        태권도 돌려차기 시 기술 수준에 따른 몸통, 골반, 지지 다리의 운동학적 변인 비교

        이 연구의 목적은 태권도 돌려차기 시 기술 수준에 따른 몸통, 골반, 지지 다리의 운동학적 변인을 비교 분석하는 데 있다. 본 연구를 위해 블랙 벨트 소지자로서 기술 수준에 따라 중급자10명, 상급자 9명이 실험에 참여하였다. 3차원 동작 분석 시스템을 사용하여 골반, 몸통, 지지다리의 방향각을 산출하였다. Start, Toeoff, MKF, Max, Impact, Min에서 그룹 간의 평균 방향각을 비교하기 위하여 독립표본 t-검정을 하였다. MKF, Impact, Min에서 골반의 좌측 기울어짐 각도가 그룹 간의 유의한 차이가 나타났다. Start와 Max에서 지지 다리의 엉덩관절 굴곡 각도와 MKF에서 지지 다리의 엉덩관절 외전 각도 사이에 유의한 차이가 있었다. 이상의 결과를 볼 때, 상급자들은 푸쉬단계에서 엉덩관절의 굴곡 각도가 증가 되었고, MKF에서는 외전 각도가 감소하였다. 또한, 스트라이크 단계에서 골반의 좌측 기울어짐 각도는 감소하였다. The study purpose was to investigate skill level differences on trunk, pelvis, and supporting leg kinematics in a Taekwondo roundhouse kick. 10 intermediate and nine expert male black-belt holders executed roundhouse kicks at a target. Orientation angles of the pelvis, trunk, and supporting -thigh, -shank, and -foot were obtained through a three-dimensional video motion analysis. Orientation angles at Start, Toeoff, Maximum Knee Flexion(MKF), Impact, Maximum angle(Max), and Minimum angle(Min) were compared between groups using independent t-tests(p<.05). Significant differences between groups were found in the pelvis left lateral tilted position at MKF, Impact, and Min; significant differences were also found in the supporting hip joint flexed position at Start and Max, and abducted position at MKF. It was concluded that higher performers increased the hip joint flexion angle in the Push phase, decreased abduction angle at MKF, and decreased the pelvis left tilted angle in the Strike phase.

        The Effect of pH and Various Cations on the GTP Hydrolysis of Rice Heterotrimeric G-protein α Subunit Expressed in Escherichia Coli

        Previously, we reported the biochemical properties of RGAl that is expressed in Escherichia coli(Seo et al., 1997). The activities of RGAl that hydrolyzes and binds guanine nucleotide were dependent on the MgCl_(2) concentration. The steady state rate constant (k_(cat)) for GTP hydrolysis of RGAl at 2 mM MgCl_(2) was 0.0075±0.0001 min^(-1). Here, we examined the effects of pH and cations on the GTPase activity. The optimum pH at 2mM MgCl_(2) was approximately 6.0; whereas, the pH at 2 mM NH_(4)Cl was approximately 4.0. The result from the cation dependence on the GTPase (guanosine 5`-triphosphatase) activity of RGAl under the same condition showed that the GTP hydrolysis rate (k_(cat)=0.0353 min^(-1)) under the condition of 2mM NH_(4)Cl at pH 4.0 was the highest. It corresponded to about 3.24-fold of the k_(cat) value of 0.0109 min^(-1) in the presence of 2mM MgCl_(2) at pH 6.0.

        정상상피세포(HaCat)와 자궁경부 암세포(SiHa)에서 GeneFishing^(TM) PCR technique을 이용한 유전자 발현의 변화

        목적 : 본 실험의 목적은 정상상피 세포와 자궁경부 암세포 사이에서 유전자의 발현 차이를 조사하였다. 연구 방법 : 정상상피 세포(HaCat)와 자궁암 세포(SiHa)를 사용하였으며, 두 세포 간에 유전자 발현 차이를 GeneFish^(TM) PCR을 이용하여 알아보았으며, BLAST serach를 통해 분석하였다. 결과 : 정상상피 세포와 자궁암 세포 비교 결과, 자궁암 세포에서 S1-2-2와 S5-1을 포함한 25개의 유전자가 발현이 증가하였고, 24개의 유전자가 감소하였다. 결론 : GeneFishing^(TM) PCR기법은 유전자의 발현 변화를 확인하는데 있어서 아주 민감하고 효과적인 방법이다. 우리는 정상상피 세포와 자궁경부 암세포에서 다르게 발현하는 유전자를 찾을 수 있었고, 앞으로는, 종양의 발생과 진행과정에 관여하는 유전자를 더 탐지하고 해당 유전자의 기능을 연구할 필요가 있다고 생각된다. Objective : The purpose of this study is investigated the differentially expressed genes between normal and cervical cancer cell line. Methods : We used normal human keratinocyte (HaCaT) as a control and HPV-16 positive cervical cancer (SiHa) cell line. Two cell lines were studied differential expressed genes by using GeneFishing^(TM) PCR and analyzed with BLAST search. Results : As compared with normal, cervical cancer cell line was showed 25 up-regulated genes including the S1-2-2, S5-1 and 24 down-regulated genes. Conclusion : GeneFish^(TM) PCR test is very sensitive and effective method for detection of changed gene expression. We could search differentially expressed genes between normal and cervical cancer cell line. In the future, we need to research various genes function to participate in the process of tumor development and progression.

