        생체 세라믹스용 마모시험장비를 제작하였다. 마모시험기는 lever system을 이용하여 하중인가가 되게 설계되었으며 step motor를 사용하여 회전수 조절이 용이하게 하였다. DAQ (Data Acquisition System)을 이용하여 하중을 측정하여 마찰계수를 측정할 수 있게 하였다. 성능평가를 위해 정방정지르코니아를 Ringer's solution에서 선속도 40 mm/s, 인가하중 10, 20, 30 N에서 마모시험 하였다. Grooving, crack networking등의 마모기구가 관찰되었다. A wear testing machine for bioceramics was designed and made, which loading and rotating parts consisted of a lower systemed and a step motor, respectively. Friction coefficient was determined by DAQ(Data Acquisition System). Wear resistance of tetragonal zirconia was measured under linear velocity of 40 mm/sec, applied load of 10, 20, 30 N in Ringer's solution, which showed grooving, crack networking on worn surfaces.

        한국절식에 대한 초등학교 영양사의 의식조사 : 서울과 인천지역을 중심으로

        The purpose of this study was to investigate perceptions and uses of Korean festive foods by dietitians in elementary school food service. Questionnaires were distributed to 217 dietitians in Seoul(117) and Inchon(100). Statistical data analysis was completed by SAS package program. Results were as follows ; 1. Most dietitians(89.8%) who answered the inquiry had knowledge about Korean festive foods. 2. Most respondents applied Korean festive foods to their menu planning. 3. More than 85% of respondents used festive foods, especially Ddukguk, Mandu, Yaksik, Yukgaejang, Galbiggim, Tangpyungchae, Patjuk, among the 43 kinds considered for their menu planning. 4. The higher the perception of traditional foods, the more the respondents applied them to their menu planning, especially Injulmi, Shikhae, Yaksik, Bamchoe, Jeongpyun, Kalkuksu, Torantang, Hobakgigim, Namul. 5. Respondents got information sources about traditional foods from books, school, home, massmedia, and etc, in the order. 6. Most respondents(88.3%) wanted to simplify festive foods' cooking method or standardize the recipes. 7. On the necessity of education about festive food for children, 81.5% of respondents and festive foods were considered to be cultivated and developed to uphold the Korean traditional dietary life.

        노인의 휴대전화 사용속도에 영향을 미치는 요인 분석

        Objective: This study is to analyze the factor affecting elderly's speed of mobile phone ability. Method: Sixty healthy senior citizens who live in Kim-hye participated in Cognition, Visual perception, Speed of mobile phone ability and the collected data was analyzed by an independent multiple regression analysis. Result: The findings can be summarized as follows; 1. Although men were faster than women while using mobile phone, there was little satisfical significance(p<0.05). 2. Although people in their sixties were faster than people in their seventies while using mobile phone, there was minial difference(p<0.05). 3. The primary factor which affect mobile phone using speed is figure ground, visual memory, visual processing and the most beneficial discovery in the entire project is figure ground(p<0.05). Conclusion: The result of study showed that in order to enhance figure ground, visual memory, visual processing in mobile phone using speed. These results will be consider useful for elderly's mobile phone ability.

        Helicobacter pylori 감염 치료 후 완전관해된 위십이지장 Mucosa Associated Lymphoid Tissue 림프종 1예

        Low-grade (LG) gastric mucosa-associated lymphoid tissue (MALT) lymphoma has been shown to be associated with Helicobacter pylori (H. pylori) infection. However, gastroduodenal MALT lymphoma is rare. We report a case of gastroduodenal MALT lymphoma occurred in a 39-year-old man. It has been regressed after eradication of H. pylori. He was admitted because of epigastric pain. Gastroduodenoscopic findings were erythema, multiple nodules, and erosions on the body and antrum, which were superficially spreading through the first and second portion of duodenum. Endoscopic biopsies revealed the proliferation of centrocyte-like (CCL) cells and lymphoepithelial lesions with lymphoid follicles. Amoxacillin and omeprazole were administrated for 2 weeks under the impression of gastroduodenal LG MALT lymphoma. Three months later, gastroduodenoscopy with biopsy showed the complete regression of LG MALT lymphoma. To the best of our knowledge, this is the first report about the regression of gastroduodenal LG MALT lymphoma after H. pylori eradication.

      • Metabolic Engineering of Saccharomyces cerevisiae for Redox Balance of Xylose Fermentation

        The bioconversion of cellulosic biomass hydrolyzates consisting mainly of glucose and xylose requires the use of engineered Saccharomyces cerevisiae expressing a heterologous xylose pathway. However, there is concern that a fungal xylose pathway consisting of NADPH-specific xylose reductase (XR) and NAD+-specific xylitol dehydrogenase (XDH) may result in a cellular redox imbalance. However, the glycerol biosynthesis and glycerol degradation pathways of S. cerevisiae, termed here as the glycerol cycle, has the potential to balance the cofactor requirements for xylose metabolism, as it produces NADPH by consuming NADH at the expense of one mole of ATP. Therefore, this study tested if the glycerol cycle could improve the xylose metabolism of engineered S. cerevisiae by cofactor balancing, as predicted by an in-silico analysis using elementary flux mode (EFM). When the GPD1 gene, the first step of the glycerol cycle, was overexpressed in the XR/XDH-expressing S. cerevisiae, the glycerol production significantly increased, while the xylitol and ethanol yields became negligible. The reduced xylitol yield suggests that enough NAD+ was supplied for XDH by the glycerol cycle. However, the GPD1 overexpression completely shifted the carbon flux from ethanol to glycerol. Thus, moderate expression of GPD1 may be necessary to achieve improved ethanol production through the cofactor balancing.

