who experience CORONA 19 pandemic anxiety.

        차문화치료 융합프로그램을 통한 노인의 관계적 공격성 완화 : 노인주간보호센터 이용 여성노인들을 대상으로

        본 연구는 관계적 공격성 완화를 위한 차문화치료 융합프로그램의 효과성을 검증하기 위하여 P시 소재 노인주간보호센터를 이용하는 70세 이상의 여성노인들 10명을 연구대상으로 하여 노인의 관계적 공격성 완화를 위한 차문화치료 융합프로그램을 적용하고 그 효과를 살펴보고자 한다. 분석결과로 반응적 관계적 공격성과 주도적 관계적 공격성 점수는 사전검사 평균보다 사후검사 평균에서 0.21 낮아져 차문화치료 프로그램에 참여한 노인들의 주도적 관계적 공격성은 프로그램 참여 전보다 감소 된 결과를 보여주고 있다. 이러한 분석결과를 바탕으로 노인주간보호센터 이용노인의 관계적 공격성 완화를 위한 실천적 제언을 제시하였다. In order to verify the effectiveness of the tea culture treatment program to mitigate the relative aggressiveness, the research will target 10 women aged 70 or older who use the P City-based Senior Citizens' Day Care Center to apply the tea culture treatment program to alleviate the relative aggressiveness of the elderly and examine its effects. As a result of the analysis, the score of reactive relational aggressiveness and leading relational aggression is 0.21 lower than the pre-exam average, indicating that the leading relational aggression of the elderly who participated in the tea culture therapy program is reduced than before the program. Based on these analysis results, practical suggestions were made for alleviating aggression among senior citizens using the Center for the Elderly.
