      • Pseudorabies Virus의 Major Capsid Protein 유전자의 클론닝과 Baculovirus Vector System에 의한 발현

        Pseudorabies is caused by Pseudorabies virus (PRV: Aujeszky's disease virus) of Herpesviridae that is characterized by 100 to 150nm in size with a linear double-stranded DNA molecule with of approximately 90 × 10 exp (6)Da. This disease affects most of domestic animals such as swine, cattle, dog, sheep, cat, chicken, etc. causing high mortality and economic losses. In swine, young piglets show high mortality and pregnant sows, reproductive failures. However the adult swine reveals no clinical signs in general. But they become a carrier state and play an importnat role for propagation of the disease. In this study, the nucleotide sequence of major casid protein gene of PRV, Yangsan strain isolated from the diseased swine in Korea was analyzed, and the recombinant MCP was produced by expression of the MCP gene in Sf-9 cell using baculovirus transfer vector system. As result, in BamHI digestion, MCP gene locus of PRV YS strain showed different from that of Indiana S strain. The patterns of enzyme mapping were also found to be unidentical each other. The sequence of the MCP gene partially analyzed showed 98.09% identity to Indiana S strain. The expression of MCP in Sf-9 cell cotransfected by pVLMCP-44 baculovirus expression vector was characterized by Southern blot hybridization, immunofluoresent and immunocytochemical tests, SDS-PAGE and Western blotting. The rMCP with M.W. 142kDa was most effectively expressed in Sf-9 cells at the 3-4th days post inoculation of the recombinant baculovirus by 2 moi.

      • Classical Swine Fever (Hog cholera) Virus 약독순화주 (Suri 주)의 gp55 Gene 염기서열 분석

        An attenuated classical swine fever virus (CSFV), Suri strain, is a variant derived from a vaccine virus, LOM strain. This study was performed to elucidate the molecular biologcal properties of CSFV Suri strain, and to obtain the basic data for molecular epidemiological approaches for the disease. The truncated form of gp55 gene without the C-terminal trasmembrane domain, in size of 1,023bp, was amplified by RT-PCR and sequenced by dye terminator cyclic sequencing method, and inserted into BamHI site of pAcGP67B baculovirus vector, establishing a cloned pAcHEG plasmid. By the nucleotide sequences determined, 341 amino acid sequences were predicted. As compard the nucleotide and amino acid sequences of gp55 of Suri with the various CSFV, Suri strain showed the high homology over 99.1% with ALD and LOM strains, but comparably the lower homology with Alfort and Brescia. In comparison of amino acid sequence in variable domain of gp55 protein, the similar tendency of homology was observed. In hydrophobicity analysis, all of four CSFV strains revealed the analogous patterns of hydrophobicity. The numbers and locations of N-glycosylation site and cysteine residuse in gp55 were analyzed, those of Suri strain being coincident with ALD and LOM strains. The results suggest that gp55 in Suri strain has the high similarity to those in ALD and LOM strains in terms of the nucleotide and amino acid sequences and the functional properties of gp55 protein.

