        맛버섯 에탄올 추출물의 생리활성

        맛버섯 10계통의 에탄올 추출물에 대한 polyphenol 및 β-glucan 함량을 분석하고, 생리활성으로 항산화 및 항암, 항고혈압, 항당뇨, 항염활성을 측정하였다. Polyphenol 함량에서는 전 계통에서 전반적으로 40 mg% 함량 이상이였고, 맛버섯 M-1548이 61.50±0.59 mg%로 가장 높았다. β-glucan 함량에서도 맛버섯 M-1548에서 37.2±1.12%로 가장 높았으며, 그 외에 맛버섯 M-900에서 36.35±1.11%, M-1630에서 36.24±1.27%의 순서로 높은 β-glucan 함량을 나타냈다. 항당뇨 활성에서도 역시 맛버섯 M-1548이 13.78±0.56%로 가장 높은 효소 저해율을 보였으나 항염 활성에서는 맛버섯 M-1630이 56.59±7.11%로 가장 높은 nitric oxide 저해율을 보였으며 맛버섯 M-1548은 26.21±0.5%로 미미한 저해율을 보였다. 전자공여능 및 ACE 저해활성, nitrite 저해활성은 효과가 없거나 미미한 활성만을 나타냈다. 세포독성 실험에서는 1 mg/mL로 처리시 폐암세포에 대해 전반적으로 30%이상의 세포 사멸율을 보였으며, 자궁경부암세포에서도 맛버섯 M-1548에서 10.05±0.44%의 세포 사멸율을 보였다. 그러나 대장암세포와 정상세포인 섬유아세포에서는 세포 사멸율이 나타나지 않았다. 따라서 맛버섯 10계통은 폐암세포와 자궁경부암세포에 세포 독성을 나타내는 것으로 보아 암세포에 대한 선택성을 갖고 있음을 알 수 있고 정상세포에 대해선 세포 독성을 나타내지 않는 걸 확인하였다. This study was conducted to investigate the physiological activities of the ethanol extracts from 10 different strains of Pholiota nameko. In addition, the β -glucan and polyphenol contents from these strains were also measured. Total polyphenol contents from all the strains were more than 40 mg% with the highest content of 61.50±0.59 mg% and β -glucan contents were 30% with the highest content of 37.20±1.12%. The highest inhibitory activities on α -amyloglucosidase activity and nitric oxide production were 13.78±0.56% and 56.59±7.11%, respectively. However, ethanol extracts from all the strains have little effects on DPPH radical and nitrite scavenging rates and the activity of angiotensin I converting enzyme. The cytotoxic activity of ethanol extracts from all the strains on A549 cell was shown upto 30% with the highest effect of 47.96±8.46% treated at 1 mg/mL concentration.

        팽이버섯 에탄올 추출물의 생리활성

        팽이버섯 11계통을 사용하여 polyphenol 및 β -glucan 함량을 분석하고, 생리활성으로 항산화 및 항암, 항고혈압, 항당뇨, 항염활성을 측정하였다. Polyphenol 함량에서는 CBMFV-65가 244.74 mg%로 가장 많은 함량을 나타내었고 전 품종에서 전반적으로 100 mg% 이상의 함량을 나타냈다. β -glucan 함량에서는 CBMFV-41에서 27.37%로 가장 높았으며, 그 다음으로 CBMFV-30 27.21%, CBMFV-65 27.11%의 순서로 높은 β -glucan 함량을 나타냈다. 전자공여능에서는 CBMFV-66이 91.74%로 가장 높은 DPPH 소거활성을 나타냈으며 전체적으로 70%의 높은 소거활성을 보였다. 세포독성 실험에서는 폐암세포에 대한 세포 저해활성이 가장 컸으며, 폐암세포와 대장암세포 모두 CBMFV-30에서 각각 76.07%, 67.05%로 가장 높은 세포 저해활성을 나타냈다. ACE 저해활성에서는 CBMFV-65에서 10.96%를 나타냈고 나머지 품종은 10%미만의 저해활성을 나타냈다. 항당뇨 활성에서는 CBMFV-41에서 63.57%의 효소 저해율이 측정되었고, CBMFV-63은 10.98%로 가장 낮은 효소 저해활성을 나타냈다. 마지막으로 항염활성에서는 CBMFV-41에서 61.44%의 nitric oxide 억제율이 측정되었다. This study was carried out to investigate the physiological activities of the ethanol extracts from 11 strains of Flammulina velutipes. In addition, the β -glucan and polyphenol contents were also measured. The contents of polyphenol and β -glucan of Flammulina velutipes were found to be more than 100 mg% and 20%, respectively, one of which showed 244.74 mg% and 27.37%, respectively. Further, The highest inhibitory activities on DPPH radical, α -amyloglucosidase activity and nitric oxide production were 91.74%, 63.57% and 61.44%, respectively. However, ethanol extracts from all the strains have little effects on Angiotensin I converting enzyme(ACE)-inhibitory effects. Finally, the highest cytotoxic activities of ethanol extracts from all the strains on A549 and HT-29 cell were 76.07% and 67.05%, respectively.

