        광학적 pH 센서를 위한 지시염료가 고정된 필름의 광학적 특성 연구

        The optical characteristics of cresol red, phenol red, and neutral red immobilized in the thin film were investigated with absorbance measurement in order to find a sensing part of a fiber-optic pH sensor. Sol-Gel method with tetramethyl orthosilicate as a precursor was used to immobilize the pH indicators in the thin film. The absorbance spectra were measured when pH indicators were immobilized in the film and were dissolved in the buffer solution. Experimental results showed that the absorbance spectra could be changed when the pH indicator is immobilized in the thin film. As compared with other pH indicators, the neutral red exhibited similar absorbance spectra regardless of physical conditions and was sensitive over whole pH range between 4 and 11. In addition, the absorbance ratio of base peak to acid peak tended to increase in proportion to the increase in pH. Experimental results indicate that the neutral red is a good pH indicator for fabrication of a sensing part of the fiber-optic pH sensor.

        고지혈증 흰쥐에 있어서 버섯 분말의 콜레스테롤 저하효과

        본 실험에서는 0.5% 콜레스테롤과 0.25% 콜산나트륨을 첨가한 콜레스테롤 식이와 콜레스테롤 식이에 4.0% 수준의 버섯분말(Lentinus edodes, Ganoderma lucidum, Pleurotus ostreatus; 5 : 3 : 2, w/w/w) 첨가 식이를 female Sprague-Dawley 흰쥐에 4주동안 섭취시켜 지질농도에 미치는 영향을 조사하였다. 혈청 콜레스테롤 농도는 버섯분말 첨가 식이군에서 57% 감소하고, 혈청 HDL-콜레스테롤 농도는 230% 증가하여 동맥경화 지수가 68.4% 감소함으로써 버섯분말에 항동맥경화 효과가 있는 것으로 사료되었다. 간장 콜레스테롤농도는 콜레스테롤 식이군에 비해 버섯분말 첨가식에서 24.7% 감소하였으나, 중성지질 농도는 50%의 유의한 증가를 나타내었다. 본 실험의 결과 콜레스테롤 식이에 버섯분말의 첨가에 의해 혈중 콜레스테롤 농도 저하효과와 동시에 항동맥경화 효과가 인정되어 버섯분말을 소재로 새로운 생리활성을 가지는 기능성식품개발의 가능성이 시사되었다. This study was performed to evaluate the effect of mushrooms powder (Lentinus edodes, Ganoderma lucidum, Pleurotus ostreatus; 5 : 3 : 2, w/w/w) of on the lipid concentrations in female Sprague-Dawley rats for 4 weeks. Experimental groups were divided into two dietary groups, the cholesterol diet (Cholesterol group) and the cholesterol diet supplemented mixed mushrooms powder (Mushroom group). The concentration of total cholesterol in serum was significantly decreased by 57.4% in mushroom group compared to cholesterol group. The concentration of HDL-cholesterol was significantly increased by 230% in mushroom group compared to cholesterol group. At the same time, atherogenic index was also significantly decreased by 68.4% in mushroom group compared to cholesterol group. The concentration of triglyceride in liver was significantly increased by 50% in mushroom group compared to cholesterol group. However, the concentrations of triglyceride and phospholipid in serum and cholesterol and phospholipid in liver had no significant difference both groups. This study suggested that mixed mushroom powders exert a cholesterole-lowering effect in hyperlipidemic female rats.

