      • Hypoxia-Responsive MicroRNA-101 Promotes Angiogenesis <i>via</i> Heme Oxygenase-1/Vascular Endothelial Growth Factor Axis by Targeting Cullin 3

        <P>Aims: Hypoxia induces expression of various genes and microRNAs (miRs) that regulate angiogenesis and vascular function. In this study, we investigated a new functional role of new hypoxia-responsive miR-101 in angiogenesis and its underlying mechanism for regulating heme oxygenase-1 (HO-1) and vascular endothelial growth factor (VEGF) expression. Results: We found that hypoxia induced miR-101, which binds to the 3 ' untranslated region of cullin 3 (Cul3) and stabilizes nuclear factor erythroid-derived 2-related factor 2 (Nrf2) via inhibition of the proteasomal degradation pathway. miR-101 overexpression promoted Nrf2 nuclear accumulation, which was accompanied with increases in HO-1 induction, VEGF expression, and endothelial nitric oxide synthase (eNOS)-derived nitric oxide (NO) production. The elevated NO-induced S-nitrosylation of Kelch-like ECH-associated protein 1 and subsequent induction of Nrf2-dependent HO-1 lead to further elevation of VEGF production via a positive feedback loop between the Nrf2/HO-1 and VEGF/eNOS axes. Moreover, miR-101 promoted angiogenic signals and angiogenesis both in vitro and in vivo, and these events were attenuated by inhibiting the biological activity of HO-1, VEGF, or eNOS. Moreover, these effects were also observed in aortic rings from HO-1(+/-) and eNOS(-/-) mice. Local overexpression of miR-101 improved therapeutic angiogenesis and perfusion recovery in the ischemic mouse hindlimb, whereas antagomiR-101 diminished regional blood flow. Innovation: Hypoxia-responsive miR-101 stimulates angiogenesis by activating the HO-1/VEGF/eNOS axis via Cul3 targeting. Thus, miR-101 is a novel angiomir. Conclusion: Our results provide new mechanistic insights into a functional role of miR-101 as a potential therapeutic target in angiogenesis and vascular remodeling. Antioxid.       • Antioxidant and cytoprotective effects of morin against hydrogen peroxide-induced oxidative stress are associated with the induction of Nrf-2-mediated HO-1 expression in V79-4 Chinese hamster lung fibroblasts

        <P>Natural phytochemicals of plant origin, including flavonoids, have been found to be potent antioxidants providing beneficial effects against oxidative stress-related diseases. The present study was carried out to investigate the antioxidant properties of morin, a flavonoid originally isolated from the flowering plants of the Moraceae family. Superoxide dismutase (SOD)-like activity and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(center dot+)) radical scavenging activity were determined. We also investigated the cytoprotective effects of morin against hydrogen peroxide (H2O2)-induced DNA damage and apoptosis in V79-4 Chinese hamster lung fibroblasts. Our results demonstrated that morin had strong scavenging effects against ABTS' radicals with enhanced SOD activity, which varied in a dose-dependent manner. Morin was found to reduce H2O2-induced intracellular reactive oxygen species generation and nuclear DNA damage, and it recovered cell viability damaged by H2O2 via inhibition of mitochondrial dysfunction-mediated apoptosis. Notably, the treatment of V79-4 cells with morin markedly enhanced the expression of heme oxygenase-1 (HO-1) but not quinone oxidoreductase-1, which was associated with the increased expression and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the downregulation of Kelch-like ECH-associated protein 1 expression. Based on our findings, we conclude that morin effectively ameliorated oxidative stress-induced DNA damage through intrinsic free radical scavenging activity and activation of the Nrf2/HO-1 pathway.</P>

      • 電流 페이저 制御方式을 이용한 高周波 인버어터에 관한 硏究

        전력용 반도체 소자를 이용한 가변전압, 가변주파수 고주파 인버터의 응용은 무정전 전원장치나 유도가열등 응용범위가 매우 넓다. 본 연구에서는 L,C공진회로로 링크된 2조의 full-bridge 인버어터회로를 병렬제어하는 기법을 제안하고 있으며 MOSFET로 구성된 각 인버어터는 한개의 등가스위치 모델로 되어 직렬공진 half-bridge형 고주파 인버어터 회로를 구성하고 있다. 게이트 제어기법으로 각 소자의 구동 주기를 분할 제어하여 출력전류 페이저를 합성하는 시분할 게이트 구동방식을 사용하고 이에 따른 인버어터의 회로동작 모드를 분석하고 해석 하였다. The proposed systems can be used power supplys for high power conversion and high frequency induction heating systems. This research proposes a high frequency resonant inverter consisting of equivalent half-bridge model in combination with two L-C linked full-bridge inverter circuits using MOSFET. As a output power control strategy, the time-sharing gate control method is applied. Also, analysis of operating MODE and state equations are described.

