      • Down-regulated MYH11 Expression Correlates with Poor Prognosis in Stage II and III Colorectal Cancer

        The MYH11 gene may be related to cell migration and adhesion, intracellular transport, and signal transduction. However, its relationship with prognosis is still uncertain. The aim of this study was to investigate correlations between MYH11 gene expression and prognosis in 58 patients with stage II and III colorectal cancer. Quantitative real-time polymerase chain reaction was performed in fresh CRC tissues to examine mRNA expression, and immunohistochemistry was performed with paraffin-embedded specimens for protein expression. On univariate analysis, MYH11 expression at both mRNA and protein levels, perineural invasion and lymphovascular invasion were related to disease-free survival (p<0.05; log-rank test). Cancers with lower MYH11 expression were more likely to have a poor prognosis. Otherwise, MYH11 expression was unrelated to patient clinicopathological features. On multivariate analysis, low MYH11 expression proved to be an independent adverse prognosticator (p<0.05). These findings show that MYH11 can contribute to predicting prognosis in stage II and III colorectal cancers.

        Quality assessment of high performance concrete using digitized image elements

        The quality of high performance concrete largely depends on water cement ratio, porosity,material composition and mix methods. The uniformity of color, texture and compressive strengths are quality indicators commonly used to assess the overall characteristics of concrete mixes. The homogeneity and share of coarse aggregates play a key role in concrete quality and must be analyzed in a microscopic point of view. This research studies the quality of high performance concrete by taking drilled cores in both horizontal and vertical directions from a 1.0 m3 specimen. The coarse aggregate, expressed in digitized 100×116 dpi resolution images are processed based on brightness in colors through commercial software converted into text files. With the image converting to text format, the share of coarse aggregate is quantified leading to a satisfactory assessment of homogeneity – a quality index of high performance concrete. The compressive strengths of concrete and the shares of coarse aggregate of the samples are also compared in this research study to illustrate its correlation in concrete quality. It is concluded that a higher homogeneity of aggregate exists in the vertical plane than that of the horizontal planes of the high performance concrete. In addition, the concrete specimen showing denser particle packing has relatively higher compressive strengths. The research methodology provides an easy-to-use, direct measurement of high performance concrete when conducting quality assessment in the construction site.

      • Research On Optimization In NC Bending Process Of Thin-walled Tube Based on Orthogonal Experiment

        In the process of research and development of tube bending forming technology, the way of improving the forming quality and establishing the optimization of forming process parameters has become an important problem that should be solved quickly. This text simulated the process of bend forming with the way of finite element, combined with the effects of geometric and physical parameters on the forming quality of bending pipe, taking the important parameters which are measurements of the pipe wall thickness variation as test indexes, made the virtual orthogonal experiment and gained the laws of the bending angle, relative bending radius, relative wall thickness, thrust, pipe friction coefficient between mode have effects on the forming quality of wall thinning rate and wall thickening. Through the comprehensive analysis and additional test, obtained the optimal parameters values, improved the accuracy of simulation and the optimization results of orthogonal test, improved the quality of tube bending forming.

      • Enhanced electrical conductivity of nanocomposites containing hybrid fillers of carbon nanotubes and carbon black.

        <P>Nanocomposites reinforced with hybrid fillers of carbon nanotubes (CNTs) and carbon black (CB) are developed, aiming at enhancing the electrical conductivity of composites with balanced mechanical properties while lowering the cost of the final product. Epoxy-based nanocomposites were prepared with varying combinations of CNTs and CB as conducting fillers, and their electrical and mechanical properties were evaluated. It was shown that the addition of CNTs in CB composites enhanced the electrical conductivity of composites: a low percolation threshold was achieved with 0.2 wt % CNTs and 0.2 wt % CB particles. The CB particles also enhanced the ductility and fracture toughness of nanocomposites, confirming the synergistic effect of CB as a multifunctional filler. The novelty of this work lies in the synergy arising from the combination of two conducting fillers with unique geometric shapes and aspect ratios as well as different dispersion characteristics, which have not been specifically considered previously.</P>

