        Possible role of Pax-6 in promoting breast cancer cell proliferation and tumorigenesis

        Pax 6, a member of the paired box (Pax) family, has been implicated in oncogenesis. However, its therapeutic potential has been never examined in breast cancer. To explore the role of Pax6 in breast cancer development, a lentivirus based short hairpin RNA (shRNA) delivery system was used to knockdown Pax6 expression in estrogen receptor (ER)-positive (MCF-7) and ER-negative (MDA-MB-231) breast cancer cells. Effect of Pax6 silencing on breast cancer cell proliferation and tumorigenesis was analyzed. Pax6-RNAi-lentivirus infection remarkably downregulated the expression levels of Pax6 mRNA and protein in MCF-7 and MDA-MB-231 cells. Accordingly, the cell viability, DNA synthesis, and colony formation were strongly suppressed, and the tumorigenesis in xenograft nude mice was significantly inhibited. Moreover, tumor cells were arrested at G0/G1 phase after Pax6 was knocked down. Pax6 facilitates important regulatory roles in breast cancer cell proliferation and tumor progression, and could serve as a diagnostic marker for clinical investigation.         Peach-specific aldehyde nonanal attracts female Oriental fruit moths, Grapholita molesta (Lepidoptera: Tortricidae)

        The oriental fruit moth (OFM), Grapholita molesta (Lepidoptera: Tortricidae), is a serious pest of stone and pome fruit trees worldwide. G. molesta can switch hosts and produce multiple generations per year. Both the primary and secondary host plants of G. molesta emit aldehydes. Here, we tested whether these aldehydes are helpful for host switching of G. molesta. SPME (Solid Phase Microextraction)-GC–MS analysis identified four aldehydes (octanal, nonanal, decanal, and benzaldehyde) from volatiles of late-season yellow peach fruits. Of the four aldehydes, eletroantennogram analysis showed that nonanal produced the strongest response from the antennae of virgin and mated females. In Y-tube bioassays, only nonanal was attractive to mated G. molesta females amongst the four aldehydes, suggesting that nonanal is important for host recognition in G. molesta. In a field trial, each of the four aldehydes was tested as an attractant for both sexes of G. molesta. The traps baited with nonanal captured more female G. molesta, as compared to traps with other aldehydes and control; the traps baited with benzaldehyde captured most male G. molesta. Our research investigations provide theoretical foundation for biological control of G. molesta females. Future G. molesta lure designs should take into account sex-specific differences in the response to individual aldehydes.

        An Innovative Collaborative Design of Shape and Performance for Super-High-Contact-Ratio Spiral Bevel Gear

        For upgrading the transmission performance of spiral bevel gear and extending the time between overhauls of the helicopter reducer, an innovative multi-objective collaborative optimization strategy of shape and performance is put up for the super-high-contact-ratio spiral bevel gear transmission with the contact path along tooth width based on ease-off. Firstly, according to the preset contact path curve along tooth width and the symmetrical transmission error curve, the pinion optimal surface satisfying the preset functions is obtained based on ease-off. Secondly, based on the basic principles of simulated annealing and gravitational search, the simulated annealing-gravity search cooperative intelligent optimization algorithm is set up for the calculation of the multi-objective collaborative optimization model of the spiral bevel gear transmission, and the modified surface of pinion satisfying the function requirements is obtained based on the collaborative optimization of shape and performance. Finally, combined with the tooth contact analysis method, meshing impact analysis method and dynamic analysis method, the loaded transmission error, meshing impact and dynamic performance of the super-high-contact-ratio spiral bevel gear transmissions are discussed. The simulation result shows the contact path of the collaborative designed spiral bevel gear transmission is along the tooth width, and the contact ratio reaches the maximum. The loaded transmission error, meshing impact, dynamic load factor and root mean square of vibration acceleration of the collaborative design spiral bevel gear transmission are all smaller than the original design and optimized design spiral bevel gear transmissions. The feasibility of the innovative multi-objective collaborative optimization strategy of shape and performance for the spiral bevel gear is verified, and the super-high-contact-ratio design can greatly cut down the contact vibration of the spiral bevel gear transmissions and better the helicopter’s performance.

