      • RTN4 3'-UTR Insertion/Deletion Polymorphism and Susceptibility to Non-Small Cell Lung Cancer in Chinese Han Population

        Nogo protein, encoded by gene reticulon-4 (RTN4), includes three major isoforms by different splicing, named Nogo-A Nogo-B and Nogo-C. Nogo proteins play an important role in the apoptosis of cells, especially in tumor cells. RTN4 single nucleotide polymorphisms (SNPs) can influence the efficiency of transcription and translation thus being related with an individual's predisposition to cancer. The CAA insertion/deletion polymorphism (rs34917480) within RTN4 3'-UTR has been reported to be associated with many cancer types. In order to investigate the relationship between this polymorphism and susceptibility to non-small cell lung cancer (NSCLC) in the Chinese population, we conducted the present case-control study including 411 NSCLC patients and 471 unrelated healthy controls. The genotype distributions were significantly different between cases and controls (p=0.014). We found that the del allele could significantly increase NSCLC risk (ins/ins vs ins/del: p=0.007, OR 1.46, 95%CI=1.11-1.93; dominant model: p=0.004, OR 1.47, 95%CI=1.13-1.92 and allele model: p=0.008, OR 1.35, 95%CI=1.08-1.67). This association was stronger in participants over 60 years old, males and smokers. We therefore conclude that the CAA insertion/deletion polymorphism (rs34917480) contributes to non-small cell lung cancer risk in Chinese population. Age, sex and environmental exposure are also related to carcinogenic effects of rs34917480.

        Separation of arenols from a low-temperature coal tar by liquid-liquid extraction

        Low-temperature coal tar (LTCT) is a sticky liquid mixture produced mainly from coal pyrolysis, which contains various value-added chemicals (VACs). Liquid-liquid extraction is considered as one of the green and effective ways to explore the organic composition and separate the VACs from LTCT. Herein, petroleum ether, methanol, and carbon disulfide were used to extract arenols from a LTCT. As a result, the relative content and absolute content of arenols extracted from the LTCT are ca. 96.3% and 85.9%, respectively. Among them, p-cresol is predominant, accounting for 22.2%. The isolated contents of arenols are up to 84.6%. Moreover, a kilogram-scale operation was carried out under the same conditions, which offers a potential application in industrial production.

      • Expression of Transcription Factor FOXC2 in Cervical Cancer and Effects of Silencing on Cervical Cancer Cell Proliferation

        Objective: Forkhead box C2 (FOXC2) is a member of the winged helix/forkhead box (Fox) family of transcription factors. It has been suggested to regulate tumor vasculature, growth, invasion and metastasis, although it has not been studied in cervical cancer. Here, we analyzed FOXC2 expression in cervical tissues corresponding to different stages of cervical cancer development and examined its correlation with clinicopathological characteristics. In addition, we examined the effects of targeting FOXC2 on the biological behavior of human cervical cancer cells. Methods: The expression of FOXC2 in normal human cervix, CIN I-III and cervical cancer was examined by immunohistochemistry and compared among the three groups and between cervical cancers with different pathological subtypes. Endogenous expression of FOXC2 was transiently knocked down in human Hela and SiHa cervical cells by siRNA, and cell viability and migration were examined by scratch and CCK8 assays, respectively. Results: In normal cervical tissue the frequency of positive staining was 25% (10/40 cases), with a staining intensity (PI) of $0.297{\pm}0.520$, in CIN was 65% (26/40cases), with a PI of $3.00{\pm}3.29$, and in cancer was 91.8% (68/74 cases), with a PI of $5.568 {\pm}3.449$. The frequency was 100% in adenocarcinoma (5/5 cases) and 91.3% in SCCs (63/69 cases). The FOXC2 positive expression rate was 88.5% in patients with cervical SCC stage I and 100% in stage II, showing significant differences compared with normal cervix and CIN. With age, pathologic differentiation degree and tumor size, FOXC2 expression showed no significant variation. On transient transfection of Hela and SiHa cells, FOXC2-siRNA inhibition rates were 76.2% and 75.7%; CCK8 results showed reduced proliferation and relative migration (in Hela cells from $64.5{\pm}3.16$ to $49.5{\pm}9.24$ and in SiHa cells from $60.1{\pm}3.05$ to $44.3{\pm}3.98$) (P < 0.05). Conclusion: FOXC2 gene expression increases with malignancy, especially with blood vessel hyperplasia and invasion degree. Targeted silencing was associated with reduced cell proliferation as well as invasion potential.