      • Molecular Recognition Enables Nanosubstrate-Mediated Delivery of Gene-Encapsulated Nanoparticles with High Efficiency

        <P/><P>Substrate-mediated gene delivery is a promising method due to its unique ability to preconcentrate exogenous genes onto designated substrates. However, many challenges remain to enable continuous and multiround delivery of the gene using the same substrates without depositing payloads and immobilizing cells in each round of delivery. Herein we introduce a gene delivery system, nanosubstrate-mediated delivery (NSMD) platform, based on two functional components with nanoscale features, including (1) DNA⊂SNPs, supramolecular nanoparticle (SNP) vectors for gene encapsulation, and (2) Ad-SiNWS, adamantane (Ad)-grafted silicon nanowire substrates. The multivalent molecular recognition between the Ad motifs on Ad-SiNWS and the β-cyclodextrin (CD) motifs on DNA⊂SNPs leads to dynamic assembly and local enrichment of DNA⊂SNPs from the surrounding medium onto Ad-SiNWS. Subsequently, once cells settled on the substrate, DNA⊂SNPs enriched on Ad-SiNWS were introduced through the cell membranes by intimate contact with individual nanowires on Ad-SiNWS, resulting in a highly efficient delivery of exogenous genes. Most importantly, sequential delivery of multiple batches of exogenous genes on the same batch cells settled on Ad-SiNWS was realized by sequential additions of the corresponding DNA⊂SNPs with equivalent efficiency. Moreover, using the NSMD platform <I>in vivo</I>, cells recruited on subcutaneously transplanted Ad-SiNWS were also efficiently transfected with exogenous genes loaded into SNPs, validating the <I>in vivo</I> feasibility of this system. We believe that this nanosubstrate-mediated delivery platform will provide a superior system for <I>in vitro</I> and <I>in vivo</I> gene delivery and can be further used for the encapsulation and delivery of other biomolecules.</P>

        First-principles study of the switching characteristics of the 15,16-dinitrile DDP/CPD-based optical molecular switch with carbon nanotube electrodes

        We have studied the switching characteristics of an optical molecular switch based on the 15,16-dinitrile dihydropyrene/cyclophanediene (DDP/CPD) molecule with two single-walled carbon nanotube (SWCNT) electrodes using first-principles transport calculations. It is shown that the DDP shows an overall higher conductance than the CPD at low bias which is independently of the SWCNT chirality. Meantime, the conductance of the molecular switch can be tuned by the chirality of the SWCNT.         Studies of the microbial metabolism of flavonoids extracted from the leaves of Diospyros kaki by intestinal bacteria

        Flavonoid glycosides are metabolized byintestinal bacteria, giving rise to a wide range of phenolicacids that may exert systemic effects in the body. Themicrobial metabolism of flavonoids extracted from theleaves of Diospyros kaki (FLDK) by intestinal bacteria wasinvestigated in vitro. High-performance liquid chromatography/linear trap quadrupole orbitrap mass spectrometrywas performed to analyze the metabolites of flavonoidsin vivo using Xcalibur2.1 software. The results showed thatthe levels of flavonoid glycosides and flavonoid aglyconesdecreased rapidly in the process of microbial metabolismby intestinal bacteria in vitro, and the metabolic rate maybe related to the concentration of intestinal bacteria in theculture solution. In vivo metabolites of FLDK weredetected in rat plasma and urine after oral administration ofFLDK. Eight flavonoids were identified in the urine, andthree were identified in the plasma; however, flavonoidaglycones were not found in the plasma.