      • Predictive Factors Determining Neoadjuvant Chemotherapy Outcomes in Breast Cancer - a Single Center Experience

        From January 1, 2008 to March 31, 2010, 101 patients with stage II-III breast cancer were enrolled in this study and subjected to an anthracycline-based neoadjuvant chemotherapy regimen with or without docetaxel. Surgery was performed after 2-6 cycles of chemotherapy, and the clinical response was determined by pathological and histochemical assessments. The clinical response rate, as indicated by complete response (CR), partial response (PR), stable disease (SD), and progressive disease (PD), were 6.9, 52.5, 36.6, and 4.0%, respectively. A multivariable correlation analysis indicated that the overall clinical response rate correlated with the number of metastatic lymph nodes, number of chemotherapy cycles, and vessel invasion status. Importantly, the CR rate was only associated with the number of chemotherapy cycles. Nonparametric tests failed to detect a correlation between HER2 or Topo $II{\alpha}$ status and clinical response to neoadjuvant chemotherapy in these patients. When they were stratified by HER2 or HR status, for HER2-positive patients the CR rate was associated with vessel invasion and Topo $II{\alpha}$ status. Based on our findings, we propose that HR, HER-2 and Topo $II{\alpha}$ are not putative predictive biomarkers of chemotherapy outcome for breast cancer patients. Topo $II{\alpha}$ expression level was only inversely correlated with CR rate among HR-positive patients. Importantly, the achievement of CR was largely related to the number of chemotherapy cycles.

        The modal characteristics of non-uniform multi-span continuous beam bridges

        According to the structure characteristics of the non-uniform beam bridge, a practical model for calculating the vibration equation of the non-uniform beam bridge is given and the application scope of the model includes not only the beam bridge structure but also the non-uniform beam with added masses andelastic supports. Based on the Bernoulli-Euler beam theory, extending the application of the modal perturbation method and establishment of a semi-analytical method for solving the vibration equation of thenon-uniform beam with added masses and elastic supports based is able to be made. In the modal subspace of the uniform beam with the elastic supports, the variable coefficient differential equation that describes the dynamic behavior of the non-uniform beam is converted to nonlinear algebraic equations. Extending the application of the modal perturbation method is suitable for solving the vibration equation of the simply supported and continuous non-uniform beam with its arbitrary added masses and elastic supports. The examples, that are analyzed, demonstrate the high precision and fast convergence speed of the method. Further study of the timesaving method for the dynamic characteristics of symmetrical beam and the symmetry of mode shape should be developed. Eventually, the effects of elastic supports and added masseson dynamic characteristics of the three-span non-uniform beam bridge are reported.

        The high Curie temperature and long-range ferromagnetism in Mn-doped 3C-SiC: a study using first-principles calculation

        Magnetic semiconductors with high Curie temper (TC) and inherent ferromagnetism, which not only provide a data-processing function via semiconductors’ properties, but also offer a data-storage function through intrinsic ferromagnetism, have become key materials in the field of spintronics. We studied the electronic structures and magnetic properties of Mn-doped cubic (3C) SiC using first-principles calculation. The calculated results revealed that MnSi and MnC substitutions can introduce 3 μB and 1 μB magnetic moments into SiC, respectively, which are both localized and extended. The extended tails of the moments indicate a significant spatial extension of the spin-polarized electron states. The interaction between the long tails, which can mediate long-range ferromagnetic (FM) coupling up to 8.55 Å, and is very different from common exchange, in which the ferromagnetism decreases sharply with increasing distance between the moments, and is hard to maintain to the next-neighbor order due to the limited spatial spin-electrons distribution. The distributions of magnetic moments become more and more localized or extended as compressive or tensile strains are increased, but FM interactions under compressive and tensile strains are weaker than those in undeformed structure. The FM coupling mechanism is explained and discussed in detail using p–d electron exchange. These results can provide a routine for engineering FM semiconductor of 3C-SiC.

        Characterization of Ecdysteroid UDP-Glucosyltransferase Gene Promoter from Bombyx mori Nucleopolyhedrovirus

        Bombyx mori nucleopolyhedrovirus (BmNPV) ecdysteroid UDP-glucosyltransferase gene (egt) promoter fragments of different lengths were amplified from BmNPV ZJ-8 genomic DNA by PCR. Reporter plasmids pBmegt542-luc, pBmegt309-luc and pBmegt159- luc with luciferase (luc) driven by egt promoters were constructed. Both in vitro and in vivo expressions showed that BmNPV egt promoter activity requires the transactivation of viral factor(s), and expression of luc was detected earliest at 24 hrs post infection (pi). BmNPV ZJ-8 homologous region 3 (hr3) increased the expression of luc by over 1,600-fold. Molting hormone of 1.0 ~ 2.0 mg/ml can dramatically down regulate expression of luc. Juvenile hormone analogue of 0.5 ~ 2.0 mg/ml increased expression of luc by 145.8% to 75.7%. Deletion assay revealed that the promoter fragment of 159 bp contains the basal promoter structure; Promoter fragments of 309 bp and 542 bp showed similar but much higher transcriptional activities than that of 159 bp, suggesting that nucleotide from -159 to -309 nt upstream the translation initiation site harbors the main cis-acting elements.
