Three flavonoid glycosides have been isolated from the aerial part of Armoracia rusticana P. (Cruciferae) in Korea and identified by means of spectral analysis as $kaempferol-3-O-{\beta}-D-xylofuranoside(l)$, $kaempferol-3-O-{\beta}-D-galactopyranoside(2)$ and $kaempferol-3-O-{\beta}-D-xylofuranosyl(1\rightarrow2)-b{\beta}-D-galactopyranoside(3)$. When 1 mg/ml of the methanol extract from the aerial part of this plant was added, lipid peroxide formation in the bromobenzene-treated rat liver decreased by 64%. Among the components isolated from title plant, compounds 2 and 3 reduced the formation of lipid peroxide by 16% and 39% respectively at the concentration of ${10}^{-1}$ mg/ml.

Regarding chemical changes in oriental drugs after food processing such as roasting, fermentation, and extrusion, fifty commonly-used medicinal plants were investigated. As a result, Aurantii nobilis Pericarpium (a tangerine peel from Citrus unshu Markovich) showed remarkably different HPLC profiles after being roasted. An increased peak was isolated by repeated chromatography and identified as 5-hydroxymethyl furfral (5-HMF) by means of instrumental analyses. The 5-HMF content of Aurantii nobilis Pericarpoum reached its maximum level after being roasted for 30 min at 225$^{\circ}C$ (49.2 mg/g extract, ca 42 times of increase over untreated control). Although there were no significant changes in in vitro biological activity such as antioxidative, anti-dementia, anti-hypertension, anti-coagulation, or cytotoxicity, before and after roasting process, our results suggested that simple heat treatment might improve the value of the above oriental drug since 5-HMF has been known to possess inhibitory activities toward nitric oxide formation, tyrosinase, and sickling of red blood cells.
해조류 곰피로부터 분리한 Phloroglucinol이 흰쥐의 아세트아미노펜 대사효소활성에 미치는 영향

실험동물에서 곰피로부터 분리한 phloroglucinol은 acetaminophen의 투여로 현저히 증가된 간조직에 있어서 지질과산화의 함량을 억제하였다. Acetaminophen투여에 따른 간 cytochrome P-450, aminopyrine N-demethylase 및 aniline hydroxylase 활성변동은 관찰할 수없었다. 곰피 성분 투여군은 glutathione S-transferase의 활성에서는 대조군의 수준에는 미치지 않으나 효소의 활성이 acetaminophen 단독 투여군보다 현저히 증가되었다. 그리고 간조직중 glutathione의 함량은 phloroglucinol을 전처리군에서 acetaminophen 단독 투여군보다 증가되었다. Glutathione reductase 활성에서는 acetaminophen투여군은 대조군보다 활성이 감소되었으며, 성분으로 전처리한 군은 acetaminophen 단독 투여군보다 증가되었다. 따라서 곰피에서 분리한 페놀성화합물인 phloroglucinol은 acetaminophen투여로 증가되던 지질과산화 함량을 감소시키며, acetaminophen 대사효소활성에서는 glutathione S-transferase의 활성이 증가되어 acetaminophen의 대사를 촉진시키는 것으로 추정된다. Phloroglucinol isolated from Ecklonia stolonifera prevented the acetaminophen (800 mg/kg, i.p.)- induced rats as evidenced by the decreased formation of lipid peroxide. The activities of cytochrome P-450, animopyrine N-demethylase and aniline hydroxylase were not changed by the treatment with phenolic compound as compared with acetaminophen-treated rats. The glutathione S-transferase activity was decreased by the treatment of acetaminophen. This enzyme activity was restored in liver of pretreated-rats given by phloroglucinol. Pretreatment of this compound marked the liver protective activities as recovering the decreased glutathione level and glutathione reductase activity in rats treated with acetaminophen. But γ-glutamylcystein synthetase activity was not changed. The results suggest that the acetaminophen- induced hepatic lipid peroxidation may be reduced by enhancing the activity of glutathione S-transferase.
택사에서 Alisol B 23 - Acetate의 분리 및 함량분석