        불소함유 교정용 레진이 법랑질 인공우식과 재석회화에 미치는 영향에 관한 연구

        To study the effect of fluroide-containing orthodontic resin on the artificial caries susceptibility and remineralization of enamel around bracket, Direct Bonding System brackets were bonded to the buccal surfaces of the 24 sound extracted teeth. 1. The findings of the artificially induced enamel lesion were; the surface zone and intact enamel showed negative birefringence while the subsruface area showed positive birefringence in both control and experimental group. 2. Experimental group of the artificially induced enamel lesion showed more intact, weak birefringence surface zone and lower depth of the subsurface demineralization area than control group. The pattern of the progression of the demineralization was regular. 3. Control group of the artificially induced enamel lesion showed partially destructed, strong birefringence surface zone and deeper depth of the subsurface demineralization area than experimental group. The pattern of the progression of the demineralization was irregular. 4. After remineralization, the experimental group showed more intact, thicker sufrace zone and lower depth of subsurface demineralized area than control group.

        Lip-protecting appliance를 이용한 Self-injurious Lip-biting의 치료증례

        Self-injurious lip-biting is one of the condition refered to as self-injurious behavior. The etiology of this behavior can be divided into organic and fuctional categories. Organic etiologic factors include metabolic and genetic disorders. functional self-injurious behavior is performed consciously, as a response to certain stimuli. The management of this unusual problem depends on the medical history of the child, the etiology of the behavior, and the severity, frequency, and method of injury involved. This report documents a case of self-injurious lip-biting in a 7 year 5 month old boy.

        In this paper, we evaluated two main QoS supporting routing protocol on wireless ad hoc network. At presence, wireless mobile communication focuses on how to efficiently support mobility of users more than QoS guarantee. However, in order to satisfy requirement of various applications which have been or will be served, QoS support service between source and destination becoming a very important issue. Of all routing protocols, DSR and AODV are very important routing protocol in MANET. So we simulated DSR and AODV QoS Routing Protocol Through simulation evaluation tool NS(Network Simulation) based on various environments.

        Chemical Modification of the Human Ether-a-go-go-related gene (HERG) $K^+$ Current by the Amino-Group Reagent Trinitrobenzene Sulfonic Acid

        We investigated the effects of trinitrobenzene sulfonic acid (TNBS), an amino-group reagent, on the human ether-a-go-go-related gene (HERG) $K^+$ channels expressed in Xenopus oocytes. TNBS neutralizes the positively charged amino-groups of peptide N-terminal and lysine residues. External application of TNBS at 10 mM for 5 min irreversibly shifted the curves for currents at the end of the pulse and tail currents of HERG to a more negative potential and decreased the maximal amplitude of the $I_{tail}$ curve $(I_{tail,max})$. TNBS had little effect on either the activated current-voltage relationship or the reversal potential of HERG current, indicating that TNBS did not change ion selectivity properties. TNBS shifted the time constant curves of both activation and deactivation of the HERG current to a more hyperpolarized potential; TNBS's effect was greater on channel opening than channel closing. External $H^+$ is known to inhibit HERG current by shifting $V_{1/2}$ to the right and decreasing $I_{tail,max}$. TNBS enhanced the blockade of external $H^+$ by exaggerating the effect of $H^+$ on $I_{tail,max}$, not on $V_{1/2}$. Our data provide evidence for the presence of essential amino-groups that are associated with the normal functioning of the HERG channel and evidence that these groups modify the blocking effect of external $H^+$ on the current.
