      • Novel Methylation Biomarker for Non-invasive Diagnostics in Lung Cancer

        To identify aberrantly hypermethylated DNA in lung cancer cells we established a genome-wide analysis for hypermethylation sites, namely Methyl DNA Isolation and Amplification (MeDIA) coupled-CpG microarray analysis. In the comprehensive methyaltion profiling analysis between human lung cancer, A549 cells and normal NHBE cells, we observed that several clusters of genes show a significant level of aberrancy in CpG island methylation pattern in cancer cells compared to normal cells. We further identified PCDHGA12 gene as a new marker of non-invasive diagnostics for lung cancer based on followings. 1) Transcription of PCDHGA12 gene is reactivated after treatment of A549 cells with demethylating agent. 2) Bisulfide clonal-sequencing reveals that CpG island of PCDHGA12 shows a distinctive differential methylation between two cell lines. 3) Pyrosequencing-based quantitative methylation assay for such region in tumor and non-tumorous tissues from lung cancer patients shows aberrant hypermethylation in 37 (92%) of the 40 tumor tissues. In clinical validation by pyrosequencingin induced-sputum of lung cancer patients (n=87) and healthy controls (n=51), we observed aberrant hypermethylation incident at significantly elevated level in samples derived from lung cancer patients. According to the optimal threshold calculated by ROC curve analysis, sensitivity and specificity of PCDHGA12 was 86.2% and 82.4%, respectively. PCDHGA12 methylation status could be a potential methylation biomarker alone or combined with others for the screen and the detection of relapse of lung cancer.

        비장에서의 악성 림프종

        We experienced a case of malignant lymphoma of the spleen on 69-year-old male patient with huge splenomegaly. The spleen represented diffuse proliferation of large lymphocytes with plasmacytoid feature by microscope and strong positively stained to B-cell marker for immunohistochemical stain and finally diagnosed as malignant lymphoma, large cell, immunoblastic, diffuse(B-cell type). So we reported this case and reviewed the related literature briefly.

        색소실조증 환아의 증례보고

        색소실조증(Incontinentia pigmenti)은 신생아기에 체간과 사지에 소수포가 발생하여 1세경이 되면 수포는 더 이상 생기지 않고 갈색반점이 나타나는 특징을 보이는 아주 드문 질환이다. Bloch-Sulzberger syndrome이라고 불리는 이 질환은 반성 우성 유전질환으로 대부분 여성에게 나타나며, 외배엽, 중배엽 기원 조직들인 눈, 치아, 피부, 손톱, 가슴, 머리카락, 신경계 등에 이상을 동반한다. 본 증례는 유치의 선천적 결손을 주소로 내원한 환아의 문진 중 색소실조증의 사실을 발견하고 검사한 결과, 다수의 유치와 영구치의 선천적 결손과 치아의 형태이상, 상악의 열성장, 치아의 형태이상과 악골 성장의 부조화 등으로 인한 교합불안정을 보이고 있었다. 전신적으로는 망막이상을 보여 수술을 시행한 상태이며, 피부에는 특징적인 갈색반점을 보였다. 현재 치아결손부의 공간관리와 심미적, 심리적인 문제의 해결, 그리고 기능의 회복을 위해 가철식 부분의치를 장착해주었으며 계속적인 관찰을 하고 있다. Incontinentia pigmenti(IP), so called Bloch-Sulzberger syndrome is a rare genodermatosis that occurs almost in female infant; usually lethal in males, X-linked dominantly inherited disorder. IP is characterized by abnormalities of mesodermal and ectodermal tissues including eye, tooth, skin, nail, breast and hair as well as neurological deficiencies. Dental problems are congenital missing of teeth, delayed eruption. abnormal crown shape and so on. Here is a case of 6 year-old female with IP. She had congenital missing of primary and permanent teeth, delayed eruption. maxillary deficiency and extra cusps, resulting in unstable occlusion. Systemically, she had a history of operating eyes due to problem of retina and hyperpigmented macules on her trunk and extremities as typical character of IP.

        중합효소연쇄반응(Polymerase Chain Reaction)을 이용한 Porphyromonas endodontalis의 동정에 대한 연구

        Porphyromonas endodontalis, and anaerobic Gram negative cocobacillus which was known to be associated with the infected root canals and periapical lesions, is very difficult to culture and to detect by the traditional method in that it requires much time to induce the specific black pigmentation, and it is very sensitive to oxygen and the antibiotics added in the culture medium. In this study, the nucleotide sequences of the 'probe h' (0.73kb), one of the specific DNA probes to P.endodontalis (ATCC 35406) which had been developed by our department, was determined and then a pair of primers for PCR amplification was fabricated to identify P. endodontalis. The plasmids containing 'probe h' were purified by Wizard^TM Midipreps DNA Purification System (Promega Corp.), and the nucleotide sequences of the 'probe h' were determined by the dideoxy chain termination method using TaqTrack Sequencing System (Promega Corp.) and detected by fluorescent labelling method. The sense/antisense PCR primers were designed with computer software (Lasergene, DNASTAR Inc.). PCR was done with a programmable GeneAmp PCR System 2400 (Perkin Elmer-Cetus Co.). Each sample containing the whole genomic DNA of P. endodontalis and other black-pigmented Bacteroides was itailly denatured at 94℃ for 5 min and then subjected to 30 cycles, each of them consisting of 60s at 94℃, 60s at 60℃, and 90s at 72℃. The amplified DNA was resolved electrophoretically in a 1.0% agarose gel in 1X TAE buffer, stained with EtBr, and photographed on a UV transilluminator. The results were as follows : 1. The nucleotide sequences of 'probe h' (743 base pairs) were obtained by dideoxy chain termination method, and from that results the specific primers to P. endodontalis (ATCC 35406), 'Primer H1/ Primer H2', were designed. 2. It has been found that 'Primer H1/H2' could detect P. endodontalis (ATCC 35406) using PCR. 3. The PCR system with this primers may be a powerful technique to amplify the specific sequences of 'probe h' of P. endodontalis (ATCC 35406) that produce distinct identification of it from other black-pigmented Bacteroides, and this could help us to determine the nature of periapical disease.

