Choline dehydrogenase interacts with SQSTM1/p62 to recruit LC3 and stimulate mitophagy.
Park, Sungwoo,Choi, Seon-Guk,Yoo, Seung-Min,Son, Jin H,Jung, Yong-Keun Landes Bioscience 2014 AUTOPHAGY Vol.10 No.11

CHDH (choline dehydrogenase) is an enzyme catalyzing the dehydrogenation of choline to betaine aldehyde in mitochondria. Apart from this well-known activity, we report here a pivotal role of CHDH in mitophagy. Knockdown of CHDH expression impairs CCCP-induced mitophagy and PARK2/parkin-mediated clearance of mitochondria in mammalian cells, including HeLa cells and SN4741 dopaminergic neuronal cells. Conversely, overexpression of CHDH accelerates PARK2-mediated mitophagy. CHDH is found on both the outer and inner membranes of mitochondria in resting cells. Interestingly, upon induction of mitophagy, CHDH accumulates on the outer membrane in a mitochondrial potential-dependent manner. We found that CHDH is not a substrate of PARK2 but interacts with SQSTM1 independently of PARK2 to recruit SQSTM1 into depolarized mitochondria. The FB1 domain of CHDH is exposed to the cytosol and is required for the interaction with SQSTM1, and overexpression of the FB1 domain only in cytosol reduces CCCP-induced mitochondrial degradation via competitive interaction with SQSTM1. In addition, CHDH, but not the CHDH FB1 deletion mutant, forms a ternary protein complex with SQSTM1 and MAP1LC3 (LC3), leading to loading of LC3 onto the damaged mitochondria via SQSTM1. Further, CHDH is crucial to the mitophagy induced by MPP+ in SN4741 cells. Overall, our results suggest that CHDH is required for PARK2-mediated mitophagy for the recruitment of SQSTM1 and LC3 onto the mitochondria for cargo recognition.

This study applies the concept of Crime Prevention Through Environmental Design (CPTED) and suggests ways to make improvements in environmental design to provide secure facilities for those who use park-and-ride structures which are increasing on a regular basis. First, the authors made a checklist for field examination after consulting related literature and related laws and holding discussion sessions with the experts of the field. After the field investigation with the quantitative and qualitative checklist, another qualitative checklist was developed to supplement the first one. With the second checklist, the authors visited five different park-and-ride structures around the city of Seoul that were diverse in layout, scale, and management system. The result showed that four out of the five sites assessed needed improvements. The analysis was divided into the following areas: location of the structure, maintenance, electronic security, signs, landscaping, and lighting. A popular method of crime prevention at the parking structures was electronic security which was in fact operated mainly to encourage the drivers to pay their parking fees.

Plasma fibronectin is an α₂-glyoprotein, which is produced by vascular endothelial cells. Raised level of plasma fibronectin has been observed in diabetic patients particularly in the presence of microvascular complications. However, no available data exist about urine level of this glycoprotein in diabetic patients. We measured urine fibronectin level by ELISA methods in 54 diabetic patients who have microvascular comlications or not. The following results were obtained. 1) Urine fibronectin level(㎍/g creatinine) in diabetic group(1740.0±678.0) is increased compaired with those in normal control group(471.0±59.0). but this results were not significantly different among two groups(P>0.05). 2) Urine fibronectin level(㎍/g creatinine) was significantly increased in patients with nephropathy (6188.0±3144.0) compared with those in normal control group(471.0±59.0) and patients without nephropathy(645.0±251.0)(P<0.001). 3) There were a significant correlation between BUN, creatinine, creatinine clearance, 24hr urine total protein and urine fibronectin level in diabetic patients. Our data suggest that urine fibronectin excretion level might be used as a sensitive guide for diabectic nephropathy.

BSCCO(Bi_(2)Sr_(2)Ca_(n-1)Cu_(n)O_(x)) superconducting phase were fabricated by an ion beam sputtering and single target dc-magnetron sputtering. Phase intergrowth among n=2-3, 3-4 and 4-5 phases was observed. The molar fraction of each phase in the mixed crystal of the deposited films was determined by X-ray diffraction analyses and investigated as a function of O_(2) gas pressure during sputtering. We investigated the changes of the superconducting properties by molar fraction of each phase. Also, the thin film surface observation was carried out by AFM. According to the result observing the surface of the thin film with SEM, even in case that the formed phase and the composition of the thin film agree, it can be known that there are a number of the precipitates on the surface.

Various approaches can be used to study a special field such as biomechanical aspects of marathon shoes. A runner who wants to buy a pair of running shoes is confronted with a wide range of models, ranging from inexpensive to expensive, from soft to hard, and from red to yellow. These marathon shoes may have special features like wedges or exchangeable damping elements which visually underline the special quality of the shoes. Advertisements for running shoes in marathoner's magazines explain their products by using words such as "support", "rearfoot control", "cushioning", "shock distributor" and "heel stabilizer". One aspect of marathon shoes biomechanical design is the protection and /or the reduction of injuries. In other words, it is aspect of forces acting on the human body, and also the effects these forces produce in this system. The problems can be studied using theoretical techniques using mathematical models and some of these are discussed int below. A marathon shoe must fit not only overall length and ball width, but with respect to several other aspects such as the instep, arch, top lines, and heel and toe curve. A marathon shoes must fit your biomechanical needs very, very well. Match the support in the shoe as closely as possible to your pronation factor, or how much motion your feet experience when on the ground. Marathon shoes must fit your feet even better than the proverbial glove. They swell from the ramped-up circulation that accompanies continuous activity, and may actually spread out a bit as the 26 bones and dozens of tendons, ligaments and muscles in each foot adapt to several hours of pounding. Marathon shoes should be racing flats only if you think you can win. What makes no sense is to think that is significant when compared with how hard your shoes have to work during the last few miles to support your rapidly fading body. Lightweight shoes mean lightweight support. A large variety of elements which have various names, such as inserts, arch supports, arch cookies, orthotics, and orthoses, can be put into a shoes. The inserts for the sports shoe market consist of many different types of plastics and E.V.A(Ethylene Vinyl Acetate). Another type of insert is the built-in insert which is incorporated into or onto the sole of the shoe. This is a rather rare form of conventional shoe construction. Changes in midsole geometry and material are only one of various strategies which can be used to alter the properties of a running shoe. Heel counters, heel stabilizers, insoles, inserts, additional wedges, and different lasts are some additional strategies used in running shoe construction. Many of them are used in combination with others which may introduce additional problems in quantifying their effects. An athlete can use external elements to influence load and stress on his body. The two most important ones are the running shoe and the surface. On the other hand, he or she can use internal elements which anre muscles, tendons, ligaments, and so forth, and his or her running style. Careful exercise and control of these elements is an important factor in the attempt to reduce running injuries.
