The vascular actions of local anesthetics are important in determining the uptake and distribution of these agents from their site of injection as well as influencing their hemodynamic effects once absorbed. Because of the importance of the endothelium in determining of modulating the vascular response of a wide variety of agents, cumulative dose-dependent vasular effects of lidocaine, mepivacaine and bupivacaine on isolated rings of thoracic aorta in normotensive rats(NTR) and spontaneously hypertensive rats(SHR) were studied in the presence and absence of intact endothelium. The results were as follows ; The body weight of NTR and SHR averaged 274.71±55.80(N = 38) and 241.43±17.73gm(N = 18) and mean arterial pressure was 74.4l±3.60 and 129.34±2.89mmHg respectively. The mean absolute value of the contraction induced by 5×l0^(-6) M phenylephrine was 3.27±0.98(N = 18) and 2.3l±50.64gm(N = 18) with intact endothelium and 3.12±0.92 and 2.46±0.87 gm without intact endothelium in aortic rings of NTR and SHively. In the response to local anesthetics in preparation with resting tension(1.0 gm), lidocaine and mepivacaine in concentration of 10^(-3) to 1.25×10^(-2) M not produced dose dependent contraction in aortic ring with intact endothelium from NTR. but bupivacaine produced dose-dependent contraction in aortic rings with intact endothelium from NTR. In the aortic rings from NTR and SHR previously contracted with phenylephrine, lidocaine in contraction of 10^(-3) to 1.25×10^(-2) M caused dose related relaxation in aortic rings with or without endothelium but in concentration of 10^(-3) to 510^(-3) M, aortic rings with endothelium were more relaxed than those af without endothelium in NTR. In SHR, aortic rings without endothelium in concentration of 5×10^(-3) to 1.25×10 M were more significantly relaxed than those of with endothelium. In aortic rings from NTR previously contracted with phenylephrine, mepivacaine caused dose-related relaxation, which was more profound in SHR. In aortic rings with endothelium froR previously contracted with phenylephrine, bupivacaine in concentration of 10^(-3) to 1.5×10^(-3) M caused a relaxation and in concentration of 2.5×10^(-3) to 7.5×10^(-3) M and 1.25×10^(-3) M caused a relaxation again. But in the aortic rings without intact endothelium, bupivacaine caused dose-related relaxation. In the aortic rings without intact endothelium, bupivacaine caused dose-related relaxation in NTR. In the aortic rings from SHR previously contracted with phenylephrine, bupivacaine caused dose-related relaxation, which was more profound than those of NTR. The local anesthetics appear to exert their relaxant effect on endothelium independently and more profoundly in SHR.

Background: Recently, as production costs have been increasing owing to rising feed prices worldwide, shortening the age of slaughter has been recognized as a way to increase farm income. In Korea, the raising period for Hanwoo steers is over 31 months with the delay of weaning and castration stated as one of the reasons for the increase in the raising period. Thus far, studies on age of weaning and castration have been conducted individually, and there have been no studies on the combined effects of weaning and castration ages on the growth performance and carcass characteristics in Hanwoo steers. Methods: Weaning ages were calculated at 80 or 130 days of age, and castration ages were calculated at 90 days and 180 days of age. Calves were allocated to one of the four treatment groups: W80C90 (weaning at 80 days of age and castration at 90 days of age), W80C180, W130C90, and W130C180. Results: For the entire experimental period, weaning and castration ages did not significantly affect growth performance of Hanwoo steers. In addition, weaning and castration ages did not affect the overall yield and quality traits of carcass in Hanwoo steers. Conclusion: Weaning and castration ages had small effects on growth performance and carcass characteristics in Hanwoo steers. Therefore, the early weaning and castration ages are recommended to reduce the slaughter age without any negative effects on meat quality grade.
제주흑우, 한우 및 수입 소 품종에서 새로운 indel 마커의 다형성과 대립인자 분포

