Targeted retardation of hepatocarcinoma cells by specific replacement of alpha-fetoprotein RNA

<P><B>Abstract</B></P><P>Although hepatocellular carcinoma (HCC) is one of the world-wide common malignancies, development of more specific and controlled therapeutic methods should be warranted. In this study, we describe a novel approach to HCC therapy that is based on <I>trans</I>-splicing ribozyme-mediated replacement of HCC-associated specific RNAs. We have developed a specific ribozyme that can target and replace human alpha-fetoprotein (AFP) RNA, which is highly expressed in HCC, with new transcript exerting therapeutic activity selectively in AFP-expressing liver cancer cells. The RNA replacement was employed via a high-fidelity <I>trans</I>-splicing reaction with the targeted residue in the AFP-expressing cells. Noticeably, the ribozyme could selectively deliver activity of suicide gene, herpes simplex virus thymidine kinase gene, into the liver cancer cells expressing the AFP RNA and thereby specifically and effectively retarded the survival of these cells with ganciclovir treatment. Simultaneously with the specific induction of therapeutic gene activity, the ribozyme reduced expression level of the targeted AFP RNA in the cells. These results suggest that the AFP RNA-targeting <I>trans</I>-splicing ribozyme could be a useful genetic agent for HCC-targeted efficient gene therapy.</P>

In this study, we describe a novel approach to achieve replicative selectivity of conditionally replicative adenovirus that is based upon trans-splicing ribozyme-mediated replacement of cancer-specific RNAs. We developed a specific ribozyme that can reprogram human telomerase reverse transcriptase (hTERT) RNA to induce adenoviral E1A gene expression selectively in cancer cells that express the RNA. Western blot analysis showed that the ribozyme highly selectively triggered E1A expression in hTERT-expressing cancer cells. RT-PCR and sequencing analysis indicated that the ribozyme-mediated E1A induction was caused via a high fidelity trans-splicing reaction with the targeted residue in the hTERTexpressing cells. Moreover, reporter activity under the control of an E1A-dependent E3 promoter was highly transactivated in hTERT-expressing cancer cells. Therefore, adenovirus containing the hTERT RNA-targeting transsplicing ribozyme would be a promising anticancer agent through selective replication in cancer cells and thus specific destruction of the infected cells.

Java Card makes it possible to easily have several applications and programs written in Java. The one of the main issues of Java Card is about memory optimization to maximize the use of limited smart card memory. In this paper, we propose a new JCVM Installer architecture. The key idea is to use RAM as much as possible by resolving indirect references in RAM, not EEPROM. It makes the Java Card more efficient in terms of speed during the post-issuance of applications. Writing in RAM is much faster than writing in EEPROM.

Seasonal incident rates were examined from the 981 pigs slaughtered in southern Kang-won and distribution of lesions and histopathological examination were conducted from 231 Enzootic pneumonia affected lungs. The results obtained were summarized as follows: 1. 39.7~50.2% of 981 slaughtered pigs showed pneumonia lesions without seasonal variation and the incidence rate of Enzootic pneumonia was 23.5 %. 2. The distribution of Lesions of Enzootic pneumonia lobes was observed. The right lobes were affected more frequently than the left, and the highest frequency of 79.7% being recorded in the right cardiac lobe followed by the left cardiac, right apical, intermediate, left apical, right diaphrogmatic and diaphrogmatic. 3. In histopatological observation lung lobes were forming lymphonodulus from perivascular, peribronchiolar lymphoid hyerplasia and it was remarkable to bronchostenosis.

질산염은토양으로부터 식물에 흉수되어 단백질합성에 이용되는 필수성븐이나 그 안전성에 대하여 논란이 되고있다. 본 연구에서는 운리나라에서 저산되는 채소류의 질산염(NO3-) 함량을조사하여 잔류실태를 과악하고 외국의 모니터링 자료와 비교 평가하고자 하였다. 전국에서 생산되는 배추, 무우, 상치, 시금치를 재배방법, 재배시기에 따라 각각 100건ㅆ기 총 400건을 채취, Ion chromatography를 이용하여 질산염 함량을 측정하였다. 조사결과 재배방법, 재배 시기에 따른 채소류의 질산염함량은 큰 차이가 없었으며, 채소종류별 질산염 함량은 시금치 311-5522(평균 2788mg/kg), 상치 542-4484(평균 .2287m9/kg), 무우 273-4151(평균 15mg/kg), 배추 362-3035(평균 1498mg/kg)으로 나타났다. 따라서 국내 채소류의 질산염함량은 외국의 모니터fi 결콰와 유사하거나 낮은 수준므로나타났다. 국래 채소류의 질산염은 식물에 정상적으로 존재하는 함량으로 건강에 위해가 없는 것으로 판단되었다. Nitrate is taken up from the soil by plants for protein synthesis and present in vegetables as a natural component and/or contaminant. The objective of this study was to estimate nitrate(NO_(3)^(-)) contents of some vegetables(Chinese cabbage, radish, lettuce, spinach) which were produced in Korea and provide a scientific basis for the evaluation of risk to public health arising from dietary exposure to nitrate. A total of 400 samples were taken over a period of one year and analysed of nitrate using Ion Chromatography. From the results, in general, nitrate levels in vegetables produced by 2 growing methods and 2 harvest seasons were not different. The minimum, maximum and mean values of nitrate are spinach 311-5522,2788; lettuce 542-4484,22871;radish 273-4151, 1551; Chinese cabbage 362-3015,1498 (mg/kg). Nitrate contents of vegetables grown in Korea are similar to the other monitoring results.
野冬靑果가 實驗的 糖尿에 미치는 影響에 관한 免疫組織化學的 硏究

This experiment was performed into investigate the effect of Syzygii cumini Fructrs on the diabetic rats induced by STZ. Experimental diabetes was induced by 55㎎/㎏ of STZ. injection and administered Syzygii cumini Fructrs extract during 14days after STZ injection. Glucagon and insulin granules in Langerhans islets were stained by use of immunohistochemical method and observed the relative amount of the each granules in Langerhans islet by light microscope and image analysis ststem. The results obtained are as follows: 1. Area % of insulin granules in Langerhans islets in Syzygii cumini Fructrs raised and showed the statistically significant difference with the control group at 14th day. 2. Area % of glucagon granules in Langerhans islets in Syzygii cumini Fructrs decreased and showed the statistically significant difference with the control group at 7th and 14th day. To conclude, it can be inferred that Syzygii cumini Fructrs has a control effect on glucagon and insulin granules in Langerhans islets of diabetic rats induced by STZ.

Non-Hodgkin's lymphoma (NHL), a disease which may involve the spine, is frequently associated with advanced disease. Radiculopathy caused by spinal root compression as the initial presentation in patients with NHL is very rare and thought to occur in less than 5% of cases. A 69-year-old woman complained of a history of low back pain with right sciatica for 1 month prior to admission. Computed tomography and magnetic resonance imaging of the lumbar spine showed a dumbbellshape epidural mass lesion extending from L2 to L3, which was suggestive of a neurogenic tumor. After paraspinal approach and L2 lower half partial hemilaminectomy, total excision of the tumor was achieved, followed by rapid improvement of back pain and radiating pain. The lesion was confirmed to be Burkitt’s lymphoma by histopathological examination. We then checked whole-body PET-CT, which showed multifocal malignant lesions in the intestine, liver, bone and left supraclavicular lymph node. Although a rare situation, Burkitt’s lymphoma should be considered in the differential diagnosis for patients presenting with back and lumbar radicular pain without a prior history of malignancy. Burkitt’s lymphoma could be the cause of dumbbell-shape spinal tumor.
