        Effects of Phytase Supplementation on the Performance of Broiler Chickens Fed Maize and Wheat Based Diets with Different Levels of Non-phytate Phosphorus

        An experiment was conducted to evaluate the effects of phytase supplementation on the growth performance, nutrients utilization and bone mineralization in broiler chickens. Day-old broiler chicks (n=480) were equally devided into eight treatment groups and fed maize or wheat based isocaloric, isonitrogenous and isocalcium diets having two non phytate phosphorus (NPP) concentrations (0.50% and 0.30%) and two phytase levels (0 and 500 phytase units/kg diet) in a 42 days growth trial. Maize based dietary treatments were MC (NPP 0.50%, MN (NPP 0.30%), MNP (MN+500 units of phytase) and MCP (MC+500 units of phytase), whereas wheat based experimental diets were WC (NPP 0.50%), WN (NPP 0.30%), WNP (WN+500 units of phytase) and WCP (WC+500 units of phytase). The NPP levels were maintained by dicalcium phosphate. Reduction in dietary NPP depressed live weight gain and feed intake and increased feed conversion ratio (FCR). Phytase supplementation to low NPP (0.30%) diets significantly (p<0.05) improved the growth performances of broilers. The supplementation to low NPP diets allowed complete, safe and economic replacement of dietary inorganic P (dicalcium phosphate) to reduce feed cost per kg live weight gain of broilers. Reduction in dietary NPP did not affect retention of nutrients except phosphorus (P) but had a significant (p<0.05) depression in tibia ash and minerals (Ca, P) concentration in serum and tibia ash. Phytase supplementation at low NPP level was effective (p<0.05) in improving the retention of dry matter, Ca and P and Ca and P concentration in serum and tibia ash. However, the supplementation was not effective at high level of NPP (0.50%). There were no significant (p>0.05) differences in carcass quality among dietary treatments. The response of phytase was greater in low NPP and maize based diets as compared with high NPP and wheat based diets, respectively. The results show that phytase supplementation to low NPP (0.30%) diets improved the growth performance, relative retention of nutrients (N, Ca and P) and minerals (Ca, P) status of blood and bone in broiler chickens, with a better efficacy in maize based diets.

        Probing the Thermodynamic and Magnetic Properties of UV-B-Emitting GdAlO3 Phosphors by ESR and Optical Techniques

        <P>GdAlO3 phosphor samples have been prepared by a solution combustion method. X-ray diffraction analysis revealed that the GdAlO3 phosphor stabilized in orthorhombic phase. The optical absorption spectrum showed bands due to Gd3+ ions as well as defect level in the host. The excitation spectrum exhibited a dominant band with maximum at 273 nm. Upon excitation at 273 nm, ultraviolet-B (UV-B) emission with maximum at 312 nm was observed from the undoped sample, being attributed to P-6(7/2) -> S-8(7/2) transition of Gd3+ ion. The UV-B emission was enhanced at liquid-nitrogen temperature (77 K), most probably due to increased ground-state population according to the Boltzmann distribution law. Electron spin resonance spectra of samples at both room and liquid-nitrogen temperature exhibited resonance signals at g ae 2, attributed to Gd3+ ions arranged with octahedral symmetry linked via oxygen bridges. Various magnetic and thermodynamic parameters for the phosphor system were evaluated by standard procedures. Correlation of these values with the optical properties suggested that the GdAlO3 phosphor can be effectively used as an UV-B light source for industrial applications at room temperature.</P>

        Production Performance and Carcass Characteristics of Malpura and Mutton Synthetic Lambs Fed Low and High Energy Rations in a Semiarid Region of India