        치아종을 동반한 석회화 치성낭의 치험례 : CASE REPORT

        석회화 치성낭은 비교적 드문 낭으로 하나의 독립된 유형의 치성 병소로 인정받고 있지만, 조직학적 소견이 다양하고 분류체계도 학자들마다 다르다. 석회화 치성낭의 이장상피는 중층편평상피로 되어 있으며, 상피층 내에 특징 세포인 유령세포가 출현한다. 유령세포들은 인접한 결합조직에 치성조직을 유도할 수 있는 능력을 가지고 있어 법랑모세포종, 법랑모세포 치아종, 법랑모세포 섬유치아종, 치아종 등의 치성종양이 합병되어 발생하기도 한다. 외과적 제거 후 재발율은 적지만 치성종양이 병발될 경우가 있기 때문에 반드시 조직병리학적 검사를 수행하여야 한다. 7세 남아가 상악 좌측 중절치의 미맹출을 주소로 원광대학교 치과병원 소아치과에 내원하였다. 방사선 사진에서 맹출되지 않은 상악 좌측 중절치 및 측절치 치관 상방에 방사선 불투과성 물질이 보였다. 초기 임상적 방사선학적 검사 결과 치아종으로 진단하였으며, 외과적 적출 및 생검을 시행하였다. 적출물은 불규칙적인 석회화 물질로 구성된 복잡 치아종 양상을 보였으며 상피조직으로 싸여 있었다. 조직병리학적 검사 결과, 이장상피에서 유령세포의 군집이 관찰되었고, 석회화 조직은 법랑질, 상아질 형태의 조식들이 혼합된 양상을 보여, 복잡 치아종을 동반한 석회화 치성낭으로 진단하였다. 본 증례에서와 같이 매복치 상방의 치아종 양상의 병소가 관찰 되었을 때 석회화 치성낭의 가능성을 염두에 두어야 할 필요가 있다. Calcifying odontogenic cyst(COC) is a rare developmental odontogenic cyst, which shows diverse classification and terminology. Cystic epithelial lining of COC is composed of basal cell layer of columnar cells and overlying layer of stellate reticulum. In the epithelium, ghost cells that might induce adjacent mesenchymal tissue to develop dental organ are shown characteristically. In spite of low rate of recurrence, we have to get a histopathological examination so that odontogenic lesions may recur without fully curettage of lining epithelium. 7-year-old male child came pediatric dentistry in wonkwang university dental hospital in order to check the delayed eruption of left maxillary central incisor. Radiographic examination revealed a well-defined radiopaque mass, overlapping impacted left central and lateral incisor crown. Enucleated mass was tooth-like features and also had epithelium lining. Results of histopathologic procedure, we saw the lots of ghost cell and proliferating hard dental tissues. Also we saw the cystic epithelium cells. It revealed diagnosis of the COC associated complex odontoma. For this reason one should consider of COC when patients present odontoma-like lesion with impacted tooth.

      • 광역수계 수질관리의 개선방안 : 한강수계를 중심으로 With Emphasis on the Water System of Han River

        최근 광역적인 환경오염문제가 점차 증가하고 있으며 중대한 현안으로 대두되고 이Tekj. 특히 광역수계의 수질·수량관리 문제로 인해 지역간 이해관계가 상충됨에 따라 갈등과 분쟁이 증대되고 있다. 이러한 광역수계문제는 새로운 관점에서 접근하여 해결해 나아가야 할 시급한 현안이라고 할 수 있다. 이러한 문제의식 하에 본 연구는 한강수계유역을 대상으로 광역수계 수질문제에 중점을 두어 우리나라의 수자원 관리체계와 선진 외국의 사례를 살펴보고, 현행 광역수계 수질관리의 실태분석을 통해 광역수계 수질관리상의 개선방안을 제시한다. Recently, with the increasingly deep concerns on environmental quality, the area-wide water conflict problem has become so large and complex that water quantity/quality management and conflict resolution should be regarded as an urgent issue calling for new approaches. In relation to this issue, this study has an intention of examining the area-wide water problem, especially, with emphasis on the area-wide water quality management problem in the water system of Han with emphasis on the area-wide water quality management problem in the water system of Han River. And then, suggests the improving devices of area-wide water quality management in the water system of Han River.

      • SDA를 이용한 마이크로스트립-슬롯 결합 급전선 해석

        The new feeding technique which is introduced for aperture coupled microstrip antenna, is analysed in this paper, in microstrip-slot coupled feeder, the microstripline is the first feeder and the narrow slotline which is on the ground is the second feeder to antenna above the slot. The reflection coefficient and the transmission coefficient caused by a slot type discontunuity is analysed using the reciprocity theorem. To analyse microstrip-slot coupled feeder, the equivalent magnetic current is obtained from the schematic of the substrate with a feed line and a slot. The exact Green's function is derived from the equivalent magnetic current and then analysed in a moment method solution using PWS mode functions assumed as an unknown E field an a slot aperture. The unknown E field can be represented by one PWS mode, since the operational frequency is in S-band. The equivalent series impedance seen by the microstrip feed line is added to the equivalent admittance of the slot. The SDA(Spectral Domain Approach) with Green' function is carried and the characteristics of microstrip-slot coupled feeder can be considered with varying many parameters of the substrates.