        APPROXIMATE OPTIMAL AUTOSAR SOFTWARE COMPONENTS DEPLOYING APPROACH FOR AUTOMOTIVE E/E SYSTEM

        The AUTOSAR has been developed as the worldwide standard for automotive E/E software systems, making the electronic components of different suppliers to be employed universally. However, as the number of component-based applications in modern automotive embedded systems grows rapidly and the hardware topology becomes increasingly complex, deploying such large number of components in automotive distributed system in manual way is over-dependent on experience of engineers which in turn is time consuming. Furthermore, the resource limitation and scheduling analysis make the problems more complex for developers to find out an approximate optimal deploying approach in system integration. In this paper, we propose a novel method to deploy the AUTOSAR components onto ECUs with the following features. First, a clustering algorithm is designed for deploying components automatically within relatively low time complexity. Second, a fitness function is designed to balance the ECUs load. The goal of our approach is to minimize the communication cost over all the runnable entities while meeting all corresponding timing constraints and balancing all the ECUs load. The experiment results show that our approach is efficient and has well performance by comparing with other existing methods in specific and synthetic data set.

        Molecular Characteristics and Extracellular Expression Analysis of Farnesyl Pyrophosphate Synthetase Gene in Inonotus obliquus

        A farnesyl pyrophosphate synthase gene was cloned from Inonotus obliquus, designated IOFPS. The IOFPS cDNA contained an open reading frame (ORF) of 972 bps, encoding a protein of 324 amino acids. The deduced amino acid sequence of IOFPS revealed moderate homology with that of other fungi, and contained four conserved domains. Phylogenetic analysis showed that IOFPS belonged to the basidiomycete group. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis indicated that the IOFPS gene was successfully expressed in a yeast recombinant cell. Enzyme catalytic experiments were carried out with purified protein (IOFPS protein), which was isolated and purified from recombinant yeast cells. The special hydrolysis product (farnesol) was then detected by liquid chromatography coupled with tandem mass spectrometry (LC-MS). These results indicated that the cloned cDNA encoded a farnesyl diphosphate synthase and the IOFPS protein maintained catalytic activity in vitro.

      • A FREE BREATHING IMPEDANCE RHEOGRAM APPARATUS MADE OF MICROPROCESSOR

        The impedance rheogram is a modern non-invasive method used to observe the disease of each part of the body such as heart, liver and lung etc. Because of the breathing interference effect, the patients have to hold their breathing when they are being tested by conventional impedance rheogram apparatus. It is not convenient to observe those patients who are difficult to cooperate with doctor such as children and poor-function-heart patients. Therefore, we have developed the free breathing impedance rheogram apparatus by using microprocessor, it can effectively eliminate the interference of the breathing to impedance rheogram, so that patients can be tested in the free breathing. By this way, it can not only much improve measuring accuracy, and also extend clinical service extant. The digital filtering system that we have developed by using microprocessor is used for automatically tracking variation of rhythm of the heart by specific program and filtering interference of the breathing by in-phase superposing-averaging the impendance signal of the lung in free breathing. The present artical introduces the hardware structure and main technical targets of the free breathing impedance rheogram apparatus, and provides application results for grown-up healthful persons and the patients having heart or lung problems in clinical service. The results show that the effect filtering interference of the breathing to impedance rheogram is very obvious. This technique can be used In impedance rheogram of heart and liver etc, and lays down a foundation for developing monitor in clinical service. In addition, it can also be used in studying of sport-medicine and space-medicine. This result has gotten United States Patent on Jun. 25,1991.

        Active Compounds from Schisandra chinensis Exhibiting Tyrosinase Activity and Melanin Content Inhibition in B16 Melanoma Cells

        Schisandra chinensis has been used as traditional medicine. The structures of isolate active compounds (schisandrin B, deoxyschisandrin, schisandrin C) from S. chinensis were characterized by physical and spectroscopic analyses. Active compounds were tested for their potential to act as anti-melanogenesis or skin-whitening agents by their abilities to inhibit tyrosinase activity in the cell-free mushroom tyrosinase assay and cellular tyrosinase derived from B16 melanoma cells. The tyrosinase inhibitory activity was correlated to the inhibition of melanin productions in α-MSH-stimulated and unstimulated B16 cells. Cellular tyrosinase kinetics were analyzed and showed by Lineweaver- Burk plot. Schisandrin B was minimally cytotoxic (cell viability: 88.99% at 0.75 μM) and the IC50 value for suppression of mushroom tyrosinase activity was estimated as 0.6 μM. Zymography analysis demonstrated schisandrin B’s concentration-dependent effects and the kinetic analysis indicated schisandrin B’s noncompetitive-inhibitory action.