      • 최근 한국의 가축질병 방제연구 동향

        최근 20년 동안 우리 나라의 축산은 급속히 근대화되고 있으며, 현재 축산물의 생산량은 농업 총생산액의 약 25%를 차지할 정도로 성장하였다. 1989년말 축산통계에 의하면 한우가 1,536,000두, 젖소가 515,000두, 돼지가 4,801,000두 그리고 닭이 61,689,000수로 기록되고 있다. 이와 같은 가축수의 증가는 역시 경제발전과정상의 개인소득 증대로 육류의 소비가 계속 증가되고 있기 때문에 가능한 것으로 생각된다. 현재 우리 나라에서는 75종 이상의 가축전염병 발생이 보고되고 있으며 농장경영 형태에 따라 질병발생 양상도 달라지고 있다. 이러한 가축전염병을 효과적으로 방제하려면 예방의학의 중요성이 선진국 수준으로 인식되어야 하겠으며 또한 각종 질병의 진단법과 백신개발 및 개량이 대학이나 연구소에서 부단히 연구되어야 할 것이다. 국내에서 개발 보급되고 있는 진단제는 약 14종이며 백신류는 30종 정도로 이러한 생물학적 제제는 가축질병 방제에 없어서는 안될 중요한 핵심기술제품이라 하겠다. 저자는 이 보고서에서 가축위생 향상을 위하여 한국에서 연구 개발되고 있는 각종 기존진단법, 단클론성 항체이용 진단법, 유전자분석진단법과 또한 각종 백신의 연구 동향을 간략하게 소개하고저 하였다. During the last two decades livestock farming in the Republic of Korea has been rapidly modernized and intensified, and current livestock products contribute to approximately 25% of the total agricultural estimated value In 1989, livestock population recorded 1,536,000 Korean native cattle, 515,000 dairy cattle, 4,801,000 pigs and 61,689,000 chickens(Table 1). Such an increase in livestock population has been generated to meet the growing demand for animal products as a result of economic growth and increased income per capita. In Koreas. as many as 75 infectious diseases of livestock have been reported to occur, showing a variety of outbreak patterns according to the change in livestock farming. It has become widely acknowledged that the control of livestock diseases by means of preventive medicine is of paramount importance in domestic livestock industries as is the case in many other countries. In attempts to control major diseases of economic importance, many researches on development or improvement of diagnostic methods and vaccines are still being actively conducted in veterinary colleges and institutions, Currently total of 30 different vaccines and 14 kinds of diagnostic reagents are being used nationwide for control and prevention of the diseases. This report, which is admittedly superficial, focused on (1) the diagnosis of infectious diseases by use of conventional reagents, monoclonal antibodies and gene characterization techniques, and (2 the trend of vaccine development to enhance animal health in Korea.

      • Unity를 이용한 아케이드 미니 게임 제작

        본 논문은 Unity3D 게임엔진을 이용하여 쉽게 접할 수 있는 아케이드 형식의 미니게임을 제작하는데 목적이 있다. 우리는 미니 게임을 통하여 창의력, 반사 신경을 향상시키도록 하였고, 또한 일상생활에서 볼 수 있는 애완동물이라는 소재를 사용하여, 게임에 접근하기 어려운 사람들에게 친근감을 주게 하는 것을 특징으로 한다. 또한 랭킹 시스템을 도입하여 유저들에게 경쟁을 유도하게 하여 유저들의 게임 참가율을 증대하도록 하였다. 본 논문에서는 Unity3D 엔진을 이용하여, 게임의 개발시간을 단축하여 개발 비용을 줄일 수 있으면서도 게임의 질은 떨어지지 않도록 하였다

        In this study, a survey was conducted to investigate the environmental degradation factors of specific areas in residential apartment underground structures an`d the required performance attributes for waterproofing material. Based on the survey, it was concluded that joint displacement of concrete is the highest cause for problems, and the required performance was determined to high wet surface adhesion performance for waterproofing materials.

      • 고분자막 이온선택성 전극에 관한 연구

        The effect of composition and structure of ion-exchanger, plasticizer, and membrane thickness on the selectivity and the response characteristics of electrode potential was studied. The nitration-selective electrode was prepared using ion-exchanger, plasticizer, and PVC as supporter. As the active material of electrode, the quaternary ammonium nitrate salts like Aliquat 336N, tetracotylammonium nitrate (TOAN), and tetradodecylamnonium nitrate(TDDAN) were used. The Nernst linear response ranges and the detection limits of those electrodes were measured and the effect of interfering anions were anions were discussed. It was concluded that the best characteristics of the Aliquat 336N-PVC membrane electrode were obtained with the ion-exchanger concentration level of 6.5-9.1 percent by weight. The optimal membrane composition was 9.09wt.% of ion-exchanger, 30.95wt.% of PVC, 60.6wt.% of plasticizer (DBP), 0.5㎜ of thickness.

      • Trioctylamine에 의한 유기산의 추출

        For elucidation of various parameter's effect on the reactive extraction of organic acidstrioctylamine(TOA)-diluents system, the association model, equilibrium constants and extraction factors(loading) were investigated . The organic acids rosed were propionic. succinic, masonic, and benzoic acid. Extractant was tertiary amine, TOA and diluents were butylaeohol, hexane, and benzene. It was concluded that the loading of monobasic acid of propionic acid were higher than that of dibasic acids of oxalic, and malonic acid with extranctant TOA. In the extraction of monobasic acid the extraction factor was not affected by the initial concentration of extractant amine, but in the extraction of dibasic acids the extraction factor was getting better with decreasing the initial concentration of amine. There was a strong effect of diluents upon the extractien factor, and the order of these factor was as shown below: xylene<Benzene<butylalcohol<choloroform<methylisobutylketone.