        수질환경에서 광섬유 센서의 구리 이온 감지 물질로서 8-Hydroxyquinoline 합성유도체의 광학적 반응 특성 연구

        수질환경에서 구리이온을 검출하기 위하여 광섬유 센서의 구리 이온 감지 물질로 8-Hydroxyquinolin-2carboxaldehyde와 4-Aminoantipyrine으로 합성된 8-Hydroxyquinoline 합성유도체에 대해 광학적 반응 특성을 조사하였다. 실험은 광섬유를 이용한 측정시스템을 이용하여 수용액에 용해된 구리 이온의 농도에 대해 합성유도체에 의한 흡광도와 관계를 평가하기 위하여 수행되었다. 8-Hydroxyquinoline 합성유도체는 수질 환경에서 다양한 금속 이온들 중에서 구리 이온에 대해 노란색에서 붉은색으로 변화되는 높은 발색 현상을 나타냈으며 530 nm의 고유 흡광 파장을 보였다. 제작된 합성유도체의 Cu 이온에 대한 선택성을 평가하기 위하여 Hg 이온의 영향을 조사하였다. Cu와 Hg 이온의 다양한 농도 비 (Cu:Hg ratio 0.05부터 20까지)에서 흡광도를 측정한 결과, Cu 이온의 농도 증가에 따라 530nm에서 흡광도가 증가하는 경향을 보였다. 구리 이온의 고유 흡광 파장인 530 nm에서 합성 유도체에 의한 흡광도 세기는 구리 이온의 로그 농도에 선형으로 비례하였다. 따라서 8-Hydroxyquinoline합성유도체는 광섬유 센서의 구리 이온 감지 물질로 우수한 특성을 보임을 실험적으로 확인할 수 있었다. An 8-hydroxyquinoline compound that was synthesized with 8-hydroxyquinoline-2-carboxaldehyde and 4-aminoantipyrine was investigated for use as the sensing material of a fiber-optic copper ion sensor in an aqueous environment. The experiment was conducted with a fiber-optic measurement system, in order to evaluate the relationship between the absorbance peak and copper ion concentration. The synthesized derivative exhibited a (highly selective) chromogenic phenomenon for copper ions among various metal ions in an aqueous environment and showed a specific absorbance peak at a wavelength of 530 nm for copper ions. The effect of mercury ions was investigated to evaluate the selectivity of the prepared synthesized derivatives toward Cu ions. The absorbance was measured at various concentration ratios of Cu and Hg ions (Cu:Hg ratios from 0.05 to 20), and it was found that the absorbance at 530 nm tended to increase with increasing Cu ion concentration. The experimental results also showed the linear relationship between the logarithmic concentration of copper ions and the specific absorbance peak at a wavelength of 530 nm. These results indicate that the synthesized 8-hydroxyquinoline compound has selectivity for copper ions and can be used as a sensing material for fiber-optic copper ion sensors.

        논문 : 기관평가지표 및 외형적 특성이 연구기관평가 결과 수용성에 미치는 영향

        본 연구에서는 경제·인문사회연구회 소관 연구기관 조직구성원들의 기관평가지표 및 외형적 특성이 연구기관평가 결과 수용성에 미치는 영향을 분석함으로써 기관평가지표의 타당 성 및 평가 수용성의 중요성에 대한 시사점을 얻고자 하였다. 경제·인문사회연구회 소관 연구기관 14개 기관의 136명을 대상으로 BSC 평가지표의 4대 관점 및 기관의 역사성, 기관의 규모, 기관장 특성을 확인적 요인분석 및 영향검증을 실시하였다. 이와 함께 BSC 평가지표의 4대 관점에 대해 종합가중치를 분석하였다. 연구결과 BSC 평가지표는 평가의 수용성에 정(+)의 방향으로 영향을 미쳤고, 외형적 특성 도 정(+)의 방향으로 영향을 미치는 것으로 나타났다. BSC 4대 관점의 가중치 결과는 재무적 관점 지표, 학습과 성장관점 지표, 내부 프로세스관점 지표, 고객관점 지표 순으로 상대적 중 요도가 나타났다. 그러나 인구통계학적 변인은 평가수용성에 영향을 미치지 못하였다. 이와 같은 문헌검토와 실증분석 연구결과를 토대로 기관평가지표의 타당성 및 평가결과 수용성의 중요성이 확인되었다. 그러므로 연구기관의 감독책임을 맡고 있는 정부 관련부처 와 경제·인문사회연구회는 기관평가지표 변경 등에는 연구기관의 의견을 최대한 수렴하는 것이 중요한 요소라 할 수 있다. The importance of institution evaluations and the needs for change of perception on such evaluations are also the case of the research institutions affiliated to the National Research Council for Economic Humanities and Social Sciences. Such circumstances are differences in functions of 23 institutions, difficulties in research performance evaluation as research tends to be long-term and intricate to evaluate, and reluctance to non-financial aspects and lack of perception. In order to analyze the influences of BSC evaluation indicators on the evaluation results acceptance of institutions, external characteristics and the institution evaluation results were additionally selected as control variables. The study results can be summarized as follows. First of all, the results shows that the BSC evaluation indicators has positive influences upon the evaluation results acceptance of institutions. Secondly, the external characteristics has positive influences as well. External characteristics such as history, size, inspection results, director of institutions are in alignment with the evaluation acceptance. Third, it is shown that the other control factors consists of research/business management evaluation results and demographics do not affect the evaluation results acceptance. Fundamentally, any further research should focus on the appropriate BSC evaluation model particularly for research institutions. Specifically, deriving the relations between and among the factors of performance measurements will enable long-term development of institutions by providing them relevant information based on scientific and systematic measurement, and ultimately, contribute to management performance enhancement.

