      • Carotenoids 식이와 알코올이 혈액학적 성분과 간조직에 미치는 영향에 관한 연구

        Alcohol is well known agent which can damage the human tissues such as liver via stimulating lipid peroxidation and storage, denaturation of macromolecules, and inhibiting protein metabolism. On the other hand, carotenoids in addition to vitamins A, C, E and lipoic acid, play important roles in protecting these oxidative damages as well as preventing the production of free radicals. This study was carried out to elucidate the precise effects of alcohol administration into rats on the antioxidative functions of dietary carotenoids and isolated β-carotene, and to find out any parameters to uncover more detailed biochemical mechanisms of these agents. For these purposes, two different approaches were performed: 1) examination of the changes in hematological parameters (e.g., total proteins, A/G ratio, ALT/AST et. al) and statistical correlations among inter- and intragroups, 2) examination of the histopathological changes by an electron microscope. The results were analyzed and summarized as following; (1) Carrot diet for one week caused a slight increase in albumin. A/G ratio and AST levels, and a slight decrease in ALT, ALP, BUN and uric acid levels. Two-week alcohol administration following carrot diet increased total proteins, albumin A/G ratio and BUN levels. However, both groups were shown to have little significant changes in cholesterol concentrations. (2) Cheese diet for one week caused a significant increase in total proteins, albumin, AST, ALP, uric acid and total cholesterol levels, and a significant decrease in A/G ratio and glucose concentrations. However, two-week β-carotene diet following cheese intake increased total proteins albumin, A/G ratio, BUN and total cholesterol levels, whereas AST, ALT, ALP and uric acid levels were decreased. (3) Alcohol administration for one week caused an increase in AST activities and a decrease in total proteins, albumin, ALT, ALP and BUN levels. A successive β-carotene diet following alcohol administration increased total proteins, albumin, ALT, ALP and BUN levels, whereas decreased AST and uric acid levels. However, there were not significant changes in A/G ratio, glucose and total cholesterol levels in both groups. (4) β-carotene diet for one week caused a slight increase in albumin, glucose, AST and uric acid levels, and a slight decrease in ALP and BUN levels. Two-weeks alcohol administration following β-carotene diet increased albumin, glucose, BUN and total cholesterol levels, whereas decreased AST, ALT, ALP and uric acid levels. (5) Although the pathological investigation on the liver did not reveal significant changes, cheese diet group (CH-BC/1-CH) was shown to have some lipid deposits. Some results were unexpected and different from typical hematological changes shown by other researchers. Nonetheless, these results strongly suggest that the liver damage or hepatism caused by alcohol intake affects many kinds of biochemical metabolisms, which results in significant changes in many hematological parameters. In addition, dietary carotenoid and isolated β-carotene were shown to have protective roles against the biochemical changes by alcohol intake.

      • 알코홀이 r-GTP 활성에 미치는 인삼의 영향

        This study was carried out to investigate the effects on the changes of the r-glutamyl transpeptidase activity of ethanol-administered white rats. There was classified into group; ethanol treated group(group A), ethanol treated group which was pretreated with ginseng extract(group B), ginseng extract treated group(group C), and ginseng extract treated group which was pretreated with ethanol(group D). White rats of four groups were treated with combined 1% ginseng extract and / or 20% ethanol for 30 days, and were measured the changes of r-GTP activity on the serum, hepatic and renal tissue sample collected at 5 days interval. Following results have been obtained. 1. The changes of serum r-GTP activity were significantly decreased in ethanol treated group B which was pretreated with ginseng extract than in ethanol treated group A. The values were highly significant at the 20th day of feeding. 2. The changes of hepatic r-GTP activity were more increased in group A and decreased in group D. The values were found significantly lowest in 10th and highest in 5th days. 3. The changes of renal r-GTP activity were more increased in group A and decreased in group D. The values were significantly highest in 10th and lowest in 20th days. Based on these results it was found that the changes of r-GTP activity between serum and hepato-renal tissue were exist and the protective action of ginseng against alcohol degradative effects was noted.

      • Thermoplasma acidophilum의 20S Proteasome에 의한 ATPase-활성적 단백질분해에 관한 연구

        The eukaryotic 26S proteasome is an ATP/ubiquitin-dependent proteolytic complex consisting of the 20S core particle and 19S ATPase complex. However, because of its complexity and unstable properties, this study was carried out to present more simple and stable model for the ATP-activated proteolytic complex in prokaryotes which can take the place of the eukaryotic 26S proteasome. For this purpose, recombinant Thermoplasma 20S proteasome (T20S) and Methanococcus MS4, a sequence homolog of one ATPase subunit in the 19S ATPase complex, were successfully isolated from Escherichia coli (E. coli). The α and β subunits of T20S expressed in E. coli could assemble for themselves, and showed the peptide-hydrolyzing activity. Whereas both T20S and R20S (the 20S complex from rabbit skeletal muscle) had the highest peptidase activity against Suc-LLVY-AMC, a good substrate for chymotrypsin-like peptidase activity, the specific activity of T20S was slightly lower than that of R20S. In addition, several reagents such as KCI, SDS, and ovalbumin were shown to have different effects on the peptidase activities between T20S and R20S. When the ATPase activity of the purified MS4 were assayed , the Km for ATP was about 0.5 mM, and casein could stimulate the activity more than 2 fold without the change in Km. This result implicates that protein-activated ATPase may induce the conformational change of casein, and therefore suggests that MS4 ATPase may activate the proteolytic activity of the 20S proteasome via accelerating the recognition and translocation of the protein substrates.

