        Production of L-Theanine Using Escherichia coli Whole-Cell Overexpressing γ-Glutamylmethylamide Synthetase with Baker’s Yeast

        L-Theanine, found in green tea leaves has been shown to positively affect immunity and relaxation in humans. There have been many attempts to produce L-theanine through enzymatic synthesis to overcome the limitations of traditional methods. Among the many genes coding for enzymes in the L-theanine biosynthesis, glutamylmethylamide synthetase (GMAS) exhibits the greatest possibility of producing large amounts of production. Thus, GMAS from Methylovorus mays No. 9 was overexpressed in several strains including vectors with different copy numbers. BW25113(DE3) cells containing the pET24ma::gmas was selected for strains. The optimal temperature, pH, and metal ion concentration were 50℃, 7, and 5 mM MnCl<sub>2</sub>, respectively. Additionally, ATP was found to be an important factor for producing high concentration of L-theanine so several strains were tested during the reaction for ATP regeneration. Baker’s yeast was found to decrease the demand for ATP most effectively. Addition of potassium phosphate source was demonstrated by producing 4-fold higher L-theanine. To enhance the conversion yield, GMAS was additionally overexpressed in the system. A maximum of 198 mM L-theanine was produced with 16.5 mmol/l/h productivity. The whole-cell reaction involving GMAS has greatest potential for scale-up production of L-theanine.

      • 柔道 밧다리 후리기의 kinematic 分析

        The purpose of the study was to analyze the Kinematics component of O-soto-gari in Judo. The subjects were 4 male college judo caliber players. They were filmed at 100f/sec. The analyzed Variables were angular velocity and angular accee ation of leg and angular change during O-soto-gari. The results were summarized as follow; 1. During Kuzushi-Tsukuri phase the stride and volocities were 1.093m and 1.497m/sec in each. 2. And during in the Tsukuri-Kake phase the maximum angular velocities in 0.2 seconds were 400 degree/sec in low leg and 350 degree/sec in upper leg. The maximum angular acceleration was -4000 degree/sec²in low leg and 4000 degree/sec²in upper leg. 3. During Kake phase maximum angular velocities were 680 degree/sec in low leg and 400 degree/sec in upper leg. And maximum angular acceleration was 9500 degree/sec² in low leg and 3000 degree/sec²in upper leg. 4. In order to analyze the Kinematic and Kinetic components of O-soto-gari accuretely, the study should be performed with 3-D and force plat-form system.

        하악전돌증의 악교정수술후의 회귀성향에 관한 연구

        저자등은 악교정수술후에 발생하는 희귀성향에 관한 연구를 위하여 계명대학교 의과대학 치과학교실에 내원하여 하악전돌증으로 진단되어 하악후퇴술을 받은 7명의 환자를 대상으로 하여 수술전, 후의 경조직의 변화를 계측, 분석하여 다음과 같은 결과를 얻었다. 수술직후 Gn(H)는 평균 8.2mm 후방이동되었고 SNB는 평균 4.4 감소하였다. SNB는 장기간 경과후 수술직후보다 평균 0.9 증가되었으며 21.1%의 희귀율을 나타내었다. Gn(H)는 장기간 경과후 수술직후보다 6례에서 평균 1.3mm 전방희귀되었으며 나머지 1례에서 4mm 후방이동되었다. SN-MP 및 gonial angle 은 장기간 경과후 수술 직후보다 각각 평균 2.2 ,5.8 증가되었다. Gn(V)는 장기간 경과후 수술직후와 비교하여 변화가 없는 경우가 4례, 증가 2례, 감소 1례로 나타났다. 상하악 중절치는 장기간 경과후 수술직후보다 각각 순측 혹은 설측으로 경사되었다. This study was undertaken to examine relapse tendency after mandibular setback. Seven cases were selected from patients who underwent orthodontic treatment and mandibular setback surgery in our department. The serial lateral cephalogram of each patient were obtained and analyzed: The result were as follows; 1. By operation, Gn(H) moved backward (average 8.2mm), SNB angle decreased(average 4.4˚). 2. When the long term follow-up measurements were compared with the immediate postop, mesurements. ·SNB angle increased on average by 0.9˚and relapse tate was 21.1%. ·GN(H) returned foward on average by 1.3mm in 6 cases, moved backward by 4mm in 1 case. ·SN-MP and gonial angle increased on average by 2.2˚, 5.8˚respectively. ·Gn(V) stayed unchanged in 4 cases, increased in 2 cases, decreased in 1 case. ·Upper and lower incisors slightly moved labially or lingually.

        The Effects of Recovery Pattern on Changes of Blood Components. The purpose of the study was to investigate effects of recovery pattern on changes of blood components in endurance running with the same work intensity and 5 different relief levels were used as protochol. Used 10 male endurance runners who belong to H University students as subjects, and the results were summarized as follows; with the same work intensity the increasing ratio of lactic acid concentration in blood was shown significant differences (p<0.01) in accordance with recovery patterns. To improve capacity of tolerence against lactic acid accumalation, jogging with 60% of individual maximal speed was superior to jogging with 90% of individual maximal speed and 2 or 4 minutes of passive recovery between work sets. Lactic acid production was significantly increased in jogging with 60% of individual maximal speed, therefore, to improve anaerobic endurance, 2 minutes of passive recovery was recommended because of more rapid break down of Lactic acid accumulation with 2 minutes, or more passive recovery.