      • The MDM2 SNP309T>G Polymorphism Increases Bladder Cancer Risk among Caucasians: a Meta-analysis

        Published studies have evaluated associations between the MDM2 SNP309T>G polymorphism and bladder cancer susceptibility. However, these generated inconsistent results. The aim of the present investigation was to quantify the strength of association between MDM2 SNP309T>G polymorphism and bladder cancer risk by conducting a meta-analysis. We searched PubMed and Embase for related studies that had been published in English before April 1, 2014 and associations were assessed by summarizing the odds ratios (ORs) with the corresponding 95% confidence intervals (CIs). Five case-control studies with a total of 972 cases and 1,012 controls were finally identified to be eligible for the meta-analysis. Overall, the results indicated that there was no significant association between the MDM2 SNP309T>G polymorphism and bladder cancer risk (for the allele model G vs. T: OR=1.08, 95% CI 0.85-1.36, p=0.54; for the co-dominant model GG vs. TT: OR=1.20, 95% CI 0.74-1.93, p=0.46; for the dominant model GG+GT vs. TT: OR=0.98, 95% CI 0.80-1.20, p=0.83; for the recessive model GG vs. GT+TT: OR=1.20, 95% CI 0.83-1.74, p=0.33). However, on subgroup analysis by ethnicity, significant associations were found in Caucasians in three models (for the allele model G vs. T: OR=1.41, 95% CI 1.10-1.81, p=0.006; for the co-dominant model GG vs. TT: OR=2.16, 95% CI 1.28-3.63, p=0.004; for the recessive model GG vs. GT+TT: OR=2.06, 95% CI 1.31-3.22, p=0.002). In summary, the present meta-analysis provides evidence that the genotype for the MDM2 SNP309T>G polymorphism may be associated with genetic susceptibility to bladder cancer among Caucasians.

      • Human Papillomavirus (HPV) Infection in Women Participating in Cervical Cancer Screening from 2006 to 2010 in Shenzhen City, South China

        Purpose: Human papillomavirus (HPV) infection plays an important role in the development of cervical cancer, but the prevalence of HPV infection in women of Shenzhen city remains unclear. The present study was performed to describe the change of cervical HPV infection in females who participated in voluntary cervical cancer screening from 2006 to 2010 in Shenzhen city, China. Methods: A total of 4, 413 women were recruited. HPV infections were genotyped by polymerase chain reaction (PCR) and reversed dot blot hybridization in Shenzhen Maternity and Child Health Hospital. Results: The prevalence of HPV infection was 13.8%. The five most commonly found HPV types were HPV16 (3.47%), HPV58 (1.68%), HPV33 (1.38%), HPV43 (1.36%) and HPV18 (1.27%). The secular trends of major HPV type-specific were diverse. Among of them, the prevalence of HPV18 increased sharply while others increased slowly or even decreased in the period. The change of total HPV, single HPV and multiple HPV infection were similar during the five years. Conclusions: Our findings suggested that HPV infection is common with HPV16 and HPV 58 as the primary subtypes in women in Shenzhen city.The prevalence of HPV 18 infection is increasing faster than any others, which will lead it to be one of the main subtypes in this city in the future.

      • Could Tumor Size Be A Predictor for Papillary Thyroid Microcarcinoma: a Retrospective Cohort Study

        Background: Central lymph node metastasis(CLNM) is common in papillary thyroid microcarcinoma (PTMC). The aim of this study was to define the pathohistologic risk grading based on surgical outcomes. Materials and Methods: Statistical analysis was performed to figure out the optimal cut-off values of size in preoperative ultrasound images for defining the risk of CLNM in papillary thyroid microcarcinoma. Receiver operating characteristic curves (ROC) studies were carried out to determine the cutoff value(s) for the predictor(s). All the patients were divided into two groups according to the above size and the clinic-pathological and immunohistochemical parameters were compared to determine the significance of findings. Results: The optimal cut-off value of tumor size to predict the risk of CLNM in papillary thyroid microcarcinoma was 0.575 cm (area under the curve 0.721) according to the ROC curves. Significant differences were observed on the multifocality, extrathyroidal extension and central lymph node metastasis between two groups which were divided according to the tumor size by the cutoff values. Patients in two groups showed different positive rate and intensity of Ki67. Conclusions: The size of PTMC in ultrasound images are helpful to predict the aggressiveness of the tumors, it could be an easy predictor for PTMC prognosis and assist us to choose treatment.