      • 이온 플레이팅법으로 제조한 TiN 박막의 미세구조

        Hard coating의 일종인 TiN 피막을 PVD 법으로 제조할 경우, 이온 충돌 양상을 조절할 수 있는 기판 Bias와 질소 분압 등이 미세 구조와 피막 특성에 많은 영향을 미치는 것으로 알려지고 있다. 본 실험에서는 Ion plating에 의한 TiN 피막 제조에서 기판 바이어스 전압과 질소 분압을 변화시키면서 TiN 피막을 제조하고, 미세 구조를 조사하였다. 공정 변수에 관계없이 얻어진 모든 피막은 δ-TiN 단일상이었다. 질소 분압이 높아짐에 따라 우선배향은 (111)면에서 (200)면으로 바뀌었으며, (111)회절상과 (222) 회절상의 반가폭의 비는 감소하였다. 그리고 결정의 크기는 감소하는 거동을 보였으며 피막의 격자 상수는 증가하는 경향을 보였다. 기판 바이어스 변화에 대해서는 전압이 증가함에 따라 (111) 회절상의 반가폭은 -100V 가지 증가하다 그 이상에서는 다시 감소하였으며, 그 때 결정이 크기는 증가하는 경향을 보였다. 피막의 격자 상수는 기판 바이어스가 증가함에 따라 150V까지 증가하다 그 이상에서는 다시 감소하였다. The action of ionized particles has to be controlled properly in ion plating method, because the action of ionized particles has an effect on the microstructure of the thin film. In this pointview, we investigated microstructure of TiN thin film that was deposited with variables of N₂partial pressure is increased, the prefered orientation was changed(1110 plane to (200) plane, the ratio of FWHM of (222) to FWHM of (111)was decreased, grain size was decrease and lattice parameter was increased. As bias voltage was increased, the FWHM of (111) peak was increased to-100V, and then decreased with the grain size was increased. Lattice parameter of film was increased to-150V, and then decreased

      • 초음파 펄스에코 데이터 기반 3차원 시각화 방법론

        The nondestructive testing using ultrasonic pulse-echo data is an effective test methodology esperially for metal structure. Typically, the ultrasonic pulse-echo data is processed and the results are shown in A-scan, B-scan or C-scan formats. Upon viewing B-scan and C-scan data the viewer is able to identify the location of faults as well as their rough dimension. In this paper, we evaluate the applicability of various 3-D visualization methods frequently used in medical imaging, namely, surface rendering (SR), volume rendering (VR) and maximum intensity projection (MIP), for nondestructive testing purposes. Such 3-D visualization of ultrasonic pulse-echo data enables easier identification of the location and dimension of faults more accurately.

      • PC를 이용한 2축 구동시스템개발

        본 연구는 X-Y테이블을 스테핑 모터로 구동하여 시일드를 파렛에 적재하기 위한 시스템을 구성하는 것으로서 X-Y테이블에 조립된 파렛의 움직임을 여러경로의 시일드적재에 적용할 수 있도록 하였다. 본 연구를 통해 차분에 의한 직선제어 방식의 알고리즘을 제안하였으며, 이 알고리즘에 의해 제어되는 복잡다양한 경로를 PC에서 다운로드 받아 처리할 수 있는 시스템을 구성하여 시일드적재장치에 적용함으로써 유연생산시스템을 구현하였다. Tow axis driving system was developed to establish flexible loading system of shield which is a part of electronic equipment, on pallet. Driving of two stepping motors is accomplished by Z-80 single board microprocessors. Moving trajectories of x-y atble are developed by personal computer and downloaded to single boad through RS232C serial communication port. Developed system has shown flexible movement as expected to laad shield on pallet automatically.

      • 구름베어링 설계용 Expert System 개발에 관한 연구

        In the study, an expert system for the design of rolling bearings has been developed by using GURU which is expert system building tool and by constructing knowledge base which consist of rules and facts. The developed system is composed of three parts : the first one is the bearing selection part that recommands the correct combination of ball and roller bearing to support a shaft for a given set of operationg conditions, as initial stage of bearing design. After requesting the minimum amout of input data from the engineer, the second part calculates the parameters and compares with data-base which constains the specifications of bearing. If the results do not satisfy the selection criterion, the search process is repeated. When the malfunction occurs, the last part displays the causes of bearing failure and the countermeasures for correction. As the result of applying this algorithm to the several real design examples, the proper bearing combination and the bearing dimensions are obtained. Throuth inserting the expertise into the knowledge base, the conceptual design process which could not easily be manipulated by conventional CAD approach could be implemented in computer.

      • 스트레스의 이론적 체계에 관한 연구

        The way to accomplish effectively the organizational objective lies in the fulfillment of goals on the organizational level as well as individual levels. But most of organizational members are, to a certain degree, confronted with stress and thus have difficulties in attaining the organizational goals. In the organizational study, however, there is no general agreement on the meaning of stress and the methods of measuring it are differently used among the researchers. The purpose of this paper is to offer theoretical framework in stress in order to faciliate a greater understanding definition of stress, general adaptation syndrome(GAS), eustress and distress, stressors, moderators of stress, outcomes of stress, and build the theoretical model. The definition of stress is an adaptive response, mediated by individual differences and/or psychological processes, that is consequences of any external(environment) action, situation, or event that places excessive psychological and/or physical demands on person. The four types of stressors are individual-level stressors, group-level stressors, organizational stressors, extraorganizational stressors. The moderators of stress are social support, coping, hardiness, cognitive complexity, and control. The effects of stress are many and varied. Some, of course, are positive such as self-motivation, stimulation to work harder, and increased inspiration to live better life. However, many are disruptive and potentially dangerous. Shuler(1980) identified three categories of potential effects of stress: physiological response, psychological response, behavior response. Finally based on the major components of stress I develop the theoretical model of stress.