        Physiological Effect of Hamcho Yogurt on Streptozotocin-Induced Diabetic Rats

        함초 추출물 분말을 첨가하여 유산 발효시킨 함초 요구르트를 streptozotocin-유발 당뇨쥐에 투여하여 항당뇨효과 및 콜레스테롤 저하 효과를 규명하기 위하여 3주간 실험하였다. 당뇨 대조군에 비해 함초 요구르트 투여군에서 혈당치 농도 및 식이 섭취량이 현저히 감소하였다. 경구당부하 실험에서도 당뇨 대조군에 비해 함초 요구르트 투여군에서 현저한 혈당상승 억제작용을 나타내었다. 당뇨쥐에 함초 요구르트와 일반 요구르트 섭취에 의해 혈중 총 콜레스테롤 농도 감소와 더불어 동맥경화지수도 감소하였다. 따라서, 함초 요구르트에 의한 혈당치 및 콜레스테롤 농도 감소효과는 고혈당증 및 고 콜레스테롤 혈중에도 유익한 작용을 할 수 있는 기능성식품 개발 가능성이 있음을 시사하였다. The present study was undertaken to investigate the hypoglycemic effect of Hamcho yogurt fermented by lactic acid bacteria containing Hamcho (Salicornia herbacea) extract powder in streptozotocin(STZ)-induced diabetic rats for 3 week period. The Hamcho yogurt given to the STZ-diabetic rats had significant effect in lowering the serum glucose concentrations and food intakes compared to the STZ-diabetic rats. The Hamcho yogurt administration increased the tolerance to oral glucose loading in the STZ-diabetic rats. The Hamcho yogurt and yogurt given to the STZ-diabetic rats were significantly decreased the serum total cholesterol concentration and the atherogenic index. Based on the beneficial effect against the blood glucose and cholesterol levels, we suggest the Hamcho yogurt can be used as functional food for the hypoglycemic and hypocholesterolemic effects.

        위암의 내시경하 생검 조직에서 CD105 (Endoglin), D2- 40, Vasc ular Endothelial Growth Fac tor- A와 D의 발현을 통한 침습성의 예측

