      • Alkaloids from Beach Spider Lily (Hymenocallis littoralis) Induce Apoptosis of HepG-2 Cells by the Fas-signaling Pathway

        Alkaloids are the most extensively featured compounds of natural anti-tumor herbs, which have attracted much attention in pharmaceutical research. In our previous studies, a mixture of major three alkaloid components (5, 6-dihydrobicolorine, 7-deoxy-trans-dihydronarciclasine, littoraline) from Hymenocallis littoralis were extracted, analyzed and designated as AHL. In this paper, AHL extracts were added to human liver hepatocellular cells HepG-2, human gastric cancer cell SGC-7901, human breast adenocarcinoma cell MCF-7 and human umbilical vein endothelial cell EVC-304, to screen one or more AHL-sensitive tumor cell. Among these cells, HepG-2 was the most sensitive to AHL treatment, a very low dose ($0.8{\mu}g/ml$) significantly inhibiting proliferation. The non-tumor cell EVC-304, however, was not apparently affected. Effect of AHL on HepG-2 cells was then explored. We found that the AHL could cause HepG-2 cycle arrest at G2/M checkpoint, induce apoptosis, and interrupt polymerization of microtubules. In addition, expression of two cell cycle-regulated proteins, CyclinB1 and CDK1, was up-regulated upon AHL treatment. Up-regulation of the Fas, Fas ligand, Caspase-8 and Caspase-3 was observed as well, which might imply roles for the Fas/FsaL signaling pathway in the AHL-induced apoptosis of HepG-2 cells.

        Cytotoxicity of polymethyl methacrylate cement on primary cultured metastatic spinal cells

        Polymethyl methacrylate (PMMA) bone cement has been commonly used for percutaneous injection into collapsed vertebral bodies due to malignant tumor. The purpose of this study was to investigate the possible mechanisms of PMMA’s cytotoxcity on primary cultured spinal metastastic cells (SMCs) in vitro. PMMA specimens were prepared in standard discs made of dough and polymerization stages, and the eluates were prepared following the ISO standard. Primary SMCs were obtained and isolated from 7 patients with spinal metastatic tumors undergoing vertebroplasty. Primary cultured SMCs were treated with PMMA specimens of different stages for 24 h, or co-cultured with extracted medium for successive 3 days. The temperatures in two locations from cement discs were recorded by K-type thermocouples. Furthermore, cell proliferation, apoptosis and cycles were determined by MTT and flow cytometry, respectively. The modulation of apoptotic proteins (bcl- 2, bax, caspase-3, caspase-8, Fas and PARP) and cell cycle proteins (cyclin D1, P21 and P27) were analyzed by western blot and real time PCR. The results of this study demonstrated that PMMA treatment was able to suppress SMCs proliferation, induce cell apoptosis and inhibit cell cycle arrest when compared to the control group in vitro (P<0.05). Simultaneously, the temperature recorded at the periphery of the PMMA specimen was not high enough to casue thermal injury. Furthermore, molecular markers of apoptosis including Fas, caspase-3 and caspase-9 activated, and Bcl-2/Bax dysregulated in the SMCs with PMMA stimulation. In addition, PMMA treatment showed decreased expression of cyclin D1 that induces cell cycle and increased epxression of inhibitory protein P21, with no significant difference of P27 expression. In summary, the present study confirms that PMMA cement can compromise the vitality and apoptosis of SMCs. This cytotoxic effect may be regulated by the biomarkers Fas, Bcl-2, Bax, caspase-3, caspase-9, cyclin D1 and P21, but not on account of thermal damage.

Compounds 1 and 2, designated as monan chosterols A abd B, resoectubely, represent the first example of steriods possessing the bicyclo[4.3.1] A/Brubg ststem from a natural source, Compounds 1-3 were investigated for their anti-inflammatory activity by evaluating their inhibitory effects on the mRna expression of 1L-6, TNF-a and COX-2 in the LPS-stimulated murine RAW264.7 macrophage cells, Compounds 2 and 3 exhibited significant inhibitpry effects on the mRNA expression of 1L-6 without notable cytotoxocotu tp tje cells in a dose-dependent manner.

