β<sub>2</sub>-adrenergic receptor (β<sub>2</sub>-AR) was expressed efficiently using Bac-to-Bac Baculovirus Expression System in Sf9 cells as a bio-recognition element for multianalyte screening of β-agonist residues in pork. Sf9 cells were selected as the expression system, and codon optimization of wild-type nucleic acid sequence and time-dependent screening of expression conditions were then carried out for enhancing expression level and biological activity. Under optimum conditions of multiplicity of infection (MOI) = 5 and 48 h post transfection, the protein yield was up to 1.23 mg/ml. After purification by chromatographic techniques, the purified recombinant protein was applied to develop a direct competitive enzyme-linked receptor assay (ELRA) and the efficiency and reliability of the assay was determined. The IC50 values of clenbuterol, salbutamol, and ractopamine were 28.36, 50.70, and 59.57 μg/l, and clenbuterol showed 47.61% and 55.94% cross-reactivities with ractopamine and salbutamol, respectively. The limit of detection (LOD) was 3.2 μg/l and the relevant recoveries in pork samples were in the range of 73.0-91.2%, 69.4-84.6%, and 63.7-80.2%, respectively. The results showed that it had better performance compared with other present nonradioactive receptor-based assays, indicating that the genetically modified β<sub>2</sub>-AR would have great application potential in detection of β-agonist residues.

Fangchinoline (Fan) inhibits cell proliferation and induces apoptosis in several cancer cell lines. The effects of Fan on cell growth and proliferation in breast cancer cells remain to be elucidated. Here, we show that Fan inhibited cell proliferation in the MDA-MB-231 breast cancer cell line through suppression of the AKT/Gsk-3beta/cyclin D1 signaling pathway. Furthermore, Fan induced apoptosis by increasing the expression of Bax (relative to Bcl-2), active caspase 3 and cytochrome-c. Fan significantly inhibited cell proliferation of MDA-MB-231 cells in a concentration and time dependent manner as determined by MTT assay. Flow cytometry analysis demonstrated that Fan treatment of MDA-MB-231 cells resulted in cell cycle arrest at the G1 phase, which correlated with apparent downregulation of both mRNA and protein levels of both PCNA and cyclin D1. Further analysis demonstrated that Fan decreased the phosphorylation of AKT and GSK-3beta. In addition, Fan up-regulated active caspase3, cytochrome-c protein levels and the ratio of Bax/Bcl-2, accompanied by apoptosis. Taken together, these results suggest that Fan is a potential natural product for the treatment of breast cancer.

Tumor necrosis factor (TNF)-alpha-induced protein 8 (TNFAIP8 or TIPE) is a recently identified protein considered to be associated with carcinogenesis. To investigate its expression pattern in pancreatic cancer patients and to analyse its correlation with clinicopathological significance and the expression levels of epithelial growth factor receptor (EGFR), immunohistochemistry was performed to detect the TNFAIP8 and EGFR proteins in pancreatic cancers, pancreatitis tissues, and healthy controls. The results showed stronger staining of TNFAIP8 protein in pancreatic cancer tissues compared with normal pancreas tissue. Furthermore, in 56 patients with pancreatic cancer, the expression levels of TNFAIP8 in patients with low tumor stage was higher than that with high tumor stage, and correlated with tumor staging and lymph node metastasis (P<0.05). Furthermore, TNFAIP8 expression positively correlated with EGFR levels (r=0.671135, P<0.05). These results indicate that TNFAIP8 may play important roles in the progression of pancreatic cancer.

In order to obtain and explore selenide composed of selenium and polysaccharide, three parameters were selected to optimize the synthesis process of selenium-Euryale ferox Salisb. polysaccharide (Se-ESPS) by Box-Behnken design. Furthermore, ESPS-B1 separated from ESPS was selenitized to Se-ESPS-B1 by the optimal synthesis process, then the characterization, and antioxidant activity in vitro of Se-ESPS-B1 were explored. The result manifested that the Se content of Se-ESPS was (2.915 ± 0.03) mg/g according to the optimal synthesis process of Se-ESPS (reaction time at 5 h, reaction temperature at 81 ℃, weight ratio of Na2SeO3 to ESPS at 0.9 g/g). A series of detection results indicated that the characterizations of Se-ESPS-B1 were apparently distinguished from that of ESPS-B1. Moreover, the antioxidant experiments in vitro demonstrated that Se-ESPS-B1 could exert antioxidant activity by scavenging DPPH, ABTS,·OH, and increasing reduction ability. In conclusion, the synthesis process is an effective approach to harvest seleniumpolysaccharide, and Se-ESPS-B1 may be utilized as a potential antioxidant or selenium nutritional supplement.