        Forty eight lambs (24 each of Malpura and Mutton synthetic breeds) were weaned at 90 days of age and divided into two groups. One group of 12 lambs from each of the two breeds was maintained on low energy (52% TDN) and the other group of 12 on high energy (58% TDN) feedlot ration until 6 of the lambs attained 20 kg and the other 6 attained 25 kg live weight in each of the two groups. Daily feed intake and weekly body weights were recorded. Conventional metabolism trials were conducted on the two breeds and the two rations. The growth rate was found to be higher for lambs on high energy ration (p<0.01). The growth rate was also higher for higher target weight groups (p<0.01). The breed did not significantly affect the live weight gains. Mutton synthetic lambs required less number of days to reach 20 kg live weights but took more time to reach 25 kg target weight as compared to Malpura lambs (p<0.01). The lambs of both the breeds reached 25 kg live weight earlier on high energy than those on low energy ration (p<0.01). The dry matter intake, irrespective of breed, was significantly higher (4.57% or 93.4 g per kgW$^{0.75}$) on low energy than that on high energy ration (4.20% or 87.2 g per kg W$^{0.75}$). The digestibility coefficients of all the nutrients excepting crude fibre were significantly higher on high energy diet irrespective of the breed. Lambs on low energy ration, however, digested the crude fibre more than those on the high energy ration (p<0.05). Although there were no significant differences in the intakes of nitrogen, calcium and phosphorus, the balance of nitrogen was higher on high energy ration. The DCP and TDN values were 9.70 and 52.76% for low energy and 9.89 and 57.68 % for high energy ration, respectively. The dressing percentages on live weight basis were 50.2 on low and 51.6 on high energy ration, 50.5 at 20kg and 51.3 at 25 kg slaughter weight and 51.0 in Malpura and 50.8 in Mutton synthetic lambs, respectively. The percent of bones in the carcass was higher on low energy ration in Mutton synthetic lambs at 20 kg slaughter weight than others. It was concluded that the performance of the lambs in respect of mutton production was significantly better on high energy ration fed upto 25 kg slaughter weight with no or little breed differences.

        Cytokines Expression and Nitric Oxide Production under Induced Infection to Salmonella Typhimurium in Chicken Lines Divergently Selected for Cutaneous Hypersensitivity

        In the present study, the impact of Salmonella Typhimurium on cell-mediated immunity (CMI) was investigated in 5 week-old immuno divergent broiler lines selected for the high and low response to phytohemagglutinin-P. The immune response was assessed in peripheral-blood mononuclear cells (PBMCs) induced with Salmonella Typhimurium at different time intervals (0 h, 0.5 h, 2 h, 4 h, 6 h, 12 h and 24 h). The differential mRNA expression patterns of IFN-${\gamma}$, IL-2 and iNOS were evaluated by quantitative real time PCR. In-vitro production of nitric oxide (NO) was also estimated in the culture supernatant and correlated with iNOS mRNA expression. Present study showed higher production of NO in the high cell-mediated line (HCMI) as compared to the low cell-mediated line (LCMI) upon stimulation with Salmonella Typhimurium. Correspondingly, higher mRNA expression of iNOS and IFN-${\gamma}$ were observed in high response birds (HCMI); but IL-2 was down regulated in this line compared to the low response birds (LCMI). Significantly (p<0.05) higher expression of iNOS, IFN-${\gamma}$ and higher production of NO in high line indicated that the selection for PHA-P response might be employed for increasing the immune competence against Salmonella Typhimurium in chicken flocks.

        Changes in Plasma Levels of Inhibin and Follicle Stimulating Hormone in Buffaloes Superovulated with eCG