        사료원료 공급량 및 스크류 회전속도를 달리하여 제조한 배합사료가 넙치(Paralichthys olivaceus)의 성장 및 체조성에 미치는 영향

        The aim of this study was to investigate the effect of diet extruder conditions, such as feed-loading rate and screw speed, on growth performance and biochemical responses in olive flounder Paralichthys olivaceus. Over 8 wks, we used four identical diets (triplicated per treatment) with differing ratios of feed-loading rate (kg/h):screw speed (rpm/min) in a laboratory-scaled twin-screw extruder of 50:640, 80:640, 120:640, and 80:400, designated as EP1, EP2, EP3, and EP4, respectively. Screw speed impacted the buoyancy of experimental diets. Diets produced at a screw speed of 640 rpm/min floated for > 24 hrs, whereas those produced at a speed of 400 rpm/min sank between 10 s and 5 min. Fish that were fed EP1 and EP4 diets grew significantly faster than those fed EP2 and EP3 diets. Fish fed EP1 diets ate and gained weight most efficiently among treatments, a result that is likely to be related to feed-loading rate, i.e., ingredients extruded at a low feed-loading rate may have more time to cook in the pre-conditioner of the extruder. A cooked diet may be easier to digest in fish. Fish fed EP4 diets also showed significant weight gain, as compared to those fed EP2 and EP3 diets. However, we found no differences among treatments in proximate compositions of dorsal muscle, liver, and viscera of fish. Our results suggest that extruder conditions, may influence feed quality, impacting feed efficiency and growth of fish.

        Development of multiplexed analysis for the photocatalytic activities of nanoparticles in aqueous suspension

        <P>A multiplexed assay technique to measure the photocatalytic activity (PCA) of nanoparticles (NPs) in aqueous suspension was developed based on the observation of TiO<SUB>2</SUB> NPs-photocatalytic oxidation rate of NADH by monitoring the fluorescence intensities. 96 sample solutions of a small volume (<150 μL) could be assayed in a single run without separation of NPs within 15 min.       • 人蔘根腐病에 關한 硏究(第Ⅵ報) : 人蔘根腐敗를 일으키는 Pseudomonas spp에 關한 形態, 培養 및 生理·生化學的 特性에 關하여 On the bacterial form, cultural, physiological and biochemical characteristics of Pseudomonas spp isolated from root rot of ginseng

        We observed that a severe root rot of bacterial type occurred and became widespread in 1973 affecting the ginseng cultivating plants around Kimpo-gun of Kyunggi province. We isolated three time 17 strains bacteria from a sample of diseased ginseng root found at Galsan-ri, Wolgot-myun, Kimpo-gun Kyunggi province. All of them were cultured to the selective medium of Pseudomonas spp. Among them, 6 strains were positive at the selective medium. In the preliminary examination their morphological, cultural and staining characteristics were similar to each other in those respects. Therefore, the 6 strains were selected to be used in a more detailed study with respect to it's morphological, cultural, physiological and biochemical characteristics. Among of them, 5 strains (No 19, 12, 4, 6, 20) were classified as a Genus Pseudomonas by the method of Bergey's Manual of Determinative Bacteriology, and also were classified as a species P.fluorescens, especially resemble to phytopathogenic P.marginalis.

      • 잔디의 立枯病菌[Curvularia geniculata(Tracy & Earle) Boedjin]의 營養要求性에 關한 硏究

        The nutritional requirements of Curvularia blight pathogens of turf (Agrostis cannina L,) causing by Curvularia geniculate were examined of various nutritional sources. The experimental results obtained summarized as follows. 1. The effects of optimum hydrogen lon concentration on the growth of this fungus in Czapek's solution medium were 6.0 and the mycelial growth was also observed in the range of pH 3.0∼9.0 2. The effects of optimum temperature on the growth of the fungus in Czapek's solution medium was 25-30℃. 3. In the effects of carbon sources on the growth of this fungus, sucrose showed the best result, and followed glucose, soluble starch, and xylose. Polyalcohol and organic acid as carbon sources affect the growth of this fungus only slightly. 4. In the effects of nitrogen sources on the growth of this fungus, ammonium nitrate and ammonium citrate affect the best results, and followed ammonium phosphate, ammonium oxalate, ammonium tartrate, and potassium nitrate. Ammonium nitrite affect the growth of this fungus only slightly. In generally, NH_4-N and NO_3-N showed the best results, whereas NO_2-N was less effective in all the nitrogens. 5. The effects of various chemicals in the formation of inhibition zone were larger in Dexon and Thiram 75, and followed PTM-V, Difolatan and Thiuramin, whereas Bla-s was not effective.

      • 人蔘根 腐病發生에 미치는 病原과 線蟲의 關聯性

        4 roots of 2 year old ginseng were planted in a small pot trateing with fungal pathogens, Fusarium solani, nematodes isolated from the ginseng (Panax ginseng C.A. Meyer) cultured soil and the two mixed inoculum respectively. Rate of infestation and nematodes distributed around the harvested host plant after 40 days was investigated. To obtained results were as follows: Each group was treated with F. solani, nematodes and mixed inoculum showed higher disease index by infestation from 3.3 to 3.8 while control group was healthy showing lower disease index of 1.2. The nematodes identified were 4 genera in control group and F. Solani inoculated group. 11 genera of nematodes including Xiphinema spp were distributed in the nematodes inoculated pot, and their numbers were higher in the population. Especially Radophalus spp, Tylenchulus spp, Meloidogyne spp, Xiphinema spp and Ditylenchus spp were numerous in their population density, but Belonolimus spp and Hemicyciophora spp were lower. Fourteen genera of nematodes were distributed in the mixed inoculation pot than only the nematodes treated group. and also the population was higher in the mixed inoculation group. The more populated genera is number were Ditylenchus spp, Paratylenchus spp, Paratylenchus spp. Hemicycliophora spp, Meloidogyne spp, Helicotylenchus spp and Tylenchorhynchus spp, but Hirschmanniella spp was less in the population density.
