Objective: The standard dose for pegylated liposomal doxorubicin (PLD) is 50 mg/m2every 4weeks. While 40 mg/m2has recently been used in clinical practice, evidence supporting thisuse remains lacking. Methods: This phase III randomized, non-inferiority study compared progression free survival (PFS) for patients with platinum-resistant ovarian carcinoma between anexperimental arm (40 mg/m2PLD) and a standard arm (50 mg/m2PLD) until 10 courses, disease progression or unacceptable toxicity. Eligible patients had received ≤2 prior lines. Stratification was by performance status and PFS of prior chemotherapy (<3 months versus ≥3months). The primary endpoint was PFS and secondary endpoints were overall survival (OS),toxicity profile, clinical response and tolerability. The total number of patients was 470. Results: The trial was prematurely closed due to slow recruitment, with 272 patients randomizedto the experimental arm (n=137) and standard arm (n=135). Final analysis was performed with234 deaths and 269 events for PFS. In the experimental arm vs. standard arm, median PFS was4.0 months vs. 4.0 months (hazard ratio [HR]=1.065; 95% confidence interval [CI]=0.830–1.366)and median OS was 14.0 months vs. 14.0 months (HR=1.078; 95% CI=0.831–1.397). Hematologictoxicity and oral cavity mucositis (≥grade 2) were more frequent in the standard arm than in theexperimental arm, but no difference was seen in ≥grade 2 hand-foot skin reaction. Conclusion: Non-inferiority of 2 PLD dosing schedule was not confirmed because the trialwas closed prematurely. However, recommendation of dose reduction of PLD should bebased both on efficacy and safety. Trial Registration: UMIN Clinical Trials Registry Identifier: UMIN000003130
MODELING OF THE ZODIACAL EMISSION FOR THEAKARI/IRC MID-INFRARED ALL-SKY DIFFUSE MAPS
Kondo, Toru,Ishihara, Daisuke,Kaneda, Hidehiro,Nakamichi, Keichiro,Takaba, Sachi,Kobayashi, Hiroshi,Ootsubo, Takafumi,Pyo, Jeonghyun,Onaka, Takashi American Astronomical Society 2016 The Astronomical journal Vol.151 No.3

The zodiacal emission, which is the thermal infrared (IR) emission from the interplanetary dust (IPD) in our solar system, has been studied for a long time. Nevertheless, accurate modeling of the zodiacal emission has not been successful to reproduce the all-sky spatial distribution of the zodiacal emission, especially in the mid-IR where the zodiacal emission peaks. Therefore, we aim to improve the IPD cloud model based on Kelsall et al., using the AKARI 9 and 18 mu m all-sky diffuse maps. By adopting a new fitting method based on the total brightness, we have succeeded in reducing the residual levels after subtraction of the zodiacal emission from the AKARI data and thus in improving the modeling of the zodiacal emission. Comparing the AKARI and the COBE data, we confirm that the changes from the previous model to our new model are mostly due to model improvements, but not temporal variations between the AKARI and the COBE epoch, except for the position of the Earth-trailing blob. Our results suggest that the size of the smooth cloud, a dominant component in the model, is about 10% more compact than previously thought, and that the dust sizes are not large enough to emit blackbody radiation in the mid-IR. Furthermore, we detect a significant isotropically distributed IPD component, owing to an accurate baseline measurement with AKARI.

ABSTRACT

Antigen-specific adhesion between T cells and antigen-presenting cells (APC) during the formation of the immunological synapse (IS) is mediated by LFA-1 and ICAM-1. Here, LFA-1–ICAM-1 interactions were measured at the single-molecule level on supported lipid bilayers. High-affinity binding was detected at low frequencies in the inner peripheral supramolecular activation cluster (SMAC) zone that contained high levels of activated Rap1 and kindlin-3. Rap1 was essential for T cell attachment, whereas deficiencies of ste20-like kinases, Mst1/Mst2, diminished high-affinity binding and abrogated central SMAC (cSMAC) formation with mislocalized kindlin-3 and vesicle transport regulators involved in T cell receptor recycling/releasing machineries, resulting in impaired T cell-APC interactions. We found that NDR1 kinase, activated by the Rap1 signaling cascade through RAPL and Mst1/Mst2, associated with and recruited kindlin-3 to the IS, which was required for high-affinity LFA-1/ICAM-1 binding and cSMAC formation. Our findings reveal crucial roles for Rap1 signaling via NDR1 for recruitment of kindlin-3 and IS organization.

Structural analysis of inhomogeneity-free poly(ethylene glycol)-poly(dimethylsiloxane) (PEG PDMS) amphiphilic conetwork gels has been performed by the complementary use of small-angle X-ray and neutron scattering. Because of the hydrophobicity of PDMS units, the PEG-PDMS gels exhibit a microphase-separated structure in water. Depending on the volume fraction of; PDMS, the microphase-separated structure varies from core-shell to lamellar. The obtained X-ray and neutron scattering profiles are reproduced well using a core-shell model together with a Percus-Yevick structure factor when the volume fraction of PDMS is small. The domain size is much larger than the size of individual PEG and PDMS unit, and this is explained using the theory of block copolymers. Reflecting the homogeneous dispersion conditions in the as-prepared state, scattering peaks are observed even at a very low PDMS volume fraction (0.2%). When the volume fraction of PDMS is large, the microphase-separated structure is lamellar and is demonstrated to be kinetically controlled by nonequilibrium and topological effects.

The upper Kayaoka sheet in the Atsumi area, Northeast Japan, has an alkaline-basaltic composition. The modal olivine and Ni enrichment in the lower part of the sheet indicates the accumulation of early formed olivine crystals. Overall, the average aspect ratio of plagioclase increases from the central part of the sheet toward both the top and bottom margins. The average aspect ratios of plagioclase and calculated crystallisation times at each height of the sheet, except for the lowermost margin, are roughly plotted near the regression line drawn based on those from many sills in previous research. However, we observed that the average aspect ratios of plagioclase in some horizons of the central part of the sheet exhibit an offset from the regression line. The olivine analcite dolerite in the central part of the sheet is rich in zeolite amygdules and glassy mesostasis with a quench texture. These observations suggest that the cooling rate in the central part of the sheet was faster compared with assuming conductive cooling only. A plausible mechanism for the rapid cooling at the centre of the sheet is heat transport by the convection of seawater that has infiltrated the columnar joint.
MODELING OF THE ZODIACAL LIGHT FOR THE AKARI MID-IR ALL-SKY DIFFUSE MAPS

The AKARI 9 and 18 $\mu$m diffuse maps reveal the all-sky distribution of the interstellar medium with relatively high spatial resolution of $\sim$6$''$. The zodiacal light is a dominant foreground component in the mid-infrared. Thus, removal of the zodiacal light is a critical issue to study low surface brightness Galactic diffuse emission. We carried out modeling of the zodiacal light based on the Kelsall model which is constructed from the COBE data. In the previous study, only a time-varying component of the zodiacal light brightness was used for determination of the model parameters. However, there remains a residual component of the zodiacal light around the ecliptic plane even after removal with the model. Therefore, instead of using a time-varying component, we use the absolute brightness of the zodiacal light and we find that the new model can better remove the residual component. As a result, the best-fit model parameters are changed from those in the previous study. We discuss the properties of the zodiacal light based on our new result.
