        Three sesquiterpene lactones suppress lung adenocarcinoma by blocking TMEM16A-mediated Ca2+-activated Cl− channels

        Transmembrane protein TMEM16A, which encodes calcium-activated chloride channel has been implicated in tumorigenesis. Overexpression of TMEM16A is associated with poor prognosis and low overall survival in multiple cancers including lung adenocarcinoma, making it a promising biomarker and therapeutic target. In this study, three structure-related sesquiterpene lactones (mecheliolide, costunolide and dehydrocostus lactone) were extracted from the traditional Chinese medicine Aucklandiae Radix and identified as novel TMEM16A inhibitors with comparable inhibitory effects. Their effects on the proliferation and migration of lung adenocarcinoma cells were examined. Whole-cell patch clamp experiments showed that these sesquiterpene lactones potently inhibited recombinant TMEM16A currents in a concentration- dependent manner. The half-maximal concentration (IC50) values for three tested sesquiterpene lactones were 29.9 ± 1.1 μM, 19.7 ± 0.4 μM, and 24.5 ± 2.1 μM, while the maximal effect (Emax) values were 100.0% ± 2.8%, 85.8% ± 0.9%, and 88.3% ± 4.6%, respectively. These sesquiterpene lactones also significantly inhibited the endogenous TMEM16A currents and proliferation, and migration of LA795 lung cancer cells. These results demonstrate that mecheliolide, costunolide and dehydrocostus lactone are novel TMEM16A inhibitors and potential candidates for lung adenocarcinoma therapy.

      • Fentanyl Increases Colorectal Carcinoma Cell Apoptosis by Inhibition of NF-κB in a Sirt1-dependent Manner

        Background: Fentanyl is used as an analgesic to treat pain in a variety of patients with cancer and recently it has become considered to also act as an antitumor agent. The study present was designed to investigate the effects of fentanyl on colorectal cancer cell growth and plausible mechanisms. Materials and Methods: The human colorectal carcinoma cell line HCT116 was subcutaneously injected into nude mice. The viability of HCT116 was tested by MTT assay, and apoptosis by flow cytometry and caspase-3 activity. The expression of Sirt1 and NF-${\kappa}B$ were evaluated by Western blotting and the levels of Sirt1 and NF-${\kappa}B$ by fluorescence method. SiRNA was used to silence and Ad-Sirt1 to overexpress Sirt1. Results: Our data showed that fentanyl could inhibit tumor growth, with increased expression of Sirt1 and down-regulation of Ac-p65 in tumors. Compared with control cells without treatment, HCT116 cells that were incubated with fentanyl had a higher apoptotic rate. Moreover, fentanyl could increase expression and activity of Sirt1 and inhibitor expression and activity of NF-${\kappa}B$, which might be mechanisms of fentanyl action. Conclusions: Fentanyl increased colorectal carcinoma cell apoptosis by inhibition of NF-${\kappa}B$ activation in a Sirt1-dependent manner.

      • Microcellular foams of thermoplastic vulcanizates (TPVs) based on waste ground rubber tire powder

        <P><B>Abstract</B></P><P>With the increased adoption of thermoplastic vulcanizates (TPVs) in automotive weather seal systems, the foams of TPVs present an important milestone in providing key applications such as trunk and door seals. In this study, microcellular foams of TPV based on waste ground rubber tire powder (WGRT) were investigated. In order to investigate the relationship between processing conditions and structure of TPV foams, we first prepared the thermoplastic vulcanizates of PP-g-MA/WGRT, then the samples were saturated with carbon dioxide and the saturated specimens were expanded during the pressure-quench process. The results indicated that the microcellular structure was dependent on the processing conditions. Cell size increased with saturation temperature, whereas cell density and relative density decreased. Different nucleation processes were produced with saturation pressure.</P>

        Two Novel 3-D Zinc Compounds with Mixture Ligands of H3BTB and 4,4'-Bipy: Crystal Structures, Blue Fluorescence and Single Point Energy Calculation

        Two 3-D zinc framework compounds, $[Zn_6(BTB)_4(4,4'-bipy)_4(H_2O)_4]{\cdot}9H_2O$ (1) and $[Zn_3(BTB)_2(4,4'-bipy)_2(H_2O)_2]{\cdot}5H_2O$ (2) ($H_3BTB$ = 1,2,3-benzenetricarboxylic acid, 4,4'-bipy = 4,4'-bipyridine), are obtained from the diffusion method and hydrothermal reaction respectively. Though 1 and 2 has the same coordination geometries of zinc atoms and coordination mode of $BTB^{3-}$, their 2-D layers are different: mirror symmetric layers in 1; parallel ones in 2, further connecting by 4,4'-bipy into 3-D frameworks. The hydrothermal reaction of 2 results in a more stable 3-D framework than the one in 1, which is supported by the single point energy calculations. 1 and 2 show similar blue fluorescence at 417 nm, which can be assigned to LMCT.

