Impaired nucleolus biogenesis during pre-implantation development of mouse nuclear transfer embryos

핵인의 가장 널리 알려진 기능 중에 하나는 리보솜의 합성을 수행한다는 것이다. 접합 유전자의 수정 이후에 핵인의 재구축은 포유 동물의 착상 전 배아의 성공적인 발달을 위해 필수 적인 요소이다. 그러나 핵 이식 배아에서 핵인의 발달이 지연 되거나 비정상적으로 발달하는 양상을 보인다. 이번 연구에서는 쥐의 난자를 이용해 정자미세주입법과 단위발생 그리고 난구세포를 이용해 핵이식을 수행했을 각각의 경우에 착상 전 배아가 발달하는 동안 핵인의 발달과 핵인의 전구체 형성 그리고 핵인의 미세구조와 핵인을 조절하는 부위의 활성화 정도를 살펴보고 비교해보기 위한 실험을 수행하였다. 결과를 살펴보면 난구세포를 이용해 핵이식을 수행한 난자에서는 정자미세주입법과 단위발생 시킨 난자보다 전핵당 전구체의 개수가 더 많은 결과를 보였다. 그리고 난구세포를 이용하여 핵이식을 수행한 난자의 4cell 단계에서 손상된 핵인이 활성화 되는 것을 살펴볼수 있었고 미세구조 단계에서부터 질이 덜 좋은 핵인이 발달하고 또 한 염색체상에서 핵인을 조절하는 부위의 활성화 정도가 떨어지는 것을 살펴볼 수 있었다. 그리고 난구세포를 이용하여 핵이식을 수행한 난자의 경우 정자미세 주입법이나 단위 발생 난자에 비해 배반포 형성 비율이 억제되는 것을 살펴볼 수 있었다. 결론적으로 핵이식 난자에서 손상된 핵인의 발달이 수행되며 핵이식 된 난자의 착상 전 발달 능력이 현저히 떨어지는 데 영향을 미치는 요인중에 하나가 핵이식 된 난자에서 손상된 핵인이 형성되기 때문이라고 설명할 수 있다. The most well-known function of nucleolus is ribosome biogenesis. The re-establishment of functional nucleoli after zygotic genome activation is essential for successful development of mammalian pre-implantation embryos. However, in somatic cell nuclear transfer (SCNT) embryos, the nucleoli always showed delayed developed or malfunction. In current study, mouse intracytoplasmic sperm injection (ICSI), pathenogenetic activated (PA) and cumulus cell nuclear transfer (CCNT) embryos were generated to compare the nucleoli development during pre-implantation development, including nucleolar precursor bodies formation, nucleolar ultrastructure and active nucleolar organizer region (NOR) numbers. The results showed that, CCNT embryos had more nucleolar precursor bodies (NPBs) per pronucleus than ICSI and PA embryos at pronuclear stage (2.82 v.s. 0.98 and 1.19). At 4-cell stage, the CCNT embryos showed impaired nucleolar activity, including inferior nucleolar development at ultrastructure level and less active NORs on chromosomes (6.68 v.s. 7.44 and 7.56). Moreover, CCNT embryos showed significant lower blastocyst rate than ICSI and PA embryos (62.0% v.s. 97.1% and 98.0%). The results demonstrated that mouse CCNT embryos had impaired nucleolar development, which may account for their low pre-implantation developmental competence.

The increasing prevalence of chronic kidney disease (CKD) is a major global public health concern. Despite the complicated pathogenesis of CKD, renal fibrosis represents the most common pathological condition, comprised of progressive accumulation of extracellular matrix in the diseased kidney. Over the last several decades, tremendous progress in understanding the mechanism of renal fibrosis has been achieved, and corresponding potential therapeutic strategies targeting fibrosis-related signaling pathways are emerging. Importantly, extracellular vesicles (EVs) contribute significantly to renal inflammation and fibrosis by mediating cellular communication. Increasing evidence suggests the potential of EV-based therapy in renal inflammation and fibrosis, which may represent a future direction for CKD therapy.

The extraction and solvent partition of the leaves ofEucalyptus citriodora, and repeated column chromatography for n-BuOH fraction yielded a new flavonoid glycoside, citrioside C (1),along with three known flavonoid glycosides (2-4). The latter wereidentified with kaempferol-3-O-β-D-glucopyranosyl (12)-α-L-rhamnoside(2), kaempferol-3-O-α-L-rhamnoside (3), and quercetin-3-O-α-Lrhamnoside(4). Their chemical structures were identified on thebasis of spectroscopic data analyses including NMR, MS, UV, andIR. All constitutents were isolated for the first time from the leavesof Eucalyptus citriodora. The potential antivirus activity of all theisolated compounds was evaluated. Compound 4 showed potentantiviral activity against respiratory syncytial virus with 50%inhibition concentration (IC50) value of 1.9 μg/mL and selectiveindex value of 9.8.

<P><B>Abstract</B></P><P> A number of germ cell-specific transcription factors essential for ovarian formation and folliculogenesis have been identified and studied. However, the role of these factors during early embryonic development has been poorly explored. In the present study, we investigated the role of SEBOX, a maternal homeobox transcription factor, during early embryonic development in porcine parthenotes. mRNA for <I>SEBOX</I> is preferentially expressed in oocytes, and expression persists until embryonic genome activation (EGA). Knockdown of SEBOX by siRNA disrupted early embryonic development, but not oocyte maturation. Many maternal genes essential for early embryonic development were upregulated in SEBOX-depleted embryos. Moreover, some pluripotency-associated genes, including <I>SOX2</I> and <I>NANOG</I>, were upregulated when SEBOX was knocked down. Therefore, our data demonstrate that SEBOX is required for early embryonic development in pigs and appears to regulate the degradation of maternal transcripts and the expression of pluripotency genes.</P>
A Secret Sharing Scheme Based on AES

In order to solve the key setting difficulty and the key security problem in the file encryption, key distributed storage technology may be a proper choice to help improve the safety of the key. In the paper, a novel secret sharing scheme is proposed by AES encryption algorithm for file confidentiality, dynamic key generation mechanism to generate keys, multi-secret-sharing ideas on key pre-treatment, using Shamir threshold scheme for secret dispersed storage. Finally, a few tests are carried out and the test results suggest that the efficiency of the whole scheme is good.