택사 MeOH 추출물에서 얻은 dichloromethane 분획물을 칼럼 크로마토그라피를 실시하여 alisol B 23-acetate를 순수히 분리하였다. Alisol B 23-acetate를 여러가지 용매와 추출 조건을 달리하여 HPLC를 실시하여 이들의 분리능을 검토한 결과 70% CH₃CN 혼합용매를 사용할 때 분리능이 가장 양호하였다. 국내 각지에서 수집한 택사 33종과 중국 현지에서 수집한 중국택사 5종에 대해 HPLC를 이용하여 함량 분석을 하였다. 국내구입 검체중 지표성분 alisol B 23-acetate의 함량은 0.25%에서 0.67%까지의 범위를 보였으며 평균값 및 표준편차가 0.47±0.11%이었다. 실험에 사용한 검체 중 재배산지에서 직접 구입한 순천시 해룡산 택사가 alisol B 23-acetate의 함량이 0.67%로서 가장 많았다. 중국에서 구입한 택사에는 alisol B 23-acetate가 포함되어 있지 않거나 0.27%에서 0.33% 함유하였다. 이 검체들은 한국에서 구입한 택사와 종이 다르거나 또는 재배조건이 상이하여 이러한 결과가 나온 것으로 추정된다. 택사의 표준화를 위해서는 지표성분 alisol B 23-acetate의 함량은 0.25% 이상으로 규정함이 타당하다고 사료된다. Alismatis Rhizoma is an oriental medicine originated from the rhizome of Alisma orientale or Alisma plang-tago-aquatica var. orientale (Alismataceae). As an standard compound of this plant, alisol B 23-acetate was isolated from the dichloromethane fraction of Alisma orientale and identified by the spectroscopic evidences. A quantitative analysis of alisol B 23-acetate using HPLC method showed that the average content was 0.47± 0.11% in 33 samples throughout the various regions of Korea.

Effects of Houttuynia cordata on the level of lipid peroxide and the enzyme activities of the liver were investigated in bromobenzene-induced rats. Lipid peroxide content in liver was increased by bromobenzene. It was decreased when the methanol extract from the aerial parts of H. cordata was treated to the rat. The methanol extract reduced the activities of aminopyrine N-demethylase and aniline hydroxylase that increased by bromobenzene, however did not affect glutathione S-transferase activity. The methanol extract recovered the activity of epoxide hydrolase activity that decreased significantly by bromobenzene. We suggest that under our experimental conditions the extract might play an important play in the prevention of hepatotoxicity by reduction of aminopyrine N-demethylase and aniline hydroxylase activities as well as enhancement of epoxide hydrolase activity. Six phenolic compounds have been isolated from H. cordata and identified by means of spectral analysis as protocatechuic acid, quercetin, apigenin, afzelin, hyperoside and quercitrin.
식용 및 약용식물 추출물과 화합물이 흰쥐의 간장조직에서 과산화지질 생성에 미치는 영향

31종의 식물추출물과 10종의 화합물들을 대상으로, 흰쥐의 간장조직 중 과산화지질 생성 효과를 시험관내에서 살펴보았다. 추출물 중 달래의 70% 아세톤 추출물은 10^(-2), 10^(-1), 1㎎/ml 농도에서 각각 3%, 17%, 33%, 동일한 농도에서 근대 아세톤 추출물은 27%, 46%, 58%, 갓의 아세톤 추출물은 38%, 49%, 62% 그리고 고추잎의 메타놀 추출물은 30%, 45%, 56% 과산화지질 억제작용을 나타내었다. 10종의 화합물에 대한 과산화지질생성에 미치는 영향으로서, quercetin, quercitrin 및 isorhamnetin은 10^(-6)㎎/ml에서 각각 21%, 4%와 14% 그리고 10^(-2)㎎/ml 농도에서는 각각 45%, 16% 및 28%의 지질과산화 억제작용이 관찰되었다. We have studied the effect of 31 extracts and 10 components from some edible and medicinal plants on the formation of lipid peroxide in the liver homogenate of rat in vitro. The 70% acetone extracts of Allium tuberosum, Beta vulgaris var. cicla and Brassica juncea var. integrifolia, and methanol extract of Capsicum annuum decreased the formation of lipid peroxide by 33%, 58%, 62% and 56% at the concentration of 1㎎/ml, respectively. And these four extracts inhibited the lipid peroxidation at the concentration of 10^(-1)㎎/ml by 17%, 46%, 49% and 45% respectively. Among the component tested, quercetin, quercitrin and isorhamnetin reduced the formation of lipid peroxide by 45%, 15% and 28% respectively at the concentration of 10^(-2)㎎/ml.