        Suppression of ICI and MAI in SC-FDMA communication system with carrier frequency offsets

        <P>Similar to other orthogonal frequency division multiplexing (OFDM)-based systems, carrier frequency offset (CFO) is a challenging problem in uplink communications of single carrier frequency division multiple access (SC-FDMA) system. It must be noticed that CFO, which is mainly due to oscillator instability and/or Doppler shift, would generate inter carrier interference (ICI) as well as multi-access interference (MAI) to disturb the received signals and seriously degrade system performance. Frequency synchronization in uplink communications is difficult because different users always experience different CFOs and one user¿s CFO correction would misalign other users. In this paper, we proposed a suppression method to overcome the multi CFOs problem. To implement this algorithm, block type pilots would be exploited, which is also utilized in LTE uplink standard. The proposed algorithm is based on the following two assumptions. Firstly the proposed algorithm is applied in this scenario, where different users start to communicate with the base station at different symbol periods, and secondly, during the pilot block and the following data blocks, the CFO of each user is quasi static, which is feasible since CFO is slow varying. Compared with other interference suppression methods, the proposed method could directly estimate the interference components from the inverse pilot matrix, thus it does not need to do the CFO estimation. Further more, since block type pilots is a common pilot pattern in wireless communication system, this algorithm can be easily extended to other communication system. Simulation results show that the proposed suppression algorithm can significantly improved system performance.</P>

      • Expression of Tumor Necrosis Factor Receptor-associated Factor 6 in Lung Cancer Tissues

        Background: Tumor necrosis factor (TNF) receptor-associated factor 6 (TRAF6) has been reported to be associated with the development of various cancers. However, the role of TRAF6 in lung cancer remains unclear. Objective: To explore the expression and clinicopathological significance of TRAF6 protein in lung cancer tissues. Materials and Methods: Three hundred and sixty-five lung cancer samples and thirty normal lung tissues were constructed into 3 microarrays. The expression of TRAF6 protein was determined using immunohistochemistry (IHC). Furthermore, correlations between the expression of TRAF6 and clinicopathological parameters were investigated. Results: The expression of TRAF6 in total lung cancer tissues (365 cases), as well as in small cell lung cancer (SCLC, 26 cases) and non-small cell lung cancer (NSCLC, 339 cases) was significantly higher compared with that in normal lung tissues. The ROC curve showed that the area under curve of TRAF6 was 0.663 (95%CI 0.570~0.756) for lung cancer. The diagnostic sensitivity and specificity of TRAF6 were 52.6% and 80%, respectively. In addition, the expression of TRAF6 was correlated with clinical TNM stage, tumor size and lymph node metastasis in all lung cancers. Consistent correlations were also observed for NSCLCs. Conclusions: TRAF6 might be an oncogene and the expression of TRAF6 protein is related to the progression of lung cancer. Thus, TRAF6 might become a target for diagnosis and gene therapy for lung cancer patients.

        Quantitative analysis and detection of abnormalities for electroencephalogram (EEG) recordings during evoked activities are essential for clinical diagnosis on neurological disorders. However, the process of interpreting EEG is time consuming and requires experienced electroencephalographers (EEGers). In this study, a method of automatic EEG interpretation during hyperventilation was proposed. The purpose is to provide a quantitative interpretation of EEG which is able to conform to EEGers’visual inspection. The final integrated results of automatic interpretation were compared with EEGers’evaluation, and showed high consistence.

      • Elevated Serum Ferritin Levels in Patients with Hematologic Malignancies

        Purpose: To retrospectively analyze variability and clinical significance of serum ferritin levels in Chinese patients with hematologic malignancies. Materials and Methods: Serum ferritin were measured by radioimmunoassay, using a kit produced by the Beijing Institute of Atomic Energy. Patients with hematologic malignancies, and treated in the Department of Hematology in Nanjing First Hospital and fulfilled study criteria were recruited. Results: Of 473 patients with hematologic malignancies, 262 patients were diagnosed with acute leukemia, 131 with lymphoma and 80 with multiple myeloma. Serum ferritin levels of newly diagnosed and recurrent patients were significantly higher than those entering complete remission stage or in the control group (p<0.001). Conclusions: Serum ferritin lever in patients with hematologic malignancies at early stage and recurrent stage are significantly increased, so that detection and surveillance of changes of serum ferritin could be helpful in assessing conditions and prognosis of this patient cohort.

      • Influence of regenerated polyurethane on the mechanical properties and thermal stability of polypropylene during multiple processing cycles

        <P>This work mainly introduces the recyclability of polypropylene (PP) modified by regenerated polyurethane (PU) that was obtained after partial thermochemical decomposition of waste PU. The influence of regenerated PU on the mechanical properties and thermal stability of PP during multiple processing cycles was evaluated by a universal testing machine, differential scanning calorimetry, and thermogravimetric analyses. The data showed that the mechanical properties of the modified PP were dependent on the content of regenerated PU with the increase in the number of processing cycles, whereas the thermal stability of the modified PP was almost independent of the content of regenerated PU.         Synergistic Inhibitory Effect of Berberine and d-Limonene on Human Gastric Carcinoma Cell Line MGC803

        This study aims at evaluating the anticancer effects of berberine hydrochloride (berberine) and d-limonene, alone and in combination, on human gastric carcinoma cell line MGC803 to determine whether berberine and d-limonene work synergistically and elucidate their mechanisms. MGC803 cells were treated with berberine and d-limonene, alone and in combination, for 24–48 h. The inhibitory effects of these drugs on growth were determined by MTT assay. The combination index and drug reduction index were calculated with the Chou–Talalay method based on the median-effect principle. Flow cytometry and laser scanning confocal microscopy were employed to evaluate the effects of both drugs on cell-cycle perturbation and apoptosis, generation of reactive oxygen species (ROS), mitochondrial membrane potential, and expression of Bcl-2 and caspase-3 in MGC803 cells. Berberine or d-limonene alone can inhibit the growth of MGC803 cells in a doseand time-dependent manner. Berberine and d-limonene at a combination ratio of 1:4 exhibited a synergistic effect on anti-MGC803 cells. The two drugs distinctly induced intracellular ROS generation, reduced the mitochondrial transmembrane potential (DCm), enhanced the expression of caspase-3, and decreased the expression of Bcl-2. The combination of berberine and d-limonene showed more remarkable effects compared with drugs used singly in MGC803 cells. The combination of berberine and d-limonene exerted synergistic anticancer effects on MGC803 cells by cell-cycle arrest, ROS production, and apoptosis induction through the mitochondria-mediated intrinsic pathway.