      • Baculovirus Vector System에 의해 발현된 재조합 Pseudorabies Virus Major Capsid Protein의 면역원성

        The recombinant pseudorabies virus major capsid protein (rMCP) was produced by expression of the MCP gene in Sf-9 cell using baculovirus transfer vector system. Following evaluation of the immunochemical properties of the rMCP, the immunogenicity of the recombinant subunit protiens were investigated in guinea pig and swine to obtain the preliminary guide line for the subunit vaccine using rMCP and gP50. It was proved that ultrasonication and 30% ammonium sulfate was most efficient to concentrate and purify the protein. The rMCP was safe in mice, guinea pigs and piglets. In guinea pigs, rMCP mixed with various adjuvants induced substantial degree of serum neutralizing antibody titers, but revealed incomplete protecivity against challenge. In swine, the combination of rMCP and gP50 showed the higher serum neutralizing antibody titers and cellular immune responses than rMCP alone. However, the protectivity was lower in comparison with the commercial gI-deleted inactivated vaccine. We expect these results to contribute to characterization of MCP gene of Korean isolate of PRV and to ultilize as preliminary information for prodution and evaluation of PRV recombinant subunit vaccines.

      • 함침액막에서 운반체의 구조에 따른 트립토판의 플럭스

        The relations between membrane potential and the flux of solutions, which were developed from the molecular structure of quaternary ammonium salts as carrier, have been analyzed for the counter transport system of tryptophan and nitrate anion across the supported liquid membrane. The liquid membrane was prepared by dissolving quaternary ammonium nitrates in toluene. The experimental results showed that the flux of tryptophan increased with increasing the carbon chain length of the alkyl group of carrier in the range of ??∼??, but in case of TDDAN, it decreased because of large membrane potential. The flux of tryptophan with the membrane structure of carrier increased in the order : Aliquat 336N, TOAN, TDDAN, and TDAN.

      • 국내분리 Aujeszky's disease virus의 실험적 감염 자돈에 대한 바이러스학적 연구 : Pathogenecity, excretion, distribution and immunogenicity of virus

        To investigate the etiology, pathogenicity and virological properties of NYJ-1-87 strain of Aujeszky's disease virus (ADV) that was isolated from the diseased piglet in Korea, the virus at 10^(6.0)TCID_50/0.1㎖ was inoculated intranasally and subcutaneously into 30 to 35 days-old piglets. Results obtained through the experiments were summarized as follows. 1. Ten of the infected piglets were clinically observed for 15 days. On the 2nd day post-inoculation(pi), the signs of pyrexia, anorexia and convulsion were noted. On the 4th 7th days pi, nervous signs of incoordination and intermittent spasm were shown in the most of piglets, and one out of 5 piglets infected intranasally was died with severe nervous signs at the 7th day pi. The signs became relieved on the 8th day pi and all of remainder were completely recovered on the 13th to 14th days pi. 2. In hematological study, prominent decrease in the number of total leukocyte and lymphocyte was shown in the ADV-infected piglets on the 6th day pi. On the 8th day pi, the cell numbers were slightly increased and returned to normal level on the 10th day pi. 3. Viral excretion of the ADV-inoculated piglets was examined by swabbing of nasal and oral cavities, and rectal feces. During the periods of the 3rd to11th days pi, the virus was excreted intermittently from nasal and oral cavities, and rectal feces. The nasal excretions were shown the highest virus concentration of 10^(5.2)TCID_50/0.1㎖ at the 5th day pi. 4. Recovery of the inoculated virus from various organs of the piglets that were died or experimentally slaughtered was attempted, and the virus was isolated from the tissues of brain and tonsil by the cultured cell-inoculation method. The highest recovery rate was noted in the tonsil. By indirect immunofluorescence antibody assay using ADV-monoclonal antibody, the viral antigens were detected in tissues of spleen and liver as brain and tonsil on the 7th to 9th days pi. The virus was not isolated from blood and the tissues of lung and kidney throughout the experiments. 5. Titers of virus neutralizing antibody in the piglets experimentally infected with ADV became increased after the 6th to 9th days pi in both of intranasal and subcutaneous inoculation showing the highest titers of 64 to 128 on the 29th day pi. When the antibody levels were measured by radial immunodiffusion enzyme assay, the reactive diameter was enlarged to be positive after the 4th to 6th days pi in both of intranasal and subcutaneous inoculation showing the largest diameter of 13 to 14mm on the 29th day pi.
