        배양 조건이 Candida albicans의 phospholipase 생성에 미치는 영향

        Background : The dimorphic yeast, Candida albicans, is considered as a dangerous opportunistic pathogen in immunocompromised hosts. Several phospholipases of C. albicans are known to be secreted into the culture medium. Phospholipases have been proposed as a virulence factor in the pathoenesis of Candida infections. Objective : In order to investigate enzyme production, we examined culture condition of secreted phospholipase production from C. albicans. Methods : C. albicans ATTCC 10231 was cultivated in various media at 37℃ for 3 days. Phospholipase activity was measured by fatty acid soap precipitaiton in plate containing 0.04T lecithin, 0.1 M citrate buffer, pH 4.2 and 1.5% noble agar. Results : Phospholipase was highly induced when C. albicans was cultivated in broth medium(containing glucose 2%, albumin 0.2% and Fe^++ ion 0.01%) and Saboulaud's dextrose agar supplemented with 0.01% sodium deoxycholate. Conclusion : Highly induction of secreted phospholipase by albumin from C. albicans may be play an importans role in tissue invasion in the pathogenesis of C. albicans.

      • 항이디오타입 종양 백신

        Anti-idiotypic antibodies (Ab2) that bind to the antigen-combining sites(paratopes) of anti-tumor antibodies (Ab1) may functionally and structurally mimic the tumor antigen defined by the Ab1. Thus, the anti-idiotypic antibodies resembling the original tumor antigen are called internal images of the tumor antigen and can be used as surrogate antigens for active specific immunotherapy. The advantages of Ab2 vaccines over conventional antigen vaccines are their high specificity, safety, ease of production, and potential to break immune tolerance to tumor antigen. The basic mechanism for immune tolerance breakage by Ab2 vaccines is a kind of cross-reaction between tumor antigen and Ab2 at T cell level. Anti-idiotypic antibodies have been shown to induce antigen-specific humoral and cellular immune reponses in experimental animals and cancer patients. In this paper, we will describe the idiotypic network, the advantages of Ab2 vaccines over conventional antigen vaccines, the mechanism of action of Ab2 vaccines, and the preclinical and clinical trials of active specific immunotherapy with Ab2 vaccines.

      • 메치실린 내성 황색포도알균의 신속한 검출을 위한 multiplex PCR

        Methicillin-resistant Staphylococcus aureus (MRSA) is a common cause of nosocomial infections as well as community acquired infections. The rapid identification of MRSA is very important for the appropriate antibiotic therapy, and MRSA must be differentiated from coagulase-negative staphylococci, which may be members of normal microbial flora. However, the conventional phenotypic methods for MRSA detection often lack sensitivity, because of the hetrogeneous expression of methicillin resistance and the dependence of resistance expression on the test conditions. In this study, we developed a multiplex PCR (MPCR) assay for rapid and accurate detection of MRSA. Two pairs of primers were used, which yielded two PCR products. One PCR product is a 310-bp mecA-based PCR fragment specific for methicllin resistance, and the other is a 108-bp S. aureus-specific DNA fragment. The multiplex PCR developed in this study showed excellent specificity and sensitivity for MRSA detection. When three methods for DNA extraction from MRSA were compared, lysostaphin methods and bead beating methods were more efficient than the heating methods. Therefore, the MPCR using two pairs of primers is thought to be a very sensitive and specific tool for rapid detection of MRSA, especially when the DNA of MRSA is extracted by bead beating method.

      • Labdane 계열 혈관신생억제 분자의 합성

        Angiogenesis, the process of new blood capillary formation, is an important component of a number of serious disease including cancer growth and metastasis, diabetic retinopathy, rheumatoid arthritis, and many others. Coronarin A, isolated from rhizomes of the Brazilian antirheumatic medicinal plant, and synthetic related compounds showed a significant cytotoxic effect against V-79 cell and sarcoma 180 ascites in Mice. In order to evaluate antiangiogenic effect of trans decaline type labdane compounds, 3, 4a, 5, 6 and 11 were synthesized from versatile chiral building blocks scalreol 1 and sclareolide 2 with appropriate synthetic methods. All tested materials have a mild growth inhibition effect on human endothelial cell line, ECV 304. We concluded that high antiangiogenic effect of coronarin A was caused by furan aromatic functional group on C-l2 position.

