연구 목적: 본 연구의 목적은 마우스에서 배양액의 용량이 배반포 배 형성과 세포수에 미치는 영향을 조사하기 위하여 실시하였다. 연구 재료 및 방법: $3{\sim}4$주령 ICR 암마우스에게 48시간 간격으로 5 IU PMSG와 hCG 주사 후 (hCG 주사 후 수컷과 동숙) $46{\sim}50$시간에 난관으로부터 총 138개의 2-세포기 배를 회수하여 2 ml (group I) 또는 $50{\mu}l$ (group II)의 배양액 (Dulbecco's Modified Eagle Medium + 20% human follicular fluid)에서 72시간 동안 배반포기까지 배양하였다. 배반포 배는 zona-intact (ZiB)와 zona-escape (ZeB)로 등급을 구분하고 나서, propidium iodide와 bisbenzimide를 이용한 differential staining 방법으로 염색하여 평균 세포수, 내세포괴(ICM) 세포수, 영양배엽(TE) 세포수, 총 세포수에 대한 ICM의 비율 (%ICM) 및 ICM:TE 비율을 조사하였다. 결과에 대한 유의성 검정은 $X^2$ test와 t-test를 이용하였으며, p<0.05일 때 통계적인 차이가 있는 것으로 하였다. 결 과: Group I과 II에서, 총 배반포 ($62.3{\pm}20.7%$ vs. $63.8{\pm}22.9%$), ZiB ($31.9{\pm}24.0%$ vs. $30.4{\pm}18.2%$)와 ZeB 형성율 ($30.4{\pm}20.8%$ vs. $33.3{\pm}22.3%$)은 차이가 없었다. 87개의 배반포 배를 염색 시도하였는데, 명확하게 differential staining된 41개의 배반포 배만을 대상으로 세포수를 조사하였다. 평균 세포수 ($61.6{\pm}19.5$ vs. $63.7{\pm}26.8$), ICM 세포수 ($13.0{\pm}10.6$ vs. $12.8{\pm}10.5$), TE 세포수 ($49.0{\pm}19.0$ vs. $47.8{\pm}18.7$), %ICM ($21.0{\pm}12.6%$ vs. $21.1{\pm}13.2%$) 및 ICM:TE 비율 (1:$3.77{\pm}4.9$ vs. 1:$3.72{\pm}4.8$)에서도 group I과 II에서 차이가 없었다. 결 론: 마우스에서 배 발생 능력의 척도로 쓰이는 배반포 배 형성율 배반포 배의 등급 세포수 및 %ICM 등이 20% 난포액을 첨가한 MEM 배양액의 용량에 따라 영향을 받지 않았다.

Amniotic fluid embolism is rare and unexpected but a life-threatening complication of pregnancy is accompanied by a high mortality rate. This syndrome usually occurs at the time of delivery. Classical clinical features are sudden onset of circulatory collapse, acute respiratory distress, disseminated intravascular coagulation and neurological impairment like seizure. The diagnosis is made by identifying clinically characteristics symptoms, and by exclusion of other cause. We report one case of a patient with amniotic fluid embolism, which showed atypical symptoms like seizure.

Angiomyofi broblastoma (AMFB) is a rare mesenchymal tumor mostly involving the female lower genital tract. Although a large number of cases of vulva and vagina has been reported, a report on a case of the uterine cervix is uncommon. Despite unknown pathogenesis of AMFB, positive immunoreactivity for hormonal receptor of AMFB allows to make an assumption that the hormonal stimuli may have a role on the development of AMFB. Therefore, it is potential that estrogenic effect of tamoxifen gives stimuli on hormonally responsible mesenchymal cells, resulting in AMFB. We report a case of AMFB of the uterine cervix in a breast cancer patient with prolonged tamoxifen treatment with brief literature review.
Cisplatin 내성 난소암 세포주로부터 2차 내성 아세포 수립과 성격규명

