      • Triton X-100이 Trichoderma koningii의 성장 및 형태에 미치는 영향

        Triton Ⅹ-100이 Trichoderma koningii의 성장 및 형태에 미치는 영향을 조사하기 위하여, Triton Ⅹ-100 첨가 여부에 따른 각 종 성장계수의 변화를 조사하여 보았다. 액체 배양시 Triton Ⅹ-100의 첨가에도 불구하고, specific growth rate과 doubling time은 변화가 없었으나, Triton Ⅹ-100을 첨가한 경우, 군체의 방사상 성장률이 감소하였다. 이러한 현상은 Triton Ⅹ-100에 의하여 균사의 분지 형성이 촉진되고, 격막형성이 촉진되어 격막 간의 간격이 짧아져 나타나는 것으로 확인되었다. We investigated the effect of Triton Ⅹ-100 on the growth and morphology of Trichoderma koningii by comparing various parameters representing the growth of mold in the presence or absence of Triton Ⅹ-100. The specific growth rate and doubling time of T. koningii were not affected by the addition of 0.05% Triton Ⅹ-100 into solid medium also resulted in decrease in the colony radial growth rate and this response was correlated with the formation of mycelia which showed increase in branching and septation in the presence of Triton Ⅹ-100.

        유화법과 분무법에 의해 제조된 경구백신용 알긴산 마이크로스피어의 평가

        Alginate microspheres, containing fluorescein isothiocyanate-bovine serum albumin (FITC-BSA) or green fluorescent protein (GFP) were prepared and used as a model drug to develop the oral vaccine delivery system. The alginate microspheres were coated with poly-L-lysine or chitosan. Two methods, w/o-emulsion and spray, were used to prepare alginate microspheres. To optimize preparation conditions, effects of several factors on the particle size and particle morphology of microsphere, and loading efficiency of model antigen were investigated. In both preparation methods, the particle size and the loading efficiency were enhanced when the concentration of sodium alginate increased. In the w/o-emulsion preparation method, as the concentration of Span 80 was increased from 0.5% to 2%, the particle size was decreased, but the loading efficiency was increased. The higher the emulsification speed was, the smaller the particle size and loading efficiency were. The concentration of calcium chloride did not show any effect on the particle size and loading efficiency. In the spray preparation method, the particle size was increased as the nozzle pressure (from 1 kgf/㎡ to 3 kgf/㎡) and spray rate was raised. Increasing calcium chloride concentration (< 7%) decreased the particle size, in contrast to no effect of calcium chloride concentration on the w/o-emulsion preparation method. Alginate microspheres prepared by two methods were different in the particle size and loading efficiency, the particle size of microspheres prepared by the spray method was about 2-6 μm, larger than that prepared by the w/o emulsion method (about 2 μm), and the loading efficiency was also higher with spray method. Furthermore, drying process for the microspheres prepared by the spray was simpler and easier, compared with the w/o emulsion preparation. Therefore, the spray method was chosen to prepare alginate microspheres for further experiments. Release pattern of FITC-BSA in alginate microspheres was evaluated in simulated intestinal fluid and PBS (phosphate buffered saline). Dissolution rate of FITC-BSA from alginate/chitosan microsphere was lower than that from alginate microsphere and alginate/poly-L-lysine microsphere. By confocal laser scanning microscope, it was revealed that alginate/FITC-poly-L-lysine microspheres were present in close apposition epithelium of the Peyei s patches of rabbits following inoculation into lumen of intestine, which proved that microspheres could be taken up by Peyer's patch. In conclusion, it is suggested that alginate microsphere prepared by spray method, showing a particle size of < 10 μm and a high loading efficiency, can be used as a model drug for the development of oral vaccine delivery system.

      • Aspergillus nidulans 에서의 핵전이에 의한 종내잡종 형성

        The nuclear transfer technique was employed to obtain intraspecific hybrids in Aspergillus nidulans. Nuclei isolated from either a wild type or an auxotrophic mutant strain (FGSC 475) were transferred into the protoplasts of a recipient strain (FGSC 514). The frequency of hybrid formation (4.8% and 10.1%, respectively) by nuclear transfer was higher than the frequency (0.6%) by protoplast fusion. Furthermore, most of the hybrids formed showed increased activity of some components of cellulase system, xylanase system, and mannanase. The hybrids were analyzed to be either diploid or aneuploid. These results suggest that nuclear transfer technique is more efficient in the formation of intraspecific hybrids than protoplast fusion method and is useful for the improvement of Aspergillus strains.