        Wogonin suppresses the LPS-enhanced invasiveness of MDA-MB-231 breast cancer cells by inhibiting the 5-LO/BLT2 cascade

        <P>Wogonin, a naturally occurring bioactive monoflavonoid isolated from <I>Scutellariae</I> radix (roots of <I>Scutellariae baicalensis</I> Georgi), has known anticancer effects. However, the molecular signaling mechanism by which wogonin inhibits invasiveness in breast cancer cells remains unclear. In the present study, it was observed that wogonin exerted an inhibitory effect on the lipopolysaccha-ride (LPS)-enhanced invasiveness of MDA-MB-231 cells. In addition, wogonin inhibited the synthesis of interleukin-8 (IL-8) and matrix metallopeptidase-9 (MMP-9), which are critical for promoting invasiveness in MDA-MB-231 cells. Wogonin also suppressed the expression of leukot-riene B4 receptor 2 (BLT2) and the synthesis of its ligand, by inhibiting 5-lipoxygenase (5-LO) in LPS-stimulated MDA-MB-231 cells. Notably, wogonin attenuated the production of IL-8 and MMP-9 by inhibiting the BLT2/extracellular signal-regulated kinase (ERK)-linked cascade. Finally, <I>in vivo</I>, LPS-driven MDA-MB-231 cell metastasis was markedly suppressed by wogonin administration. Overall, the present results suggested that wogonin inhibited the 5-LO/BLT2/ERK/IL-8/MMP-9 signaling cascade and demonstrated that this cascade may be an important target through which wogonin exerts its anticancer effects in breast cancer.</P>

        The Effect of Inhibition of Uncaria rhynchophylla as an Inhibitor of Melanogenesis and an Antioxidant in B16F10 Melanoma Cells

        최근 모든 연령대의 사람들은 미용적인 이유로 더 밝은 피부를 원하고 있으며, 천연 제품은 화학적으로 합성된 화합물보다 더 많은 관심을 받고 있다. 조구등은 아시아에서 전통 한약재로 널리 사용되어 왔다. 새로운 피부미백제를 찾기 위해 본 연구에서는 조구등의 항산화 활성과 잠재적인 tyrosinase 억제 작용을 확인하였다. 조구등을 70% 에탄올로 추출하여 항산화 활성을 분석하고 tyrosinase 활성과 melanin 합성에 미치는 영향을 평가했다. 총 mRNA 발현은 RT-PCR을 사용하였다. 그 결과 조구등 추출물은 B16F10 세포에서 뛰어난 항산화 능력과 상당한 수준의 폴리페놀 및 플라보노이드 화합물을 함유하였다. 또한, 세포 내 tyrosinase 활성을 억제하고 처리된 세포에서 melanin의 양을 감소시켰다. Tyrosinase의 mRNA 발현은 1 mg/ml 농도에서 현저히 감소하였다. 이와 같은 결과는 조구등이 미백 효과가 있는 화장품의 천연 소재로 사용될 수 있는 높은 잠재력을 가지고 있음을 시사한다. Many people of all ages wish to have lighter skin for cosmetic reasons, and natural products attract more attention than chemically synthesized compounds. Uncaria rhynchophylla is widely used in Asia as a traditional herbal medicine. In order to find novel skin whitening agents, the present study evaluated the antioxidant activity and potential tyrosinase-inhibiting properties of U. rhynchophylla. Specifically, this study analyzed the antioxidant capacity of a 70% ethanolic extract of U. rhynchophylla as well as its effects on tyrosinase activity and melanin synthesis. Total mRNA levels were examined using reverse transcription polymerase chain reaction. The results revealed that U. rhynchophylla extracts exhibit great antioxidant capacity and significant levels of polyphenol and flavonoid compounds. U. rhynchophylla extracts can also powerfully inhibit tyrosinase activity. This same capacity was observed in melanoma B16F10 cells; that is, U. rhynchophylla extracts suppressed intracellular tyrosinase activity and reduced the amount of melanin in treated cells. In addition, a 1 mg/ml concentration of U. rhynchophylla extract significantly reduced the mRNA expression levels of tyrosinase. U. rhynchophylla extracts decrease tyrosinase and inhibit melanogenesis in B16F10 cells. This finding suggests that U. rhynchophylla has great potential as a natural whitening agent in skincare products.

