      • Association between Pax8-PPARγ1 Rearrangement and Follicular Thyroid Cancer: a Meta-Analysis

        Li, Hang-Yu,Xie, Zhi-Hao,Xu, Cong-Hui,Pu, Mei-Ling,Chen, Zi-Yan,Yu, Miao,Wang, Heng-Shu,Zhou, Chen-Ming,Pu, Chao-Yu,Liu, Wei Asian Pacific Journal of Cancer Prevention 2016 Asian Pacific journal of cancer prevention Vol.17 No.9

Background: Pax8 and peroxisome proliferator-activated receptor gamma 1 gene (Pax8-$PPAR{\gamma}1$) are important factors in tumors. Several studies have suggested that follicular thyroid cancer may arise from Pax8- $PPAR{\gamma}1$ rearrangement. In order to have a better understanding of the association between Pax8-$PPAR{\gamma}1$ rearrangement and follicular thyroid cancer, we conducted the presenmt meta-analysis. Materials and Methods: The information was extracted from PubMed, EMBASE and Web of Science. Statistic analysis was performed with Stata12.0 software. Odds ratios (ORs) were calculated using a fixed-effects model. We also performed heterogeneity and publication bias analyses. Results: Nine studies including 198 follicular thyroid cancer patients and 268 controls were considered eligible. The frequency of Pax8-$PPAR{\gamma}1$ rearrangement was significantly higher in the follicular thyroid cancer group than in the control group, with a pooled OR of 6.63 (95%CI=3.50-12.7). In addition, through subgroup analysis, the OR between Pax8-$PPAR{\gamma}1$ rearrangement and follicular thyroid cancer was 6.04 (95%CI = 3.18-11.5) when using benign tumor tissues as controls. The OR for the method subgroup was 9.99 (95% CI =4.86-20.5) in the RT-PCR. Conclusions: The final results demonstrated that Pax8-$PPAR{\gamma}1$ rearrangement has significant association with follicular thyroid cancer.

The Beneficial Effects of Dietary Astragali Radix Are Related to the Regulation of Gut Microbiota and Its Metabolites

        Zhi Chai,Zhen-Yu Li,Ling Wang,Lan Yang,Xiao-Ling Cheng,Xue-Mei Qin 한국식품영양과학회 2024 Journal of medicinal food Vol.27 No.1

Astragali Radix (AR) or its extract has been used as an herbal medicine and dietary supplement in China,Europe, and the United States. The gut microbiota could provide new insights for exploring dietary supplements’ underlyingmechanism on organisms. However, no reports have focused on the regulatory effect of AR on the gut microbiota as a dietarysupplement. In this study, healthy ICR mice of either sex were divided into AR and control (CON) groups and given AR waterextract (4.55 mg/kg$day-1) or saline by gavage for 14 days, respectively. Then 16S rRNA gene sequencing and ultra-highperformanceliquid chromatography quadrupole time-of-flight tandem mass spectrometry-based fecal metabolomics wereintegrated to investigate the benefits of dietary AR. Weighted gene coexpression network analysis was also introduced toinvestigate the metabolites with highly synergistic changes. AR supplementation influenced the structure of intestinal microflora, especially enriching short-chain fatty acid-producing bacteria g_Coprobacillus, g_Prevotella, and g_Parabacteroides. AR also significantly altered the fecal metabolome, mainly related to amino acid metabolism, nucleotidemetabolism, and bile acid (BA) metabolism. Moreover, the increased secondary BAs and BA-sulfates might closely relate tointestinal microflora. These findings provide valuable insights for future research of dietary AR as a functional food.

      • Elevated Circulating CD19+ Lymphocytes Predict Survival Advantage in Patients with Gastric Cancer

Yu, Qi-Ming,Yu, Chuan-Ding,Ling, Zhi-Qiang Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.5

Background: Circulating lymphocyte subsets reflect the immunological status and might therefore be a prognostic indicator in cancer patients. Our aim was to evaluate the clinical significance of circulating lymphocyte subset in gastric cancer (GC) cases. Methods: A retrospective study on a prevalent cohort of 846 GC patients hospitalized at Hospital from Aug 2006 to Jul 2010 was conducted. We calculated the patient's disease free survival (DFS) after first hospital admission, and hazard ratios (HR) from the Cox proportional hazards model. Results: Our findings indicated a significantly decreased percentage of CD3+, and CD8+ cells, a significantly increased proportion of $CD4^+$, $CD19^+$, $CD44^+$, $CD25^+$, NK cells, and an increased $CD4^+/CD8^+$ ratio in GC patients as compared with healthy controls (all P < 0.05). Alteration of lymphocyte subsets was positively correlated with sex, age, smoking, tumor stage and distant metastasis of GC patients (all P<0.05). Follow-up analysis indicated significantly higher DFS for patients with high circulating $CD19^+$ lymphocytes compared to those with low $CD19^+$ lymphocytes (P=0.037), with $CD19^+$ showing an important cutoff of $7.91{\pm}2.98%$ Conclusion: Circulating lymphocyte subsets in GC patients are significantly changed, and elevated CD19+ cells may predict a favorable survival.