      • 인삼 추출액이 알코홀대사와 간조직에 미치는 영향

        This study was carried out to investigate the effect of ginseng extract on ethanol metabolism and histopathological changes of liver tissues n alcohol administered rats. White rats of three groups(untreated normal control, A,B) were pretreated with ginseng extract (10ml/㎏ B.W) for 15 days, and then were given with combined ginseng extract-40% alcohol(group A) and 40% alcohol(group B) by polyethylene catheter through mouth for another 30 days. The description was based on the analysis of blood sample collected by 5 days interval. The results obtained were summarized as follow : 1. The mean concentrations of serum alcohol were significantly higher in group A and group B than those of control during total experimental periods. The mean concentrations of serum alcohol were significantly lower in group A than those of group B. 2. The activities of GOT and ADH, concentration of α₂-globulin, cholesterol and triglyceride were increased than those of group A, and the more lengthy period alcohol adminstered, the more increased change observed. But in alcohol-ginseng extract administered group A, these were significantly decreased in similar to those of normal control group. 3. The blood components which is markedly influenced by administration of ginseng extract were found in the followings : enzymatic activities such as GOT and ADH, contents of α-₂globulin and cholesterol. 4. With microscopic observation, hepatic tissues of group B were observed to have mild infiltration of lymphocytes from the feeding 10th day. However the moderate destruction of hepatic architecture, prominent ballooning degeneration and mild infiltration of lymphocytes were apparent from the feeding 20th day. But mild ballooning degeneration of hepatocytes was observed in periportal area in group A, whereas at the feeding 30th day, the mild infiltration of chronic inflammatory cell in the portal tract was less severe than that of group B. Based on these results It can be suggested that the activities of serum GOT and liver ADH which is most sensitively influenced by ginseng extract may have the parameter of the protective action against alcohol, in addition to protective action of ginseng against alcohol degradative effects.

      • 알코올에 의한 α-Tocopherol이 혈액성분에 미치는 영향

        This study was done in order to observe the effects of ethanol and a-tocopherol on the serum ALP, BUN and uric acid concentrations. The white Sprague-Dawley rats were classified with 4-grops : 0.9% NaCl administration was group A control, 25% ethanol administration for the each 5, 10 and 15 days was group B(Bl, B2 and B3), 25% ethanol administration for the each 5, 10 and 15 days and 50% a-tocopherol for the 2 and 7 days administration was group C (Cl and C2) and 50% a-tocopherol pretreated for the each 5, 10 and 16 days and 25% ethanol for the 2, 7 and the 10 days administration was group D(Dl, D2 and D3) The following results were obtained. 1. The changes of serum ALP activity was significantly increased in ethanol treated group. The group C and D was decreased. 2. The changes of serum BUN concentration between each group were not significant and however B2, B3, C2 and Dl was significantly increased. 3. The changes of serum uric acid concentration between each group were not significant. However group Cl and Dl was slightly decreased patterns.

        신부식전 유치 법랑질표면의 기계적 처리가 레진접합력에 미치는 영향에 관한 연구

        The purpose of this study was to determine the effect of the grinding method on the enamel of primary tooth on the bond strength of composite resin. Methods of tooth preparation investigated were 1.unground enamel, exposure to 37% phosphoric acid for 60 sec. 2.unground enamel, exposure to 37% phosphoric acid for 120 sec. 3.ground enamel with a diamond bur, exposure to phosphoric acid for 60 sec. 4.ground enamel with a carbide bur, 37% phosphoric acid for 60 sec. 5.ground enamel with a green stone, 37%phosphoric acid for 60 sec. 6.ground enamel with a disk, 37% phosphoric acid for 60 sec. The shear bond strength of composite resin was tested a Instron and surface tomography was examined with a SEM. The results were as follows: 1.With grinding of outer enamel surface,etch-time to produce a favorable bonding surface in primary teeth was shortened. 2.In SEM study,unground surface often had a prismless layer obscuring the rod pattern.Ground surface showed more effective removal of prismless layer more uniformly etched prism patterns. 3.Grinding enhanced the bond strength the enamel surface and composite resin. 4.Grinding enamel surface with a coarse diamond bur resulted in the highest bond strength.There was no significanct difference in the bond strength between other grinding methods.