        Resilient Control for Wireless Networked Control Systems

        This paper proposes an approach to quantify the concept of resiliency in terms of Quality of Control (QoC) of a control system. Based on this concept, an intelligent resilient control algorithm (RCA) is presented for wireless networked control systems (WNCS) to maintain operational normalcy in face of wireless interference incidents, such as Radio Frequency (RF) jamming and signal blocking. The proposed algorithm closes the control loop with wireless sensors feasible by significantly increasing control system’s tolerance to data packet loss and delay caused by wireless interference. The proposed algorithm, along with other well developed wireless technologies, has the potential to enable implementing wireless sensors widely in the next generation of industrial automation and control sys-tems.

      • Suppressive Effect of Sinomenine Combined with 5-Fluorouracil on Colon Carcinoma Cell Growth

        It is reported that sinomenine (SIN) and 5-fluorouracil (5-FU) both are effective for colon cancer, but their cooperative suppressive effects and toxicity remain to be clarified in detail. This study aimed to determine suppressive effects and toxicity of sinomenine (SIN) plus 5-fluorouracil (5-FU) on LoVo colon carcinoma cells in vitro and in vivo. CCK-8, Hoechst 33258 staining and an annexin V-FITC/PI apoptosis kit were used to detect suppressive effects. Western blotting was applied to investigate the essential mechanism underlying SIN and 5-FU-induced apoptosis. SIN or 5-FU or both were injected into nude mice, and then suppressive effects and side effects were observed. SIN plus 5-FU apparently inhibited the proliferation of LoVo cells and induced apoptosis. Moreover the united effects were stronger than individually (p<0.05). The results of annexin V-FITC/PI staining and Hoechst 33258 staining showed that the percentage of apoptotic cells induced by SIN and 5-FU combined or alone was significantly higher than the control group (p<0.05). Expression of Bax and Bcl-2 was up-regulated and down-regulated respectively. SIN or 5-FU significantly inhibited effects on the volume of tumour xenografts and their combined suppressive effects were stronger (p<0.05). No obvious side effects were observed. It was apparent that the united effects of SIN and 5-FU on the growth of colorectal carcinoma LoVo cells in vitro and in vivo were superior to those using them individually, and it did not markedly increase the side effects of chemotherapy.

        Optimization of image reconstruction method for dual-particle time-encode imager through adaptive response correction

        Time-encoded imagers (TEI) are important class of instruments to search for potential radioactive sources to prevent illicit transportation and trafficking of nuclear materials and other radioactive sources. The energy of the radiation cannot be known in advance due to the type and shielding of source is unknown in practice. However, the response function of the time-encoded imagers is related to the energy of neutrons or gamma-rays. An improved image reconstruction method based on MLEM was proposed to correct for the energy induced response difference. In this method, the count vector versus time was first smoothed. Then, the preset response function was adaptively corrected according to the measured counts. Finally, the smoothed count vector and corrected response were used in MLEM to reconstruct the source distribution. A one-dimensional dual-particle time-encode imager was developed and used to verify the improved method through imaging an AmeBe neutron source. The improvement of this method was demonstrated by the image reconstruction results. For gamma-ray and neutron images, the angular resolution improved by 17.2% and 7.0%; the contrast-to-noise ratio improved by 58.7% and 14.9%; the signal-to-noise ratio improved by 36.3% and 11.7%, respectively.

        Biphasic effects of TGFβ1 on BMP9-induced osteogenic differentiation of mesenchymal stem cells

        We have found that the previously uncharacterized bone morphogenetic protein-9 (BMP9) is one of the most osteogenic factors. However, it is unclear if BMP9 cross-talks with TGFβ1 during osteogenic differentiation. Using the recombinant BMP9 adenovirus, we find that low concentration of rhTGFβ1 synergistically induces alkaline phosphatase activity in BMP9-transduced C3H10T1/2 cells and produces more pronounced matrix mineralization. However, higher concentrations of TGFβ1 inhibit BMP9-induced osteogenic activity. Real-time PCR and Western blotting indicate that BMP9 in combination with low dose of TGFβ1 potentiates the expression of later osteogenic markers osteopontin, osteocalcin and collagen type 1 (COL1a2), while higher concentrations of TGFβ1 decrease the expression of osteopontin and osteocalcin but not COL1a2. Cell cycle analysis reveals that TGFβ1 inhibits C3H10T1/2 proliferation in BMP9-induced osteogenesis and restricts the cells in G0/G1 phase. Our findings strongly suggest that TGFβ1 may exert a biphasic effect on BMP9-induced osteogenic differentiation of mesenchymal stem cells. [BMB Reports 2012; 45(9): 509-514]