Random vibration analysis of train-slab track-bridge coupling system under earthquakes

        Zhi-ping Zeng,Xian-feng He,Yan-gang Zhao,Zhi-wu Yu,Ling-kun Chen,Wen-tao Xu,Ping Lou 국제구조공학회 2015 Structural Engineering and Mechanics, An Int'l Jou Vol.54 No.5

This study aimed to investigate the random vibration characteristic of train-slab track-bridge interaction system subjected to both track irregularities and earthquakes by use of pseudo-excitation method (PEM). Each vehicle subsystem was modeled by multibody dynamics. A three-dimensional rail-slabgirder-pier finite element model was created to simulate slab track and bridge subsystem. The equations of motion for the entire system were established based on the constraint condition of no jump between wheel and rail. The random load vectors of equations of motion were formulated by transforming track irregularities and seismic accelerations into a series of deterministic pseudo-excitations according to their respective power spectral density (PSD) functions by means of PEM. The time-dependent PSDs of random vibration responses of the system were obtained by step-by-step integration method, and the corresponding extreme values were estimated based on the first-passage failure criterion. As a case study, an ICE3 high-speed train passing a fifteen-span simply supported girder bridge simultaneously excited by track irregularities and earthquakes is presented. The evaluated extreme values and the PSD characteristic of the random vibration responses of bridge and train are analyzed, and the influences of train speed and track irregularities (without earthquakes) on the random vibration characteristic of bridge and train are discussed.

Realgar bioleaching solution suppress ras excessive activation by increasing ROS in Caenorhabditis elegans

        Zhi, De Juan,Feng, Na,Liu, Dong Ling,Hou, Rong Li,Wang, Mei Zu,Ding, Xiao Xia,Li, Hong Yu 대한약학회 2014 Archives of Pharmacal Research Vol.37 No.3

Although realgar bioleaching solution (RBS) has been proved to be a potential candidate for cancer therapy, the mechanisms of RBS anticancer are still far from being completely understood. Dosed with RBS in C. elegans, the multivulva phenotype resulting from oncogenic ras gain-of-function was inhibited in a dose dependent manner. It could be abrogated by concurrent treatment C. elegans with RBS and the radical scavenger DMSO. However, RBS could not induce DAF-16 nuclear translocation in TJ356 or the increase of HSP 16.2 expression in CL2070, which both could be aroused visible GFP fluorescent variation to represent for oxidative stress generation. Treatment C. elegans with superoxide anion generator paraquat, similar results were also obtained. Our results indicated that RBS suppress excessive activated ras by increasing reactive oxygen species (ROS) in C. elegans. Secondly, ROS induced by RBS significantly accumulated on a higher level inC. elegans with amutational ras than that with wild ras, thus leading to oxidative stress on ras gain-of-function background rather than on normal ras context. Our results firstly demonstrated that using C. elegans as amodel organism for evaluating prooxidant drug candidates for cancer therapy.

Age-stage, two-sex life tables of the maple poison moth (Lymantria nebulosa Wileman) at different ambient temperatures

        Lei Ling,Huang Hui Jing,Yi Jin Yu,Qiu Lin,Ding Wen Bin,He Hua Liang,Yi Tu Yong,Li You Zhi,Xue Jin 한국곤충학회 2021 Entomological Research Vol.51 No.4