      • 메치실린 내성 포도상구균에 대한 SK-302의 시험관내 및 생체내 효과

        SK-302 is the active compound purified from culture filtrate of a streptomyces species (streptomyces sp. 302). The in vitro and in vivo antibacterial activities of SK-302 were evaluated against 6 clinically isolated MRSA strains and compared with the activities of vancomycin. SK-302 had node in virto antistaphylocoal activity stronger than that of vancomycin exhibiting MIC's, while vancomycin showed very weak 4 to 8fold lower than that of vancomycin. In SK-302 showed killing activity at the MIC, so SK-302 was superior to vancomycin. Furthermore, in acute systemic infections in mice, the ED_50s of SK-302 against infections by high-resistance MRSA were 30 to 40-feold less than that of vancomycin, therefore SK-302 showed potent in vive therapeutic efficacy against MRSA infection, These results suggest that SK-302 has excellent in virto in and in vivo activities against high-resistance MRSA.

      • 저산소 스트레스에 노출된 생쥐의 면역기능 변동에서 IL-4의 역활

        This study was aimed to investigate the role of IL-4 in the alterations of immune function in mice after chronic, repeated exposure of different levels of hypoxic stress. Four experimental groups were studied: one for home cage control (NH), others for environmental control (20%) and hypoxia-exposed groups (15% or 10%). Different levels of normoxic or hypoxic environment (20%, 15%, or 10% of oxygen will balanced nitrogen, respectively) were subjected to adult male BALB/c mice daily for an hour, 5 days per week, over 10 weeks. At the end of hypoxic exposure period, sera of the mice were taken and their IL-4 levels were measured by a sandwich ELISA method. The changes of body weight of the animals showed significantly lower increase in the 10% group, suggesting that the repeated hypoxic exposure worked as a stressful environment to them. The IL-4 levels, however, showed no differences among the experimental groups. These results indicate that IL-4 may not play a significant role in the mediation of stress effects to immunologic outcomes in this stress paradigm, and that studies about the role of Th1-type cytokines may be necessary fur further investigation.

        방선균 WCM-9가 생산하는 Polyene 항진균성 항생물질(AF1)의 생물학적 성상

        Background: Pathogenic fungi infect humans, especially immunocompromised patients, with superficial or deeply invasive pattern. In the past 20 years, fungal infections have been increased dramatically resulted by increment of organ transplantation, cancer, AIDS patients, or use of broad-spectrum antibacterial agents. Fungal infections are now important causes of morbidity and mortality of hospitalized patients. but there is no effective antifungal antibiotics as well as antibacterial antibiotics Objective: Effective new antifungal antibiotics are needed for the treatment of mycosis. So in an effort to develop effective antifungal antibiotics, we screened over 600 isolates of Streptomyces spp. from soil. Methods: Antifungal producing strain was selected using disk diffusion method, An antifungal substance (AF1) was purified with ethyl acetate extraction, silica gel column chromatography and reverse phase HPLC. MICs of AF1 were detected by agar dilition method. Results: The compound showed UV maxima of 307, 321, 340, 359 nm indicating methylpentaene. Minimum inhibitory concentrations of the AF1 were 3.7 ㎍/ml against mold, and 3.7 - 7.4 ㎍/ml against Candida species. AF1 was also active against Cryptococcus neoformans, with MIC of 0.9 ㎍/ml. The concentration of AF1 for K^+ ion release from human red blood cell and hemolysis were 5 ㎍/ml. Conclusion: The antibiotic purified from culture broth of Streptomyces sp. WCM-9 was a polyene antifungal antibiotic which have broad spectrum antifungal activity.

        The Effect of Rabbit Serum on the Expression of Cell Surface Hydrophobicity in Candida albicans

        세포표면 소수성은 부착에 중요한 역할을 한다고 알려져 있으며 여러 가지 환경조건에 따라 세포표면 소수성의 표현이 달라진다. 그러나 혈청에 의해 유도되는 세포표면 소수성이 다른 조건에서 유도되는 세포표면 소수성과 같은지는 보고되지 않았으므로 혈청에 의한 칸디다균의 세포표면소수성을 관찰하였다. 토끼 혈청과 YEPD 배지에서 칸디다균을 37℃에서 배양하였을 때 초기 1시간 동안 세포표면 소수성이 급격하게 증가하다가 이후 서서히 감소하였다. 혈청을 희석하였을 때와 포도당을 첨가하여 배양하였을 때 칸디다균의 세포표면 소수성은 큰 차이가 없었으나 혈청을 많이 회석할수록 소수성이 높게 유지되었다. 칸디다균을 단백질분해효소와 lyticase로 처리하였을 때 칸디다균의 세포표면 소수성은 감소하였다. 칸디다균의 세포벽을 lyticase로 처리하여 전기영동으로 분석한 결과 단백질 양상이 소수성 균주간에도 모두 일치하지 않았다. 이러한 결과는 혈청에 의해 유도되는 세포표면 소수성과 YEPD 배지에서 유도되는 세포표면 소수성이 같지 않음을 의미한다.