        Plasma metabolites associated with physiological and biochemical indexes indicate the effect of caging stress on mallard ducks (Anas platyrhynchos)

        Objective: Cage rearing has critical implications for the laying duck industry because it is convenient for feeding and management. However, caging stress is a type of chronic stress that induces maladaptation. Environmental stress responses have been extensively studied, but no detailed information is available about the comprehensive changes in plasma metabolites at different stages of caging stress in ducks. We designed this experiment to analyze the effects of caging stress on performance parameters and oxidative stress indexes in ducks. Methods: Liquid chromatography tandem mass spectrometry (LC/MS-MS) was used to determine the changes in metabolites in duck plasma at 5 (CR5), 10 (CR10), and 15 (CR15) days after cage rearing and traditional breeding (TB). The associated pathways of differentially altered metabolites were analyzed using Kyoto encyclopedia of genes and genomes (KEGG) database. Results: The results of this study indicate that caging stress decreased performance parameters, and the plasma total superoxide dismutase levels were increased in the CR10 group compared with the other groups. In addition, 1,431 metabolites were detected. Compared with the TB group, 134, 381, and 190 differentially produced metabolites were identified in the CR5, CR10, and CR15 groups, respectively. The results of principal component analysis (PCA) show that the selected components sufficiently distinguish the TB group and CR10 group. KEGG analysis results revealed that the differentially altered metabolites in duck plasma from the CR5 and TB groups were mainly associated with ovarian steroidogenesis, biosynthesis of unsaturated fatty acids, and phenylalanine metabolism. Conclusion: In this study, the production performance, blood indexes, number of metabolites and PCA were compared to determine effect of the caging stress stage on ducks. We inferred from the experimental results that caging-stressed ducks were in the sensitive phase in the first 5 days after caging, caging for approximately 10 days was an important transition phase, and then the duck continually adapted.         Progranulin Controls Sepsis via C/EBPα-Regulated <i>Il10</i> Transcription and Ubiquitin Ligase/Proteasome-Mediated Protein Degradation

        <P>Progranulin (PGRN) is a widely expressed, pleiotropic protein that is involved in diverse biological processes, including cellular proliferation, neuron development, and wound healing. However, the role of PGRN in the regulation of pathogen-induced systemic inflammation and the mechanisms involved have not been established. In this study, we show that PGRN-deficient mice display heightened mortality in models of polymicrobial sepsis and endotoxinemia, with increased tissue levels of inflammatory cytokines and reduced IL-10 production. Conversely, administration of rPGRN decreases the susceptibility of PGRN-deficient mice to LPS-induced endotoxemic shock and augments IL-10 production by LPS-activated macrophages in a TNFR-dependent manner. Molecular analysis reveals a direct role of the transcription factor C/EBP alpha in PGRN-regulated IL-10 expression. C/EBP alpha-deficient macrophages produce less IL-10 in response to LPS. Furthermore, mice deficient in C/EBP alpha in hematopoietic cells are highly vulnerable to LPS-induced septic shock. Lastly, the defective IL-10 production by PGRN-deficient cells is primarily due to reduced C/EBP alpha protein stability via the E3 ubiquitin-conjugating enzyme E6AP and proteasome-mediated degradation. To our knowledge, this study provides the first evidence that PGRN is a nonredundant regulator of systemic inflammation via modulating the levels and activity of C/EBP alpha, IL-10, and the ubiquitin-proteasome proteolysis pathway. The results bear strong and profound implications for PGRN insufficiency and its mutation-associated systemic and organ-specific inflammatory human diseases.</P>

        Micellization behavior of binary mixtures of amino sulfonate amphoteric surfactant with different octylphenol polyoxyethylene ethers in aqueous salt solution: Both cationic and hydrophilic effects

        The effect of different valent inorganic cations and the hydrophilic group of surfactant on themicellization behavior of binary surfactant mixtures constituted by an amphoteric surfactant, sodium3-(N-dodecyl ethylenediamino)-2-hydropropyl sulfonate (C12AS), and three nonionic surfactantsoctylphenol polyoxyethylene ether (OP-n) with different numbers of oxyethylene glycol ethers (n),namely, OP-10, OP-7, and OP-4 was investigated in aqueous solution. These inorganic cations includeNa+, K+, Mg2+, Ca2+, Ba2+, Al3+ and Fe3+. Both the tensiometry and the UV–vis spectrophotometry usingpyrene as a probe were adopted to determine the critical micelle concentrations (cmc) of individual ormixed surfactants. Interaction parameters between two surfactants and other parameters were obtainedbased on the regular solution theory, the pseudophase separation model, Rubingh’s model, etc. Thermodynamic parameters including Gibbs energy of micellization and thermodynamic stability werealso calculated by both the equation proposed by Molyneux et al. and Maeda’s treatment, respectively. The effect of different valent cations on both the mixed cmc of surfactant mixtures and the molar fractionof C12AS in mixed micelle (X1) can be explained theoretically by the salting-out effect, electrostaticinteraction and steric effect. For three binary surfactant mixtures, the chain length of hydrophilic groupof surfactant results in different micellization behaviors. On adding salts, the interaction parametersshow that there exists a synergistic effect between two surfactants, and the effect increases withincreasing the ratio of valence (Z) and atom radius (R) of cation, especially, for the C12AS/OP-4 mixture. With increasing the Z/R value of cation, a deviation of X1 from the ideal value shows a divergence forthree binary surfactant mixtures because of different hydrophilicities of surfactant. Thermodynamicparameters indicate that the addition of cations with a large Z/R value can be contributive moreeffectively to the formation of stable mixed micelle than the case in the presence of cations with a smallZ/R value.