        The present study was undertaken to investigate the effect of stimulation of follicular development with eCG on the peripheral levels of inhibin and FSH in Murrah buffaloes. Estrus was synchronized in five normally cycling females by insertion of Crestar (Intervet, Boxmeer, Holland) implants for nine days. Estradiol valerate was administered i.m. on the day of implant insertion. On the 10th day of the induced estrous cycle a single dose of 3000 IU eCG (Folligon, Intervet, Boxmeer, Holland) was given, followed by treatment with 25 mg of $PGF_2$ alpha (Lutalyse, Upjohn, Belgium) 48 h later. Blood samples were obtained during the induced estrus, on cycle day 10 (luteal phase), at the superovulatory estrus (43 h after PGF) and during the periovulatory period (64 h after PGF). Ultrasonography was done daily to monitor follicular development. Plasma concentrations of inhibin and FSH were determined by specific radioimmunoassays. Differences between $mean{\pm}SEM$ values of different phases of the cycle were compared by ANOVA. The mean number of small (2-5 mm), medium (6-9 mm) and large (>10 mm) follicles observed two days after eCG treatment and on the day of superovulatory estrus was $2.8{\pm}0.31$, $5.2{\pm}0.30$ and $1.4{\pm}0.09$ and $1.9{\pm}0.21$, $2.8{\pm}0.40$ and $5.0{\pm}0.83$, respectively. The mean number of ovulations was $3.6{\pm}0.37$ and the mean number of unovulated follicles was $6.1{\pm}0.47$. Most of the follicles >10 mm in diameter had ovulated (72%). The mean ${\pm}SEM $ of plasma inhibin concentration $(2584.15{\pm}17.92pg/ml)$ during the superovulatory estrus was significantly higher $(p{\leq}0.05)$ than during the induced estrus $(749.87{\pm}17.29pg/ml)$, the luteal phase $(1099.54{\pm}24.98pg/ml)$ and periovulatory period $(1682.71{\pm}29.88pg/ml)$, respectively. $Mean{\pm}SEM$ plasma FSH concentration during the induced estrus $(10.35{\pm}0.41ng/ml)$ was not different from that during the superovulatory estrus $(8.52{\pm}0.39ng/ml)$, but was significantly higher $(p{\leq}0.05)$ than during the luteal phase $(2.81{\pm}0.42ng/ml)$ and periovulatory period $(5.7{\pm}0.28ng/ml)$. These data indicate that treatment with eCG in buffaloes for inducing superovulation results in a significant elevation in plasma inhibin levels and a decrease in plasma FSH levels during the superovulatory estrus. Thus, we suggest that the elevated plasma inhibin coming from fully developed follicles continued for a long time which results in inhibition of FSH leading to poor ovulation in the remaining follicles, which may be the cause of suboptimal superovulatory response.

        Monocrotophos Induces the Expression of Xenobiotic Metabolizing Cytochrome P450s (CYP2C8 and CYP3A4) and Neurotoxicity in Human Brain Cells

        <P>Expression of various cytochrome P450s (CYPs) in mammalian brain cells is well documented. However, such studies are hampered in neural/glial cells of human origin due to nonavailability of human brain cells. To address this issue, we investigated the expression and inducibility of CYP2C8 and CYP3A4 and their responsiveness against cyclophosphamide (CPA) and organophosphorus pesticide monocrotophos (MCP), a known developmental neurotoxicant in human neural (SH-SY5Y) and glial (U373-MG) cell lines. CPA induced significant expression of CYP2C8 and CYP3A4 in both types of cells in a time-dependent manner. Neural cell line exhibited relatively higher constitutive and inducible expression of CYPs than the glial cell line. MCP exposure alone could not induce the significant expression of CYPs, whereas the cells preexposed to CPA showed a significant response to MCP. Similar to the case of CPA induced expressions, neural cells were found to be more vulnerable than glial cells. Our data indicate differential expressions of CYPs in cultured human neural and glial cell lines. The findings were synchronized with protein ligand docking studies, which showed a significant modulatory capacity of MCP by strong interaction with CYP regulators-CAR and PXR. Similarly, the known CYP inducer CPA has also shown significant high docking scores with the two studied CYP regulators. We also observed a significant induction in reactive oxygen species (ROS), lipid peroxides (LPO), micronucleus (MN), chromosomal aberration (CA), and reduction in reduced glutathione (GSH) and catalase following the exposure of MCP. Moreover, the expressions of apoptotic markers such as caspase-3, caspase-9, Bax, and p53 were significantly upregulated, whereas the levels of antiapoptotic marker, Bcl2, was downregulated after the exposure of MCP in both cell lines. These findings confirm the involvement of ROS-mediated oxidative stress, which subsequently triggers apoptosis pathways in both human neural (SH-SY5Y) and glial (U373-MG) cell lines following the exposure of MCP.</P>

