피부세포에서 옥돔 비늘로부터 추출한 펩타이드의 UVB에 대한 산화적 손상 및 광 노화 억제

생체에서 가장 많은 비율로 분포하고 있는 콜라겐 펩타이드는 동물의 뼈와 해양 생물의 비늘에 많이 함유되어 있다. 콜라겐은 동물 생체의 여러 결합조직에서 구조 단백질로 흔하게 발견된다. 또한, 이들은 생물 의료 자재, 제약, 화장품, 식품 및 가죽 산업에 널리 이용된다. 각종 어류 비늘에서 추출된 펩타이드는 UVB 조사에 의해 유도된 피부의 손상 및 광 노화에 대한 보호 효과가 있었다. 그럼에도 불구하고, UVB 조사에 대한 옥돔 비늘 유래의 펩타이드 특성은 명확히 알려져 있지 않다. 이 연구에서는 옥돔 비늘 추출물에서 분리된 1 kDa 이상(HMP)과 1 kDa 이하(LMP)의 펩타이드를 이용하여 UVB 조사에 의해 유도된 피부 손상과 광 노화에 대한 효과를 연구하였다. 이들 펩타이드는 농도 의존적으로 DPPH 라디칼 소거능을 보였으며 LMP는 HaCaT 인간 피부세포에서 UVB 조사에 의해 유도된 세포 지질 과산화 산물인 8-isoprostane 생성을 억제하였다. 그리고 LMP와 HMP는 B16F10 마우스 흑색종 세포에서 tyrosinase 활성 및 melanin 함량을 감소시켰으며 또한 HaCaT 세포에서 UVB로 유도된 elastase 활성을 감소시켰고 matrix metalloproteinase-1의 활성을 감소시켰다. 이러한 결과는 옥돔 비늘에서 유래된 펩타이드가 미백효과, 항산화제 및 광 노화 억제제로서 유용한 물질이 될 것으로 기대된다. Collagen peptides, which are found at high concentrations in the human body, are present in animal bones and the skin of marine organisms, namely, fish scales. Collagen is the most abundant structural protein of various connective tissues in animals. Furthermore, it is widely used in biomedical material, pharmaceutical, cosmetic, food, and leather industries. Peptides extracted from scales of various fish protect against ultraviolet B (UVB)-induced skin damage and photo-aging. However, the protective effects of collagen peptides derived from the scales of Branchiostegus japonicus against UVB exposure are unclear. This study investigated the effects of peptides larger than 1 kDa (high-molecular weight peptides [HMP]) and smaller than 1 kDa (low-molecular weight peptides [LMP]), derived from extracts of B. japonicus scales, against UVB-induced skin damage and photo-aging. These peptides scavenged 1,1-diphenyl-2-picrylhydrazyl radicals in a dose-dependent manner. In UVB-exposed HaCaT human keratinocytes, LMP inhibited 8-isoprostane generation, a marker of cellular lipid peroxidation. The peptides also suppressed the UVB-induced increase in tyrosinase activity and melanin content in B16F10 mouse melanoma cells. In addition, the LMP and HMP treatment suppressed UVB-induced elastase and matrix metalloproteinase-1 activities in the HaCaT cells. These results indicate that peptides derived from B. japonicus scales have antioxidant, antiphoto-aging, and skin-whitening effects.
피부각질형성세포에서 자외선 B 조사가 야기한 염증반응에 대한 괭생이모자반 초음파 에탄올 추출물의 억제 효능 평가