        Multiple-wavelength radiation promotes hair growth by enhancing the early stages of hair follicle development in human dermal papilla cells and C57BL/6 mice

        Background: We aimed to clarify the safety and efficacy of simultaneous skin exposure to blue, red, and infrared light. The purpose of this study was to confirm the mechanism by which multiple wavelengths increase hair development both in vivo and in vitro.Methods: Cultured human dermal papilla cells (hDPCs) were exposed to a 470/655/850 nm light-emitting diode (LED) array with a fixed energy density of 3.0 mW/cm2. We analyzed alkaline phosphatase (ALP) staining and activity. The relative expressions of ALP, VEGF, Shh, and OPN3 were examined using reverse transcriptase-polymerase chain reaction arrays 48 hours post-exposure and the protein levels related to extracellular signal-regulated kinase (ERK)/protein kinase B (AKT)/glycogen synthase kinase 3 (GSK3)β signaling were assessed by western blotting. Next, we used H&E staining, hair growth scoring, skin thickness measurement, and the immunohistochemical analysis of the dorsal skin of C57BL/6 mice to investigate the effects of the mono- or combined-photobiomodulation (PBM) groups.Results: According to our findings, simultaneous irradiation with multi-wavelength LEDs at 470/655/850 nm increased the proliferation of hDPCs. Also, compared to the control group, the red wavelength and combined PBM groups had significantly improved skin thickness measurements. Overall, we concluded that the combined PBM therapy successfully induced the early onset of anagen and stimulated hair growth.Conclusion: These results suggest that PBM therapy regulates hair growth by activating the ERK/AKT/GSK3β signaling pathway. Thus, multiple-wavelength radiation from devices combining radiation emitted by low-power lasers and LEDs could be a new approach for promoting PBM-induced beneficial effects.

        Repeated-batch fermentations of xylose and glucose–xylose mixtures using a respiration-deficient <i>Saccharomyces cerevisiae</i> engineered for xylose metabolism

        <P><B>Abstract</B></P><P>Xylose-fermenting <I>Saccharomyces</I> strains are needed for commercialization of ethanol production from lignocellulosic biomass. Engineered <I>Saccharomyces cerevisiae</I> strains expressing <I>XYL1</I>, <I>XYL2</I> and <I>XYL3</I> from <I>Pichia stipitis</I>, however, utilize xylose in an oxidative manner, which results in significantly lower ethanol yields from xylose as compared to glucose. As such, we hypothesized that reconfiguration of xylose metabolism from oxidative into fermentative manner might lead to efficient ethanol production from xylose. To this end, we generated a respiration-deficient (RD) mutant in order to enforce engineered <I>S. cerevisiae</I> to utilize xylose only through fermentative metabolic routes. Three different repeated-batch fermentations were performed to characterize characteristics of the respiration-deficient mutant. When fermenting glucose as a sole carbon source, the RD mutant exhibited near theoretical ethanol yields (0.46gg<SUP>−1</SUP>) during repeated-batch fermentations by recycling the cells. As the repeated-batch fermentation progressed, the volumetric ethanol productivity increased (from 7.5 to 8.3gL<SUP>−1</SUP>h<SUP>−1</SUP>) because of the increased biomass from previous cultures. On the contrary, the mutant showed decreasing volumetric ethanol productivities during the repeated-batch fermentations using xylose as sole carbon source (from 0.4 to 0.3gL<SUP>−1</SUP>h<SUP>−1</SUP>). The mutant did not grow on xylose and lost fermenting ability gradually, indicating that the RD mutant cannot maintain a good fermenting ability on xylose as a sole carbon source. However, the RD mutant was capable of fermenting a mixture of glucose and xylose with stable yields (0.35gg<SUP>−1</SUP>) and productivities (0.52gL<SUP>−1</SUP>h<SUP>−1</SUP>) during the repeated-batch fermentation. In addition, ethanol yields from xylose during the mixed sugar fermentation (0.30gg<SUP>−1</SUP>) were higher than ethanol yields from xylose as a sole carbon source (0.21gg<SUP>−1</SUP>). These results suggest that a strategy for increasing ethanol yield through respiration-deficiency can be applied for the fermentation of lignocellulosic hydrolyzates containing glucose and xylose.</P>

        Dendritic Cell Activation by Glucan Isolated from Umbilicaria Esculenta

        Background: Lichen-derived glucans have been known to stimulate the functions of immune cells. However, immunostimulatory activity of glucan obtained from edible lichen, Umbilicaria esculenta, has not been reported. Thus we evaluated the phenotype and functional maturation of dendritic cells (DCs) following treatment of extracted glucan (PUE). Methods: The phenotypic and functional maturation of PUE-treated DCs was assessed by flow cytometric analysis and cytokine production, respectively. PUE-treated DCs was also used for mixed leukocyte reaction to evaluate T cell-priming capacity. Finally we detected the activation of MAPK and NF-${\kappa}B$ by immunoblot. Results: Phenotypic maturation of DCs was shown by the elevated expressions of CD40, CD80, CD86, and MHC class I/II molecules. Functional activation of DCs was proved by increased cytokine production of IL-12, IL-$1{\beta}$, TNF-${\alpha}$, and IFN-${\alpha}/{\beta}$, decreased endocytosis, and enhanced proliferation of allogenic T cells. Polymyxin B, specific inhibitor of lipopolysaccharide (LPS), did not affect PUE activity, which suggested that PUE was free of LPS contamination. As a mechanism of action, PUE increased phosphorylation of ERK, JNK, and p38 MAPKs, and enhanced nuclear translocation of NF-${\kappa}B$ p50/p65 in DCs. Conclusion: These results indicate that PUE induced DC maturation via MAPK and NF-${\kappa}B$ signaling pathways.