목적: 난소암에서 1차 항암치료로 백금제제를 사용한 저항성 재발 난소암 환자는 2차 항암제에 반응율이 낮고 또한 재발율도 높다. 본 논문에서는 2차 항암제에 대한 내성 난소암 아세포주를 수립하여 항암제 내성기전을 규명고자 하였다. 방법: 2차 항암제 내성 아세포주 SNU-8/Fac를 수립하여 MTT assay로 항암제 감수성을 측정하였고 RT-PCR방법과 western blot방법으로 내성 기전을 규명하였다. 결과: 2차 항암제 내성 아세포주 SNU-8/Fac를 수립하여 항암제 감수성을 측정한 결과 multidrug resistance를 보였다. 내성 관련 유전자를 관찰한 결과 mRNA수준에서는 LRP는 발현 정도를 관찰할 수 없었고 MRP1, BCRP, TS와 MT는 SNU-8/Fac에서 SNU-8/WT보다 더 과발현되지 않았으나 MDR1와 ATP7B는 더 과발현되었고 protein수준에서는 P-gp와 ATP7B가 더 과발현되었다. 결론: 본 연구에서는 cisplatin 내성 난소암세포로부터 항암제 5-FU에 내성이 있는 세포주 SNU-8/Fac을 수립하였고 수립한 세포에서 P-gp/MDR1과 ATP7B유전자가 과발현되는 것을 관찰하였다. Objective: Multidrug resistance to chemotherapy is a major obstacle in attempts to improve the clinical outcome of ovarian carcinoma patients. The aim of this study is to establish secondary anticancer drug resistant cell line from original SNU-8/WT resistant to cisplatin and characterize it. Methods: After establishing secondary drug resistant cell line (SNU-8/Fac), drug sensitivity was measured by MTT assay. Multidrug resistance (MDR) was analyzed by RT-PCR and western blotting analysis. Results: MTT assay data demonstrated that MDR was expressed in SNU-8/Fac. In addition, mRNA expression of MDR1, ATP7B in SNU-8/Fac was increased. However, overexpression of MRP1, BCRP, TS and MT mRNA was not observed. At the protein level, protein P-gp, ATP7B were overexpressed in SNU-8/Fac. Conclusion: We established a new anticancer drug resistant cell line from original SNU-8/WT. Overexpression of P-gp and ATP7B was observed in SNU-8/Fac.

Neurological manifestation of pre-eclampsia is not so rare. But isolated abducens nerve palsy in pre-eclampsia is very rare. We report a case of severe pre-eclampsia with isolated abducens nerve palsy.

체질성 8삼염색체 모자이시즘은 비교적 드문 염색체 이상으로 특이한 두개안면 모양 (변형 두개골, 돌출된 이마, 낮거나 변형된 귀), 골격계 기형, 심장 기형, 신장 기형, 잠복고환, 다양한 정도의 발달지체 등의 특징적인 표현형을 갖는다. 그 표현형과 세포유전학적 표현의 다양성 때문에 특정 질환이 생기기 전까지 진단이 늦어지기도 한다. 전형적인 표현형 없이 속발성 무월경을 동반한 28세 여성에서 발견된 체질성 8삼염색체 모자이시즘(47,XX,+8[9]/46,XX[41])을 보고한다. Constitutional trisomy 8 mosaicism (CT8M) is a relatively rare aneuploidy in humans with characteristic phenotypes including typical craniofacial feature (such as deformed skull, prominent forehead, low-set and/or dysplastic ears), skeletal malformation, cardiac anomaly, renal malformation, cryptochidism, varying degree of developemental delay. Due to the extremely variable phenotypic and cytogenetic expression, CT8M has gone undiagnosed in certain patients. We report a 28-year-old women with secondary amenorreha without characteristic CT8M phenotype. Chromosomal analysis showed a CT8M (47,XX,+8[9]/46,XX[41]).

Objective : To evaluate degree of bone mineral density and prevalence rate of osteoporosis according to age and to search the preventive strategy for osteoporosis, the factors that affect on bone density were studied in pre- and postmenopausal women in korea. Methods : The subjects were 961 premenopausal and 1,167 postmenopausal women who have examined in health promotion center of Yeungnam University Hospital in Daegu city from January, 1999 to January, 2001. The diet pattern of subjectives was analysed through interview with questionnaire. Results : The bone mineral density was statistically higher in women who have taken more calorie, protein, fat, or carbohydrate than recommended amount compared with women who have taken less calorie, protein, fat, or carbohydrate, than recommended amount in pre- and postmenopausal. In the effects of mineral and vitamin intake on bone mineral density, the higher intake of vitamin B12 and niacin increased bone mineral density statistically significantly in postmenopausal women, the women who have taken calcium more than 500 mg/day were revealed increased bone mineral density compared with less intake women in pre- and postmenopausal, and the women who have taken vitamin C more than 100 mg/day were revealed increased bone mineral density compared with less intake women in pre- and postmenopausal. Conclusion : These results suggested that to prevent osteoporosis, the systemized program including balanced diet for promoting anti-osteoporotic factors have to be developed and apply from the thirties whom have maximal bone mineral density.