본 연구에서는 UVB 조사에 의해 야기된 HaCaT 세포 내 염증 반응에 대한 괭생이모자반 초음파 에탄올 추출물(USHE)의 억제 효능에 대한 연구를 수행하였다. USHE는 세포독성 없이 UVB 조사에 의해 증가된 세포 내 NO 생성량을 감소시킴으로써, 세포 생존율을 유의적으로 향상시켰다. 또한, USHE 처리는 MAPKs 및 NFκB 신호전달경로의 활성화를 억제함으로써 UVB 조사가 야기한 세포 내 cytokines 및 chemokine의 생성량을 감소시켰다. 뿐만 아니라, USHE는 UVB 조사에 의해 증가된 염증 관련 매개인자인 iNOS 및 COX-2의 발현량을 감소시켰다. 모든 결과로부터 본 연구는 USHE가 UVB 조사에 의해 야기된 염증 반응을 효과적으로 억제시켜 HaCaT 세포를 보호한다는 것을 확인할 수 있었고 부작용을 가지지 않은 천연소재로서 이용가치가 매우 높다는 것을 제시한다. The purpose of this study was to investigate the inhibitory effects of a ultrasonic ethanol extract from Sargassum horneri (USHE) against ultraviolet B (UVB)-induced skin inflammation in HaCaT cells, a human keratinocyte. USHE markedly increased the cell viability by reducing the generation of nitric oxide (NO) in UVB-exposed HaCaT cells. Also, USHE effectively down-regulated the expression levels of iNOS and COX-2 known as inflammatory mediators. Additionally, USHE decreased the production of inflammatory cytokines (interleukin (IL)-6, 8, tumor necrosis factor (TNF)-α and thymic stromal lymphopoietin (TSLP)) and chemokines (macrophage-derived chemokine (MDC) and thymus and activation-regulated chemokine (TARC)) by suppressing the activation of mitogen-activated protein kinase (MAPK) and nuclear factor (NF)κB signaling in UVB-exposed HaCaT cells. Taken together, these results suggest that USHE inhibited the inflammatory response in UVB-exposed HaCaT cells.

Docosahexaenoic acid (DHA), a principal of mackerel-derived fermented fish oil, increases the proliferation of dermal papilla cells (DPCs) via the upregulation of cell cycle-associated proteins such as cyclin D1 and cdc2 p34, and might promote hair-growth. However, the intracellular mechanisms that underlie the action of DHA in the proliferation of DPCs have not been investigated fully. In this study, we addressed the action mechanisms of DHA to trigger the activation of anagen in DPCs. DHA activated β-catenin signaling by the increased phosphorylation at serine 552 and serine 675 as well as the translocation and accumulation of activated β-catenin into the nucleus. In the other hand, DHA inhibited canonical TGF-β/Smad signaling by the decreased phosphorylation of Smad2/3. Taken together, the results indicate that DHA might stimulate anagen signaling via the activation of Wnt/β-catenin pathway, while the inactivation of canonical TGF-β signaling pathway in DPCs.

In this study, we investigated the bioactive substances of the Alcalase hydrolysate obtained from fishery processing by-products in Jeju by measuring bioactivities including radical scavenging acitivty, cytoprotective activity against 2,2-azobis-(2-amidino-propane) dihydrochloride (AAPH), and ACE inhibitory activity. This study is important because of utilization of unused fishery processing by-products in Jeju. The Alcalase hydrolysate was prepared through the hot water extraction and enzymatic hydrolysis, and then further separation of the Alcalase hydrolysate was performed by ultrafiltration using 10 kDa molecular weight cut-off membrane. The Alcalase hydrolysate showed the relatively higher DPPH and peroxyl radical scavenging activity ($IC_{50}$ value; 1.30 mg/ml and 0.888 mg/ml, respectively). Also, the Alcalase hydrolysate showed the ACE inhibitory activity with 1.87 mg/ml of $IC_{50}$ value. These biological activities are increased over 1.2 or 2.5 times through the ultrafiltration of the Alcalase hydrolysate. Therefore, the Alcalase hydrolysate obtained from fishery processing by-products in Jeju and the different molecular weight fractions should be given consideration for food and cosmetics ingredient. Furthermore, this research on the utility of fishery processing by-products might be a useful tool into the industry.
