Biochemical and antimicrobial susceptibility tests were conducted on 40 strains of Pseudomonas aeruginosa originated from diseased chicken submitted for diagnosis to this Institute during 1978-80. An extensive study of the biochemical properties revealed that the tested strains can be identified with Pseudomonas aeruginosa. Antibiogram showed that all the strains were susceptible to gentamicin, colistin and amikacin but resist to nitrofurantoin, trimethoprim+sulfamethoxazole, ampicillin, methicillin and kanamycin, and had varing degreed of resistance to other antimicrobials including carbenicillin, sulfomamides, neomycin, streptomycin, tetracycline and chloramphenicol. Three of the most frequent resitance patterns observed were FM SXT AM ME KM CM TC SM NE SSS Pattern, FM SXT AM ME KM CM TC SM NE SSS CB Pattern and FM SXT AM EKM CM TC SM Pattern, and these resistance patterns contained 72.5% of the tested strains.

The occurrence of inclusion body hepatitis was confirmed for the first time in Korea from chickens submitted for diagnosis to this Institute from a farm located in the vicinity of Anyang. The chickens showed no specific clinical signs except for moderate anemic conditions. At autopsy, livers were swollen and mottled with numerous stellate subcapsular hemorrhages and necrotic foci. Severe nephritis and catarrhal enteritis were also seen. The most notable microscopic changes were seen in the liver. These included eosinophilic intra-nuclear inclusion bodies in the hepatocyes, massive hemorrhages and necrosis and fatty changes in the liver parenchema.
SefA 유전자 PCR에 의한 Salmonella serogroup D1의 특이적 검출

Sal enteritidis thin fimbriae, SEF14, were found to be restricted to the predominantly poultry-associated members of the Salmonella serogroup D1 that are considered as the important pathogens in poultry industry. SefA together with sefB and sefC encode the proteins involved in SEF14 biosynthesis. In order to develop the rapid and specific detection methods for Salmonella serogroup D1, a PCR technique for the amplification of sefA gene was established, and its specificity and sensitivity were investigated with various microorganisms. The bacterial genomic DNA was extracted by colony-picking and rapid boiled-lysate technique. In comparison of Sef I and Sef II primers used in the PCR, Sef I primer for sefA gene of 513bp showed higher specificity than that of Sef II. The established PCR was as sensitive as to detect 1pg of Sal enteritidis DNA. When 73 strains in 28 genera including the reference strains and the field isolates of various Salmonella serotypes, Bacillus subtilis, Bordetella bronchisepdca, E coli, Listeria spp., Micrococcus luteus, Rhodococcus equi, Staphylococcus spp., Streptococcus spp., Vibrio parahemolyticus, Yersinia spp. were studied, the established PCR yielded specifically positive results with only Salmonella serogroup D1. The results suggested that the PCR for sefA gene could be a potential candidate among the specific detection methods for Salmonella serogroup D1.

The antimicrobial drug susceptibility of 65 isolates of Salmonella recovered from chicken submitted for diagnosis at the Poultry Disease Diagnostic Service Laboratory during 1980 to 1981 was determined by the disc diffusion method. All isolates of Salmonella tested were susceptible to ampicillin, colistin, chloramphenicol, gentamicin, kanamycin and trimethoprim-sulfamethoxazole. A higher percentages of them were resistant to streptomycin(70.8%), sulfonamides(53.9%) and tetracycline(41.5%) while only 4.6% was resistant to nitrofurantoin. Among 65 isolated tested, 61.5% were resistant to 2 or more drugs and a total of 9 resistance patterns were observed. The most frequent resistance pattern of them was Sm Su pattern and foll-lowed by Sm Tc pattern and Sm pattern; these resistance patterns comprised more then half(57.0%) of the total isolates.

Serotyping of 48 isolates of Pseudomonas aeruginosa isolated from diseased chicken submitted for diagnosis to this institute was carried out by slide agglutination test with live cells using Difco’s 17 serotyping sera prerared by the Difco Company based on Liu’s serotyping system. Fourty-two isolates out of 48 (87.5%) could be classified into 8 different serotypes and the remaining 6 (12.5%) were untypable. The most frequently isolated serotypes of Pseudomonas aeruginosa in order of prevalence were Type 6(39.6%), Type 3(16.7%), Type 9(10.4%) and Type 4(8.3%). Type 1(4.2%), Type 5(4.2%), Type 10(2.1%) and Type 16(2.1%) were also isolated.

Infectious coryza is one of the important acute respiratory diseases causing a significant egg drop and retarded growth in chicken. An attempt for the isolation of etiologic agent was made by utilizing SPF sentinel birds housed in commercial farms and the results obtained are as follows. Fifteen isolates of Haemophilus gallinarum were tested for their biological and serological characteristics with reference strains, 221 and Modesto and subsequently classified into two serotypes. Of them, isolates immunolocally identical to the standard strains were also selected as vaccine strains for future studies.

It has long been believed for the presence of infectious coryza affecting serious economic loss in domestic poultry industry. However, the etiologic agent has not been isolated until quite recently. From 1979, several strains of Haemophilus-like organism were isolated from chickens with symptoms similar to infectious coryza, and their colonial morphology, growth requirement, biochemical properties and pathogenicity were assessed. In addition, serological properties of the isolates by cross hemagglutination inhibition test was also investigated. The results indicated that all the isolates were identified as Haemophilus gallinarum which had similar characteristics to the reference strains.
