Sphingomonas sp. KS 301의 Superoxide Dismutase 정제 및 특성

Sphingomonas sp. KS 301, which was isolated from oil contaminated soil, was shown to have five different SODs (SODI, II, III, IV, V) which can be separated by DEAE-Sepharose chromatography, and SOD III was finally purified in this study by ammonium sulfate precipitation, DEAE-Sepharose chromatography, Superose 12 gel filtration and Uno-Q1 ion exchange chromatography. The molecular weight of SOD III was 23 kDa as determined by SDS-PAGE and the apparent molecular weight of the native enzyme was estimated to be approximately 71 kDa by Superose-12 gel filtration chromatography. These data suggest that the purified SOD consists of at least two subunits. The specific activity of the SOD III was higher than Mn type or Fe type SOD of Escherichia coli by 5 fold. To determine the type of SOD III, inhibitory effects of $NaN_{3},\;H_{2}O_{2},\;KCN$ were examined. 10 mM $NaN_{3}$ was able to inhibit 56% of the SOD III activity, which indicates that this SOD is Mn type. The optimum pH of the SOD III was 7.0 and the optimum temperature was $20^{\circ}C$. N-terminal amino acid sequence of purified SOD III was most similar to those of Psudomonase ovalis and Vibrio cholerae among bacteria. 유류오염 토양에서 난분해성 물질인 PAH (polycyclic aromatic hydrocarbon)들을 잘 분해하는 균주 중 SOD (superoxide dismutase) 활성이 높은 균주인 Sphingomonas sp. KS 301의 SOD특성을 알아보기 위하여 Ammonium sulfate 침전, DEAE-Sepharose 크로마토그래피, Superose-12 겔 여과 크로마토그래피, Uno-Q1 이온교환 크로마토그래피를 이용하여 SOD 단백질을 정제하였다. Sphingomonas sp. KS 301은 DEAE-Sepharose 크로마토그래피로 분석한 결과, 기존의 알려진 세균들과는 달리 서로 다른 5가지의 SOD 활성을 가지고 있는 것으로 나타났으며 본 연구에서는 그중 SOD III를 부분 정제하였다. 정제한 SOD III는 Mn type 및 Fe type Escherichia coli SOD와 비교했을 때 비활성도(specific activity)가 5배로 높게 나타났다. SOD III의 분자량은 SDS-PAGE에서는 23 kDa으로 측정되었으며 Superose-12겔 여과 크로마토그래피 후 native 상태의 분자량은 71 kDa으로 정제한 SOD는 3개의 소단위체로 구성되어 있는 것으로 보여진다. 정제한SOD III의 최적 pH는 7.0 이었고 $20^{\circ}C$에서 최적의 활성을 보였다. 또한 SOD의 종류를 알 수 있는 억제물질 $NaN_{3},\;H_{2}O_{2},\;KCN$를 이용한 억제효과를 살펴보았더니 $NaN_{3}$에만 억제되어 Mn type의 SOD임을 알 수 있었다. 또한 이 효소의 아미노 말단의 아미노산 서열은 Psudomonase ovalis 및 Vibrio cholerae의 SOD와 가장 유사하였다.
자기회귀모형에서의 로버스트한 모수 추정방법들에 관한 연구

본 논문에서는 가장 많이 사용되는 시계열 모형중의 하나인 자기회귀모형에서 모수를 추정하는 방법으로 최소 절대 편차 추정법(least absolute deviation estimation)을 포함한 로버스트한 추정방법 (robust estimation)의 사용을 제안하고 모의 실험을 통하여 이러한 방법들을 기존의 최소 제곱 추정 방법과 예측의 관점에서 비교 검토하여 시계열 자료분석에서의 로버스트한 모수 추정 방법의 유효성을 확인해 보고자 한다. In this paper, we study several parameter estimation methods used for autoregressive processes and compare them in view of forecasting. The least square estimation, least absolute deviation estimation, robust estimation are compared through Monte Carlo simulations.
에탄올 발효방법에 따른 음식물류 폐기물의 바이오에탄올 생산성 비교

