        Effect of Suckling Systems on Serum Oxytocin and Cortisol Concentrations and Behavior to a Novel Object in Beef Calves

        We investigated differences between effects of natural- and bucket-suckling methods on basal serum oxytocin (OT) and cortisol concentrations, and the effect of OT concentration on affiliative and investigative behavior of calves to a novel object. Ten Japanese Black calves, balanced with birth order, were allocated evenly to natural-suckling (NS) and bucket suckling (BS) groups. Blood samples were collected at the ages of 1 and 2 months (1 week after weaning) calves, and serum OT and cortisol concentrations were measured using enzyme-linked immunosorbent assay and enzymeimmunoassay tests, respectively. Each calf at the age of 2 months (2 weeks after weaning) was released into an open-field with a calf decoy, and its investigative and affiliative behaviors were recorded for 20 minutes. In 1-month-old calves, the basal serum OT concentration ($25.5{\pm}4.9$ [mean${\pm}$standard deviation, pg/mL]) of NS was significantly higher than that of BS ($16.9{\pm}6.7$) (p<0.05), whereas the basal cortisol concentration ($5.8{\pm}2.5$ [mean${\pm}$standard deviation, ng/mL]) of NS was significantly lower than that in BS ($10.0{\pm}2.8$) (p<0.05). Additionally, a negative correlation was noted between serum OT and cortisol concentrations in 1-month-old calves (p = 0.06). Further, the higher serum OT concentration the calves had at 1 month old, the more investigative the calves were at 2 months old but not affiliative in the open-field with a calf decoy. Thus, we concluded that the natural suckling method from a dam elevates the basal serum OT concentration in calves, and high serum OT concentrations induce investigative behavior and attenuate cortisol concentrations.

        Role of ghrelin in the pancreatic exocrine secretion via mitogen-activated protein kinase signaling in rats

        Background: This study was performed to investigate the impact of exogenous ghrelin on the pancreatic α-amylase outputs and responses of pancreatic proteins to ghrelin that may relate to pancreatic exocrine. Methods: Sprague-Dawley male rats (9 weeks old, 300 ± 10 g) were injected with ghrelin via intraperitoneal (i.p.) infusion at dosage of 0, 0.1, 1.0 and 10.0 μg/kg body weight (BW), respectively. The plasma ghrelin and cholecystokinin (CCK) level were determined using enzyme immunoassay kit; the mRNA expression of ghrelin receptor (GHSR-1α) and growth hormone (GH) receptor were assessed by reverse transcription PCR; the expressions of pancreatic α-amylase activity, extracellular-signal-regulated kinases (ERK), phosphorylated extracellular-signal-regulated kinases (pERK) and c-Jun N-terminal kinase (JNK) were evaluated by western blotting; moreover the responses of pancreatic proteins to ghrelin were analyzed using the two-dimensional gel electrophoresis system. Results: The exogenous ghrelin (1.0 and 10.0 μg/kg BW) elevated the level of plasma ghrelin (p < 0.05), and suppressed the expression of pancreatic α-amylase at a dose of 10.0 μg/kg BW (p < 0.05). No difference in the level of plasma CCK was observed, even though rats were exposed to any dose of exogenous ghrelin. In addition, a combination of western blot and proteomic analysis revealed exogenous ghrelin (10.0 μg/kg BW) induced increasing the JNK and ERK expressions (p < 0.05) and four proteins such as Destrin, Anionic trypsin-1, Trypsinogen, and especially eukaryotic translation initiation factor 3 in rat pancreas. Conclusions: Taken together, exogenous ghrelin by i.p. infusion plays a role in the pancreatic exocrine secretion via mitogen-activated protein kinase signaling pathway.

        Production of Lean Beef Containing a High Content of <i>trans-10</i>, <i>cis-12</i> Conjugated Linoleic Acid by Feeding a High-Temperature-Micro-Time-Treated Diet with Extruded Soybean

        <P>The effects were observed of feeding a high-temperature-micro-time (HTMT)-treated diet of high-quality extruded soybean (ESB) on <I>trans-10</I>, <I>cis-12</I> conjugated linoleic acid (CLA) incorporation in intra-muscular fat and on the lipid metabolism of twelve Holstein steers. The animals were randomly allocated to three groups, the control group being fed on a normal diet, and the other two groups being fed on a diet containing high-quality ESB with or without the HTMT treatment. The experimental period was four months. The content of total CLA in both the HTMT and non-HTMT-treated diets was higher than that in the control diet (<I>P</I><0.05). The concentration of <I>trans-10</I>, <I>cis-12</I> CLA was significantly higher in the HTMT-treated diet than in the non-treated diets at <I>P</I><0.05, and the HTMT-treatment decreased the intra-muscular fat content (<I>P</I><0.05). The expression of fatty acid synthase (FAS) and the plasma insulin-like growth factor-1 (IGF-1) level in the HTMT-treated group were lower than those in the control group (<I>P</I><0.05). Lipoprotein lipase (LPL) mRNA level in the adipose tissue was lower in the non-HTMT-treated group than in the control (<I>P</I><0.05). These results show that the HTMT treatment increased the level of <I>trans-10</I>, <I>cis-12</I> CLA in the intra-muscular fat. This increased level of <I>trans-10</I>, <I>cis-12</I> CLA may inhibit lipogenesis in the intra-muscular fat of Holstein steers.</P>