        LED 광원을 이용한 마스트 항해등 조명광학계 개발

        This paper dealt with the LED optical illumination system design for the Masthead navigation light to replace halogen lamps. We made Fresnel lens satisfy luminous intensity distribution of “Convention on the International Regulation for Preventing Collisions at Sea(COLREG)”. The optical system is designed by classifying three parts: light source, lens, and cut off plate. The source of light has been made to have the uniform horizontal and vertical light distribution by placing 6 LEDs at intervals of 54°, and as the cylindrical Fresnel lens, the lens has been designed to achieve the uniform horizontal and vertical light distribution in the range of plain light. Finally, the cover has been designed to block the light from the outside of plain light and ultimately met the standards for light distribution of navigation lights. In addition, the validity of design has been verified with manufacturing a trial product.

        Two bacterial strains, BTF1 and BTF2, and one yeast strain, BTF3, were isolated from the starter of fermented black tea customarily called "Black tea fungus". The morphological, physiological and biochemical characteristics of the strains were analyzed. Consequently, the bacterial strains, BTF1 and BTF2, were identified as Acetobacter pasteurianus subsp. xylinum and Acetobacter pasteurianus, respectively, and the yeast strain, BTF3, were as Schizosaccharomyces pombe.

        전방향으로 발광하는 LED Bulb를 위한 2차 광학계 설계

        Secondary optical system designed for LED bulb which emits light in all directions was simulated with Energy Star standards. Components of the optical system were LED light source, the cover of the LED bulb and reflector which is to confirm the diffuser plate and LightTools software was used to design the illumination optics. The main points of the secondary optical system design are the location of the LED light source, the shape of the LED bulb cover, the location of the reflection plate, and the scattering properties of the diffusing plate. Mechanism of the LED bulb is that the light emitted from the light source move on to the backward after reflected by the coated light cover from the inside and then the reflected light is scattered by the diffuser plate. The LED bulb was designed to satisfy the standard light distribution and color specifications of the Energy Star(IES LM-79-08).

      • Cloning of Two chitin Synthase Gene Fragments from Penicillium diversum

        The PCR fragments of two distinct chitin synthase genes, PdCHS1 and PdCHS2, were cloned from Penicillium diversum KCTC 6786. The nucleotide sequences of PdCHS1 and PdCHS2 contained uninterrupted open reading frames (ORFs) of 570 bp excluding the prime sequence. The similarity analysis of the deduced amino acid sequences using BLASTP indicated that the possible evolutionary relationship between P. diversum and ascomycetous fungi. Multialignment of the deduced amino acid sequences of PdCHSs using CLASTAL W revealed that the PdCHSs fell into two different classes: PdCHS1 into Class I and PdCHS2 into Class Ⅱ of chitin synthase defined by Bowen et al. (1992). By Southern blot analysis, it was shown that each of the two genes is present as a single copy in the genome of P. diversum KCTC 6786.

      • 세포주기 연구를 위한 분열 효모의 이용 및 응용

        Cell growth is an important phenomenon for self-renewal, organogenesis, and tumor expansion. This cell growth is tightly regulated on cell cycle by very complex and huge amount factors. Cell cycle is consisted by G (Gap) phase, M (mitosis) phase, and S (Synthesis), which contain specific regulation factors, cyclin and cyclin-dependent kinase (cdk). In other mechanism to regulate cell cycle, cell cycle-checkpoints indicate steps to regulate cell cycle on DNA damage checkpoint and spindle checkpoint. Fission yeast has been used as good model system for investigation of eukaryotic cell cycle. We have known the complete fission yeast genomic sequence and haven plentiful biochemical and proteomic information. On the basis of the information, we can make several mutants to analysis the roles of cell cycle regulatory factors and identify new factors. Here, we introduce how to use yeast model to discover the relation between mutants and cell cycle phenotypes and briefly explain the results from our functional study.

        Cell Cycle-dependent Expression of Chitin Synthase Genes in Aspergillus nidulans

        The transcription of the chitin synthase genes (chss) was cell cycle-regulated in Aspergillus nidulans and the expression pattern was classified into two groups. Group one, containing chsA and chsC, showed decreasing transcription level upon entry into the S-phase and no further variation during the remainder of the cell cycle. However, group two, containing chsB, chsD, and csmA showed a sharp decrease of mRNA level upon entry into the G2-phase and an increase during the M-phase. Our results suggested that the chss, belonging to same group with the similar expression pattern during the cell cycle are functionally linked and that chsD may play a role in hyphal growth and development in A. nidulans.