In order to improve bio-ethanol productivity by various cultivation methods in this paper, the culture modes using food wastes, such as batch culture, high-cell-density fermentation, SSF (simultaneous saccharification and fermentation) by fill & draw, continuous culture by fill & draw were performed and their productivities were compared. SSFs by fill & draw were performed by continuous decompression using 1 L evaporator system, and by 10 L bioreactor without decompression. In addition, the continuous cultures by fill & draw mode using SFW (saccharafied food wastes) medium were performed by changes of 40% culture broth with intervals of 12 h (0.03 h<SUP>-1</SUP>), 6 h (0.07 h<SUP>-1</SUP>), 3 h (0.13 h<SUP>-1</SUP>). Consequently, productivities of bio-ethanol were 2.52 g/L-h and 1.30 g/L-h in batch culture and high- cell-density fermentation, respectively. The productivities of SSF by fill & draw showed 2.24 g/L-h and 2.03 g/L-h in continuous decompression with 1 L evaporator and 10 L bioreactor without decompression, respectively. Also, the productivities in continuous culture by fill & draw modes showed 2.02 g/L-h, 4.07 g/L-h and 6.25 g/L-h by medium change with intervals of 12 h, 6 h, and 3 h, respectively. In conclusion, the highest ethanol productivity was obtained in the continuous culture mode by fill & draw with dilution rate of 0.13 h<SUP>-1</SUP>.

To investigate the design effects of a heat exchanger on a tube type, a tube circuit and a fin pitch, an experimental study on the heat transfer characteristics of finned-tube evaporators was performed. The refrigerant was R-22. A refrigerant loop was established to measure the heat transfer rate, the air heat transfer coefficient. The experimental results showed that the heat transfer characteristics of the evaporators were affected by the design parameters. And the heat transfer rate of the slit fin was better about 25%, compared to those of the louver fin. In the present experimental range, the heat transfer performance with the straight tube circuit was more remarkable than that of the zigzag tube circuit, as seen from temperature variations of the evaporator exit. j -factor on the tube type, the tube circuit and the fin pitch decreased, as increasing Reynolds number.
지열원 히트펌프 시스템에 적용되고 있는 난방용 적산열량계의 신뢰성 평가에 관한 연구

A ground source heat pump system should be equipped with devices to measure the generated heating or cooling heat amount in Korea. Generally, the heat measuring devices have been developed to estimate consumed heat amount in residential or commercial buildings from a central air-conditioning system or a district heating system. In this study, two representive heat measuring devices used for buildings were selected, and the accuracy of them were experimentally estimated at the ground source heat pump operating conditions. The obtained heat amounts from the heat measuring devices were deviated within 4.3% comparing with the precise values calculated from an accredited test facility. Even though the accumulated heat amount values of the heat measuring devices had a small difference comparing with the precise values, the temperatures of heat measuring devices showed greatly different values comparing with the precise temperature. Therefore, it is highly recommended to develop the heat measuring devices which is appropriate for the ground source heat pump systems.

Purposes: This study aims to investigate the policy effect of mandatory application of DRG for 7 disease groups in general and tertiary hospitals. Methodology: As DRG was fully implemented in July 2013, this study compares two periods before and after the change(from July 2012 to June 2013, and from July 2013 to June 2014). The benefit claim data of the National Health Insurance Service was used for the comparison. Target patients were those who visited general or tertiary hospitals between July 2012 to June 2014. For pharmaceutical consumption, Interrupted Time Series (ITS) analysis was used to see the effect of DRG mandatory application. Findings: The number of drugs prescribed per patient and pharmaceutical expenditure both showed significant reduction compared to before the DRG implementation. Practical Implications: This study used 2 sets of 1 year period data from before and after the full implementation of DRG to analyze pharmaceutical consumption. When the comparison data accumulates further, it would be possible to conduct more diverse analysis to assess policy effect and to provide way forward for the future.

This study analyzed the international & korea standards and trend of the certification for the heat metering unit, and the consumer can be supplied with better quality products as necessary for the water source cooling and heating system. This study analyzed the national standards and certification for heat metering unit and discussed current status and future subjects for domestic.