The maple poison moth (MPM), Lymantria nebulosa (Wileman), is a Lepidopteran defoliator and one of the most important pests of Liquidambar formosana Hance. The objective of this study was to determine the effects of temperature on the population growth and reproduction of this species. MPM were raised in artificial climate chambers in a laboratory at 21 ± 1°C, 24 ± 1°C, 27 ± 1°C, 30 ± 1°C, 33 ± 1°C and 36 ± 1°C, under a relative humidity (RH) of 70% ± 10% and a 14 L:10D photoperiod, and age-stage, two-sex life tables complied for insects at each temperature. The developmental duration, adult preoviposition period and overall preoviposition period decreased with temperature in a time-dependent manner. The growth curves of the different temperature groups overlapped considerably. The highest larval survival rate (80%) was recorded at 30 ± 1°C. At 36°C only a few eggs hatched and the first instar could not complete the first molt. We consequently regard 36°C as the lethal temperature threshold for this species. Life expectancy decreased with age under all six temperatures. The endogenous growth rate was significantly lower at 21 ± 1°C than at the other temperatures, and was highest in 30 ± 1°C group. The net growth rate of the 30 ± 1°C group was also significantly higher than others. Reproductive parameters were highest at 27 ± 1°C and 30 ± 1°C. Although environmental factors absent from the laboratory environment may limit the generality of our results, they nonetheless provide a theoretical basis for developing integrated control methods for MPM in the wild.

Curcumin Alleviates the Functional Gastrointestinal Disorders of Mice In Vivo

        Jing Yu,Wen-hua Xu,Wei Sun,Zhi-li Guo,Xiao-ling Yu 한국식품영양과학회 2017 Journal of medicinal food Vol.20 No.12

Curcumin is a natural polyphenol extracted from the turmeric rhizome, which has a wide range of biological activities, but until now the effects of curcumin on the gastrointestinal peristalsis have not been fully understood. In vivo study, we observed the effects of curcumin on gastric emptying and intestinal propulsion rates of mice in normal state and in delayed state by atropine (ATR) or nitric oxide precursor L-arginine (L-Arg). An in vitro study explored the direct effects of curcumin on the intestinal contractility, but were studied through measuring spontaneous contraction of isolated jejunum of mice. Our results showed that intragastric administration of curcumin (200 mg/kg/day) for 10–20 days significantly improved gastric emptying and intestinal propulsion rates of mice delayed by ATR. Moreover, intragastric administration of curcumin (200 mg/kg/day) for 15 days also significantly improved mice gastric emptying and intestinal propulsion rates delayed by L-Arg. There was no significant effect on normal gastrointestinal propulsion of mice after intragastric administration of curcumin (200 mg/kg/day) for 1–20 days. When normal isolated jejunum of mice were incubated with curcumin in vitro, the amplitude of the spontaneous contractile waves of jejunum was reduced in a concentration-dependent manner. Moreover, curcumin reduced the amplitude of the contractile waves of jejunum in both contracted and relaxed state induced by acetylcholine or ATR individually. Taken together, our results suggest that curcumin has quite different effects on gastrointestinal peristalsis in vivo and in vitro. Moderate dose of curcumin by intragastric administration for more than 10 days can alleviate the functional gastrointestinal disorders of mice, but cannot affect normal gastrointestinal propulsion.

      • Expression of Endogenous Hypoxia Markers in Vulvar Squamous Cell Carcinoma

        Li, Yu-Zhu,Li, Shu-Ling,Li, Xia,Wang, Li-Jie,Wang, Jiu-Ling,Xu, Jia-Wen,Wu, Zhi-Hong,Gong, Li,Zhang, Xiao-Dan Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.8

Objective: To investigate the expression of endogenous hypoxia-related markers identified as being involved in vulvar squamous cell carcinoma (VSCC). Methods: We performed immunohistochemical staining of hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$), glucose transporter-1 (GLUT-1), carbonic anhydrase 9 (CA-9) and vascular endothelial growth factor (VEGF), on tissue sections of 25 VSCC patients, 10 vulvar intraepithelial neoplasia (VIN) patients and 12 healthy controls. Results: HIF-$1{\alpha}$ expression was found in all sections, with no significant difference between controls, VIN and VSCC sections (all P<0.05). Glut-1 expression was found in 25% of control, 90% of VIN and 100% of VSCC sections. A significant difference between control and VIN or VSCC was observed (all P<0.05), while no difference was found between VIN and VSCC sections (P>0.05). CA-9 expression was negative in control sections, but it was found in 30% of VIN sections and 52% of VSCC sections with strong staining. Similarly, CA-9 expression also showed obvious differences between controls and VIN or VSCC sections (all P<0.05). However, there was no significant difference between VIN and VSCC (P>0.05). There were only 25% of control sections with weak VEGF expression, while strong staining was found in about 60% of VIN sections and 25% of VSCC sections (all P<0.05). In addition, a difference was also found between VIN and VSCC sections (P<0.05). Conclusion: Expression of endogenous hypoxia markers (HIF-$1{\alpha}$, GLUT-1, CA-9 and VEGF) might be involved in the malignant progression of VSCC.