        Purpose: CD105 (endoglin) has been shown to be a more useful marker to identify the proliferating endothelium involved in tumor angiogenesis than are the panendothelial markers. The monoclonal antibody D2-40 is a specific lymphatic endothelial marker. Methods: We investigated CD105, lymphatic vessel marker (D2-40), vascular endothelial growth factor (VEGD)-A and the VEGF-D expressions as possible prognostic markers in the endoscopic biopsy tissue of stomach cancer patients. The pre-operative endoscopic biopsies and surgical biopsies from 73 patients were immunostained for CD105, D2-40, VEGF-A and VEGF-D. Positively stained microvessels were counted in densely vascular foci (hot spots) at a ×200 field in each specimen. Results: The microvessel density (MVD) and lymphatic vessel density (LVD), according to the CD105 and D2-40 expressions of the endoscopic biopsies, showed a statistically significant correlation with the surgical biopsies. The MVD via CD105 a showed statistically significant correlation with the histologic differentiation, T-stage, nodal metastasis and stage in the endoscopic biopsies and surgical biopsies, respectively. The lympathic vessel density (LVD) via D2-40 showed a statistically significant correlation with T-stage, nodal metastasis and stage in the endoscopic biopsies. The expressions of VEGF-A and VEGF-D showed a statistically significant correlation with the MVD and LVD. Conclusion: The MVD, as determined by the CD105 expression and the LVD as determined by the D2-40 expression may be useful markers for predicting the invasiveness with using a pre-operative endoscopic biopsy of stomach cancer.

        Proteome Analysis of Drosophila melanogaster Used 2-DE and MALDI- TOF-MS

        초파리는 유전학, 발달 생물학, 행동 유전학, 노화 연구에 이르기까지 수많은 연구에 이용 되어져 왔다. 최근 초파리 전체 유전자의 염기서열이 보고되었으며, 유전자의 기능 분석과 발현 단백질에 대한 연구가 진행되고 있다. 본 연구에서는 야생형 초파리에서 추출한 단백질을 이차원 전기영동을 통해 400여개 이상의 단백질 스폿으로 분리하였으며, 각 스폿을 적출하여 트립신으로 처리하여 얻어진 펩타이드 단편을 MALDI-TOF-MS를 이용한 펩타이드 지문 검색으로 질량을 측정하였다. 측정된 질량을 초파리 데이터 베이스를 이용하여 분리한 단백질을 동정함으로써 59개의 유전자에서 발현되는 65개의 단백질 스폿을 동정하였다. 이러한 결과는 향후의 발생단계, 외부 자극, 노화 등에 관련되는 특이적 단백질 연구의 기초 자료로 활용될 수 있을 것이다. With the completely discovery of the Drosophila genome sequence, the next great challenge is to extract its biological information by systematic expression and to perform functional analysis of the gene. Here we reported a proteome analysis of D. melanogaster with two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption ionization time-of-flight mass spectrometer (MALDI-TOF-MS). The cell extracts of D. melanogaster, $200{\mu}g$ were resolved to more than 400 silver-stained spots by 2-DE. The most abundant protein spots were ranged from 4.0-7.5 of pI and from 15-90 kDa of molecular weight. The excised spots were destained and in-gel digested by trypsin. The masses of the resulting peptide mixtures were measured by MALDI-TOF-MS. Identified proteins were compared with measured peptide mass and a dynamic peptide searching database which is accessible via the internet. The results revealed that identified proteins were produced by 59 genes derived from 65 protein spots.

        Background and Objectives: Salivary gland tumors are relatively rare and constitute 3% to 4% of all head and neck neoplasms. Their relative infrequency, inconsistent classification, and highly variable biologic behavior make some difficulty, but some general features can be drawn regarding the incidence, pathology, and pattern of behavior of the various benign and malignant tumors of salivary glands. The present study aims to provide a clinical characteristics of salivary gland tumors. Materials and Methods: We analyzed retrospectively clinical features of the 138 patients who were treated surgically at Chonbuk National University Hospital from 1992 through 2002. Results: We found 107 benign and 31 malignant tumors. Among the patients, 58 were males and 80 were females. The most common age group was the fifties. The most common site of both benign and malignant was parotid gland in major salivary glands and palatal region in minor salivary glands. The most common presenting symptom was palpable mass in both benign and malignant tumor. Histopathologically, the most common type was pleomorphic adenoma in benign tumor and mucoepidermoid carcinoma in malignant tumor. All cases were treated surgically and the most common postoperative complications was transient facial nerve weakness. Conclusion: 138 cases of salivary gland tumors were presented with respect to their clinical features.