By using the extended version of the Su-Schrieffer-Heeger (SSH) tight-binding model and a non-adiabatic dynamic evolution method, we present a study of the dynamic process of oppositely charged carriers in a metal/two coupled poly(p-phenylene vinylene) chains/metal structure. Electrons and holes can be injected into the polymer chains from metal electrodes by applying voltage biases. The behavior of the injected charged carriers depends closely on the electric field and the interchain coupling. The moderate intensity of electric field is in favor of forming stable wave packets, and the charge carriers localized in wave packets occur to transport, hopping, collide and recombine. With increasing the electric field, part of the carriers can be ejected into the metal electrodes.
A FEEDBACK-DRIVEN BUBBLE G24.136+00.436: A POSSIBLE SITE OF TRIGGERED STAR FORMATION

<P>We present a multi-wavelength study of the IR bubble G24.136+00.436. The J = 1-0 observations of (CO)-C-12, (CO)-C-13, and (CO)-O-18 were carried out with the Purple Mountain Observatory 13.7 m telescope. Molecular gas with a velocity of 94.8 km s(-1) is found prominently in the southeast of the bubble, shaped as a shell with a total mass of similar to 2 x 10(4) M-circle dot. It was likely assembled during the expansion of the bubble. The expanding shell consists of six dense cores, whose dense (a few of 10(3) cm(-3)) and massive (a few of 10(3) M-circle dot) characteristics coupled with the broad linewidths (>2.5 km s(-1)) suggest that they are promising sites for forming high-mass stars or clusters. This could be further consolidated by the detection of compact H II regions in Cores A and E. We tentatively identified and classified 63 candidate young stellar objects (YSOs) based on the Spitzer and UKIDSS data. They are found to be dominantly distributed in regions with strong molecular gas emission, indicative of active star formation, especially in the shell. The H II region inside the bubble is mainly ionized by a similar to O8V star(s), of the dynamical age of similar to 1.6 Myr. The enhanced number of candidate YSOs and secondary star formation in the shell as well as the timescales involved, indicate a possible scenario for triggering star formation, signified by the 'collect and collapse' process.</P>
NMAAP1 Expressed in BCG-Activated Macrophage Promotes M1 Macrophage Polarization

Macrophages are divided into two subpopulations: classically activated macrophages (M1) and alternatively activated macrophages (M2). BCG (Bacilli Calmette-$Gu{\acute{e}}rin$) activates disabled $na{\ddot{i}}ve$ macrophages to M1 macrophages, which act as inflammatory, microbicidal and tumoricidal cells through cell-cell contact and/or the release of soluble factors. Various transcription factors and signaling pathways are involved in the regulation of macrophage activation and polarization. We discovered that BCG-activated macrophages (BAM) expressed a new molecule, and we named it Novel Macrophage Activated Associated Protein 1 (NMAAP1). 1 The current study found that the overexpression of NMAAP1 in macrophages results in M1 polarization with increased expression levels of M1 genes, such as inducible nitric oxide synthase (iNOS), tumor necrosis factor alpha (TNF-${\alpha}$), Interleukin 6 (IL-6), Interleukin 12 (IL-12), Monocyte chemoattractant protein-1 (MCP-1) and Interleukin-1 beta (IL-$1{\beta}$), and decreased expression of some M2 genes, such as Kruppel-like factor 4 (KLF4) and suppressor of cytokine signaling 1 (SOCS1), but not other M2 genes, including arginase-1 (Arg-1), Interleukin (IL-10), transforming growth factor beta (TGF-${\beta}$) and found in inflammatory zone 1 (Fizz1). Moreover, NMAAP1 overexpression in the RAW264.7 cell line increased cytotoxicity against MCA207 tumor cells, which depends on increased inflammatory cytokines rather than cell-cell contact. NMAAP1 also substantially enhanced the phagocytic ability of macrophages, which implies that NMAAP1 promoted macrophage adhesive and clearance activities. Our results indicate that NMAAP1 is an essential molecule that modulates macrophages phenotype and plays an important role in macrophage tumoricidal functions.