      • 담배 박각시나방 Manduca sexta의 계획된 세포죽음동안의 Proteasome의 변화

        모든 진핵세포의 주요 단백질 분해요소인 proteasome은 변성되거나 상해된 불필요한 단백질을 제거할 뿐만 아니라, 세포주기를 비롯한 다양한 대사과정들의 주요 효소들을 필요 시 적절히 분해함으로써 항상성을 유지시키는 필수적인 효소로 잘 알려져 있다. 또한 proteasome은 특정 포유동물 조직의 계획된 세포죽음(programmed cell death 또는 apoptosis)와 밀접하게 관련되어 있다고 보고된 바, 본 연구에서는 담배 박각시나방(Manduca sexta)의 변태과정에서 일어나는 마디간 근육(intersegmental muscle)의 급격한 파괴과정동안의 proteasome의 변화양상을 발현 및 단백질 분해활성을 중점으로 조사하였다. 정상근육과 파괴직전의 근육으로부터 proteasome을 분석한 결과 단백질 분해활성이 최소한 6배 이상 증가하였으며, proteasome의 주요 구성성분들의 발현 변화를 각각의 항체(antibody)를 이용하여 조사한 결과 세포죽음 직전에 급격히 증가함을 보였다. 이러한 구성성분들의 발현증가가 proteasome 복합체 형성을 통해 활성의 증가를 일으키는지 조사한 결과, 세포내에서 실질적으로 단백질 분해활성을 담당한다고 여겨지는 26S proteasome(2,000 kDa)의 위치에서 커다란 활성의 변화와 구성성분의 증가를 볼 수 있었다. 따라서, 담배 박각시나방의 계획된 세포죽음 현상은 특정 호르몬의 신호에 의해 proteasome의 발현이 급격히 증가하여 26S proteasome의 복합체 형태로 세포 내 대부분의 단백질들을 분해함으로써 30시간이내 근육조직을 완전히 소멸시키는 것을 알 수 있었다. 한편, proteasome을 구성하는 특정 구성성분의 과다 발현이 세포의 단백질 분해현상에 어떠한 영향을 미치는 지 조사하기 위하여 쥐의 fibroblast cell에 26S proteasome의 ATPase 구성성분인 m56 유전자를 삽입한 후 세포의 추출물로부터 proteasome의 양과 활성의 변화를 관찰하였다. Proteasome을 초원심분리로 추출하여 관찰한 결과, 담배 박각시나방의 경우와는 달리 유전자 삽입 세포는 모든 실험에서 정상 세포와 전혀 차이를 보이지 않았다. 이 결과로부터 세포의 생명 유지에 필수적이고 매우 다양한 기능을 가진 proteasome은 40여개의 구성성분들의 발현이 매우 정교하게 조절되고 있으며, 특정 세포죽음 자극(apoptotic signal)이 왔을 때 일련의 구성 성분들의 발현과 26S proteasome 복합체 형성이 증가함으로써 그 기능을 수행하는 것을 알 수 있으며, 보다 자세한 연구는 비정상적 단백질 분해나 세포죽음을 동반하는 여러 질환들의 원인과 현상을 규명할 수 있으며, 또한 그 활성을 조절함으로써 궁극적인 치료의 방향도 제시 가능할 것으로 기대된다.

      • β-Carotene 이 혈중 효소활성도에 미치는 영향

        This study was carried out in order to observe the effects of ethanol and β-carotene on the cholesterol, Triglyceride, LDL-cholesterol, HDL-cholesterol, glutathione peroxidase and catalase activities. The normal white S.D rats were classified into 4 group: 0.9% NaCl administration and 20% ethanol administration were referred to as group A, 20% ethanol and 50% β-carotene mixture administration was group B and 50% β-carotene administration was group C for the 30 day-feeding experiment. The results were obtained as following: 1. The blood lipid concentration was most highly increased in group A, was almost normal in group B, but significantly decreased in goup C. 2. The blood antioxidant enzyme activities were significantly decreased in group A, but significantly increased in goup B and C. 3. The blood β-carotene concentration were significantly decreased in group A, and slightly decreased in group B. Surprisingly it was increased in group C.

        This paper achieves a research about avoidance of obstacles avoidance based on ultrasonic sensor. The performance of the proposed obstacle avoidance robot controller in order to determine the exact dynamic systems modeling system that uncertainty is difficult for nomadic controlled robot direction angle by ultrasonic sensors throughout controlled performance tests. In additionally, this study is an in different ways than the self-driving simulator in the development of ultrasonic sensors and unmanned remote control techniques used by the self-driving robot controlled driving through an unmanned remote controlled unmanned realize the performance of factory automation.

      • Apolipoprotein A-I에 특이적인 단일클론항체 제조 및 특성에 관한 연구

        본 연구는 HDL을 구성하는 아포단백 A-I에 특이적인 단일클론항체 A-I30 및 A-I4를 제조하여 이의 특성을 분석하였다. 아포단백 A-I을 BALB/c 생쥐에 면역한후 얻어진 비장과 Sp2/O세포주와 융합시켜 얻어진 hybridoma를 생쥐의 복강내에 주입, 복수를 채취하여 Protein A-Sepharose CL4B크로마토그래피를 통해 단일클론항체를 분리하였다. 항체의 특이성은 ELISA 및 Western blotting으로 확인하였으며 ELISA방법으로 얻어진 해리상수(Kd)치는 A-I30 은 2.25 x 10의 마이너스8승, A-I4는2.15 x 10의 마이너스 8승 이었다.