        Oxidative Stress and Antioxidant Status during Transition Period in Dairy Cows

        The study was conducted on 20 Holstein X Sahiwal cross bred dairy cows, with an average milk production of $2,752{\pm}113.79$ liters in $284{\pm}5.75$ days during a single lactation, that were divided in to two groups of 10 animals. We investigated the oxidative stress and antioxidant status during the transition period in dairy cows. In this study, plasma level of MDA was considered as an indicator of lipid peroxidation and SOD, catalase, GSH and GSHPx as antioxidants. The lipid peroxidation was significantly (p<0.001) higher in cows during early lactation as compared to the cows in advanced pregnancy. A significant positive correlation (r = +0.831, p<0.01) was determined between MDA and catalase in early lactating cows. In early lactating cows, blood glutathione was significantly lower than in advanced pregnant cows. However, early lactating cows showed non-significant negative correlation for all antioxidant enzymes with lipid peroxidation. In conclusion, dairy cows seemed to have more oxidative stress and low antioxidant defense during early lactation or just after parturition than advanced pregnant cows, and this appears to be the reason for their increased susceptibility to production diseases (e.g. mastitis, metritis, retention of fetal membranes etc.) and other health problems.

        Antioxidants Supplementation on Acid Base Balance during Heat Stress in Goats

        The effects of vitamin C and vitamin E with selenium on acid-base balance and some stress hormones were evaluated during heat stress in goats. Goats, 1.5 years of age, were divided into control, heat stress and antioxidant treatment groups 1, 2 and 3. Except for the control, all groups were exposed to a temperature of $40{\pm}2^{\circ}C$ with a relative humidity of 30% for 5 h/d for 21 days in a psychrometric chamber. Rectal temperature and respiratory rates were recorded daily post exposure. Blood samples were collected on every 3rd day for estimation of plasma vitamins C and E, total antioxidant activity and hormones, and separate blood samples were taken to estimate acid-base status. The rectal temperature and respiratory rates were increased (p<0.05) in the heat stress group only. Except for pH and $pO_2$, which were increased significantly (p<0.05) other parameters of acid-base balance such as $pCO_2$, $HCO_3^-$, $TCO_2$, BEb, BEcef, PCV and Hb were significantly decreased (p<0.05) in the heat stress group. An improvement in acid-base status was noted in the antioxidant supplemented groups. Prolactin and cortisol levels were significantly (p<0.05) higher and free T3 and T4 levels were significantly (p<0.05) lower in the heat stress group. Levels of prolactin and cortisol were decreased and free T3 and T4 were increased in antioxidant treatment groups. Different levels of antioxidant supplementation resulted in similar protection against heat stress.

        A Study on Effect of Carrying FecB Gene on Body Weight in Garole and Garole×Malpura Sheep

        High prolificacy in Garole sheep is due to existence of FecB mutation in an autosomal gene, bone morphogenetic protein receptor. The mutation enhances ovulation rate and in turn litter size in Garole sheep. Garole sires were crossed with non-prolific Malpura ewes with the aim to introduce prolificacy into Garole${\times}$Malpura (G${\times}$M) crosses through FecB introgression programme. In the present study, the effect of carrying booroola allele on litter size and live body weight was analyzed. The average litter size at birth was found to be 1.87 and 1.48 in the Garole and the G${\times}$M crosses, respectively. At weaning, 6-month, 9-month and 12-month of age, body weights were not affected by the presence of booroola allele (p>0.05); however, a significant effect (p<0.05) was found on body weight at birth in G${\times}$M crosses. In Garole sheep, no significant effect of FecB was observed on live weights in any age group. The interaction between the genetic group and the FecB genotype was also found to be non-significant.
