Tyrosinase inhibitors of Galla Rhois and its derivative components

Two hundred forty-three oriental herbs were screened for their tyrosinase inhibitory activity. Among them, methanol extract of Galla Rhois was shown to be the most potent tyrosinase inhibitory activity. The IC50 values for Galla Rhois, kojic acid, ascorbic acid, arbutin and 5 HMF were 0.163, 0.019, 0.316, 1.520 and 2.511 mg/mL, respectively. Anti-tyrosinase activity of Galla Rhois is higher about two, nine and 15-fold of anti-tyrosinase activity of ascorbic acid, arbutin and 5-HMF while lesser than kojic acid. To isolate active compounds, the 50% aqueous methanol extracts was evaporated to vacuo, and the fractionated by the Diaion HP-20, Sepadex LH-20 column chromatography and repeated the preparative HPLC procedure. Three major phenolic compounds were isolated, and characterized, one of them exhibiting tyrosinase inhibitory activity stronger than L-ascorbic acid (IC50 = 90 μg/mL) was identified as Methyl gallate ((IC50 = 40 μg/mL) by UV, IR, C-NMR, and FAB-MS spectroscopy. The compound isolated from Galla Rhois exhibited non-competitive inhibition against oxidation of l-3-4-dihydroxyphenylalanine (l-DOPA) by mushroom tyrosinase. As a naturally occurring tyrosinase inhibitor, methyl gal-late may be useful as a new agent to inhibit the oxidation of l-DOPA by mushroom tyrosinase. This is the first HPLC analysis containing ascorbic acid, 5-HMF and arbutin, Galla Rhois exert varying degrees of inhibition on tyrosinase-dependent and these result suggesting that Galla Rhois may serve as candidates for depigmenting agents. Further studies on the application of Methyl gallate is needed in cosmetics and as anti-melanogenic agents.

Moutan cortex (MC) is one of the most widely used Oriental herbal medicines for treating inflammatory diseases. In this study, the efTect of MC on lipopolysaccharide (LPS) and rccombinant interferon-gamma(rIF)-indiiced productioii ofnitric oxide (NO) and tumor necrosis factor (TNFγ)-aIpha were examined using mouse perituneal macrophages. MC inhibited the LPS/rIFN-γ-induced expression of inducible nitric oxide synthase (iNOS) and TNF-alpha release. To clarify the mechanism involved, the effect of MC on the activation of nuclear factor (NF)-kappaB was examined. The LPS/rIFN-γ-induced activation of NF-kappaB was almost completely blocked by MC at 0.5 mg/ml. These findings demonstrate that the inhibition of the LPS/rlFN-γ-induced production of NO and TNF-atpha by MC is due to the inhibition of NF-kappaB activation.
Effects of Nelumbinis Semen on Contractile Dysfunction in Ischemic and Reperfused Rat Heart

Nelumbinis Semen (NS), or lotus seed, is one of the most well-known traditional herbal medicines and is frequently used to treat cardiovascular symptoms in Korea. The anti-ischemic effects of NS on ischemia-induced isolated rat heart were investigated through analyses of changes in blood pressure, aortic flow, coronary flow, and cardiac output. The subjects in this study were divided into two groups: a control, untreated ischemia-induced group, and an ischemia-induced group treated with NS. There were no significant differences in perfusion pressure, aortic flow, coronary flow and cardiac output between the groups before ischemia was induced. The supply of oxygen and buffer was stopped for ten minutes to induce ischemia in isolated rat hearts, and NS was administered during ischemia induction. NS treatment significantly prevented decreases in perfusion pressure, aortic flow, coronary flow and cardiac output under ischemic conditions (p<0.01). In addition, the mechanism of the anti-ischemic effects of NS was also examined through quantitation of intracellular calcium content in rat neonatal cardiomyocytes. NS significantly prevented intracellular calcium increases induced by isoproterenol (p<0.01). These results suggest that NS has distinct anti-ischemic effects through calcium antagonism.