본 연구는 소 유전자 database들에 대한 사전 비교연구에서 발견된 삽입/결실(indel) marker들의 다형성과 각각의 유전자형의 분포를 확인하고자 수행하였다. 먼저, 소의 유전체 서열과 발현서열표식(EST) database 간의 생물정보학적 비교를 통해 전체 51 종의 indel marker들을 검출하였다. 이 중에서 42 종을 평가하여 최종적으로 9 종의 정보력이 있는 marker들을 집단분석을 위해 선발하였다. 각각의 marker들에 대한 염기서열을 재분석하였으며, marker의 다형성을 한국 재래소 품종인 한우와 제주흑우(JBC), Holstein, Angus, Charolais, Hereford 등 6 품종에서 조사하였다. 본 연구에서 이용한 소 6 품종은 8 종의 marker들에 대해 다형성을 나타내었으나, Indel_15의 경우 Holstein과 Charolais에서 다형성이 발견되지 않았다. JBC 집단에 대한 분석에서는 관찰된 이형 접합자 빈도는 HW_G1 (0.600)에서 가장 높고, Indel_29 (0.274)에서 가장 낮았다. Marker에 대한 다형정보량의 수준은 HW_G4 (0.373)에서 가장 높고, Indel_6 (0.305)에서 가장 낮은 수준을 보였다. 본 연구에서 조사한 새로운 indel marker들은 특히 제주흑우 집단의 생산성 향상을 위한 분자육종 체계의 개발뿐만 아니라 친자확인이나 생산이력추적을 위한 유전정보를 제공하는데 유용할 것으로 기대된다. The aim of this study was to screen the polymorphisms and distribution of each genotype of insertion/deletion (indel) markers which were found in a preliminary comparative study of bovine genomic sequence databases. Comparative bioinformatic analyses were first performed between the nucleotide sequences of Bovine Genome Project and those of xpressed sequence tag (EST) database, and a total of fifty-one species of indel markers were screened. Of these, forty-two indel markers were evaluated, and nine informative indel markers were ultimately selected for population analysis. Nucleotide sequences of each marker were re-sequenced and their polymorphic patterns were typed in six cattle breeds: Holstein, Angus, Charolais, Hereford, and two Korean native cattle breeds (Hanwoo and Jeju Black cattle). Cattle breeds tested in this study showed polymorphic patterns in eight indel markers but not in the Indel-15 marker in Charolais and Holstein. The results of analysis for Jeju Black cattle (JBC) population indicated an observed heterozygosity (Ho) that was highest in HW_G1 (0.600) and the lowest in Indel_29 (0.274). The PIC value was the highest in HW_G4 (0.373) and lowest in Indel_6 (0.305). These polymorphic indel markers will be useful in supplying genetic information for parentage tests and traceability and to develop a molecular breeding system for improvement of animal production in cattle breeds as well as in the JBC population.

<P><B>Abstract</B></P> <P>A sensitive and specific swarm primer-based reverse transcription loop-mediated isothermal amplification (sRT-LAMP) assay for the detection of serotype O foot-and-mouth disease virus (FMDV) was developed and evaluated. The assay specifically amplified the <I>VP3</I> gene of serotype O FMDV, but did not amplify the <I>VP3</I> gene of other serotype FMDVs or any other viruses. The limit of detection of the assay was 10<SUP>2</SUP> TCID<SUB>50</SUB>/mL or 10<SUP>3</SUP> RNA copies/μL, which is 100 times lower than that of the RT-LAMP assay without swarm primers. The new assay is 10 times more sensitive than reverse transcription-polymerase chain reaction (RT-PCR) and is comparable to the sensitivity of real time RT-PCR (qRT-PCR). Evaluation of the assay using different serotypes of FMDV strains showed 100% agreement with the RT-PCR results. The previously reported serotype O FMDV-specific RT-LAMP assay did not detect 20 out of 22 strains of serotype O FMDVs, probably due to multiple mismatches between the primer and template sequences, showing that it is not suitable for detecting the serotype O FMDVs circulating in Pool 1 region countries, including Korea. In contrast, the developed sRT-LAMP assay with improved primers can rapidly and accurately diagnose serotype O FMDVs circulating in Pool 1 region countries and will be a useful alternative to RT-PCR and qRT-PCR.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A swarm primer-applied RT-LAMP assay was developed for the detection of serotype O FMDV. </LI> <LI> The assay was 10 times more sensitive than RT-PCR and comparable to the sensitivity of qRT-PCR. </LI> <LI> The swarm primer RT-LAMP assay showed 100% agreement with conventional RT-PCR. </LI> </UL> </P>