      • FCT 2-3 Hyaluronic acid injection via a pneumatic microjet device to improve forehead wrinkles

        Background: Hyaluronic acid (HA) fillers are thought to exert short- and long-term effects on wrinkle. Needleless microjet energy allows HA to be injected into the dermis at high speed using pressurized air and enables an even dispersion. With the multi-needle approach, HA can be delivered in small doses by very fine needles directly at the target depth. Objectives: We investigated the efficacy of a pneumatic microjet device that delivers HA into the dermis using either pneumatic needleless action or multi-needle injections for improvement of forehead wrinkles. Methods: Twenty-four enrolled subjects were randomized to receive HA injections or placebo via the needleless or multi-needle method. Our evaluation was based on changes in dermal thickness using ultrasonography and the Wrinkle Severity Rating Scale (WSRS); post-treatment data were compared with baseline. Adverse events were also evaluated. Results: Post-treatment evaluation points showed increased dermal thickness and WSRS score was significantly greater among patients who received HA. However, there was no significant difference in dermal thickness increase between the needleless method and the multi-needle method. No severe adverse events were reported Conclusion: Injection of HA via a pneumatic microjet device can be a safe and effective treatment for improving forehead wrinkles.

      • The effect of Cinnamomi Cortex ex tract on melanin synthesis and anti-inflammation in cells

        Background: Cinnamomi Cortex has been widely used as a herbal medicine in Asia and Europe. In traditional Chinese medicine, Cinnamomi Cortex has been claimed for improving blood circulation. Objectives: This study confirmed that inhibitory effects of Cinnamomi Cortex extract for melanogenesis through B16 melanoma cells and for inflammation reaction through RAW 264.7 cells. Methods: We investigated the effect of Cinnamomi Cortex extracts on メ-MSH-induced melanogenesis in B16 melanoma cells and on LPS-induced activation of iNOS, COX-2, NO and PGE2 expression in RAW 264.7 macrophages. Results: Cinnamomi Cortex extracts inhibited melanin synthesis, tyrosinase activity, MITF and tyrosinase expression by ERK and CREB gene regulation in メ -MSH-induced B16 melanoma cells. In addition, Cinnamomi Cortex extracts inhibited NO, PGE2 expression and pro-inflammatory cytokines in LPS-induced RAW 264.7 cells. Conclusion: We anticipate that good effects of Cinnamomi Cortex extract in skin diseases by stress including UV light based on inhibitory effects of Cinnamomi Cortex extract for melanin synthesis and inflammation response in cells.

        개에서 내인성 산화질소계 억제가 국소 심기능 및 혈역학에 미치는 효과

        Background : The present study was aimed (1) to assess the effects of nitric oxide (NO) synthesis inhibitor on regional myocardial function and systemic and pulmonary hemodynamics; (2) to determine whether the blockade of the cyclo-oxygenase (COX) pathway modifies these effects on the variables, and (3) to investigate the mechanism of cardiac depression following NO synthesis inhibition in an open-chest canine model. Methods : Twenty-five dogs of either sex were acutely instrumented under 1.6% ethrane anesthesia to measure aortic, pulmonary arterial and left ventricular pressure, pulmonary (cardiac output) and left circumflex coronary flow, and subendocardial segment length. NG-nitro-L-arginine methyl ester (L- NAME) at doses of 0.3, 1.0, 3.0, or 10.0 mg/kg i.v. was administered alone (control dogs, n = 10) or in the presence of COX inhibitor, indomethacin (10 mg/kg i.v., n = 10). Seven dogs (n = 7) received phenylephrine at doses of 0.1, 0.3, 1.0, or 3.0 μg/kg/min i.v. to compare its hemodynamic effects wih those of L-NAME. The preload recruitable stroke work slope (Mw) and percent systolic shortening (%SS) as an index of regional myocardial contractility, and the maximum segment lengthening rate (dL/dt max) and percent post-systolic shortening (%PSS) as an index of regional diastolic function, were evaluated. Results : L-NAME dose-dependantly attenuated both regional systolic (Mw and %SS) and diastolic functions (dL/dt max and %PSS), whereas it caused an increase of coronary flow. L-NAME dose- dependently increased systemic blood pressure and vascular resistance as well as pulmonary arterial pressure and vascular resistance. L-NAME also reduced cardiac and stroke volume indices. Pretreatment with indomethacin did not affect the regional myocardial and systemic hemodynamic responses to L-NAME, but did blunt the coronary flow and pulmonary pressure responses. The magnitude of decreases in cardiac and stroke volume indices and Mw was greater with L-NAME than with phenylephrine (P < 0.05), despite the comparble blood pressure increases. Conclusions : These results suggest (1) that NO plays a significant role in cardiac function as well as in systemic and pulmonary but not coronary, vasomotor activities, and (2) that COX products are involved in pulmonary hemodynamic responses to NO synthesis inhibition. It is also suggested that the decline in cardiac output following the NO synthesis inhibition results from a direct myocardial depressant effect of the drug. (Korean J Anesthesiol 1999; 37: 902∼912)
