Trichinella spiralis 感染이 마우스의 免疫反應에 미치는 影響

Male, IcR mice weighing 20 to 25 gm were infected experimentally by esophageal intubation of 300 Trichinella spiralis infective muscle larvae prepared by acid-pepsin digestion of infected mice. Delayed type hypersensitivity and hemagglutinin antibody reactions to unrelated antigen, sheep red blood cell (SRBC) were measured at 37 and 83 days after infection, employing footpad and ear swelling reactions and microtitration technique. All mice were sensitized i.v. with SRBC. It was found that DTH and hemagglutinin responses to SRBC were depressed in infected mice when observed at 37 and 83 days postinfection, and degree of immunosuppression was more profound in mice observed at 83 days than in those at 37 days. The secondary cell-mediated immune response of DTH to SRBC were suppressed in Trichinella - infected mice, indicating the development of immunological memory cell is affected. In addition, immunodepression to SRBC in mice infected with T. spiralis was eliminated by pretreating infected mice by subcutaneous injection of cyclosphosphamide(100㎎/㎏) 2 days prior to infection. These results suggest that the immuno suppression in trichinellosis can be due to suppressor T cells.
장티푸스균에 대한 Ampicillin, Chloramphenicol 및 Trimethoprim-Sulfamethoxazole의 단독 및 병합작용

Salmonella typhi H901 및 O901과 본 교실에서 환자로부터 분리한 S. typhi 4주를 공시하여 ampicillin(AP), chloramphenicol(CM) 및 trimethoprim-sulfame-thoxazole(TP-SX)의 단독 혹은 병합작용을 checkerboard 수기와 time kill 검사법으로 실지하였던 바, minimal inhibitory concentration(MIC)의 AP와 CM의 병합은 길항작용을, TP_Sx와 AP또는 CM의 병합은 대체적으로 상가 또는 상승작용을 나타냈다. 그러나 checkerboard 실험결과 나타난 길항작용과는 달리 MIC 이상 농도의 AP와 CM을 병합하여 time kill수기로 검사했을때는 상가작용을 나타냈다. 이상의 실험 결과는 장티푸스의 치료에 AP와 CM을 병합요법은 금기이나, TP-Sx와 AP 또는 CM의 병합요법은 유효함을 시사한다고 사료되었다. The effect of combination of chloramphenicol(CM) with ampicillin(AP) or trimethoprim-sulfamethoxazole(TP-SX) and the effect of combination of AP with TP-SX against standard strains, Salmonella typhi H_(901) and 4 clinical isolates of S. typhi was examined by using checkerboard technique and time kill curve at various combination of minimal inhibitory concentration(MIC) or above MIC of respective antibiotic Fractional inhibitory concentration indices, time kill curves or both demonstrated that the combination of AP with CM at their MIC or above MIC of either antibiotic produced antagonism against S. typhi H_(901) and Owl, and all of the isolates, but the combination of AP and CM at their higher concentration (above MIC) than MIC of either drug revealed additive effect against the same strains. The combination of AP with TP-SX at their MIC or above MIC was found to act synergistically against 2 strains of the isolates and was additive against all standard strains and 2 strains of the isolates. The combination of CM with TP-SX produced additive effects against all standard strains and 3 isolates, and was synergistic against one strain of the isolates. These results suggest that the combination of AP and CM may be contraindicated for the treatment of typhoid fever, and the combination of TP-SX with AP or CM may be recommended as combined antibiotic therapy for typhoid fever, and that combination chemotherapy deserves further study.
Generation of Monoclonal Antibody against Hepatitis B virus(HBV)Pre - S2 Peptide

Monoclonal antibody against HBV pre-S2 peptide was generated. Six clones among 70 hybridomas which were survived in HAT media produced anti pre-S2 antibody. All six clones has been identified to produce IgM-type monoclonal antibody which has kappa light chain after ELISA subtyping analysis. After purification of IgM monoclonal antibody, heavy and light chain of immunoglobulin has been identified whose molecular weights are 79KD and 26.5KD, respectively. These pre-S2 antibodies were confirmed using immunoblotting techniques. IgM type pre-S2 monoclonal antibody were bound to 55 amino acid pre-S2 antigen whose M.W. is 55KD.
Generation of Monoclonal Antibody Against Interleukin 2 Peptide

Interleukin 2 (IL-2) is a glycoprotein of 15500 daltons molecular weight produced by T-lymphocytes, which stimulates cells from hepatopoietic lineages which express specific IL-2 receptors. Monoclonal antibody against IL-2 peptide was generated 104 wells among 218 which were survived in HAT media produced anti IL-2 antibody. All 11 clones which were recloned from 104 positive wells have been identified to produce IgG₁ type monoclonal antibody which has kappa light chain after ELISA subtyping analysis. After purification of IgG₁ monoclonal antibody, heavy and light chain of immunoglobulin has been identified whose molecular weight are 51 KD, 25 KD, respectively. These IL-2 antibodies were confirmed using immunoblotting techniques. IgG₁ type Il-2 monoclonal antibody were bound to IL-2 antigen whose molecular weight is 15.5 KD.
